Systemic Colonization of Potato Plants by Verticillium dahliae Leads to Infection of Tubers and Sprouting Buds.

A green fluorescent protein (GFP)-tagged isolate of Verticillium dahliae was used to study its colonization in potato plants and tubers. Three-week-old potato plants of the highly susceptible cultivar 'Shepody' were inoculated with a conidial suspension of a GFP-tagged isolate of V. dahliae using a wound inoculation method. Colonization was studied using confocal microscopy combined with tissue sections. Conidia germinated and hyphae grew along the root hairs, elongation zones, and root caps between 24 and 96 h postinoculation (HPI). At 7 days postinoculation (DPI), the pathogen advanced to co

Verticillium Wilt in Oilseed Rape-the Microbiome is Crucial for Disease Outbreaks as Well as for Efficient Suppression.

Microbiome management is a promising way to suppress verticillium wilt, a severe disease in Brassica caused by Verticillium longisporum. In order to improve current biocontrol strategies, we compared bacterial Verticillium antagonists in different assays using a hierarchical selection and evaluation scheme, and we integrated outcomes of our previous studies. The result was strongly dependent on the assessment method chosen (in vitro, in vivo, in situ), on the growth conditions of the plants and their genotype. The most promising biocontrol candidate identified was a Brassica endophyte Serratia

Cotton polyamine oxidase is required for spermine and camalexin signalling in the defence response to Verticillium dahliae.

Verticillium dahliae is a destructive, soil-borne fungal pathogen that causes vascular wilt disease in many economically important crops worldwide. A polyamine oxidase (PAO) gene was identified and cloned by screening suppression subtractive hybridisation and cDNA libraries of cotton genotypes tolerant to Verticillium wilt and was induced early and strongly by inoculation with V. dahliae and application of plant hormone. Recombinant cotton polyamine oxidase (GhPAO) was found to catalyse the conversion of spermine (Spm) to spermidine (Spd) in vitro. Constitutive expression of GhPAO in Arabidops

Reduced Dosage of Methyl Bromide for Controlling Verticillium Wilt of Potato in Experimental and Commercial Plots.

The use of gas-impermeable films to reduce the dosage of methyl bromide (MB) required to control Verticillium wilt in potatoes was examined in field experiments, conducted in soils naturally infested with Verticillium dahliae. The incidence and severity of Verticillium wilt were significantly reduced (by 74 to 94%) by fumigation with MB at 50 g/m2 under standard low density polyethylene (LDPE) or at 25 g/m2 under gas-impermeable films. Fumigation at 25 g/m2 under LDPE was less effective. Disease severity was inversely correlated (r2 = 0.89 to 0.91) with chlorophyll content in the leaves. Fumig

Multi-source UAV remote sensing for cotton Verticillium wilt resistance grading using TPAC-Net.

INTRODUCTION: Cotton Verticillium wilt resistance grading under field conditions remains challenging because disease symptoms are spatially heterogeneous within plots, local pathological cues are unevenly distributed across high-resolution unmanned aerial vehicle (UAV) orthomosaics, and disease-related responses are not expressed with the same sensitivity or spatial consistency across RGB, raw multispectral (MS), and vegetation-index (VI) representations. METHODS: To address these challenges, this study proposes TPAC-Net, a UAV-based Tri-source Pathological Alignment and Coupling Network for

Cotton SNARE complex component GhSYP121 regulates salicylic acid signaling during defense against Verticillium dahliae.

Syntaxin of plant (SYP) plays a crucial role in SNARE-mediated membrane trafficking during endocytic and secretory pathways, contributing to the regulation and execution of plant immunity against pathogens. Verticillium wilt is among the most destructive fungal diseases affecting cotton worldwide. However, information regarding SYP family genes in cotton is scarce. Through genome-wide identification and transcriptome profiling, we identified GhSYP121, a Qa SNARE gene in Gossypium hirsutum. GhSYP121 is notably induced by Verticillium dahliae, the causal agent of Verticillium wilt in cotton, and

Volatiles emitted by Pseudomonas aurantiaca ST-TJ4 trigger systemic plant resistance to Verticillium dahliae.

Verticillium dahliae is among the most devastating fungal pathogens, causing significant economic harm to agriculture and forestry. To address this problem, researchers have focused on eliciting systemic resistance in host plants through utilizing volatile organic compounds (VOCs) produced by biological control agents. Herein, we meticulously measured the quantity of V. dahliae pathogens in plants via RTqPCR, as well as the levels of defensive enzymes and pathogenesis-related (PR) proteins within plants. Finally, the efficacy of VOCs in controlling Verticillium wilt in cotton was evaluated. Fo

ATP-citrate lyase B (ACLB) negatively affects cell death and resistance to Verticillium wilt.

BACKGROUND: ATP-citrate lyase (ACL) plays a pivotal role in histone acetylation and aerobic glycolysis. In plant, ACL is a heteromeric enzyme composed of ACLA (45 kD) and ACLB (65 kD). So far, the function of ACL genes in cotton still remains unknown. RESULTS: Here, we identified three ACLA homologous sequences and two ACLB homologous in each genome/sub-genome of cotton species. Silencing ACLB in cotton led to cell death at newly-grown leaves and stem apexes. Simultaneously, in ACLB-silenced plants, transcription factors related to senescence including SGR, WRKY23 and Osl57 were observed to b

High-throughput qPCR phenotyping of Verticillium stripe resistance in Brassica napus under natural field infection.

Verticillium stripe, caused by Verticillium longisporum, is an emerging threat to Brassica napus production in North America, with increasing field incidence and yield loss reported in affected regions. Although visual ratings are useful for rapid field assessment, resistance breeding is constrained by the lack of scalable, objective phenotyping methods that can quantify pathogen colonization across large genotype panels under field conditions. Here, we adapted and validated an established OLG70/OLG71-based SYBR Green qPCR assay within a high-throughput workflow combining freeze-dried hypocoty

PPtase-activated production of borrelidin from extremophilic actinobacteria against multidrug-resistant cotton pathogen Verticillium dahliae.

As the most devastating disease in cotton crops, Verticillium wilt caused by Verticillium dahliae led to fiber quality reduction and extensive yield loss. In recent years, Verticillium wilt has been increasingly serious in Xinjiang, China, the world's largest cotton production area. In this study, 52 actinobacterial strains were selected for the discovery of NPs with anti-V. dahliae activity, from our strain collection isolated from three types of extreme habitats (high salinity, high temperature or plant endophytes) in Xinjiang. In the culture broth extracts of phosphopantetheinyl transferase

Co-Infecting Mycoviruses VdPV1 and VdMoV1 Attenuate Verticillium dahliae and Are Transmitted Vertically and Horizontally.

Cotton verticillium wilt caused by Verticillium dahliae severely restricts global cotton production. Mycovirus-induced hypovirulence provides a promising strategy for sustainable disease management. In this study, V. dahliae isolates from Xinjiang were screened by virome sequencing and RT-PCR, and strain 121-11C-1 co-infected with Verticillium dahliae partitivirus 1 (VdPV1) and Verticillium dahliae magoulivirus 1 (VdMoV1) was identified. Virus-free isogenic strains were obtained via single-conidium purification, verifying that both viruses can be vertically transmitted through fungal conidia.

Preparation of a Fluxapyroxad Nanoformulation with Strong Plant Uptake for Efficient Control of Verticillium Wilt in Potato.

Potato (Solanum tuberosum L.) is ranked as the fourth largest staple crop in China. However, potato production is increasingly threatened by Verticillium wilt (VW) caused by the fungus Verticillium dahliae in various provinces. In the present study, we explored the application of star polycation (SPc) nanocarrier to improve the effectiveness of the fungicide fluxapyroxad (Flu) in combating VW. The SPc self-assembled with Flu through hydrogen bonds and van der Waals forces to form the Flu/SPc complex spontaneously, which exhibited strong intermolecular interactions, as indicated by a high affin

Defense Mechanisms of Cotton Fusarium and Verticillium Wilt and Comparison of Pathogenic Response in Cotton and Humans.

Cotton is an important economic crop. Fusarium and Verticillium are the primary pathogenic fungi that threaten both the quality and sustainable production of cotton. As an opportunistic pathogen, Fusarium causes various human diseases, including fungal keratitis, which is the most common. Therefore, there is an urgent need to study and clarify the resistance mechanisms of cotton and humans toward Fusarium in order to mitigate, or eliminate, its harm. Herein, we first discuss the resistance and susceptibility mechanisms of cotton to Fusarium and Verticillium wilt and classify associated genes b

Transcriptome profiling of Gossypium barbadense inoculated with Verticillium dahliae provides a resource for cotton improvement.

BACKGROUND: Verticillium wilt, caused by the fungal pathogen Verticillium dahliae, is the most severe disease in cotton (Gossypium spp.), causing great lint losses worldwide. Disease management could be achieved in the field if genetically improved, resistant plants were used. However, the interaction between V. dahliae and cotton is a complicated process, and its molecular mechanism remains obscure. To understand better the defense response to this pathogen as a means for obtaining more tolerant cultivars, we monitored the transcriptome profiles of roots from resistant plants of G. barbadense

Verticillium dahliae alters Pseudomonas spp. populations and HCN gene expression in the rhizosphere of strawberry.

The production of hydrogen cyanide (HCN) by beneficial root-associated bacteria is an important mechanism for the biological control of plant pathogens. However, little is known about the biotic factors affecting HCN gene expression in the rhizosphere of plants. In this study, real-time reverse transcription PCR (qRT-PCR) assays were developed to investigate the effect of the plant pathogen Verticillium dahliae on hcnC (encoding for HCN biosynthesis) gene expression in Pseudomonas sp. LBUM300. Strawberry plants were inoculated with Pseudomonas sp. LBUM300 and (or) V. dahliae and grown in pots

Restructuring tilth layers suppresses cotton Verticillium wilt through the niacinamide-mediated enrichment of beneficial Pseudomonas.

Restructuring tilth layers (RTL) is an innovative tillage practice that involves the vertical exchange of topsoil and subsoil while the deeper layer is loosened, and this practice has been verified to significantly reduce the incidence of cotton Verticillium wilt. However, the ecological mechanisms underlying disease suppression remain unclear. In this study, we integrated field experiments, metagenomic sequencing, untargeted metabolomics, and functional validation to elucidate the effects of RTL on the rhizosphere ecosystem from the perspectives of microbe and metabolite interactions. The complex subunit VdAP-2α regulates polar growth, ergosterol metabolism, and virulence in Verticillium dahliae.

The normal polar growth and morphogenesis represent fundamental factors in development, reproduction survival and host invasion of phytopathogenic fungi. The heterotetramerized AP-2 complex is widely present in eukaryotes and plays a specific role in clathrin-mediated vesicle formation and transport at the plasma membrane in mammals and plants. However, the functions of AP-2 complex in growth, metabolism and virulence of plant pathogenic fungi remain unclear. In this study, the conserved VdAP-2α subunit was identified in Verticillium dahliae by screening of a T-DNA insertion library. VdAP-2α w

Verticillium dahliae Secretory Aspartyl Protease VdSAP Targets Cotton GhARP to Modulate Plant Defence and Defoliation.

Verticillium dahliae isolates causing devastating vascular wilt in cotton plants can be divided into defoliating and nondefoliating pathotypes. The mechanisms underlying how V. dahliae uses secretory proteins to manipulate plant physiological processes and suppress immunity in cotton plants have received renewed research focus over the past several years. Here, we describe the role of a secretory protein named VdSAP (secretory aspartyl protease) from the defoliating V. dahliae strain XJ592 in virulence and defoliation. Deletion of VdSAP in strain XJ592 led to significantly reduced virulence an

Salicylic acid-related cotton (Gossypium arboreum) ribosomal protein GaRPL18 contributes to resistance to Verticillium dahliae.

BACKGROUND: Verticillium dahliae is a phytopathogenic fungal pathogen that causes vascular wilt diseases responsible for considerable decreases in cotton yields. The complex mechanism underlying cotton resistance to Verticillium wilt remains uncharacterized. Identifying an endogenous resistance gene may be useful for controlling this disease. RESULTS: We cloned the ribosomal protein L18 (GaRPL18) gene, which mediates resistance to Verticillium wilt, from a wilt-resistant cotton species (Gossypium arboreum). We then characterized the function of this gene in cotton and Arabidopsis thaliana pla
