Vitellogenin receptor mediates heat adaptability of oocyte development in mud crabs and zebrafish.

Climate-driven warming affects the reproduction of oviparous ectotherms. However, whether oviparous ectotherms possess a protection mechanism against heat stress for oocyte development, which is essential for maintaining the continuity of animal populations, is largely unknown. Under high temperatures, female mud crabs (Scylla paramamosain) typically have well-formed ovaries, while a few crabs were found to experience oocyte development failure. To investigate the heat stress protection mechanism of oocyte development in mud crabs, we construct a chromosome-level genome of this species and ide

Ectopic synthesis of vitellogenin in testis and epididymis of estrogen-treated lizard Podarcis sicula.

In oviparous vertebrates, vitellogenin (VTG) is the major yolk precursor synthesized in the liver of sexually mature females during the reproductive period. In males, the VTG gene is silent, but it may be activated by estradiol-17β (E2) or estrogen-like substances. Until now, extra-hepatic expression and synthesis of VTG after estrogen exposure has been reported only for aquatic vertebrates. This study demonstrates the ability of testis and epididymis of the terrestrial oviparous lacertid Podarcis sicula to synthesize VTG following E2 exposure. The results of in situ hybridization and immunohi

Exposure to Bisphenol AF disrupts sex hormone levels and vitellogenin expression in zebrafish.

Bisphenol AF (BPAF) is widely used in food-contact products, electronic devices, and as a cross-linking reagent in fluoroelastomers. There are growing concerns about its toxicity and endocrine-disrupting effects based on its structural similarity with bisphenol A (BPA). The endocrine-disrupting effects of BPAF were studied by exposing 2-month-old zebrafish to 0, 0.05, 0.25, or 1 mg/L BPAF for 28 days and evaluating the effect on growth, histopathology, hormone levels, enzyme activity, and gene expression. The overall fitness was not significantly affected. There were no apparent alterations in

Vitellogenin and yolk protein processing in Bothrops jararaca (Wied), a tropical venomous snake.

The plasma vitellogenin of Bothrops jararaca is composed of two subunits. The larger subunit (160 kDa) is phosphate rich and carbohydrate poor, while the smaller (110 kDa) is highly glycosylated and less phosphorylated. As in other vertebrates, the vitellogenin of B. jararaca is synthesized in the liver under estrogen control. The newly synthesized vitellogenin molecule is a 270 kDa polypeptide. This polypeptide originates the two subunits of the plasma vitellogenin by proteolytic cleavage. In the eggs of B. jararaca six main yolk polypeptides have been detected (113, 107, 104, 72, 27.2 and 20

Bioinformatic and functional characterization of the vitellogenin receptor in Dipetalogaster maxima, a vector of Chagas disease.

Reproduction in insects is a tightly regulated process that relies on the accumulation of yolk protein precursors (YPPs) to support embryonic development. Vitellogenin, the main YPP, is synthesized in the fat body-analogous to the liver and adipose tissue of vertebrates-and transported through the hemolymph to the ovarian follicles, where it is internalized by developing oocytes via the vitellogenin receptor (VgR). Despite its essential role in vitellogenesis, the functional characterization of insect VgR remains incomplete. Triatomines are hematophagous insects and vectors of Trypanosoma cruz

Chlorpyrifos causes histopathological changes and modulates vitellogenin levels in zebrafish (Danio rerio).

Chlorpyrifos is the most widely used insecticide worldwide for domestic and agricultural purposes. It is also an endocrine-disrupting chemical that affects non-target organisms even at low concentrations. The present study aimed to investigate sublethal concentration of chlorpyrifos in male zebrafish, focusing on its endocrine-disrupting effects on vitellogenin levels and tissue-level impact, as determined by histopathology. In our study, zebrafish were exposed to chlorpyrifos at concentrations of 10 and 100 µg/L for 24 h and 7 days, and compared to a control group. The results of vitellogenin

Vitellogenin in the honey bee brain: Atypical localization of a reproductive protein that promotes longevity.

In comparative gerontology, highly social insects such as honey bees (Apis mellifera) receive much attention due to very different and flexible aging patterns among closely related siblings. While experimental strategies that manipulate socio-environmental factors suggest a causative link between aging and social signals and behaviors, the molecular underpinnings of this linkage are less well understood. Here we study the atypical localization of the egg-yolk protein vitellogenin (Vg) in the brain of the honey bee. Vg is known to influence honey bee social regulation and aging rate. Our findin

Development of an enzyme-linked immunosorbent assay for vitellogenin of Morelet's crocodile (Crocodylus moreletii).

The purpose of this study was to develop an immunoassay for vitellogenin in Morelet's crocodile (Crocodylus moreletii). Blood was collected from wild-caught crocodiles in Belize. Plasma samples from adult females taken during the breeding season were used for vitellogenin purification and samples from adult males were used for comparison. No differences were detected between males and females for plasma total protein concentration, as measured by Coomassie assay. However, denaturing polyacrylamide gel electrophoresis (SDS-PAGE) revealed that female plasma contained a 210-kDa protein, presumabl

Characterization of the low-density lipoprotein receptors family and identification of vitellogenin receptors in Mugil cephalus.

Vitellogenin receptors (VtgRs) are pivotal to the successful reproductive event in oviparous species by mediating the uptake of vitellogenin (Vtg) into developing oocytes, ensuring proper yolk formation and embryonic development. In this study, we characterized the low-density lipoprotein receptor (LDLR) family in flathead mullet Mugil cephalus, to which VtgRs belong, and placed a focus on identifying and analyzing vitellogenin receptors. Using a comprehensive approach that combined LDLR orthology inference across 13 species, protein domain analysis and 3D structure prediction, synteny evaluat

Male vitellogenin regulates gametogenesis through a testis-enriched big protein in Chrysopa pallens.

In insects, vitellogenin (Vg) is generally viewed as a female-specific protein. Its primary function is to supply nutrition to developing embryos. Here, we reported Vg from the male adults of a natural predator, Chrysopa pallens. The male Vg was depleted by RNAi. Mating with Vg-deficient male downregulated female Vg expression, suppressed ovarian development and decreased reproductive output. Whole-organism transcriptome analysis after male Vg knockdown showed no differential expression of the known spermatogenesis-related regulators and seminal fluid protein genes, but a sharp downregulation

Development of specific chemiluminescent immunoassays for three subtypes of vitellogenin in grey mullet (Mugil cephalus).

Chemiluminescent immunoassays (CLIAs) were developed for each of three subtypes of vitellogenin (VtgAa, VtgAb and VtgC) in grey mullet, primarily for use in monitoring estrogenic pollution of the environment. The working range of VtgAa-CLIA and VtgAb-CLIA was from 0.975 to 1,000 ng/ml, while that of VtgC-CLIA was from 0.487 to 1,000 ng/ml. Each CLIA appeared to be specific to the targeted Vtg subtype. Intra- and inter-assay coefficients of variation in the developed CLIAs were lower than 10%. In male serum, VtgAa, VtgAb and VtgC were detected in ranges from 0.01 to 0.38, 0.02 to 1.01, and 0.01

Purification, partial characterization and peptide sequences of vitellogenin from a reptile, the tuatara (Sphenodon punctatus).

Vitellogenin (Vg), a major precursor to egg yolk proteins, was purified from plasma of an estradiol-treated female tuatara (Sphenodon punctatus) by MgCl2-EDTA precipitation and DEAE-cellulose chromatography. The amino acid composition of tuatara Vg is similar to that of other vertebtate Vgs and contains a large proportion of serine (13.7 mol/100 mol of total amino acid). The amino acid sequences of the N-terminus of mature Vg (33 residues) and of several trypsin- and CNBr-generated peptides were determined. Six peptide sequences obtained from tuatara Vg could be aligned with Vg sequences from

Culex tarsalis vitellogenin gene promoters investigated in silico and in vivo using transgenic Drosophila melanogaster.

INTRODUCTION: Genetic modification, or transgenesis, is a powerful technique to investigate the molecular interactions between vector-borne pathogens and their arthropod hosts, as well as a potential novel approach for vector-borne disease control. Transgenesis requires the use of specific regulatory regions, or promoters, to drive expression of genes of interest in desired target tissues. In mosquitoes, the vast majority of described promoters are from Anopheles and Aedes mosquitoes. RESULTS: Culex tarsalis is one of the most important vectors of arboviruses (including West Nile virus) in No

Effects of illegal cyanide fishing on vitellogenin in the freshwater African catfish, Clarias gariepinus (Burchell, 1822).

The effects of cyanide, used in illegal fishing, on one of the most economically important Nile fishes, the African catfish (Clarias gariepinus), were studied. Cyanide impacts were evaluated in terms of biochemical, molecular and histopathological characteristics. After exposure to sublethal concentration (0.05mg/l) of potassium cyanide (KCN) for two and four weeks, GOT (glutamate oxaloacetate transaminase) was significantly increased in both male and female, while GPT (glutamate pyruvate transaminase), total plasma protein, phosphoprotein phosphorus (Vgt) in serum, vitellogenin gene expressio

The major yolk protein vitellogenin interferes with the anti-plasmodium response in the malaria mosquito Anopheles gambiae.

When taking a blood meal on a person infected with malaria, female Anopheles gambiae mosquitoes, the major vector of human malaria, acquire nutrients that will activate egg development (oogenesis) in their ovaries. Simultaneously, they infect themselves with the malaria parasite. On traversing the mosquito midgut epithelium, invading Plasmodium ookinetes are met with a potent innate immune response predominantly controlled by mosquito blood cells. Whether the concomitant processes of mosquito reproduction and immunity affect each other remains controversial. Here, we show that proteins that de

Tick vitellogenin receptor reveals critical role in oocyte development and transovarial transmission of Babesia parasite.

A cDNA encoding the vitellogenin receptor of the ixodid tick, Haemaphysalis longicornis Neumann (HlVgR) was cloned and characterized. The full-length cDNA is 5631 bp, including an intact ORF encoding an expected protein with 1782 amino acids. The deduced amino acid sequence of the HlVgR cDNA revealed two ligand-binding domains with four class A cysteine-rich repeats in the first domain and eight in the second domain similar to those of insect VgRs. The immunoblot analysis detected approximately 197 kDa protein in both tick ovary and egg. The developmental expression profile demonstrated that H

Reduced Vitellogenin Expression is Associated with Impaired Oogenesis in Female Triploid Pacific Oysters (Crassostrea gigas).

Triploidy is widely applied in oyster aquaculture because of its association with reduced gametogenesis, yet female triploid Pacific oyster (Crassostrea gigas) exhibit pronounced variability in ovarian development. The mechanisms underlying ovarian impairment in triploid females remain poorly understood. In this study, we investigated the role of vitellogenin (VTG) in oogenesis and gonadal development by comparing diploid females with β triploid females exhibiting severe gonadal arrest. Quantitative PCR and in situ hybridization analyses revealed that the expression of VTG was significantly re

Vitellogenin and its receptor characterize exogenous vitellogenesis mode in the Pacific white shrimp Litopenaeus vannamei.

Vitellogenesis is a pivotal process of animal reproduction, characterized by yolk protein formation and accumulation in developing oocytes. Vitellogenin (Vg), the precursor of vitellin (Vn), serves as the primary yolk component. While animals employ both exogenous and endogenous vitellogenesis, the precise synthetic locus of Vg in the Pacific white shrimp Litopenaeus vannamei, the globally most important cultured shrimp, remains unclear. To characterize vitellogenesis and its regulatory mode, the present study examined ovarian morphology, gonadosomatic index (GSI) dynamics, hepatopancreatic an
