Genetic and molecular characterization of the polar flagellum of Vibrio parahaemolyticus.

Vibrio parahaemolyticus possesses two alternate flagellar systems adapted for movement under different circumstances. A single polar flagellum propels the bacterium in liquid (swimming), while multiple lateral flagella move the bacterium over surfaces (swarming). Energy to rotate the polar flagellum is derived from the sodium membrane potential, whereas lateral flagella are powered by the proton motive force. Lateral flagella are arranged peritrichously, and the unsheathed filaments are polymerized from a single flagellin. The polar flagellum is synthesized constitutively, but lateral flagella

Phytochemical analysis and antibacterial activity of palm waste extract against Vibrio harveyi and Vibrio parahaemolyticus.

Vibrio harveyi and Vibrio parahaemolyticus are species of the Vibrio genus that often cause disease and mass mortality in crustaceans. If not handled quickly and appropriately, these diseases can cause considerable losses to farmers. Therefore, it is necessary to find a solution with safe and environmentally friendly disease prevention technology using natural ingredients, among others from plants, namely oil palm. Some parts of oil palm, namely leaves, fronds, fibres and oil palm pulp, which are palm waste, contain antibacterial compounds. This study aimed to assess the antibacterial activity

Molecular and pathogenic characterization of Vibrio parahaemolyticus isolated from seafood.

Gastroenteritis infections in humans are mainly associated with consumption of Vibrio parahaemolyticus contaminated shellfish, which causes health and economic loss. Virulence factor production, antibiotic resistance profile, and biofilm-forming capacity of Vibrio parahaemolyticus isolates on food and food contact surfaces at 30 °C were investigated to evaluate the antibiotic sensitivity and pathogenic level. Strains of V. parahaemolyticus were isolated from shellfish (e.g., Crassostrea gigas, Venerupis philippinarum, Mytilus coruscus, Anadara kagoshimensis) in Korea. When examined for 17 viru

Vibrio parahaemolyticus infectious for both humans and edible mollusk abalone.

The aims of this study are to report evidence of the first laboratory-acquired infection of Vibrio parahaemolyticus associated with handling experimentally infected abalones and to describe the virulence of the two bacterial strains tested in these animals. Two strains of V. parahaemolyticus, one from the stool of a patient with acute gastroenteritis (strain 880713) and the other from the hemolymph of a diseased small abalone Haliotis diversicolor supertexta (strain 880915), were identified and characterized. Both strains were lethal to small abalone, with similar LD(50) values (8.36-8.41 x 10

Global metabolic regulation in Vibrio parahaemolyticus under polymyxin B stimulation.

Vibrio parahaemolyticus is responsible for infection diseases of people who consume the contaminated seafood, but its metabolic regulation profile in response to colistin, the last treatment option for multidrug-resistant Gram-negative bacteria, remains unclear. In this study, the metabolic regulation profile of V. parahaemolyticus ATCC33846 under polymyxin B stimulation has been investigated. V. parahaemolyticus exposed to polymyxin B resulted in 4597 differentially transcribed genes, including 673 significantly up-regulated genes and 569 significantly down-regulated genes. In V. parahaemolyt

Gp8 mediates adsorption of bacteriophage vB_VpP_jzsmvpa to the host Vibrio parahaemolyticus.

In this study, we aimed to identify the principal tail protein responsible for mediating adsorption of bacteriophage vB_VpP_jzsmvpa to its host bacterium Vibrio parahaemolyticus, a significant pathogen in aquaculture. The structure and function of three candidate proteins-the tail tubular protein Gp8, tail completion protein Gp20, and tail fibre protein Gp41-were predicted through bioinformatic analysis. Recombinant plasmids were constructed and the three tail proteins were successfully expressed and purified. Competitive adsorption assays demonstrated an 81.3% reduction in bacteriophage adsor

New insights into the resistome-mobilome-biofilm relationship in Vibrio parahaemolyticus.

Vibrio parahaemolyticus is a major foodborne pathogen frequently isolated from seafood, notable for its biofilm-forming capacity, which enhances its persistence in environments and increases resistance to disinfection. This study investigated the virulome, the associations between the resistome and mobilome of V. parahaemolyticus and its biofilm forming ability to better understand the genomic determinants of persistence. Among 845 isolates from imported seafood (mainly shrimp), 20.6% showed acquired resistance to at least one clinically important antibiotic class, most commonly tetracycline,

Prevalence, antibiotic susceptibility and characterization of Vibrio parahaemolyticus isolates in China.

Vibrio parahaemolyticus is a marine and estuarine bacterium that poses a major threat to human health worldwide. In this study, from 2017 to 2019, we evaluated 900 food samples collected from China in 2017, with the aim of determining the incidence and features of V. parahaemolyticus in ready-to-eat (RTE) foods, shrimp and fish in China. The contamination rates in these were 3.67, 19.33 and 10.67%, respectively, and the prevalence of V. parahaemolyticus was higher in summer than in winter. In addition, 101 V. parahaemolyticus strains were isolated. Our results suggested that most of the isolat

Galleria mellonella as an infection model to investigate virulence of Vibrio parahaemolyticus.

Non-toxigenic V. parahaemolyticus isolates (tdh-/trh-/T3SS2-) have recently been isolated from patients with gastroenteritis. In this study we report that the larvae of the wax moth (Galleria mellonella) are susceptible to infection by toxigenic or non-toxigenic clinical isolates of V. parahaemolyticus. In comparison larvae inoculated with environmental isolates of V. parahaemolyticus did not succumb to disease. Whole genome sequencing of clinical non-toxigenic isolates revealed the presence of a gene encoding a nudix hydrolase, identified as mutT. A V. parahaemolyticus mutT mutant was unable

Features of cholera and Vibrio parahaemolyticus diarrhoea endemicity in Calabar, Nigeria.

The clinical and epidemiological features of acute vibrio diarrhoeal disease were studied in 881 patients seen at the University of Calabar Teaching Hospital (UCTH), Calabar, Nigeria, between January and December 1989. Stools and rectal swabs of patients and randomly-selected control subjects were microscopically and culturally examined for the presence of enteric pathogens. Households of vibrio diarrhoea cases and matched controls were visited for ecologic studies. Of a total of 108 (12.3%) culturally-confirmed bacterial diarrhoeas, 47 (43.5%) were due to Escherichia coli, 33 (30.6%) to Vibri

Ultrasound-assisted blue light killing Vibrio parahaemolyticus to improve salmon preservation.

Vibrio parahaemolyticus is a typical marine bacterium, which often contaminates seafood and poses a health risk to consumers. Some non-thermal sterilization technologies, such as ultrasonic field (UF) and blue light (BL) irradiation, have been widely used in clinical practice due to their efficiency, safety, and avoidance of drug resistance, but their application in food preservation has not been extensively studied. This study aims to investigate the effect of BL on V. parahaemolyticus in culture media and in ready-to-eat fresh salmon, and to evaluate the killing effectiveness of the UF combi

ALF-like mediates Penaeus vannamei innate immunity by recognizing Vibrio parahaemolyticus.

The coordination between reactive oxygen species (ROS) and antimicrobial peptide (AMP) responses in invertebrate immunity remains poorly understood. Here, we identify the anti-lipopolysaccharide factor-like protein (ALF-like) from Penaeus vannamei as a functional hub that bridges these two defense layers. We show that ALF-like directly recognizes bacterial components and exerts broad-spectrum bactericidal activity. More importantly, in vivo knockdown and rescue experiments reveal that ALF-like not only restricts bacterial dissemination but also prevents excessive ROS production and modulates t

BfrR-mediated biofilm regulation in the seafood-borne pathogen Vibrio parahaemolyticus.

Vibrio parahaemolyticus is a major seafood-associated pathogen whose biofilm formation enhances environmental persistence, antibiotic resistance, and food safety risks. In this study, we identified a LysR-type regulator, BfrR (VPA1687), as a critical mediator of biofilm development in V. parahaemolyticus. Deletion of bfrR severely impaired biofilm formation, switching colony morphology from wrinkled to smooth and reducing extracellular matrix components (exopolysaccharides (EPS), proteins, and extracellular DNA). BfrR also modulated metabolic activity in biofilm-embedded cells and exhibited du

Genetic and virulence characterisation of Vibrio parahaemolyticus isolated from Indian coast.

BACKGROUND: V. parahaemolyticus is autochthonous to the marine environment and causes seafood-borne gastroenteritis in humans. Generally, V. parahaemolyticus recovered from the environment and/or seafood is thought to be non-pathogenic and the relationship between environmental isolates and acute diarrhoeal disease is poorly understood. In this study, we explored the virulence potential of environmental V. parahaemolyticus isolated from water, plankton and assorted seafood samples collected from the Indian coast. RESULTS: Twenty-two V. parahaemolyticus isolates from seafood harboured virulenc
