We evaluated the effects of Tetrascorpin-1 from Androctonus australis (AaTs-1), a tetrapeptide obtained from scorpion venom, previously hypothesized to bind the formyl peptide receptor like-1 (FPRL-1) known as formyl peptide receptor-2 (FPR-2) in vitro, on pain responses and cytokines, neuronal and glial morpho-functional alterations in the spinal cord of mice with formalin-induced long-lasting pain hypersensitivity. Due to the peptide chemical nature and for favoring its penetration into the central nervous system, AaTs-1 was daily administered intranasally for 10 days. In formalin-injected m

BACKGROUND: Severe anaphylaxis to honeybee or vespid stings is associated with a variety of risk factors, which are poorly defined.
OBJECTIVE: Our aim was to evaluate the association of baseline serum tryptase concentrations and other variables routinely recorded during patient evaluation with the frequency of past severe anaphylaxis after a field sting.
METHODS: In this observational multicenter study, we enrolled 962 patients with established bee or vespid venom allergy who had a systemic reaction after a field sting. Data were collected on tryptase concentration, age, sex, culprit insect,

BACKGROUND: During a systemic hypersensitivity reaction (SR), an increase in serum tryptase compared to the baseline value is an indicator of mast cell activation, most often due to an IgE-mediated mechanism.
OBJECTIVE: To study the relevance of an increase in serum tryptase below the upper normal value of 11.4 ng/mL.
METHODS: Serum tryptase levels were measured in 35 patients with Hymenoptera venom hypersensitivity before and during venom exposure. Of these, 20 developed SR to stings or following venom injections during immunotherapy (reactors), while 15 tolerated reexposure to stings or ve

14 patients clinically hypersensitive to vespid sting by history, 6 patients with large local reactions and 10 control subjects were skin tested with serial two- and tenfold dilutions of venom and whole-body extract of yellow jacket, wasp, and white-faced hornet antigens. Radioallergosorbent test determinations with the same antigens were also made. Determination of minimal reactive dose by skin testing distinguished control and hypersensitive patients groups for all three antigens using either venom or whole body extract. Yellow jacket venom skin testing detected 12 of 14 patients considered

BACKGROUND: The common striped scorpion, Centruroides vittatus, and the imported fire ant (IFA) are endemic to the south-central United States. There is evidence of venom-specific IgE in patients experiencing hypersensitivity reactions to scorpion stings. The infrequency of repeated scorpion stings and the presence of immediate reactions to an initial sting suggest prior sensitization.
OBJECTIVE: In the present study we evaluated the cross-reactivity of C vittatus venom with IFA whole-body extract (WBE).
METHODS: Sera were obtained from patients with symptoms of immediate hypersensitivity to

BACKGROUND: The common striped scorpion, Centruroides vittatus, is endemic to the southwestern United States and causes thousands of human stings annually. Immediate hypersensitivity reactions to C. vittatus venom have been reported.
OBJECTIVES: To identify specific IgE in 11 patients with immediate hypersensitivity to C. vittatus and to characterize the allergens present in the venom.
METHODS: Skin testing to dialyzed, filtered venom was performed in 5 patients. Immunoglobulin E immunoblots to whole milked venom was accomplished with serum samples from 8 patients. Enzymatic properties of wh

BACKGROUND: It is hypothesized that because of higher mast cell numbers and mediator release, mastocytosis predisposes patients for systemic immediate-type hypersensitivity reactions to certain drugs including non-steroidal anti-inflammatory drugs (NSAID).
OBJECTIVE: To clarify whether patients with NSAID hypersensitivity show increased basal serum tryptase levels as sign for underlying mast cell disease.
METHODS: As part of our allergy work-up, basal serum tryptase levels were determined in all patients with a diagnosis of NSAID hypersensitivity and the severity of the reaction was graded.

Hypersensitivity to the stings of the Hymenoptera has been described since antiquity. The hypersensitive reactions to insect stings vary from minor skin reactions to severe and sometimes fatal anaphylaxis. Concerns about sting hypersensitivity have been increasing because of many incidents of allergic reactions of patients to the fire ant in the southern area of the United States as well as the harvester ant in some areas. We experienced one unique case with severe allergic reactions by ant of the Ectomomyrmex spp. of the subfamily Ponerinae, which is not a harvester ant. For three years the p

Hymenoptera stings are a common occurrence worldwide, with a significant proportion of the general population experiencing at least one sting during their lifetime. The order Hymenoptera includes several species, such as honey bees, hornets, bumblebees, and yellow jackets, that cause hypersensitivity reactions. The most common form of hypersensitivity is systemic sting reaction mediated by specific immunoglobulin E (sIgE) to venom components. The prevalence of systemic sting reaction in the general population is estimated to be approximately 3%, while gross local reactions are reported in 2.4-

BACKGROUND: There are scarce data about the prevalence of Hymenoptera venom allergy in the general population. The aim of this study was to determine the prevalence of Hymenoptera venom allergy in the general adult population of Istanbul.
METHODS: A total of 17,064 randomly selected telephone numbers were contacted and 11,816 (69.25%) individuals who agreed to participate completed a questionnaire. Those who disclosed hypersensitivity reactions due to Hymenoptera stings in this initial survey were called again and given another questionnaire. Those who were suspected of experiencing hypersens

Data are summarized in this Hymenoptera venom study (HVS) article on the safety of skin testing with venom extracts. Of the 3236 subjects studied, 89% had experienced an historical sting systemic reaction (SSR). Seventy-four percent of all subjects and 76% of subjects who had experienced an historical SSR had a positive skin test to at least one venom. More subjects tested positive to yellow jacket venom (51.8%) than to any other venom. There were no significant differences of the wheal and erythema sizes associated with different venoms or different historical sting reactions. Forty-five perc

The developmental and pharmacological characteristics of pain responses induced by the experimental scorpion BmK (Buthus martensi Karsch) sting were detailed in this study. Following the unilateral intraplantar injection of BmK venom into rat hind paw, it was found: 1) BmK venom induced an edematogenic response, spontaneous pain and pain hypersensitivity in a dose-dependent manner; 2) the paw edema and flare were induced rapidly and restricted at the injected paw for about 24-48 h; 3) the monophasic tonic spontaneous pain manifested as continuous paw flinching and lifting/licking of the inject

Background: Hymenoptera venom allergy is an immunoglobulin (Ig) E mediated hypersensitivity reaction to Hymenoptera venoms. Obvious identification of the culprit insect that causes the clinical symptoms and, hence, the accurate selection of venom for curative treatment, is of great importance for the effectiveness and safety of venom immunotherapy. Objective: In this study, the contribution of component-resolved diagnostics (CRD) is evaluated in the diagnosis of Hymenoptera venom allergy. Method: Ninety-three patients from four different centers in Turkey were included in the study. Convention

PURPOSE OF REVIEW: Allergy to insect stings remains a hazard worldwide and is the object of updated guidelines on management. This paper reviews the various clinical responses that may occur following an insect sting.
RECENT FINDINGS: Although the general population is at slight risk, certain groups are more susceptible, including occasionally stung adult male agricultural workers, hobby honey beekeepers and family members of beekeepers. Individuals with systemic mastocytosis are especially reactive to stings. The body of evidence attesting to the marked beneficial effect that 3-5 years of ve

Thirty-four patients with a history of immediate hypersensitivity to the sting of the imported fire ant were evaluated in a study designed to compare the diagnostic usefulness of fire ant whole body extract (WBE) preparations with that of fire ant venom (IFAV). Ninety-one percent (31/34) of the hypersensitive patients skin tested with IFAV at a maximal concentration of 1:5 X 10(3), v/v, demonstrated a wheal equal to or greater than the histamine control. Fifty-three percent (18/34) of the group were skin test positive to a WBE preparation. When the criteria for a positive skin test were relaxe

BACKGROUND: The increased incidence of bee-sting allergy among beekeepers is well known, whereas Hymenoptera hypersensitivity among forestry workers, who are frequently stung by Hymenoptera, is not well understood.
OBJECTIVE: To elucidate the nature of Hymenoptera hypersensitivity among forestry workers and analyze the mechanism involved in Hymenoptera-induced systemic reactions.
METHODS: A questionnaire was administered and yellow jacket venom-specific IgE antibody titers measured in 323 forestry workers and 100 age-matched office workers with little occupational exposure to Hymenoptera sti

We studied occupational injuries with captive lance-headed vipers (Bothrops moojeni) that occurred in a snake farm in south-eastern Brazil from February 1981 to May 1999. The risk of injury, taking into account 13 cases of snake-associated injuries (12 of them snake bites) was 2.73 per 10,000 person-days of work, and 3.51 per 100,000 venom extractions. Thirteen cases of injury occurred in seven workers, whereas 18 workers were never injured, suggesting that some individuals have a higher risk of injury than others perhaps due to lack of concentration or overconfidence. Eight episodes of occupa

Up to 5% of the population suffer from systemic, 19% from local allergic hypersensitivity reactions to stinging insects. Even though specific immunotherapy is very effective in treating allergy to insect venom, new concepts of treatment strategies with only the disease eliciting allergen in recombinant form, along with antigen application via a less invasive route might be suggested for enhanced treatment efficacy and compliance. In the present study we aimed (i) to establish a mouse model of wasp venom allergy, mimicking the natural mode of sensitization, and (ii) to develop a prophylactic tr
