Vitamins B(12), B(6), and folic acid for cognition in older men.

OBJECTIVE: To investigate whether supplementing older men with vitamins B(12), B(6), and folic acid improves cognitive function. METHODS: The investigators recruited 299 community-representative hypertensive men 75 years and older to a randomized, double-blind controlled clinical trial of folic acid, vitamin B(6), and B(12) supplementation vs placebo over 2 years. The primary outcome of interest was the change in the cognitive subscale of the Alzheimer's Disease Assessment Scale (ADAS-cog). A secondary aim of the study was to determine if supplementation with vitamins decreased the risk of co

Clinical use of cyclooxygenase inhibitors impairs vitamin B-6 metabolism.

BACKGROUND: A low circulating vitamin B-6 concentration, which is an independent risk factor for cardiovascular disease, is commonly seen in human inflammation. OBJECTIVE: We investigated whether cyclooxygenase inhibitors alter vitamin B-6 metabolism. DESIGN: To investigate whether subjects taking a cyclooxygenase inhibitor had an altered vitamin B-6 profile, we conducted a cross-sectional study that involved 150 rheumatoid arthritis patients, with and without cyclooxygenase inhibitor treatments. C57BL/6J mice and hyperlipidemic Syrian hamsters received drug regimens that reflected clinical

Vitamin B-6 malnutrition among elderly Europeans: the SENECA study.

Inadequate vitamin B-6 status is common among elderly people. It is still unclear to what extent factors other than reduced vitamin B-6 intake are responsible for this. We studied the vitamin B-6 intake and status [measured as plasma pyridoxal 5'-phosphate (PLP)] in 546 elderly Europeans, aged 74-76 years, with no known vitamin B-6 supplement use. In addition, we examined interrelations and associations with other dietary and lifestyle factors, including indicators of physical health. Overall, 27% of the males and 42% of the females had dietary vitamin B-6 intakes below the mean minimum requir

Humoral immunity in chicks experiencing marginal vitamin B-6 deficiency.

An economical vitamin B-6-deficient ration that was palatable to broiler chickens was prepared and fed to 1-day-old chicks. The experimental ration was a typical soy-glucose ration. Vitamin B-6 was removed by washing the soybean meal with water. Microbiological analysis revealed that the washed ration contained 0.45 mg vitamin B-6 per kilogram. Experimental rations were formulated to contain 0.5, 1.0 and 3.0 mg supplemental pyridoxine x HCl per kilogram of ration. These supplemental levels produced the following total pyridoxine concentrations in the diet: 0.95, 1.48 and 3.18 mg pyridoxine x H

Longitudinal association of vitamin B-6, folate, and vitamin B-12 with depressive symptoms among older adults over time.

BACKGROUND: B-vitamin deficiencies have been associated with depression; however, there is very little prospective evidence from population-based studies of older adults. OBJECTIVE: We examined whether dietary intakes of vitamins B-6, folate, or vitamin B-12 were predictive of depressive symptoms over an average of 7.2 y in a community-based population of older adults. DESIGN: The study sample consisted of 3503 adults from the Chicago Health and Aging project, an ongoing, population-based, biracial (59% African American) study in adults aged > or =65 y. Dietary assessment was made by food-fr

Influence of pyridoxine supplementation on vitamin B-6 levels in milk of rats deficient in the vitamin.

Levels of vitamin B-6 in milk from pyridoxine deficient dams were used as an indicator of the ability of pyridoxine to protect offspring against the effects of the deficiency. Sprague Dawley rats were fed a basal diet containing 30.0 (control) or 1.2 (deficient) mg pyridoxine-HC1/kg diet from weaning throughout growth, gestation and until 5 days postpartum. At this time, deficient dams were supplemented by a single intraperitoneal injection of 600 mug pyridoxine-HC1, or by adding 30 or 60 mg pyridoxine-HC1/kg to the diet. The vitamin B-6 content in milk form the group supplemented by injection

Plasma vitamin B(6) and risk of myocardial infarction in women.

BACKGROUND: We prospectively evaluated the relationships between fasting plasma levels of vitamin B(6), as pyridoxal phosphate, and subsequent myocardial infarction risk in women. METHODS AND RESULTS: Among 32 826 women who provided blood samples between 1989 and 1990 (27% of the original 1976 cohort), 239 were diagnosed with incident myocardial infarction (fatal and nonfatal) after blood collection but before July 1998. Of these women, 144 had provided a sample after fasting >10 hours. Cases were matched 1:2 by age, cigarette smoking status, and month of and fasting status at the time of blo

In vitro evidence for a relationship between magnesium and vitamin B-6.

Vitamin B-6 and magnesium appear to interact in vivo and it has been suggested that vitamin B-6 may enhance transport or accumulation of magnesium in cells. The purpose of this study was to evaluate the possibility of the formation of B-6 vitamer-Mg complexes in vitro using spectrophotometric analysis of ultraviolet or visible spectra or change in fluorescence intensity of B-6 vitamers. Pyridoxal phosphate but not pyridoxal appears to form a complex with Mg as evidenced by a change in the ultraviolet and visible spectra with an increase in absorption and spectral shift in the visible maxima fr

Evidence for increased catabolism of vitamin B-6 during systemic inflammation.

BACKGROUND: Plasma concentrations of PL 5'-phosphate (PLP), which is the active coenzyme form of vitamin B-6, are reduced during inflammation. The underlying mechanisms may include altered tissue distribution or increased catabolism via pyridoxal (PL) to pyridoxic acid (PA). Recently, we showed that catabolic enzyme activity could be assessed by substrate product ratios measured in plasma. OBJECTIVE: We evaluated the ratios PA:PL, PA:PLP, and PA:(PL + PLP) as possible markers of vitamin B-6 catabolism. DESIGN: Cross-sectional and longitudinal data were derived from the Western Norway B-Vitam

Connective tissue integrity is lost in vitamin B-6-deficient chicks.

The objective of the present investigation was to characterize further the connective tissue disorder produced by pyridoxine (vitamin B-6) deficiency, as previously evidenced by electron microscopy. Following the second post-natal week, fast growing male chicks were deprived of pyridoxine for a 1-mo period. Six weeks post-natally, blood concentrations in the experimental deficiency group had declined to deficiency levels as registered by low concentrations of pyridoxal phosphate (coenzyme form) in erythrocytes, but did not reach levels associated with neurological symptoms. Light microscopic s

Effect of physical activity on thiamine, riboflavin, and vitamin B-6 requirements.

Because exercise stresses metabolic pathways that depend on thiamine, riboflavin, and vitamin B-6, the requirements for these vitamins may be increased in athletes and active individuals. Theoretically, exercise could increase the need for these micronutrients in several ways: through decreased absorption of the nutrients; by increased turnover, metabolism, or loss of the nutrients; through biochemical adaptation as a result of training that increases nutrient needs; by an increase in mitochondrial enzymes that require the nutrients; or through an increased need for the nutrients for tissue ma

Contribution of vitamin B 6 deficiency to anemia in children on regular hemodialysis.

BACKGROUND: Anemia is prevalent among pediatric patients diagnosed with end-stage kidney disease (ESKD). In addition, erythropoiesis-stimulating agents (ESA) and iron supplementation are considered the cornerstones in the management of anemia. However, a significant proportion of patients remain anemic. Vitamin B6 plays a vital role in the biosynthesis of heme and may be deficient in children with chronic kidney disease (CKD), particularly those on regular hemodialysis (HD). We aimed to evaluate serum vitamin B6 concentration in those children and determine its correlation with anemia indices.

Vitamin B-6 and depressive symptomatology, over time, in older Latino adults.

Background: Low vitamin B-6 status has been linked to depressive symptomatology. We examined the longitudinal association of vitamin B-6 status with depressive symptomatology across 3-time points over ∼5-7 years in a cohort of older Hispanic adults. Methods: We used two-level hierarchical linear regression models for continuous outcomes. Vitamin B-6 status was associated with depressive symptomatology across these time points. Results: Plasma pyridoxyl-5-phosphate (PLP) concentration, a time-varying predictor, was significantly associated with depressive symptomatology. Study participants with

Functional and metabolic consequences of vitamin B-6 deficiency in the rat heart.

The cardiac functional and metabolic consequences of pyridoxine deficiency were studied in rats maintained on a pyridoxine-deficient diet for 10 weeks. Because food intake was diminished in the pyridoxine-deficient rats, a second group of animals was fed a diet restricted to the intake of the pyridoxine-deficient animals. The inotropic response (developed pressure) to an isoproterenol or Ca2+ concentration response curve was measured simultaneously with high energy phosphate levels using a modified Langendorf apparatus and 31P nuclear magnetic resonance spectroscopy. The inotropic response to

Genetic variants modify the associations of concentrations of methylmalonic acid, vitamin B-12, vitamin B-6, and folate with bone mineral density.

BACKGROUND: Elevated plasma homocysteine has been found to be associated with an increased risk of osteoporosis, especially hip and vertebral fractures. The plasma concentration of homocysteine is dependent on the activities of several B vitamin-dependent enzymes, such as methylenetetrahydrofolate reductase (MTHFR), methionine synthase (MTR), methionine synthase reductase (MTRR), and cystathionine β-synthase (CBS). OBJECTIVES: We investigated whether genetic variants in some of the genes involved in 1 carbon metabolism modify the association of B vitamin-related measures with bone mineral den
