An Efficient Colony PCR Method for Construct Verification in the Basidiomycete Ustilago maydis.

Rapid advances in AI-driven annotation and automation are increasing the demand for fast and reliable screening protocols for reverse genetic approaches. Traditional methods for genomic DNA extraction from the basidiomycete Ustilago maydis are often time-consuming and require expensive reagents and hazardous chemicals, making them unsuitable for large-scale mutant screening. Existing colony PCR protocols using crude lysates without DNA precipitation are fast but often fail in terms of reproducibility because cellular components can interfere with polymerase activity or DNA stability. These lim

Herbicides as fungicides: Targeting heme biosynthesis in the maize pathogen Ustilago maydis.

Pathogens must efficiently acquire nutrients from host tissue to proliferate, and strategies to block pathogen access therefore hold promise for disease control. In this study, we investigated whether heme biosynthesis is an effective target for ablating the virulence of the phytopathogenic fungus Ustilago maydis on maize plants. We first constructed conditional heme auxotrophs of the fungus by placing the heme biosynthesis gene hem12 encoding uroporphyrinogen decarboxylase (Urod) under the control of nitrogen or carbon source-regulated promoters. These strains were heme auxotrophs under non-p

Conditional Gene Deletion in Ustilago maydis Via CRE Recombinase In Planta.

Site-specific recombinases have been extensively utilized in higher eukaryotes, particularly in animals, for various applications such as chromosomal translocations, large deletions, site-specific integration, and, notably, tissue-specific conditional knockouts. This technique has also been applied in simpler eukaryotes, including fungi and protozoa. However, in fungal research, especially involving phytopathogenic fungi, its use has often been limited to the recycling of marker genes during transformation experiments. In this study, detailed instructions are provided for generating conditiona

Endogenous hydrogen sulfide reprograms mitochondrial respiration and modulates pathogenicity in Ustilago maydis.

Sulfur metabolism is a central determinant of microbial physiology and adaptation to environmental conditions. Hydrogen sulfide (H2S), a reactive sulfur species produced during cysteine metabolism, has recently emerged as a key regulator of mitochondrial function and redox signaling. However, its role in phytopathogenic fungi and its potential impact on fungal pathogenic behavior remain poorly understood. Here, we show that metabolic conditions that enhance endogenous H2S production in the maize phytopathogen Ustilago maydis trigger coordinated cellular remodeling. Elevated intracellular H2S l

Insertion Pool Sequencing (iPool-Seq) for theHigh-Throughput Quantification of Ustilago maydis Mutant Fitness.

Insertion pool sequencing (iPool-Seq) is a robust functional genomic technique that combines insertion mutagenesis with next-generation sequencing to comprehensively analyze the genome-wide gene function in maize infecting fungus Ustilago maydis. This high-throughput method tracks the abundance of individual mutants in pooled populations under defined selective conditions. It relies on the construction of saturated mutant libraries, where each mutant carries a unique insertion, followed by tagmentation for efficient DNA fragmentation, tagging with unique molecular identifiers (UMIs), and affin

Immunodetection of Cell-Surface-Associated Proteins in Ustilago maydis and Infected Maize Leaves.

Immunolocalization relies on the sequential binding of a specific primary antibody to its target antigen, followed by detection with a fluorophore-conjugated secondary antibody to visualize the subcellular localization of proteins. This technique enables precise and high-resolution mapping of protein localization and distribution patterns after secretion. This chapter presents a detailed protocol for the immunolocalization of secreted proteins from Ustilago maydis, both in vitro and in infected maize leaves.

Maize requires arogenate dehydratase 2 for resistance to Ustilago maydis and plant development.

Maize (Zea mays) smut is a common biotrophic fungal disease caused by Ustilago maydis and leads to low maize yield. Maize resistance to U. maydis is a quantitative trait. However, the molecular mechanism underlying the resistance of maize to U. maydis is poorly understood. Here, we reported that a maize mutant caused by a single gene mutation exhibited defects in both fungal resistance and plant development. maize mutant highly susceptible to U. maydis (mmsu) with a dwarf phenotype forms tumors in the ear. A map-based cloning and allelism test demonstrated that 1 gene encoding a putative aroge

Phosphate limitation enhances malic acid production on nitrogen-rich molasses with Ustilago trichophora.

BACKGROUND: An important step in replacing petrochemical products with sustainable, cost-effective alternatives is the use of feedstocks other than, e.g., pure glucose in the fermentative production of platform chemicals. Ustilaginaceae offer the advantages of a wide substrate spectrum and naturally produce a versatile range of value-added compounds under nitrogen limitation. A promising candidate is the dicarboxylic acid malic acid, which may be applied as an acidulant in the food industry, a chelating agent in pharmaceuticals, or in biobased polymer production. However, fermentable residue s

N-glycosylation of the protein disulfide isomerase Pdi1 ensures full Ustilago maydis virulence.

Fungal pathogenesis depends on accurate secretion and location of virulence factors which drive host colonization. Protein glycosylation is a common posttranslational modification of cell wall components and other secreted factors, typically required for correct protein localization, secretion and function. Thus, the absence of glycosylation is associated with animal and plant pathogen avirulence. While the relevance of protein glycosylation for pathogenesis has been well established, the main glycoproteins responsible for the loss of virulence observed in glycosylation-defective fungi have no

The Use of Yeast Two-Hybrid Analysis to Study Protein-Protein Interactions for Ustilago maydis.

Yeast two-hybrid (Y2H) analysis is an important first step in identifying protein-protein interactions and has been used in a variety of biological systems. In fungi, this method has been employed to evaluate proteins that interact within the fungal cell, often as part of signaling pathways. For fungi that infect or are in relationships with other organisms, the method has also been used to identify and characterize host targets for fungal effector proteins, often used in establishing and maintaining the relationship. Here we describe the Y2H method, as it has been applied to work with Ustilag

Construction and analysis of gene co-expression network in the pathogenic fungus Ustilago maydis.

INTRODUCTION: Biological systems respond to environmental disturbances and a wide range of compounds through complex gene interaction networks. The enormous growth of experimental information obtained using large-scale genomic techniques such as microarrays and RNA sequencing led to the construction of a wide variety of gene co-expression networks in recent years. These networks allow the discovery of clusters of co-expressed genes that potentially work in the same process linking them to biological processes often of interest to industrial, medicinal, and academic research. METHODS: In this

Degradation of the plant defence hormone salicylic acid by the biotrophic fungus Ustilago maydis.

Salicylic acid (SA) is a key plant defence hormone which plays an important role in local and systemic defence responses against biotrophic pathogens like the smut fungus Ustilago maydis. Here we identified Shy1, a cytoplasmic U. maydis salicylate hydroxylase which has orthologues in the closely related smuts Ustilago hordei and Sporisorium reilianum. shy1 is transcriptionally induced during the biotrophic stages of development but not required for virulence during seedling infection. Shy1 activity is needed for growth on plates with SA as a sole carbon source. The trigger for shy1 transcripti

Chloramphenicol-induction of a second cyanide-and azide-insensitive mitochondrial pathway in Ustilago maydis.

Growth of wild type Ustilago maydis in presence of chloramphenicol results in increased resistance of cyanide-insensitive respiration to hydroxamic acid derivatives. The biphasic nature of the Dixon plots for hydroxamate inhibition of mitochondrial alternative respiration suggested involvement of two cyanide-sensitive systems. This hypothesis was confirmed by the analysis of kinetic data. One of the two systems exhibits normal sensitivity to hydroxamates and appears similar to that present in mitochondria from cells grown in control medium. In contrast, the inducible system is characterised by

Unlocking the potentials of Ustilago trichophora for up-cycling polyurethane-derived monomer 1,4-butanediol.

Plastic usage by microbes as a carbon source is a promising strategy to increase the recycling quota. 1,4-butanediol (BDO) is a common monomer derived from polyesters and polyurethanes. In this study, Ustilago trichophora was found to be an efficient cell-factory to valorize BDO. To investigate product formation by U. trichophora, we refined the traditional ion exclusion liquid chromatography method by examining eluent, eluent concentrations, oven temperatures, and organic modifiers to make the chromatography compatible with mass spectrometry. An LC-UV/RI-MS2 method is presented here to identi

Rum1 and Cdc73 integrate transcriptional repression and autophagy to control teliospore timing in Ustilago maydis.

In the biotrophic smut fungus Ustilago maydis, teliospore formation depends on mating and host cues, but the mechanisms that restrain this program during vegetative growth remain unclear. We show that loss of the conserved PAF1 complex subunit UmCdc73 triggers spontaneous teliospore-like differentiation under axenic conditions. The resulting structures undergo cellular remodeling, wall thickening and melanization, acquire increased heat tolerance and limited germination capacity, and induce meiosis-related genes during germination. UmRum1 directly binds to the UmCDC73 promoter and represses it

Induction Medium to Analyze In Vitro Morphological Changes and Biotrophic Features in Sporidia of Ustilago maydis.

Biotrophic fungal pathogens such as Ustilago maydis rely on a host to complete their lifecycle. U. maydis has the advantage, in contrast to obligate biotrophs such as rust fungi, that the saprophytic yeast-like sporidia can be cultivated in artificial media, while the dikaryotic filamentous and infectious stage depends on the host to complete the sexual phase of the lifecycle. Key phenotypes during plant infection include filamentous growth after mating, an increase of fungal biomass in developing tumors, production of an extracellular matrix (ECM) around fungal cells, and melanin formation du

Phytohormone sensing in the biotrophic fungus Ustilago maydis - the dual role of the transcription factor Rss1.

The phenolic compound salicylic acid (SA) is a key signalling molecule regulating local and systemic plant defense responses, mainly against biotrophs. Many microbial organisms, including pathogens, share the ability to degrade SA. However, the mechanism by which they perceive SA is unknown. Here we show that Ustilago maydis, the causal agent of corn smut disease, employs a so far uncharacterized SA sensing mechanism. We identified and characterized the novel SA sensing regulator, Rss1, a binuclear zinc cluster protein with dual functions as putative SA receptor and transcriptional activator r

Endoplasmic reticulum glucosidases and protein quality control factors cooperate to establish biotrophy in Ustilago maydis.

Secreted fungal effectors mediate plant-fungus pathogenic interactions. These proteins are typically N-glycosylated, a common posttranslational modification affecting their location and function. N-glycosylation consists of the addition, and subsequent maturation, of an oligosaccharide core in the endoplasmic reticulum (ER) and Golgi apparatus. In this article, we show that two enzymes catalyzing specific stages of this pathway in maize smut (Ustilago maydis), glucosidase I (Gls1) and glucosidase II β-subunit (Gas2), are essential for its pathogenic interaction with maize (Zea mays). Gls1 is r

Loss of Cohesin Subunit Rec8 Switches Rad51 Mediator Dependence in Resistance to Formaldehyde Toxicity in Ustilago maydis.

DNA-protein cross-links (DPCs) are frequently occurring lesions that provoke continual threats to the integrity of the genome by interference with replication and transcription. Reactive aldehydes generated from endogenous metabolic processes or produced in the environment are sources that trigger cross-linking of DNA with associated proteins. DNA repair pathways in place for removing DPCs, or for bypassing them to enable completion of replication, include homologous recombination (HR) and replication fork remodeling (FR) systems. Here, we surveyed a set of mutants defective in known HR and FR
