Modeling Lipid Vesicles' Dynamic Response to Osmotic Pressure to Quantify Small-Molecule Transport.

Lipid membrane permeability is fundamental for cellular homeostasis, signaling, and drug transport. Experimental methods exist to characterize it, from cell-based assays to more reproducible artificial, filter-supported systems. Although they closely mimic physiological environments, these assays pose challenges for quantitative analysis. Simpler models, such as supported lipid bilayers and liposomes, offer better control of experimental variables and membrane composition, making them suitable for detailed physicochemical investigations. The dynamic response of unilamellar lipid vesicles to os

99mTechnetium-stannous oxinate as marker of liposome formulations.

Liposomes associated with tin(II) dioxinate were prepared from egg yolk phosphatidylcholine and cholesterol as sterile and pyrogen-free multilamellar or unilamellar vesicles. Complexing of liposomal tin(II) dioxinate with 99mTc attained 98% of the added radioactivity. Thirty percent 99mTc were released during 24-h incubation in biological fluids. The absence of tin colloids seen by electron microscopy and the stability of liposomal phospholipid and tin(II) dioxinate during 72-h incubation at 37 degrees C in plasma and cerebrospinal fluid would allow safe and reliable scintigraphic liposome pha

Membrane Formulation Impacts Vesicle-Based Artificial Cell Functions.

Engineering vesicle-based artificial cells capable of mimicking cellular processes is a central objective in synthetic biology. However, while giant unilamellar vesicles (GUVs) have become a popular artificial cell platform, the impact of fundamental design features remains obscure. Here, we assembled GUVs composed primarily of two phospholipids: POPC, a phosphatidylcholine widely employed in GUV preparation, and DOPC, a phosphatidylcholine with a lower melting temperature and higher fluidity. We found that the phospholipid mixture leads to an increase in average GUV size (compared to POPC onl

Rearrangement of GUV-confined actin networks in response to micropipette aspiration.

Although diverse actin network architectures found inside the cell have been individually reconstituted outside of the cell, how different types of actin architectures reorganize under applied forces is not entirely understood. Recently, bottom-up reconstitution has enabled studies where dynamic and phenotypic characteristics of various actin networks can be recreated in an isolated cell-like environment. Here, by creating a giant unilamellar vesicle (GUV)-based cell model encapsulating actin networks, we investigate how actin networks rearrange in response to localized stresses applied by mic

Functionalization and higher-order organization of liposomes with DNA nanostructures.

Small unilamellar vesicles (SUVs) are indispensable model membranes, organelle mimics, and drug and vaccine carriers. However, the lack of robust techniques to functionalize or organize preformed SUVs limits their applications. Here we use DNA nanostructures to coat, cluster, and pattern sub-100-nm liposomes, generating distance-controlled vesicle networks, strings and dimers, among other configurations. The DNA coating also enables attachment of proteins to liposomes, and temporal control of membrane fusion driven by SNARE protein complexes. Such a convenient and versatile method of engineeri

Drug-fortified liposomes as carriers for sustained release of NSAIDs: The concept and its validation in the animal model for the treatment of arthritis.

Drug-fortified cationic liposomes of 6‑methoxy‑2‑naphthylacetic acid (6‑MNA) were prepared and characterized by various techniques. The residence time of drug-fortified liposomes in joint cavity was evaluated by intra-articular (IA) administration of the radio-labeled (99mTc) liposomal formulation in the inflamed joints in rats. The cationic liposomal formulation composed of 6‑MNA (3) as an active agent, its double salt (4) with the lipid 1,2‑distearoyl‑sn‑glycero‑3‑phosphoethanolamine (DSPE), and pharmaceutically acceptable excipients such as hydrogenated soyabean phospatidylcholine (HSPC) an

Mechanism of cationic amphiphilic drug inhibition of purified lysosomal phospholipase A1.

Cationic amphiphilic drugs like chlorpromazine, propranolol, and chloroquine inhibit lysosomal phospholipase A in vitro. Some workers have proposed that cationic amphiphilic drugs inhibit the activity of phospholipase A1 by forming substrate-drug complexes which cannot be degraded while others have reported competitive inhibition implying drug effects on the enzyme. To analyze the mechanism of inhibition, we examined the binding ability of these drugs to unilamellar vesicles of dioleoylphosphatidylcholine and correlated these results with a detailed kinetic analysis of phospholipase A. Polysaccharide functionalization reduces lipid vesicle stiffness.

The biophysical properties of lipid vesicles are important for their stability and integrity, key parameters that control the performance when these vesicles are used for drug delivery. The vesicle properties are determined by the composition of lipids used to form the vesicle. However, for a given lipid composition, they can also be tailored by tethering polymers to the membrane. Typically, synthetic polymers like polyethyleneglycol are used to increase vesicle stability, but the use of polysaccharides in this context is much less explored. Here, we report a general method for functionalizing

Quantification of cholesterol incorporation in giant unilamellar vesicles produced by a modified cDICE method.

Cholesterol is an essential component of eukaryotic cell membranes, influencing membrane packing, fluidity, and domain formation. Replicating these properties in model membranes is critical for reconstitution studies, but common emulsion-based methods for producing giant unilamellar vesicles (GUVs) fail to incorporate cholesterol efficiently. Here, we use methyl-β-cyclodextrin-cholesterol (MβCD-CL) complexes to deliver cholesterol into GUVs produced by the emulsion droplet interface crossing encapsulation (eDICE) method and demonstrate a convenient way to quantify the degree of cholesterol inc

Kinetics of demetallation of a zinc-salophen complex into liposomes.

A Zn-salophen complex has been incorporated into POPC large unilamellar liposomes (LUV) obtained in phosphate buffer at pH 7.4. Fluorescence optical microscopy and anisotropy measurements show that the complex is located at the liposomal surface, close to the polar headgroups. The interaction of the POPC phosphate group with Zn(2+) slowly leads to demetallation of the complex. The process follows first order kinetics and rate constants have been measured fluorimetrically in pure water and in buffered aqueous solution. The coordination of the phosphate group of monomeric POPC with salophen zinc

Preparation and characterization of polymer coated small unilamellar vesicles.

Glucose oxidase was entrapped in small unilamellar vesicles composed of phosphatidylcholine, dicetyl phosphate and cholesterol. Prediction of the enzyme content of liposomes by calculations based on input concentrations of lipid and protein, dimensions of the lipids and the liposomes yielded one protein per vesicle. The entrapment efficiency was experimentally determined to be about 13%. On the other hand the entrapment efficiency for the small chromate ions was found to be significantly lower (0.1%). The liposomes were then coated with a polymer, poly(1,4-pyridinium diylethylene salt). It was

Shadow electrochemiluminescence imaging of giant liposomes opening at polarized electrodes.

In this work, the release of giant liposome (∼100 μm in diameter) content was imaged by shadow electrochemiluminescence (ECL) microscopy. Giant unilamellar liposomes were pre-loaded with a sucrose solution and allowed to sediment at an ITO electrode surface immersed in a solution containing a luminophore ([Ru(bpy)3]2+) and a sacrificial co-reactant (tri-n-propylamine). Upon polarization, the electrode exhibited illumination over its entire surface thanks to the oxidation of ECL reagents. However, as soon as liposomes reached the electrode surface, dark spots appeared and then spread over time

A simple method for preparation of immuno-magnetic liposomes.

A simple and readily manoeuverable method for preparing immuno-magnetic liposomes that indigenously contain binding sites for attaching other molecules like antibodies on their exterior surface is described. In this method magnetic unilamellar vesicles are prepared from a mixture of phosphatidylcholine, cholesterol, small amounts of a linear chain aldehyde and colloidal particles of magnetic iron oxide, using a reverse phase evaporation technique. The aldehyde (dedecanal) molecules align themselves among the lipid molecules in the bilayer with their aldehyde groups exposed to the aqueous phase

Interaction of chlorpromazine and imipramine with model membranes.

The aim of the present study is to examine the effect of phospholipids on the lipid destabilization induced by chlorpromazine (CPZ) and imipramine (IP) at different levels of pH. The large unilamellar vesicles (LUV) are formed in the presence of calcein (60 mM). The vesicles containing calcein have been incubated in the presence of CPZ and IP at pH 4.5 and pH 8. At pH 4.5 CPZ and IP induce a rapid release of the calcein encapsulated in the liposomes. Calcein release, at equal concentrations of pharmacological agent, is more important by CPZ than by IP. At pH 8, the calcein release was more imp

Advancing psoriasis drug delivery through topical liposomes.

Psoriasis, recognized as a chronic inflammatory skin disorder, disrupts immune system functionality. Global estimates by the World Psoriasis Day consortium indicate its impact on approximately 130 million people, constituting 4 to 5 percent of the worldwide population. Conventional drug delivery systems, mainly designed to alleviate psoriasis symptoms, fall short in achieving targeted action and optimal bioavailability due to inherent challenges such as the drug's brief half-life, instability, and a deficiency in ensuring both safety and efficacy. Liposomes, employed in drug delivery systems,

Preparation of Membrane Protein-Functionalized Polymer and Polymer/Lipid Hybrid Large and Giant Unilamellar Vesicles.

Vesicles formed from amphiphilic copolymers, alone or blended with phospholipids, offer superior mechanical and chemical stability compared to conventional lipid vesicles. This makes them an attractive chassis, one that can be further developed and expanded to enable specific applications, such as drug delivery, diagnostic biosensing, construction of artificial cells, and bioinspired micro- and nanoreactors. In particular, functionalization through membrane protein incorporation is essential for many of these applications. Here, we present a protocol for preparing membrane protein-functionaliz

Rotational dynamics of bound pairs of bacteria-induced membrane tubes.

We present experiments demonstrating tube formation in giant unilamellar vesicles that are suspended in a bath of swimming E. coli bacteria. We chose the lipids such that the bacteria form no adhering interactions with the membrane. The tubes are generated by the pushing force exerted by the bacteria on the membrane of the vesicles. Once a tube is generated, the bacterium is confined within it, resulting in long-lived tubes that protrude into the vesicle. We show that such tubes interact to form stable bound pairs that orbit each other. We speculate that the tubes are maintained by the persist

Nuclear Assembly in Giant Unilamellar Vesicles Encapsulating Xenopus Egg Extract.

The reconstitution of a cell nucleus in a lipid bilayer-enclosed synthetic cell makes great strides in bottom-up synthetic biology. In this study, a method for assembling a nucleus in giant unilamellar vesicles (GUVs) is proposed. To induce reconstitution of the nucleus, the interphase egg extract of African clawed frogs Xenopus laevis is utilized, known as a biochemically controllable cell-free system capable of transforming an added sperm chromatin into a nucleus in vitro. The GUV formation efficiency is enhanced by the inverted emulsion method through incorporating prolonged waiting time an

Efficient electroformation of supergiant unilamellar vesicles containing cationic lipids on ITO-coated electrodes.

Giant unilamellar vesicles (GUVs) represent a versatile in vitro system widely used to study properties of lipid membranes and their interaction with biomacromolecules and colloids. Electroformation with indium tin oxide (ITO) coated coverslips as electrodes is a standard approach to GUV production. In the case of cationic GUVs, however, application of this approach leads to notorious difficulties. We discover that this is related to aging of ITO-coated coverslips during their repeated use, which is reflected in their surface topography on the nanoscale. We find that mild annealing of the ITO-
