Association study of Wnt signaling pathway genes in bipolar disorder.

CONTEXT: The Wnt signaling pathways promote cell growth and are best known for their role in embryogenesis and cancer. Several lines of evidence suggest that these pathways might also be involved in bipolar disorder. OBJECTIVE: To test for an association between candidate genes in the Wnt signaling pathways and disease susceptibility in a family-based bipolar disorder study. DESIGN: Two hundred twenty-seven tagging single- nucleotide polymorphisms (SNPs) from 34 genes were successfully genotyped. Initial results led us to focus on the gene PPARD, in which we genotyped an additional 13 SNPs f

Epigenetics and Expression of the Wnt Signaling Pathway in Ulcerative Colitis.

BACKGROUND/AIMS: Secreted frizzled-related proteins (SFRPs) are antagonists that bind Wnt and inhibit signaling through this pathway. Secreted frizzled-related proteins are silenced by promoter methylation and cause hyperactivation of the Wnt pathway. In this study, the aim was to evaluate the relationship between methylation and expression of genes involved in the Wnt signaling pathway and the risk of cancer development in inflammatory bowel disease. MATERIALS AND METHODS: The patient group consisted of 20 individuals who were diagnosed with left-side ulcerative colitis and underwent surveil

Hedgehog and Wnt Signaling Pathways Regulate Tail Regeneration.

Urodele amphibians have a tremendous capacity for the regeneration of appendages, including limb and tail, following injury. While studies have focused on the cellular and morphological changes during appendicular regeneration, the signaling mechanisms that govern these cytoarchitectural changes during the regenerative response are unclear. In this study, we describe the essential role of hedgehog (Hh) and Wnt signaling pathways following tail amputation in the newt. Quantitative PCR studies revealed that members of both the Hh and Wnt signaling pathways, including the following: shh, ihh, ptc

Wnt pathway in Dupuytren disease: connecting profibrotic signals.

A role of Wnt signaling in Dupuytren disease, a fibroproliferative disease of the hand and fingers, has not been fully elucidated. We examined a large set of Wnt pathway components and signaling targets and found significant dysregulation of 41 Wnt-related genes in tissue from the Dupuytren nodules compared with patient-matched control tissue. A large proportion of genes coding for Wnt proteins themselves was downregulated. However, both canonical Wnt targets and components of the noncanonical signaling pathway were upregulated. Immunohistochemical analysis revealed that protein expression of

The canonical Wnt/β-catenin signaling pathway regulates Fgf signaling for early facial development.

The canonical Wnt/β-catenin signaling pathway has implications in early facial development; yet, its function and signaling mechanism remain poorly understood. We report here that the frontonasal and upper jaw primordia cannot be formed after conditional ablation of β-catenin with Foxg1-Cre mice in the facial ectoderm and the adjacent telencephalic neuroepithelium. Gene expression of several cell-survival and patterning factors, including Fgf8, Fgf3, and Fgf17, is dramatically diminished in the anterior neural ridge (ANR, a rostral signaling center) and/or the adjacent frontonasal ectoderm of

Klotho Attenuates Glucocorticoid-Induced Osteoblast Cytotoxicity Via Wnt Signaling Pathway Modulation.

Glucocorticoids are commonly prescribed in clinical settings; however, their prolonged use at high doses can adversely affect human health. One significant complication following glucocorticoid therapy is glucocorticoid-induced osteoporosis (GIO), which is second in incidence only to senile osteoporosis. Based on previous research indicating that Klotho alleviates dexamethasone-induced osteoblast cytotoxicity through the NF-kB pathway, we aimed to explore the underlying mechanisms in greater depth. We assessed the impact of Lithium chloride (LiCl), a Wnt pathway activator, on glucocorticoid-in

Autophagy controls neuronal differentiation by regulating the WNT-DVL signaling pathway.

Macroautophagy/autophagy dysregulation is associated with various neurological diseases, including Vici syndrome. We aimed to determine the role of autophagy in early brain development. We generated neurons from human embryonic stem cells and developed a Vici syndrome model by introducing a loss-of-function mutation in the EPG5 gene. Autophagy-related genes were upregulated at the neuronal progenitor cell stage. Inhibition of autolysosome formation with bafilomycin A1 treatment at the neuronal progenitor cell stage delayed neuronal differentiation. Notably, WNT (Wnt family member) signaling ma

Role of Wnt signaling pathway in joint development and cartilage degeneration.

Osteoarthritis (OA) is a prevalent musculoskeletal disease that affects approximately 500 million people worldwide. Unfortunately, there is currently no effective treatment available to stop or delay the degenerative progression of joint disease. Wnt signaling pathways play fundamental roles in the regulation of growth, development, and homeostasis of articular cartilage. This review aims to summarize the role of Wnt pathways in joint development during embryonic stages and in cartilage maintenance throughout adult life. Specifically, we focus on aberrant mechanical loading and inflammation as

Potential Roles of the WNT Signaling Pathway in Amyotrophic Lateral Sclerosis.

The WNT signaling pathway plays an important role in the physiological and pathophysiological processes of the central nervous system and the neurodegenerative disease amyotrophic lateral sclerosis (ALS). We reviewed the literature pertinent to WNT/β-catenin signaling in ALS from cellular studies, animal models, and human clinical trials. WNT, WNT receptors, and other components of the WNT signaling pathway are expressed in both ALS patients and transgenic mice, and are involved in the pathogenesis of ALS. Studies have shown that abnormal activation of the WNT/β-catenin signaling pathway is re

PiRNA-63049 inhibits bone formation through Wnt/β-catenin signaling pathway.

Bone remodeling is a dynamic process between bone formation mediated by osteoblasts and bone resorption mediated by osteoclasts. Disrupted bone remodeling is a key factor in postmenopausal osteoporosis, a metabolic disorder characterized by deteriorated bone microarchitecture and increased risk of fracture. Recent studies have shown that piwi-binding RNA (piRNA) is involved in the pathogenesis of certain diseases at the post-transcriptional level. Here, we analyzed piRNA-63049 (piR-63049), which may play an essential role in bone remodeling. The expression of piR-63049 significantly increased

Experimental agents to improve fracture healing: utilizing the WNT signaling pathway.

The process of bone healing largely recapitulates bone development in the embryo and ideally achieves complete restoration of bone shape and structure. However, because successful fracture healing requires tight interactions of numerous cell types and signaling molecules, any disruption of this highly coordinated processes can result in delayed healing or even non-union formation. The rate of fracture healing complications in orthopedic patients is reported to be 5-20%. Therefore, there is a need for new therapeutic strategies to improve fracture healing in patients with healing complications.

How the Wnt signaling pathway protects from neurodegeneration: the mitochondrial scenario.

Alzheimer's disease (AD) is the most common neurodegenerative disorder and is characterized by progressive memory loss and cognitive decline. One of the hallmarks of AD is the overproduction of amyloid-beta aggregates that range from the toxic soluble oligomer (Aβo) form to extracellular accumulations in the brain. Growing evidence indicates that mitochondrial dysfunction is a common feature of neurodegenerative diseases and is observed at an early stage in the pathogenesis of AD. Reports indicate that mitochondrial structure and function are affected by Aβo and can trigger neuronal cell death

Wnt signaling pathway: Therapeutic target of natural products for cancer therapy.

The Wnt signaling pathway is an evolutionarily conserved system crucial for embryonic development, tissue homeostasis and various diseases. Wnt signaling is generally categorized into two main branches: the canonical (β-catenin-dependent) pathway and the non-canonical (β-catenin-independent) pathway, including the Wnt/planar cell polarity (PCP) and Wnt/Ca2+ pathways. The dysregulated Wnt signaling pathway is highly associated with the pathogenesis of multiple cancers, such as proliferation, apoptosis, autophagy, metastasis, immune evasion, drug resistance and maintenance of cancer stem cells.

MBP1 inhibits AML proliferation through downregulating noncanonical Wnt/Ca2+ signaling pathway.

Acute myeloid leukemia (AML) remains therapeutically challenging, highlighting an urgent need for novel therapeutic targets. Our prior work showed ENO1 promotes AML, but the role of its short variant MBP1 was unknown. Here, we demonstrated significant downregulation of MBP1 alongside ENO1 upregulation in primary AML patient samples compared to healthy donors, establishing an imbalanced ENO1/MBP1 ratio. Functionally, restoring MBP1 expression in AML cell lines (KG1, OCI-AML3) inhibited cell proliferation, suppressed colony formation, induced cell apoptosis, and triggered G1-phase cell cycle arr

Expression Levels of WNT Signaling Pathway Genes During Early Tooth Development.

It is known to all that Wnt signaling pathway plays an important role in the early development of tooth. Our previous research found that Wnt signaling pathway played crucial roles in dental development, and mutations in antagonist of Wnt signaling pathway may lead to the formation of supernumerary teeth. However, the expression pattern of Wnt signaling molecules in early development of tooth, especially genes with stage specificity, remains unclear. Hence, we applied RNA-seq analysis to determine the expression levels of wnt signal molecules at five different stages of rat first molar tooth g
