Intervertebral disc degeneration (IDD) is a major contributor to chronic low back pain, yet current treatments primarily offer symptom relief without addressing the underlying pathology. In this study, we demonstrate that vanillic acid (VA), a natural phenolic compound, protects nucleus pulposus (NP) cells from inflammation-induced pyroptosis, oxidative stress, and extracellular matrix (ECM) degradation. VA markedly reduced both intracellular and mitochondrial reactive oxygen species (ROS) levels and inhibited NLRP3 inflammasome-mediated pyroptosis. Transcriptomic analysis identified KLF2 as a

Clerodendrum petasites S. Moore has been prescribed in Thai traditional medicine for over 30 years for the treatment of ailments including asthma, inflammation, fever, cough, vomiting, and skin disorders. The phytochemicals from this plant have been identified as phenolic acids, flavones, flavone glycosides, glycosides, phenylpropanoid, and diterpenoid. The pharmacological activities both in vitro and in vivo have mostly been reported from crude extracts and not from pure compounds. This review, therefore, brings together information on the specific phytochemicals found in C. petasites in orde

Preclinical Research Vanillic acid (VA) is a dihydroxybenzoic acid derivative widely used as a flavoring agent. It has chemopreventive effects on experimentally-induced carcinogenesis and in ulcerative colitis. The object of the present study was to investigate the effects of VA, alone and in combination with methylprednisolone (MP), on cationic bovine serum albumin (cBSA induced immune-complex glomerulonephritis in female BALB/c mice. Pre-immunization was carried out with cBSA in BALB/c mice and repeated (cBSA, 13 mg/kg, 3 times/week, i.v.) for 6 weeks to induce glomerulonephritis which was c

The damage of integrated epithelial epithelium is a key pathogenic factor and closely associated with the recurrence of ulcerative colitis (UC). Here, we reported that vanillic acid (VA) exerted potent therapeutic effects on DSS-induced colitis by restoring intestinal epithelium homeostasis via the inhibition of ferroptosis. By the CETSA assay and DARTS assay, we identified carbonic anhydrase IX (CAIX, CA9) as the direct target of VA. The binding of VA to CA9 causes insulin-induced gene-2 (INSIG2) to interact with stromal interaction molecule 1 (STIM1), rather than SREBP cleavage-activating pr

Leishmaniasis is caused by infection with the protozoan parasites Leishmania. It is classified as one of the most significant neglected tropical diseases. It remains a significant global public health concern. Current treatments include the use of pentavalent antimonial, amphotericin B, pentamidine, miltefosine, and paromomycin. However, several limitations such as toxicity, side effect, and resistance to these drugs of certain species are of concern. To combat this disease, effective chemotherapy is urgently required for its treatment and management. In this study, we synthesized a series of

In recent years, there has been a growing emphasis on packaging made from biodegradable materials. These materials not only help to reduce environmental impact, but also extend the shelf life of food products, thereby contributing to a significant reduction in food waste. In order to develop novel biodegradable polymeric films for use as active packaging, polylactide materials filled with vanillic acid were prepared. Analyses were conducted to determine the influence of vanillic acid on the structural morphology and key food storage properties of the resulting films, including water vapor resi

Development of resistance to antibiotics is one of the major reasons of difficulties in treatments of diseases caused by antibiotic-resistant bacteria, and this resistance makes the investigation of alternative antimicrobials a key priority. Phenolic acids are plant- and fungi-originating natural antimicrobial products, and there is no known bacterial resistance after exposure to them. The purpose of this study was to investigate the resistance ability of bacteria against phenolic acids. Therefore, the ability of methicillin-resistant Staphylococcus aureus and methicillin-susceptible S. aureus

Thermally processed meat products are popular among consumers, but high-temperature processing generates heterocyclic amines (HCAs), particularly the highly toxic 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), which impairs liver and intestinal function. Research has demonstrated that vanillic acid (VA) possesses potent antioxidant and anti-inflammatory activities. However, its protective mechanism against IQ-induced liver injury remains unknown. This study aims to explore VA's mitigation effects and underlying mechanisms. Results reveal that VA alleviates IQ-induced colonic ferroptosis and int

Cancer cachexia is a cancer-associated disease characterized by gradual body weight loss due to pathologic muscle and fat loss, but effective treatments are still lacking. Here, we investigate the possible effect of vanillic acid (VA), known for its antioxidant, anti-inflammatory, and anti-obesity effects, on mitochondria-mediated improvement of cancer cachexia. We utilized cachexia-like models using CT26 colon cancer and dexamethasone. VA improved representative parameters of cancer cachexia including body weight loss and increased serum intereukin-6 levels. VA also attenuated muscle loss in

Vanillic acid (VA), widely used in industrial and therapeutic applications, also presents potential ecological and biological risks. This study systematically investigated the developmental and neurotoxic effects of VA in zebrafish and explored its mechanistic pathways within an Adverse Outcome Pathway (AOP) framework. VA exposure resulted in concentration-dependent toxicity, with an LC50 of 638.3 μM. High VA concentrations induced developmental abnormalities, including malformations of the eyes, otic vesicles, craniofacial structures, and tails. Behavioral assessments revealed a significant r

This study examined the potential of vanillic acid (VA) to protect against renal oxidative stress and inflammation caused by sodium arsenite (SA) in mice. Mice were assigned to five groups: control, VA (100 mg/kg), SA (50 ppm in drinking water for 8 weeks), and SA + VA (50 and 100 mg/kg orally in the 7th and 8th weeks). After the experiment was ended, the Mice were sacrificed and serum and renal tissue samples were collected for additional assessments. Treatment with VA suppressed SA-induced increase in blood urea nitrogen and creatinine in the serum. Furthermore, renal histological damage ind

Huntington disease (HD), an autosomal dominant neurodegenerative disorder characterized by involuntary choreatic movements with cognitive and behavioral disturbances. HD striatum has increased conversion of kynurenine to quinolinic acid (QA) which activates NMDA receptors leading to activation of microglia and increased levels of nuclear factor kappa B (NF-κB) leading to elevated transcription of inducible nitric oxide synthase (iNOS) and various cytokines causing neuronal death via neuroinflammation, oxidative stress, mitochondrial dysfunction and apoptosis. Therefore, inhibiting IKK-NF-κB pa

Non-alcoholic fatty liver disease (NAFLD), a growing health issue around the world, is defined as the presence of steatosis in the liver without any other detectable byproducts such as alcohol consumption, which includes a wide spectrum of pathologies, such as steatohepatitis, cirrhosis, and hepatocellular carcinoma. A growing body of evidence indicates that the reduction in the 5' adenosine monophosphate-activated protein kinase (AMPK) activity, which could be activated by the consumption of the drugs, hormones, cytokines, and dietary restriction, is related to some metabolic disorders such a

Cartilage endplate (CEP) plays essential roles in the initiation and progression of intervertebral disc degeneration. Vanillic acid (VA) is widely used as a food additive and has been shown to exert cardioprotective, anti-inflammatory, and antioxidant effects. Nevertheless, the specific effects and underlying mechanisms of VA on CEP chondrocytes have not been fully elucidated. The present study aimed to explore the impacts of VA on CEP degeneration and clarify its potential molecular mechanisms. To simulate the pathological microenvironment of IVDD, tumor necrosis factor-α (TNF-α) and tert-but

BACKGROUND: The aromatic compounds vanillin and vanillic acid are important fragrances used in the food, beverage, cosmetic and pharmaceutical industries. Currently, most aromatic compounds used in products are chemically synthesized, while only a small percentage is extracted from natural sources. The metabolism of vanillin and vanillic acid has been studied for decades in microorganisms and many studies have been conducted that showed that both can be produced from ferulic acid using bacteria. In contrast, the degradation of vanillin and vanillic acid by fungi is poorly studied and no genes
