Characterization of pain and pharmacologic responses in an animal model of lumbar adhesive arachnoiditis.

STUDY DESIGN: The study used a small animal laminectomy model with kaolin to investigate the relationship of cauda equina pathology to pain, and the effects of systemic and intrathecal drugs to reduce pain. OBJECTIVE: To determine if higher amounts of cauda equina adhesions produce increased pain-related behavior following laminectomy and epidural kaolin application. SUMMARY OF BACKGROUND DATA: Clinically, the relationship of cauda equina pathology to pain is controversial. An established animal model produces cauda equina adhesions. The correlation of this anatomic pathology with pain-relat

Localization and visual verification of a complex minke whale vocalization.

A recently described population of minke whales (Balaenoptera acutorostrata) offered a unique opportunity to study its acoustic behavior. The often-inquisitive dwarf minke whale is seen on the Great Barrier Reef nearly coincident with its suspected calving and breeding seasons. During drifting encounters with whales, a towed hydrophone array was used to record sounds for subsequent localization of sound sources. Shipboard and in-water observers linked these sounds to the closely circling minke whale. A complex and stereotyped sound sequence, the "star-wars" (SW) vocalization, was recorded duri

Ultrasonic vocalization changes and FOXP2 expression after experimental stroke.

Speech impairments affect one in four stroke survivors. However, animal models of post-ischemic vocalization deficits are limited. Male mice vocalize at ultrasonic frequencies when exposed to an estrous female mouse. In this study we assessed vocalization patterns and quantity in male mice after cerebral ischemia. FOXP2, a gene associated with verbal dyspraxia in humans, with known roles in neurogenesis and synaptic plasticity, was also examined after injury. Using a transient middle cerebral artery occlusion (MCAO) model, we assessed correlates of vocal impairment at several time-points after

Social isolation alters ultrasonic vocalizations but not thyroarytenoid neuromuscular junctions in old rats.

OBJECTIVE: Age-related muscle atrophy of the laryngeal muscles contributes to presbyphonia. Remodeling of the neuromuscular junction is one aspect underlying age-related muscle atrophy. Although muscle disuse has been shown to exacerbate age-related neuromuscular changes in the limb muscles, it is unknown if reduced vocal use has a similar effect in the laryngeal muscles. The objective of this study was to examine the use of social isolation as a novel method to reduce vocal use in old rats-and the impact of that reduced vocal use on ultrasonic vocalization acoustics and neuromuscular junction

Deficits in ultrasonic vocalization development and production following neonatal hypoxic ischemic insult.

Neonatal hypoxic ischemia encephalopathy (HIE) leads to major deficits in language development. While clinically there is a known correlation in the degree of HIE injury and subsequent language disability, there are no treatments beyond speech and language therapy; therefore, experimental studies with a HIE animal model to test new interventions and therapeutics are warranted. Neonatal rodents normally ultrasonically vocalize at postnatal day 7 (PND 7) to PND 14 in response to removal from their mothers. At 6-8 weeks of age juvenile male rodents ultrasonically vocalize in response to exposure

VoICE: A semi-automated pipeline for standardizing vocal analysis across models.

The study of vocal communication in animal models provides key insight to the neurogenetic basis for speech and communication disorders. Current methods for vocal analysis suffer from a lack of standardization, creating ambiguity in cross-laboratory and cross-species comparisons. Here, we present VoICE (Vocal Inventory Clustering Engine), an approach to grouping vocal elements by creating a high dimensionality dataset through scoring spectral similarity between all vocalizations within a recording session. This dataset is then subjected to hierarchical clustering, generating a dendrogram that

Influence of sedation on onset and quality of euthanasia in sheep.

The purpose of this study was to determine if dexmedetomidine administered IV prior to euthanasia in sheep affected the speed or quality of euthanasia. Twenty clinically healthy Dorset-cross adult ewes between 1 and 3years of age were enrolled in a randomized blinded experimental trial. The subjects were randomly assigned to receive dexmedetomidine 5μg/kg IV or an equivalent volume of saline. Five minutes later, euthanasia was accomplished with a pentobarbital/phenytoin overdose given IV. The time to apnea, asystole, cessation of audible heartbeat, and absence of corneal reflex were recorded b

Characterization of Efficacy and Animal Safety across Four Caprine Disbudding Methodologies.

There is a strong industry demand for technically simple and highly efficacious alternatives to heat cautery disbudding in goat kids that can be performed as a stand-alone procedure without adjunct anesthesia, and that result in improved overall welfare through reduced acute pain, reduced tissues healing interval, and a consistent safety record. The objective of this study was to consider the net effect of disbudding techniques on goat welfare by examining vocalization frequency, long-term efficacy and animal safety associated with four alternative caprine disbudding methods against sham-disbu

From songbird to humans: The multifaceted roles of FOXP2 in speech and motor learning.

Motor learning involves a complex network of brain structures and is crucial for tasks like speech. The cerebral cortex, subcortical nuclei, and cerebellum are involved in motor learning and vocalization. Vocal learning has been demonstrated across species. However, it is a task that should be further studied and reevaluated, particularly in species considered non-vocal learners, to potentially uncover new insights. FOXP2, a transcription factor, has been implicated in speech learning and execution. Several variants have been involved in speech and cognitive impairments; the most studied is th

Effect of 5-HT5A antagonists in animal models of schizophrenia, anxiety and depression.

The few available data on the pharmacological effect of 5-HT5A receptors suggest that antagonists may have anxiolytic, antidepressant and antipsychotic activity. The aim of our study was to verify these suggestions in relevant animal models. Two 5-HT5A antagonist ligands, SB-699551-A (N-[2-(dimethylamino)ethyl]-N-[[4'-[[(2-phenylethyl)amino]methyl][1,1'-biphenyl]-4-yl]methyl]cyclopentanepropanamide dihydrochloride) (3-60 mg/kg, intraperitoneally) and A-843277 (N-(2,6-dimethoxybenzyl)-N'[4-(4-fluorophenyl)thiazol-2-yl]guanidine) (3-30 mg/kg, intraperitoneally), were examined in the open-field t

Speech-induced suppression and vocal feedback sensitivity in human cortex.

Across the animal kingdom, neural responses in the auditory cortex are suppressed during vocalization, and humans are no exception. A common hypothesis is that suppression increases sensitivity to auditory feedback, enabling the detection of vocalization errors. This hypothesis has been previously confirmed in non-human primates, however a direct link between auditory suppression and sensitivity in human speech monitoring remains elusive. To address this issue, we obtained intracranial electroencephalography (iEEG) recordings from 35 neurosurgical participants during speech production. We firs

Neuroinflammatory Mechanisms and Vocalization Biomarkers in the Pink1-/- Preclinical Model of Parkinson's Disease.

Parkinson's disease is a progressive neurodegenerative disorder with an unclear etiology and a long preclinical phase. The multitude of challenges of studying this in humans highlight the need for robust preclinical animal models that accurately reflect the disease's preclinical progression. While traditional neurotoxin models fail to capture the slow, genetic nature of PD, genetic models, such as the phosphatase and tensin homolog-induced putative kinase 1 (Pink1-/-) knockout rat, offer a preclinical, translational approach. This review synthesizes the current understanding of the Pink1-/- ra

How small could a pup sound? The physical bases of signaling body size in harbor seals.

Vocal communication is a crucial aspect of animal behavior. The mechanism which most mammals use to vocalize relies on three anatomical components. First, air overpressure is generated inside the lower vocal tract. Second, as the airstream goes through the glottis, sound is produced via vocal fold vibration. Third, this sound is further filtered by the geometry and length of the upper vocal tract. Evidence from mammalian anatomy and bioacoustics suggests that some of these three components may covary with an animal's body size. The framework provided by acoustic allometry suggests that, becaus

Chronic recording of cortical activity underlying vocalization in awake minipigs.

BACKGROUND: Investigating brain dynamics underlying vocal production in animals is a powerful way to inform on the neural bases of human speech. In particular, brain networks underlying vocal production in non-human primates show striking similarities with the human speech production network. However, despite increasing findings also in birds and more recently in rodents, the extent to which the primate vocal cortical network model generalizes to other non-primate mammals remains unclear. Especially, no domestic species has yet been proposed to investigate vocal brain activity using electrophy

Valproic acid exposure impairs ultrasonic communication in infant, adolescent and adult rats.

Persistent deficits of social communication are a hallmark of autism spectrum disorders (ASD). Communication disabilities can be experimentally modeled using rodents' ultrasonic vocalizations (USVs). Although prenatal exposure to valproic acid (VPA) is one of the most widely used animal models of ASD, little is known about communication impairments in this model. We performed a longitudinal study to characterize VPA-induced socio-communicative deficits in male and female rats. USVs were recorded in neonatal rats during maternal separation, in adolescent rats during social play, and in adult ra

High channel count microphone array accurately and precisely localizes ultrasonic signals from freely-moving mice.

BACKGROUND: An integral component in the assessment of vocal behavior in groups of freely interacting animals is the ability to determine which animal is producing each vocal signal. This process is facilitated by using microphone arrays with multiple channels. NEW METHOD AND COMPARISON WITH EXISTING METHODS: Here, we made important refinements to a state-of-the-art microphone array based system used to localize vocal signals produced by freely interacting laboratory mice. Key changes to the system included increasing the number of microphones as well as refining the methodology for localizin

Increase in sexual motivation throughout adolescence in the cycling female rat.

Sexual behavior in the female rat is a highly motivated behavior first displayed during adolescence, a developmental period when neural circuits underlying motivation are not mature. This study characterizes the natural development of sexual motivation and behavior of female rats. We compared the incentive value of the male for mid-adolescent (PNDs:39-43), late adolescent (PNDs:49-53), and adult (PNDs:90-115) cycling females, using a male-female preference task and an ultrasonic vocalization emission test following exposure to a male or female stimulus animal. Furthermore, display of sexual an

Vocalization of echolocation-like pulses for interindividual interaction in horseshoe bats (Rhinolophus ferrumequinum).

Although much is known about the echolocation of horseshoe bats (Rhinolophus spp.), little is known about the characteristics and function of their communication calls. This study focused on a stereotyped behavior of a bat approaching a companion animal in the colony, and examined their interaction and vocalization during this behavior. The bats emit echolocation-like vocalizations when approaching each other and these vocalizations contain a "buildup" pulse sequence, in which the frequency of the pulse increases gradually to normal echolocation pulse frequencies. The results suggest that the

The Effect of Fibroblast Growth Factors in Grafted Fascia into the Vocal Fold of Rabbits.

Introduction  The human larynx is a very important organ for communication. Many conditions lead to scarring of the vocal folds, decreasing voice quality. Objective  We aimed to determine whether fibroblast growth factors (FGFs) may influence tissue integration of grafted fascia into the vocal folds of an animal model. Methods  This is an experimental animal study with 12 adult rabbits that were submitted to a grafting fragment obtained from superficial cervical fascia into the vocal fold lamina propria, bilaterally. The right vocal fold was injected with FGFs. The animals were sacrificed afte
