Visna/Maedi virus genetic characterization and serological diagnosis of infection in sheep from a neurological outbreak.

An extensive outbreak characterized by the appearance of neurological symptoms in small ruminant lentivirus (SRLV) infected sheep has been identified in Spain, but the genetic characteristics of the strain involved and differential diagnostic tools for this outbreak remain unexplored. In this work, 23 Visna-affected naturally infected animals from the outbreak, 11 arthritic animals (both groups presenting anti-Visna/Maedi virus serum antibodies), and 100 seronegative animals were used. Eight of the Visna-affected animals were further studied post-mortem by immunohistochemistry. All had lesions

[Neurotoxicity in mice due to cysteine-rich parts of visna virus and HIV-1 Tat proteins].

The trans-activating visna virus and HIV-1 Tat proteins share, at their amino-acid sequence level, a significant 60% analogy on 17 consecutive residues. These homologous sequences are also found in a part of the short neurotoxin sequence from snake venom. Synthetic peptides representative of the two analogous viral sequences are, after intracerebroventricular injection at doses of 200 micrograms per 20 g mouse, responsible for the death of the injected animal in few hours. The HIV-1 recombinant Tat protein has the same effect. Such observation suggests a direct role of the Tat lentiviral prote

Treatment of visna virus infection in lambs with the acyclic nucleoside phosphonate analogue 9-(2-phosphonylmethoxyethyl)adenine (PMEA).

Nucleoside and nucleotide analogues, which are inhibitors of human immunodeficiency virus reverse transcriptase, are highly active inhibitors of visna virus replication in cell cultures. One such analogue, the acyclic nucleoside phosphonate PMEA, has also been found to have a prophylactic effect on visna virus infection in lambs. In the present study, lambs were injected subcutaneously with 10 mg/kg PMEA three times a week starting 4 weeks after inoculation with visna virus, when brain infection had been established. After 3 weeks of treatment there was a reduction in the amount of virus isola

The visna virus genome: evidence for a hypervariable site in the env gene and sequence homology among lentivirus envelope proteins.

The complete nucleotide sequence of the visna virus 1514 genome was determined. Our sequence confirms the relationship of visna virus and other lentiviruses to human immunodeficiency virus (HIV) both at the level of sequence homology and of genomic organization. Sequence homology is shown to extend to the transmembrane proteins of lentivirus env genes; this homology is strongest in the extracellular domain, suggesting that close structural and functional similarities may also exist among these envelope proteins. Comparison of our data with the sequence of visna virus LV1-1, an antigenic varian

Retroviral arthritis: phenotypic analysis of cells in the synovial fluid of sheep with inflammatory synovitis associated with visna virus infection.

A phenotypic analysis on synovial fluid cells from the carpal and tarsal joints of sheep with visna virus-induced inflammatory synovitis was performed. The results showed increased representation of cells bearing lymphocyte, macrophage, and dendritic cell markers compared to equivalent synovial fluid cells from normal uninfected age-matched controls. In infected sheep, CD8+ T cells tended to predominate over CD4+ cells, while the numbers of gamma delta T cells varied from being absent in some samples to constituting the major T cell subset in others. B cells were found in relatively smaller nu

Alternative Molecular Tools for the Fight against Infectious Diseases of Small Ruminants: Native Sicilian Sheep Breeds and Maedi-Visna Genetic Susceptibility.

Maedi-visna (MV) is a disease caused by small ruminant lentiviruses. It is included in the list of notifiable terrestrial animal diseases due to economic losses and animal welfare harm in the sheep sector. To date, control programs remain the onliest approach to avoiding infection. The allelic variant p.Glu35Lys (E35K) of the TMEM154 gene has been strongly associated with host vulnerability to MV illness. The present study aimed to investigate the association of TMEM154 E35K allele frequencies with MV susceptibility in native Sicilian sheep breeds. More than 400 animals from 14 local sheep wer

[Results of a serological study of the natural spread of maedi-visna virus infection in a sheep herd during an observation period of 30 months].

In a sheep population consisting of animals of different breeds the epidemiologic development of the Maedi-Visna-infection was evaluated by means of monthly Immune Diffusion Test (ID) for a period of 30 months. At the beginning the herd consisted of eight animals, where four of them showed clinical symptoms indicative for a MV-infection with positive serological results. The other 4 animals were negative. The herd increased in number by including the offspring and buying in other animals. During the time of 30 months only in one animal a seroconversion could be observed. In nine of 13 lambs bo

[Proposal of the DVG Specialist Group "Diseases of Small Ruminants and New World Camelids" for a program to combat Maedi-Visna infection in sheep herds and caprine arthritis-encephalitis infection in goat herds].

ABSTRACT: The control of small ruminant lentivirus (SRLV) infections - Maedi, Visna, and caprine arthritis and encephalitis (CAE) - is based on the establishment of SRLV-free closed herds, achieved by serologically testing of all small ruminants aged one year and older for antibodies against SRLV. After four completely negative serological test results within a period of at least 24 months, a farm is recognized as SRLV-free. Maintaining SRLV-free status is based on regular serological testing of all small ruminants in the herd over one year of age. To reduce the administrative and financial bu

Retroviral interference with neuronotrophic signaling in human motor neuron disease?

We tested 13/32 patients with non-familial amyotrophic lateral sclerosis (ALS) positive (41%) for human spuma retrovirus (HSRV) seroreactivity. Half the non-ALS HSRV-positive neurologic patients (12/25) reported multiple fasciculations within the last two years. Sera from four HSRV-env negative ALS patients measurably competed with visna antibodies on maedi-visna antigen. The serologies for bovine leukemia virus, Borna disease virus and GM1IgM-antibodies remained negative. Both visna and foamy virus sequences share epitopes with motor nervous system signal polypeptides, suggesting a retroviral

[In vitro fusion ability of agents causing slow diseases of the central nervous system].

The authors study cell fusion in vitro induced by two kinds of slow viruses : visna virus of sheep and scrapie of mouse and Creutzfeldt-Jakob disease of man agents. Cell-fusion induced by visna virus is either slow and late occuring (fusion from within) or early and rapid (fusion from without) according to experimental conditions. Whereas, cell-fusion induced by spongiform encephalopathies agents is only of slow appearance, looking an endogenous phenomenon.

Human T-cell lymphotropic virus type III shares sequence homology with a family of pathogenic lentiviruses.

The etiologic agent of the acquired immune deficiency syndrome, human T-cell lymphotropic virus type III (HTLV-III), has recently been shown to morphologically resemble and share sequence homology with visna virus, a pathogenic lentivirus. Molecular hybridization, heteroduplex mapping, and DNA sequence analyses were used to compare HTLV-III to other lentiviruses of domestic animals, including visna, caprine arthritis encephalitis, and equine infectious anemia viruses. Hybridization results showed that a substantial amount of sequence homology exists between each of these viruses and HTLV-III.

Infectious pathogens potentially transmitted by semen of the black variety of the Manchega sheep breed: Health constraints for conservation purposes.

Conservation of genetic resources from endangered breeds may be conducted through germinal banks. Preservation of healthy samples is paramount to avoid preserving pathogens shed with germinal products. The black variety of Manchega sheep (BMS), and endangered breed endemic to south-central Spain, is the subject of a conservation program; a germinal bank has been recently established. However, several pathogens circulating in BMS flocks may be shed with semen and threaten BMS preservation. Therefore, we investigated the sanitary status of BMS flocks and semen samples from 4 of the 17 flocks in

A review of the sheep-multiple sclerosis connection.

This paper reviews a notion that the prevalence of multiple sclerosis is high in global areas where sheep populations are concentrated. Pilot studies are reported to serum antibodies in humans to three sheep diseases; focal symmetrical encephalomalacia (FSE), maedi visna and sarcocystis. In MS patients and controls antibodies were not found to the epsilon neurotoxin of the FSE organism, Clostridium welchii type D and to a caprine retrovirus that is closely related to maedi-visna virus. However, 34% of MS and control patients had antibodies to the protozoan parasite Sarcocystis spp., tissue cys

Isolation and identification of a South African lentivirus from jaagsiekte lungs.

In the course of attempts to grow the jaagsiekte retrovirus in cell culture, a typical lentivirus was isolated for the first time in South Africa from adenomatous lungs. Morphologically the virus could not be distinguished from other lentiviruses, but serologically it was shown to be more closely related to visna virus than to caprine arthritis-encephalitis virus. However, a preliminary restriction enzyme analysis of the linear proviral DNA of this new lentivirus (SA-DMVV) revealed that it is significantly district from visna virus and CAEV and therefore may represent a third type of lentiviru
