A review of the sheep-multiple sclerosis connection.

This paper reviews a notion that the prevalence of multiple sclerosis is high in global areas where sheep populations are concentrated. Pilot studies are reported to serum antibodies in humans to three sheep diseases; focal symmetrical encephalomalacia (FSE), maedi visna and sarcocystis. In MS patients and controls antibodies were not found to the epsilon neurotoxin of the FSE organism, Clostridium welchii type D and to a caprine retrovirus that is closely related to maedi-visna virus. However, 34% of MS and control patients had antibodies to the protozoan parasite Sarcocystis spp., tissue cys

Amplicon sequencing with Oxford nanopore technologies as a diagnostic alternative for small ruminant lentiviruses in sheep.

In Europe, Maedi-Visna disease has high prevalence rates at the individual and flock levels, respectively, and is regarded as one of the most significant infectious disease in sheep. The lack of treatment or a commercial vaccine underscores the need for accurate and reliable diagnostic tools to support control programs. Conventional methods, including ELISA and qPCR, provide useful but incomplete information due to the genetic variability of small ruminant lentiviruses (SRLVs) and the heterogeneous host immune response. In this work, third-generation sequencing was assessed as a diagnostic str

Autoimmune reactivity in sheep induced by the visna retrovirus.

Sheep, either naturally or experimentally infected with visna virus, were examined for the production of autoantibodies. Elevated titres of IgM and IgG antiglobulins and rheumatoid factors as well as antibodies to ssDNA, cardiolipin or histones were found in serum of both groups of animals using ELISA techniques. IgA autoantibodies were usually absent or present in low titres. The specificity of the autoantibody reactivities was confirmed by inhibition assays, affinity chromatography, Western blotting and agglutination methods. Antiglobulin titres were slightly elevated in synovial fluids, alt

[Disease concept of the slow virus infection].

This article gives a brief history of the terminology of slow virus infection, the conceptual change that occurred in it, the features common to slow infection and the current concept of slow virus infection. Björn Sigurdsson from the field of veterinary medicine proposed slow virus infection as unique mode of infection in 1954. Its initial concept was remodeled along with the general acceptance of prion theory of sheep scrapie that was proposed in 1982. The features common to slow infection include very long latency, unanimous poor prognosis, central nervous system involvement, etc. Currently

Pulmonary adenomatosis and maedi-visna in Ethiopian central highland sheep: a microscopic study.

Microscopic examination of pneumonic lungs of the Ethiopian highland sheep (n = 35) was made and compared with the pneumonic lungs from ten sheep and 66 goats from the lowlands. Lesions compatible with sheep pulmonary adenomatosis (SPA; 8/35, 22.8%), and maedi-visna (MV; 9/35, 25.7%) were recorded only in sheep from the central highlands. Interstitial pneumonia (43.2%), bronchopneumonia (35.1%), and verminous pneumonia (6.3%) were recorded in both sheep and goats from the high- and the lowlands. SPA was documented for the first time in sheep from Ethiopia in this report. We believe that MV and

Molecular characteristics and prevalence of small ruminant lentiviruses in goats in Japan.

Small ruminant lentiviruses (SRLVs), which comprise caprine arthritis-encephalitis virus (CAEV) and maedi-visna virus (MVV), are prevalent in goats and sheep worldwide, including in Japan. However, little is known about the molecular characteristics of goat lentiviruses in Japan. In this study, a molecular and phylogenetic analysis of the long gag region was performed. The phylogenic tree demonstrated that all samples belonged to SRLV subtype B1. Two clusters were identified, with one cluster distinct from previously reported strains of subtype B1. In addition, several alterations in the amino

Pathological, molecular, and serological study of small ruminant lentiviruses in Jordan.

BACKGROUND AND AIM: Maedi-visna is a chronic viral disease of sheep with worldwide distribution causing substantial economic losses to the small ruminant industry. Pneumonia and mastitis are the main manifestations of the disease. This study aimed to investigate the occurrence of maedi-visna virus (MVV) in sheep using histopathology and nested polymerase chain reaction (PCR) techniques and also to estimate the seroprevalence of small ruminant lentiviruses (SRLVs) in sheep and goats using commercially available enzyme-linked immunosorbent assay (ELISA). MATERIALS AND METHODS: Lung tissue sampl

Pathology of the Mammary Gland in Sheep and Goats.

The recognition of lesions of the mammary gland in small ruminants is a useful diagnostic procedure that can aid in the identification of several udder diseases. This article reviews the main pathological lesions in this organ in sheep and goats. Mastitis is, by far, the most commonly diagnosed change. Acute clinical mastitis is associated with bacterial infections, mainly Staphylococcus aureus or Mannheimia haemolytica. Lesions related to subclinical and chronic mastitis are also described, either as localized cases or as a part of systemic diseases such as contagious agalactia, maedi-visna o

[Interferon in sheep].

There is only limited information on sheep interferon available. Recent publications have reported on: 1. an interferon (IFN) alpha subtype, which is secreted by the fetal trophectoderm into the lumen of the uterus between the 10th and 21st day of gestation. It was therefore named ovine trophoblast protein (oTP-1), and is responsible for signalling pregnancy to the ewe via high affinity receptors in the endometrium. It is thought that oTP-1 acts by directly influencing prostaglandin metabolism. 2. the role of lentivirus-induced interferon (LV-IFN) in the pathogenesis of Maedi/Visna. The result

Clinical and pathological investigation of Maedi-visna outbreak in a farm from Bulgaria.

BACKGROUND: In the autumn of 2024, an outbreak of Maedi-visna (MV) was identified in Bulgaria in the village of Sigmen, Burgas district. The disease is a slowly progressive, persistent, usually subclinical viral infection in sheep, distributed worldwide. Difficulties in early diagnosis are the long period between infection and the dynamics of the antibody titer (seroconversion), the emission of an infectious onset without clinical signs, the lack of vaccines, as well as some ongoing misunderstandings in the epidemiology and the transmission mechanisms, which determine the complex nature of the

[Proposal of the DVG Specialist Group "Diseases of Small Ruminants and New World Camelids" for a program to combat Maedi-Visna infection in sheep herds and caprine arthritis-encephalitis infection in goat herds].

ABSTRACT: The control of small ruminant lentivirus (SRLV) infections - Maedi, Visna, and caprine arthritis and encephalitis (CAE) - is based on the establishment of SRLV-free closed herds, achieved by serologically testing of all small ruminants aged one year and older for antibodies against SRLV. After four completely negative serological test results within a period of at least 24 months, a farm is recognized as SRLV-free. Maintaining SRLV-free status is based on regular serological testing of all small ruminants in the herd over one year of age. To reduce the administrative and financial bu

Isolation and identification of a South African lentivirus from jaagsiekte lungs.

In the course of attempts to grow the jaagsiekte retrovirus in cell culture, a typical lentivirus was isolated for the first time in South Africa from adenomatous lungs. Morphologically the virus could not be distinguished from other lentiviruses, but serologically it was shown to be more closely related to visna virus than to caprine arthritis-encephalitis virus. However, a preliminary restriction enzyme analysis of the linear proviral DNA of this new lentivirus (SA-DMVV) revealed that it is significantly district from visna virus and CAEV and therefore may represent a third type of lentiviru

[Antibodies to certain infections on large goat farms in the Czech Republic].

On five newly established large goat farms the incidence of antibodies to some chronic and latent infections (arthritis and encephalitis, Q-fever, caseous lymphadenitis and toxoplazmosis) was investigated in the first six months of the year 1994. Agar-gel immunodiffusion did not reveal any antibodies to arthritis and encephalitis of goats (CAE). Complement fixation test did not demonstrate any antibodies to Q-fever. Neither agar-gel immunodiffusion nor neutralization test confirmed any antibodies to caseous lymphadentitis. Complement fixation test (titer 1:8 and more) revealed antibodies to to

[Amyotrophic lateral sclerosis--indications of increased antiretroviral seroreactivity without obvious epidemiology].

Of 25 male and 13 female patients diagnosed as sporadic cases of amyotrophic lateral sclerosis between 1989 and 1992, 17 had reproducible serum antibodies against human foamy virus (HFV = human spuma retrovirus, HSRV). HFV-positive ALS patients had higher IgG3 concentrations than HFV-negatives (P < 0.05) and competed better on maedi-visna retroviral antigen than HFV-negatives or controls (P < 0.05), but did not differ otherwise. Two HFV-positive patients were living in the same building; two other ALS patients lived within 300 m of one another, and three HFV gag reactive men (2 ALS, 1 control)

Slow viruses and chronic disease of the central nervous system.

Although recognized since the 1930s, slow infections have only recently received considerable attention. There are three types, group A (related to the type C RNA viruses), group B (bizarre agents such as scrapie and kuru) and group C (viruses such as measles which normally produce acute infections but which are behaving here in an unusual fashion). These viruses are only united in that they produce disease with excessively long incubation periods. Many slow infections result in neurological diseases and these will be discussed, together with some possible explanations of their action.

Ultrasonography and digital radiography findings in sheep with clinical disease associated with small ruminant lentivirus infection.

Digital radiography and ultrasonographic images were used in this case series to evaluate 4 ewes from a single flock for chronic weight loss and ill-thrift. On examination, all displayed tachypnea, dyspnea, coughing, and normothermia with abnormal thoracic auscultations. Three of the 4 animals were diagnosed with chronic respiratory disease associated with Maedi-visna (MV) infection confirmed via serologic testing. Diagnostic thoracic imaging identified characteristics consistent with pathological lesions associated with interstitial pneumonia in the 3 MV affected animals; these findings were

Phylogenetic analysis of small ruminant lentiviruses in Mongolian sheep supports an ancient east-west split for the genotype A.

The ovine maedi-visna virus (MVV) and caprine arthritis-encephalitis virus (CAEV) are small ruminant lentiviruses (SRLVs) with striking genetic and structural similarities. The presence of SRLV in Mongolian sheep and goats was serologically demonstrated more than a decade ago; however, the viral genotype remains unknown. In total, 329 blood samples were collected from two sheep breeds (i.e., Khalkha and Sumber) in Tov, Govisumber, Arkhangay, Dornogovi, Zavkhan, and Sukhbaatar provinces, Mongolia. Serological and phylogenetic analyses were performed regardless of any apparent clinical signs, al

A polytropic caprine arthritis encephalitis virus promoter isolated from multiple tissues from a sheep with multisystemic lentivirus-associated inflammatory disease.

Caprine arthritis encephalitis virus (CAEV) is a lentivirus that infects both goats and sheep and is closely related to maedi-visna virus that infects sheep; collectively, these viruses are known as small ruminant lentiviruses (SRLV). Infection of goats and sheep with SRLV typically results in discrete inflammatory diseases which include arthritis, mastitis, pneumonia or encephalomyelitis. SRLV-infected animals concurrently demonstrating lentivirus-associated lesions in tissues of lung, mammary gland, joint synovium and the central nervous system are either very rare or have not been reported.

Study of compartmentalization in the visna clinical form of small ruminant lentivirus infection in sheep.

BACKGROUND: A central nervous system (CNS) disease outbreak caused by small ruminant lentiviruses (SRLV) has triggered interest in Spain due to the rapid onset of clinical signs and relevant production losses. In a previous study on this outbreak, the role of LTR in tropism was unclear and env encoded sequences, likely involved in tropism, were not investigated. This study aimed to analyze heterogeneity of SRLV Env regions--TM amino terminal and SU V4, C4 and V5 segments--in order to assess virus compartmentalization in CNS. RESULTS: Eight Visna (neurologically) affected sheep of the outbreak
