Hyperuricemia causes gouty arthritis, kidney disease, heart disease, and other diseases. Xanthine oxidase (XOD) and urate transporters play important roles in urate homeostasis. Numerous plants have been identified as XOD inhibitors. Longan seeds are known to contain high levels of polyphenols such as corilagin, gallic acid and ellagic acid. We examined the effect of longan seed extract on XOD inhibition and urate transporters GLUT1 and GLUT9 using both in vitro and in vivo assays. The results showed that dried longan seed extract (LSE) and its active components inhibited XOD dose-dependently

Our previous study has shown that uric acid is metabolized by recombinant rat urate oxidase (UOX) and generates hydrogen peroxide leading to DNA damage and cell transformation. However, in transformed cells protein levels of UOX were reduced compared with the control cell. To investigate the characterization and the mechanisms responsible for the degradation of UOX, a controllable gene expression system has been used to switch on/off controlled expression of rat UOX in vitro. Chinese hamster ovary cells were double transfected with regulatory and responsive plasmids, pCMV-tTA and pTRE-rUOX, re

Gout in heart transplant recipients is common and poses a significant therapeutic challenge. Concomitant administration of azathioprine and allopurinol therapy carries a high risk of leukopenia. Uricosuric agents can cause renal lithiasis and/or acute renal failure in patients with renal failure and/or high urinary levels of uric acid. We report our experience with urate-oxidase in three heart transplant recipients with severe polyarticular and tophaceous gout, a history of leukopenia under allopurinol and unresponsiveness or contraindications to uricosuric agents. Urate-oxidase was given pare

BACKGROUND: Urate oxidase (EC 1.7.3.3 or UOX) catalyzes the conversion of uric acid and gaseous molecular oxygen to 5-hydroxyisourate and hydrogen peroxide, in the absence of cofactor or particular metal cation. The functional enzyme is a homo-tetramer with four active sites located at dimeric interfaces.
RESULTS: The catalytic mechanism was investigated through a ternary complex formed between the enzyme, uric acid, and cyanide that stabilizes an intermediate state of the reaction. When uric acid is replaced by a competitive inhibitor, no complex with cyanide is formed.
CONCLUSION: The X-ra

Compatible osmolytes like trimethylamine N-oxide (TMAO), known for preferentially stabilizing proteins in their folded states, offer a potential solution; however, the precise molecular mechanisms underlying their stabilizing effects are not fully understood. On the other hand, the poor thermal stability of pharmaceutical enzymes such as urate oxidase (UOX) in aqueous solutions remains a significant challenge. This study explored how TMAO modulates the enzyme's environment to enhance UOX's catalytic efficiency and stability. By combining experimental and computational approaches, we evaluated

INTRODUCTION: Hyperuricaemia in tumour lysis syndrome (TLS) can cause acute renal failure (ARF), necessitating dialysis. Recombinant urate oxidase (rasburicase) converts uric acid to soluble allantoin, which is excreted easily.
CASE REPORT: An 8-year-old boy with stage 3 Burkitt's lymphoma, TLS was successfully treated with hyper-hydration, diuretics and rasburicase, without dialysis. This is the first paediatric case in Kandang Kerbau Women's & Children's Hospital (KKH) in which rasburicase was used. We review the literature on the effectiveness of urate oxidase in avoiding dialysis in TLS.

Urate oxidase (Uox) has great potential in biotechnology. However, the exploration of Uox with superior stability and its simple preparation with high yield remains challenging. By fusing human ferritin with the Uox from the extremophile Deinococcus radiodurans, a novel 24-polymeric Uox (DrUox-F) with excellent properties was achieved. The DrUox-F with 98 % purity was obtained through low-speed centrifugation with the yield of 890 mg/L, representing the highest yield currently reported. Remarkably, DrUox-F preserved 100 % activity after 35 days at pH 10, which was significantly superior to DrU

Microbial urate oxidase has emerged as a potential source of therapeutic properties for hyperuricemia in arthritic gout and renal disease. The thermostability and long-term thermal tolerance of the enzyme need to be established to prolong its therapeutic effects. Here, we present the biochemical and structural aspects of a hyperthermostable urate oxidase (TbUox) from the thermophilic microorganism Thermobispora bispora. Enzymatic characterization of TbUox revealed that it was active over a wide range of temperatures, from 30 to 70 °C, with optimal activity at 65 °C and pH 8.0, which suggests i

Hyperuricemia (HUA), a metabolic disorder associated with gout and cardiometabolic diseases, has become an important target for the development of food-derived functional ingredients. Paeonia lactiflora Pall, a widely consumed edible medicinal plant, has attracted attention as a potential source of urate-modulating compounds; however, its key active compounds and their urate-related activities remain unclear. In this study, through comprehensive phytochemical profiling, 20 compounds were isolated from P. lactiflora and systematically evaluated using xanthine oxidase (XO) inhibition and HK-2 ce

Urate oxidase (uricase; EC 1.7.3.3) catalyzes the oxidation of uric acid to allantoin, a highly soluble metabolite that is readily excreted. Lack of functional urate oxidase in human's leads to hyperuricemia, which is associated with gout, tumor lysis syndrome and renal complications. The present study reports statistical optimization, purification, and comprehensive characterization of a novel extracellular urate oxidase secreted by Arthrobacter creatinolyticus SA1. Plackett-Burman Design screening followed by Central Composite Design identified inoculum volume, initial pH, peptone, and yeast

BACKGROUND: Effective xanthine oxidoreductase inhibition (XOI) urate-lowering treatment (ULT) to target significantly reduces gout flare burden and synovitis between 1-2 years therapy, without clearing all monosodium urate crystal deposits. Paradoxically, treat to target ULT is associated with increased flare activity for at least 1 year in duration on average, before gout flare burden decreases. Since XOI has anti-inflammatory effects, we tested for biomarkers of sustained, effective ULT that alters gouty inflammation.
METHODS: We characterized the proteome of febuxostat-treated murine bone

A high plasma urate concentration (PUA), related to loss of urate oxidase in evolution, is postulated to protect humans from oxidative injury. This hypothesis has broad clinical relevance, but support rests largely on in vitro data and epidemiologic associations. Pegloticase therapy generates H(2)O(2) while depleting urate, offering an in vivo test of the antioxidant hypothesis. We show that erythrocytes can efficiently eliminate H(2)O(2) derived from urate oxidation to prevent cell injury in vitro; during therapy, disulfide-linked peroxiredoxin 2 dimer did not accumulate in red blood cells, i

Along with hydration and urinary alkalinization, allopurinol has been the standard agent for the management of hyperuricemia in patients with a high tumor burden at risk of tumor lysis syndrome; however, this agent often fails to prevent and treat this complication effectively. Rasburicase (recombinant urate oxidase) has been shown to be effective in reducing uric acid and preventing uric acid accumulation in patients with hematologic malignancies with hyperuricemia or at high risk of developing it. Rasburicase acts at the end of the purine catabolic pathway and, unlike allopurinol, does not i

Background: Hyperuricemia (HU) results from excessive uric acid (UA) production/impaired excretion and is associated with gout and renal dysfunction. Gut microbiota dysbiosis may impair intestinal UA excretion, disrupt the gut barrier, and promote inflammation. Traditional Chinese medicine (TCM), with multitarget and microbiota-modulating effects, may offer a therapeutic strategy for HU. In this study, the rhizome of Phragmites communis, a TCM used for clearing heat and promoting diuresis, was investigated. Specifically, we evaluated whether P. communis could alleviate HU by suppressing xanthi

BACKGROUND: Tumor lysis syndrome (TLS) is an oncologic emergency commonly seen in children with hemato-lymphoid malignancies. Recombinant urate oxidase (RUO) is used in both the prophylaxis and treatment of TLS. However, in resource-constrained countries, its role is mostly limited to the treatment of established TLS and data regarding the use of RUO and its outcome is sparse.
OBJECTIVE: To describe the outcome of Pediatric TLS following the use of a fixed - dose of RUO.
METHODS: A retrospective chart review of all children <15 years of age admitted in the Department of Paediatric Oncology,

PURPOSE OF REVIEW: The purpose of this study is to give an overview of recently published articles covering the impact of anti-inflammatory therapies, xanthine oxidase inhibitors and other urate-lowering therapies on cardiovascular diseases in gout.
RECENT FINDINGS: In patients with gout, long-term xanthine oxidase inhibition might reduce some cardiovascular comorbidities because of the dual effect of lowering serum uric acid levels and reducing free-radical production during uric acid formation. Among the anti-inflammatory therapies, colchicine has been shown to reduce some major cardiovascu

Gouty arthritis (GA) is a common arthritis characterized by chronic inflammation and monosodium urate (MSU) crystal deposition in articular structures. Current clinical medications mainly target to alleviate the inflammation, but do less to the deposited MSU. Although administration of urate oxidase (UOx) could degrade the MSU, the toxic by-product hydrogen peroxide (H2O2) generated during the degradation would aggravate the inflammation. To surmount this, we developed a smart stimuli-responsive drug delivery platform (termed UBC) for the treatment of acute GA, where urate oxidase (UOx) was in
