Multiple organ injury in septic rats under weightlessness.

INTRODUCTION: As human space exploration advances, the risk of infection faced by astronauts has also become apparent. However, the impact of sepsis on the body under weightlessness remains unclear. METHODS: A rat model of sepsis under simulated weightlessness was preliminarily established by combining hindlimb unloading with cecal ligation and puncture. The effects of simulated weightlessness on the early progression of sepsis in rats were systematically evaluated through assessments of mortality, hematological parameters, inflammatory markers, coagulation function, organ function, and histo

Human autonomic responses to actual and simulated weightlessness.

Orthostatic dysfunction occurs after exposure to microgravity, and is not completely understood. The authors developed a device for stimulating carotid baroreceptors to test the hypothesis that exposure to microgravity leads to impairment of arterial baroreflex mechanisms. Data obtained before and after two head-down bedrest studies and before and after brief Space Shuttle missions indicate that baroreceptor-cardiac reflex control is impaired by simulated or actual weightlessness. The authors speculate that arterial baroreflex derangements combine with blood volume reductions and increased ven

Acute hemodynamic responses to weightlessness during parabolic flight.

Pilots and astronauts experience fluid shifts in variable gravity. Acute effects of fluid shifts on the cardiovascular system were monitored on NASA's KC-135 aircraft during parabolic flight. The variability of R-R intervals in the electrocardiogram was measured as an indication of vagal cardiac neural activity. R-R intervals were measured during the gravity transition from 2-G to 0-G produced by parabolic flight to assess the involvement of the autonomic nervous system in regulating the acute effects of fluid shifts. In seven subjects, a BoMed noninvasive continuous cardiac output monitor (NC

Skeletal abnormalities in rats induced by simulated weightlessness.

A hypokinetic model has been developed which attempts to simulate the weightlessness experienced during space flight. Male rats (approximately 130 g) were suspended from the model with a head-down tilt for a two-week period. Total mechanical unloading of the hind limbs and partial unloading of the fore limbs occurred. In comparison to pair-fed control rats, the skeletal alterations in the proximal tibial and humeral metaphyses of suspended rats were determined to be a diminished rate of longitudinal bone growth, a reduced mass of mineralized tissue, and an accumulation of marrow fat. Also, sus

Cardiovascular studies during and following simulation and weightlessness.

Two major characteristics of space flight may be simulated to some degree by bed rest and by water immersion. These are inactivity and the lack of the hydrostatic stress on the systematic circulation. Studies prior to the advent of space flight led to the prediction of orthostatic intolerance, muscular weakness, and calcium loss. Prolonged space flight has heightened the interest in these simulations to provide information concerning the temporal cause of the "deconditioning" or disuse syndrome and the value of simulation for testing the effectiveness of remedial measures. Musculoskeletal adaptations to weightlessness and development of effective countermeasures.

A Research Roundtable, organized by the American College of Sports Medicine with sponsorship from the National Aeronautics and Space Administration, met in November 1995 to define research strategies for effective exercise countermeasures to weightlessness. Exercise was considered both independently of, and in conjunction with, other therapeutic modalities (e.g., pharmacological nutritional, hormonal, and growth-related factors) that could prevent or minimize the structural and functional deficits involving skeletal muscle and bone in response to chronic exposure to weightlessness, as well as

[Ultrastructure of the submandibular glands in rats kept in weightlessness].

Submandibular salivary glands of male rats weighing 330-350 g were examined after space flight and ground-based control study. Light microscopy was carried out using hematoxylin-eosin staining and PAS-reaction. Electron microscopy was performed using glutaraldehyde and osmium tetroxide fixation and contrasting according to Reynolds. Light microscopy revealed no destructive changes in the gland parenchyma; differences between flight and control rats remained within physiological limits. Electron microscopy of acinar cells of flight animals showed chromatin condensation, darkening of cells and n

General remarks on the role of the vestibular system in weightlessness.

Different methods are described to experimentally achieve weightlessness. Since the function of the otolith system depends on the presence of contact forces opposing gravity, it is disabled in weightlessness and may send misleading positional information to the brain. Without the contributions of the otolith system it is difficult in space to distinguish self-motion from object motion. Furthermore, the disintegration of information from the neck position receptors from those of the otolith system can lead to additional illusory positional sensations. Since the function of the semicircular cana

Spatial orientation in weightlessness and readaptation to earth's gravity.

Unusual vestibular responses to head movements in weightlessness may produce spatial orientation illusions and symptoms of space motion sickness. An integrated set of experiments was performed during Spacelab 1, as well as before and after the flight, to evaluate responses mediated by the otolith organs and semicircular canals. A variety of measurements were used, including eye movements, postural control, perception of orientation, and susceptibility to space sickness.

Formaldehyde initiates memory and motor impairments under weightlessness condition.

During space flight, prolonged weightlessness stress exerts a range of detrimental impacts on the physiology and psychology of astronauts. These manifestations encompass depressive symptoms, anxiety, and impairments in both short-term memory and motor functions, albeit the precise underlying mechanisms remain elusive. Recent studies have revealed that hindlimb unloading (HU) animal models, which simulate space weightlessness, exhibited a disorder in memory and motor function associated with endogenous formaldehyde (FA) accumulation in the hippocampus and cerebellum, disruption of brain extrace

Context dependence of manual grasping movements in near weightlessness.

BACKGROUND: We have shown before that human subject grasping performance differs in an everyday-like context with that observed in a laboratory context. The purpose of the present study was to determine whether reported deficits in weightlessness are more pronounced when grasping is performed as part of everyday-like behavior rather than as an isolated laboratory-type response. METHODS: The grasping performance of 12 participants (ages 29 +/- 5 yr) during periods of near weightlessness in parabolic flights was compared. Subjects performed a typical laboratory task (L) where grasping was repet

Space Sled--a device for the investigation of the physiological effects of weightlessness.

Space Sled is a device for providing controlled linear acceleration stimuli in the microgravity environment of orbital flight. The scientific objectives of the experiments which used Space Sled on the D-1 Spacelab mission were to study aspects of otolith organ (that is, that part of the inner ear which transduces linear accelerations) function and adaptation in weightlessness. Space Sled comprises electrical and mechanical sub-systems. The latter is made up of a carriage running on twin rails that are fixed to the floor of Spacelab. The assembly is 6 m long with a working section of 3.5 m. The

[Labyrinthine and extralabyrinthine mechanisms of the development of motion sickness in weightlessness].

This paper describes an important problem of aerospace medicine, i. e. space motion sickness. The review surveys relevant Soviet and foreign publications. On their basis, including their own data, the authors discuss possible mechanisms of the sickness symptom-complex in the weightless state. The authors put forth their own ideas concerning current hypotheses of space motion sickness and discuss potential applicability of the theoretical concepts with respect to the prevention of this adverse symptom-complex. The authors describe selected outlines in the study of mechanisms of space motion sic

Cardiovascular deconditioning effects of long-term simulated weightlessness in rats.

The aim of our serial work was to investigate the cardiovascular deconditioning effect of long-term simulated weightlessness and to elucidate its mechanisms. Our research goal was established in view of the following three reasons. Firstly, even after several decades of extensive research, there still exist significant gaps in our knowledge concerning microgravity induced cardiovascular effect. Secondly, to guarantee the health and safety of astronauts in the future prolonged missions, it is important to understand the cardiovascular adaptation to long-term weightlessness. Thirdly, the reporte

Central venous pressure in humans during short periods of weightlessness.

Central venous pressure (CVP) was measured in 14 males during 23.3 +/- 0.6 s (mean +/- SE) of weightlessness (0.00 +/- 0.05 G) achieved in a Gulfstream-3 jet aircraft performing parabolic flight maneuvers and during either 60 or 120 s of +2 Gz (2.0 +/- 0.1 Gz). CVP was obtained using central venous catheters and strain-gauge pressure transducers. Heart rate (HR) was measured simultaneously in seven of the subjects. Measurements were compared with values obtained inflight at 1 G with the subjects in the supine (+1 Gx) and upright sitting (+1 Gz) positions, respectively. CVP was 2.6 +/- 1.5 mmHg

Review of the biological effects of weightlessness on the human endocrine system.

Studies from space flights over the past two decades have demonstrated that there are basic physiological changes in humans during space flight. These changes include cephalad fluid shifts, loss of fluid and electrolytes, loss of muscle mass, space motion sickness, anemia, reduced immune response, and loss of calcium and mineralized bone. The cause of most of these manifestations is not known but the general approach has been to investigate systemic and hormonal changes. However, data from the 1973-1974 Skylabs, Spacelab 3 (SL-3), Spacelab D-I (SL-DI), and now the new SLS-1 missions support a

Skeletal response to simulated weightlessness: a comparison of suspension techniques.

The skeletal response to simulated weightlessness was studied in rats subjected to two different methods of suspension. Skeletal unloading of the hind limbs for a two week period was achieved by use of either a back harness or tail traction. In comparison to pair-fed control rats, back-suspended rats failed to gain weight whereas tail-suspended rats exhibited normal weight gain. Quantitative bone histomorphometry revealed marked skeletal abnormalities in the proximal tibial metaphysis of back-suspended rats. Loss of trabecular bone mass in these animals was due to a combination of depressed lo
