Colonial architecture modulates the speed and efficiency of multi-jet swimming in salp colonies.

Salps are marine pelagic tunicates with a complex life cycle including a solitary and colonial stage. Salp colonies are composed of asexually budded individuals that coordinate their swimming by multi-jet propulsion. Colonies develop into species-specific architectures with distinct zooid orientations. These distinct colonial architectures vary in how frontal area scales with the number of zooids in the colony. Here, we address how differences in frontal area drive differences in swimming speed and the relationship between swimming speed and cost of transport in salps. We (1) compared swimming

Preparation and characterization of a novel high barrier mulching film with tunicate cellulose nanocrystals/sodium alginate/alkali lignin.

In this study, the base film (CSL) was prepared by blending tunicate cellulose nanocrystals (TCNCs) extracted from tunicate shells, with sodium alginate (SA) and alkali lignin (AL). Then, the mulching film (CSL-WK) was prepared using water-borne polyurethane (WPU) as binder to install low-energy Kaolin on the surface of CSL film. The influences of composition with different concentrations on mechanical properties were studied. The tensile strength and elongation at break of CSL-WK film could reach 86.58 MPa and 50.49 %, respectively. The mulching films were characterized by degradability test,

Evolutionary traces of miniaturization in a giant-Comparative anatomy of brain and brain nerves in Bathochordaeus stygius (Tunicata, Appendicularia).

Appendicularia comprises 70 marine, invertebrate, chordate species. Appendicularians play important ecological and evolutionary roles, yet their morphological disparity remains understudied. Most appendicularians are small, develop rapidly, and with a stereotyped cell lineage, leading to the hypothesis that Appendicularia derived progenetically from an ascidian-like ancestor. Here, we describe the detailed anatomy of the central nervous system of Bathochordaeus stygius, a giant appendicularian from the mesopelagic. We show that the brain consists of a forebrain with on average smaller and more

Surgical fusion between incompatible colonies of the compound ascidian, Botrylloides fuscus.

In the compound ascidian Botrylloides fuscus, allorecognition responses were observed at the colonial margin (natural growing edge) as necrotic reactions in the subcuticular region of the facing tunic. However, artificial cut surface contacts between incompatible colonies resulted in the formation of chimeras with common vascular systems, termed "surgical fusion." The occurrence of surgical fusion in B. fucus contrasts with rejection occurring under the same conditions in other botryllids. Together the two observations suggest that self-nonself recognition may be a specialized function of tuni

Colonial allorecognition, hemolytic rejection, and viviparity in botryllid ascidians.

Allorecognition is a fundamental system that animals use to maintain individuality. Although embryos are usually semiallogeneic with their mother, viviparous animals are required to allow these embryos to develop inside the maternal body, but must also eliminate an "invasion" by nonself. In colonial ascidians of the family Botryllidae, when two colonies are brought into contact at their growing edges, a hemolytic rejection reaction occurs between allogeneic colonies. Morula cells, a type of hemocyte, are the major effector cells in the hemolytic rejection. Morula cells infiltrate and aggregate

Involvement in Fertilization and Expression of Gamete Ubiquitin-Activating Enzymes UBA1 and UBA6 in the Ascidian Halocynthia roretzi.

The extracellular ubiquitin-proteasome system is involved in sperm binding to and/or penetration of the vitelline coat (VC), a proteinaceous egg coat, during fertilization of the ascidian (Urochordata) Halocynthia roretzi. It is also known that the sperm receptor on the VC, HrVC70, is ubiquitinated and degraded by the sperm proteasome during the sperm penetration of the VC and that a 700-kDa ubiquitin-conjugating enzyme complex is released upon sperm activation on the VC, which is designated the "sperm reaction". However, the de novo function of ubiquitin-activating enzyme (UBA/E1) during fert

Cell Communication-mediated Nonself-Recognition and -Intolerance in Representative Species of the Animal Kingdom.

Why has histo-incompatibility arisen in evolution and can cause self-intolerance? Compatible/incompatible reactions following natural contacts between genetically-different (allogeneic) colonies of marine organisms have inspired the conception that self-nonself discrimination has developed to reduce invasion threats by migratory foreign germ/somatic stem cells, in extreme cases resulting in conquest of the whole body by a foreign genome. Two prominent model species for allogeneic discrimination are the marine invertebrates Hydractinia (Cnidaria) and Botryllus (Ascidiacea). In Hydractinia, self

A new species of Polyandrocarpa (Ascidiacea, Styelidae) in the Mediterranean Sea.

Polyandrocarpa arianae n. sp. was collected repeatedly by trawls deeper than 90m from several parts of the Mediterranean Sea. It is described and compared to the shallow species Polyandrocarpa zorritensis (Van Name, 1931) considered as invasive in the Mediterranean Sea. Among the colonial Styelidae the other species which belong to the same genus differ by the colony and zooid shape, the presence/absence of endocarps and of a caecum in the gut loop and by the location and structure of the polycarps.

Integrated multi-approaches reveal unique metabolic mechanisms of Vestimentifera to adapt to deep sea.

BACKGROUND: Vestimentiferan tubeworms are deep-sea colonizers, in which chemoautotrophic symbiosis was first observed. These animals are gutless and depend on endosymbiotic bacteria for organic compound synthesis and nutrition supply. Taxonomically, vestimentiferans belong to Siboglinidae and Annelida. Compared with other siboglinids, vestimentiferans are distinguished by high tolerance of the prevailing hydrogen sulfide in hydrothermal vents, rapid growth in local habitats, and a physical structure consisting of a thick chitinous tube. The metabolic mechanisms contributing to these features r

Molecular evolution of the ankyrin gene family.

Ankyrins are membrane adaptor molecules that play important roles in coupling integral membrane proteins to the spectrin-based cytoskeleton network. Human mutations of ankyrin genes lead to severe genetic diseases such as fatal cardiac arrhythmias and hereditary spherocytosis. To elucidate the evolutionary history of ankyrins, we have identified novel ankyrin sequences in insect, fish, frog, chicken, dog, and chimpanzee genomes and explored the phylogenetic relationships of the ankyrin gene family. Our data demonstrate that duplication of ankyrin genes occurred at two different stages. The fir

New putative phenol oxidase in ascidian blood cells.

The phenol oxidase system is ancient and ubiquitously distributed in all living organisms. In various groups it serves for the biosynthesis of pigments and neurotransmitters (dopamine), defence reactions and tissue hardening. Ascidians belong to subphylum Tunicata, which is considered the closest living relative to Vertebrates. Two phenol oxidases previously described for ascidians are vertebrate-like and arthropod-like phenol oxidases. In our present study, we described a new ascidian protein, Tuphoxin, with putative phenol oxidase function, which bears no sequence similarity with two enzymes

Contact area-dependent cell communication and the morphological invariance of ascidian embryogenesis.

Marine invertebrate ascidians display embryonic reproducibility: Their early embryonic cell lineages are considered invariant and are conserved between distantly related species, despite rapid genomic divergence. Here, we address the drivers of this reproducibility. We used light-sheet imaging and automated cell segmentation and tracking procedures to systematically quantify the behavior of individual cells every 2 minutes during Phallusia mammillata embryogenesis. Interindividual reproducibility was observed down to the area of individual cell contacts. We found tight links between the reprod

Wettability and Substrate Selection in the Larval Settlement of the Solitary Ascidian Phallusia philippinensis (Phlebobranchia: Ascidiidae).

For marine benthic animals, the selection of substrate by larvae is important for their survival, with early post-settlement mortality often being affected by the microenvironment where they settle. We tested the substrate preference of the larvae of the ascidian Phallusia philippinensis toward nine commercially available substrates. In the present assay, the larvae settled on one of four substrates for seven substrate combinations with different wettabilities; we counted the number of settled larvae on each of the four substrates, and Manly's selection indices were compared to determine the p

Unique Tunicate Traits Possibly Encoded by Horizontally Transferred Genes.

While horizontal gene transfer (HGT) is common among prokaryotes, its prevalence and impact among metazoans are debatable. A clear example of HGT in animals is the cellulose synthase gene in tunicates, invertebrate chordates that possess the unique ability to produce a cellulose-containing tunic, despite sharing a conserved larval body plan with vertebrates. To investigate evolutionary roles of HGT in animals, we surveyed the genome of a tunicate and identified eight groups of candidate genes that are unlikely to have been vertically transferred. Notably, these candidates encode proteins poten

A simple inland culture system provides insights into ascidian post-embryonic developmental physiology.

Maintenance and breeding of experimental organisms are fundamental to life sciences, but both initial and running costs, and hands-on zootechnical demands can be challenging for many laboratories. Here, we first aimed to further develop a simple protocol for reliable inland culture of tunicate model species of the Ciona genus. We cultured both Ciona robusta and Ciona intestinalis in controlled experimental conditions, with a focus on dietary variables, and quantified growth and maturation parameters. From statistical analysis of these standardized datasets, we gained insights into the post-emb

Fertilization mechanisms in hermaphroditic ascidians and nematodes: Common mechanisms with mammals and plants.

Most animals have male and female, whereas flowering plants are hermaphrodites. Exceptionally, a small population of invertebrates, including ascidians and nematodes, has hermaphrodite in reproductive strategies. Several ascidians exhibit strict self-sterility (or self-incompatibility), similar to flowering plants. Such a self-incompatibility mechanism in ascidian has been revealed to be very similar to those of flowering plants. Here, we describe the mechanisms of ascidian fertilization shared with invertebrates and mammals, as well as with plants. In the nematode Caenorhabditis elegans, havi

Evolutionary volatile Cysteines and protein disorder in the fast evolving tunicate Oikopleura dioica.

Cysteine (Cys) is regarded as the most conservative amino acid in nature, something that does not occur in the tunicate Oikopleura dioica, where this amino acid is one of the fastest evolving. In this work we analyze some of the causes of this intriguing absence of conservation. Considering the well-known stabilizing role of Cys, it was first investigated whether the lack of conservation was accompanied by an increase in intrinsic protein disorder. In contrast to expectations, it was found that O. dioica is the chordate that has the lowest levels of intrinsic disorder, while vertebrates (repre

Budding by oozooids in the polystyelid ascidian Metandrocarpa tayori Huntsman.

Larvae of the stolidobranch ascidian Metandrocarpa taylori molt a thin sheath upon settling, then metamorphose and radiate a larval complement of vascular ampullae upon the substrate. These ampullae thereafter regress, "rest" in a reduced condition for several weeks, and then regrow into the oozooid's definitive array of vascular ampullae in accompaniment to the development of the oozooidal vascular nest of test-vessels. Pallial buds emerge some four months after the larva settles; the oozooid has by then grown to a length of at least 2 mm and its vascular nest is surrounded by at least 16 vas

Development and circuitry of the tunicate larval Motor Ganglion, a putative hindbrain/spinal cord homolog.

The Motor Ganglion (MG) is a small collection of neurons that control the swimming movements of the tunicate tadpole larva. Situated at the base of the tail, molecular and functional comparisons suggest that may be a homolog of the spinal cord and/or hindbrain ("rhombospinal" region) of vertebrates. Here we review the most current knowledge of the development, connectivity, functions, and unique identities of the neurons that comprise the MG, drawn mostly from studies in Ciona spp. The simple cell lineages, minimal cellular composition, and comprehensively mapped "connectome" of the Ciona MG a
