Development of a Transcriptional Biosensor for Hydrogen Sulfide that Functions under Aerobic and Anaerobic Conditions.

Hydrogen sulfide (H2S) is a gaseous gut metabolite with disputed effects on gastrointestinal health. Monitoring H2S concentration in the gut would provide insight into its role in disease, but is complicated by sulfide's reactivity and volatility. Here we develop a transcriptional sulfide biosensor in E. coli. The sensor relies on enzymatic oxidation of sulfide catalyzed by a sulfide:quinone reductase (Sqr) to polysulfides, which bind to the repressor SqrR, triggering unbinding from the promoter and transcription of the reporter. Through promoter engineering and improving soluble SqrR expressi

Gut microbial differences in healthy and sick farmed American alligators (Alligator mississippiensis) during a disease outbreak.

American alligators (Alligator mississippiensis) in production systems within the United States experience occasional disease outbreaks and die-offs. However, outbreak management is limited by an incomplete understanding of common causes of mortality in commercially raised crocodilians. In this study, we examined the gut microbiota of healthy (n = 18) and sick alligators (n = 17) during an active outbreak on an alligator farm in Southeast, USA. Sick alligators were all exhibiting generalized signs of disease. The gut microbial composition of alligators with disease differed significantly from

Microbiota associated with residual clefts and neighboring teeth in patients with cleft lip, alveolus, and palate.

Twenty patients with residual clefts or pronounced soft tissue grooves, treated for uni- or bilateral cleft lip, alveolus, and palate were included in this study. Ten patients were recalled for dental prophylaxis at regular intervals, 10 patients were not. One microbiologic sample was obtained from the cleft area and two samples from a tooth adjacent to the cleft (sites adjacent and distant to the cleft). Between the recall and the nonrecall group there were notable differences in the presence of anaerobic Gram-negative organisms. Fusobacterium spp., Prevotella melaninogenica, and P. intermedi

The octahaem MccA is a haem c-copper sulfite reductase.

The six-electron reduction of sulfite to sulfide is the pivot point of the biogeochemical cycle of the element sulfur. The octahaem cytochrome c MccA (also known as SirA) catalyses this reaction for dissimilatory sulfite utilization by various bacteria. It is distinct from known sulfite reductases because it has a substantially higher catalytic activity and a relatively low reactivity towards nitrite. The mechanistic reasons for the increased efficiency of MccA remain to be elucidated. Here we show that anoxically purified MccA exhibited a 2- to 5.5-fold higher specific sulfite reductase activ

Purification of Vibrio fischeri nitrite reductase and its characterization as a hexaheme c-type cytochrome.

Dissimilatory nitrite reductase was isolated from anaerobically nitrate-grown Vibrio fischeri cells and purified to electrophoretic homogeneity. The enzyme catalyzes the six-electron reduction of nitrite to ammonia. Upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis, under either nonreducing or reducing conditions, the purified nitrite reductase migrated as a single protein band of Mr 57,000. Gel filtration chromatography revealed a native molecular weight of 58,000, indicating the enzyme as isolated to be present in the monomeric form. Purified nitrite reductase exhibited typical

Enrichment and Isolation of Rumen Bacteria That Reduce trans- Aconitic Acid to Tricarballylic Acid.

Bacteria from the bovine rumen capable of reducing trans-aconitate to tricarballylate were enriched in an anaerobic chemostat containing rumen fluid medium and aconitate. After 9 days at a dilution rate of 0.07 h, the medium was diluted and plated in an anaerobic glove box. Three types of isolates were obtained from the plates (a crescent-shaped organism, a pleomorphic rod, and a spiral-shaped organism), and all three produced tricarballylate in batch cultures that contained glucose and trans-aconitate. In glucose-limited chemostats (0.10 h), trans-aconitate reduction was associated with a dec

Structure and function of formate-dependent cytochrome c nitrite reductase, NrfA.

Cytochrome c nitrite reductase, NrfA, catalyzes the six-electron reduction of nitrite, NO(2)(-), to ammonium, NH(4)(+), as the final enzymatic step in the dissimilatory metabolic pathway of nitrite ammonification within the biogeochemical nitrogen cycle. NrfA is a 55-65kDa protein that binds five c-type heme groups via thioether bonds to the cysteines of conserved CXXCH heme attachment motifs. Four of these heme groups are considered to be electron transfer centers, with two histidine residues as axial ligands. The remaining heme group features an unusual CXXCK-binding motif, making lysine the

Reductive activation of the heme iron-nitrosyl intermediate in the reaction mechanism of cytochrome c nitrite reductase: a theoretical study.

Cytochrome c nitrite reductase catalyzes the six-electron, seven-proton reduction of nitrite to ammonia without release of any detectable reaction intermediate. This implies a unique flexibility of the active site combined with a finely tuned proton and electron delivery system. In the present work, we employed density functional theory to study the recharging of the active site with protons and electrons through the series of reaction intermediates based on nitrogen monoxide [Fe(II)-NO(+), Fe(II)-NO·, Fe(II)-NO(-), and Fe(II)-HNO]. The activation barriers for the various proton and electron t

Experimental study on the mechanism of biological hydrogen sulfide generation from organic sulfur-rich coal.

Whether of primary or secondary origin, the presence of hydrogen sulfide (H2S) in coalbed methane (CBM) is commonly attributed to sulfate reduction facilitated by sulfate-reducing bacteria (SRB). However, the sulfate content in high-sulfur coal is exceptionally low, insufficient to function as a substrate for sulfate-reducing bacteria (SRB). In this study, an anaerobic digestion experiment was conducted with high-organic-sulfur coal collected from the Late Permian Longtan Formation in Guangxi Province as both the carbon and sulfur sources. The formation mechanism of H2S is revealed from the ev

Molecular, pathological and immunohistochemical investigation of Helicobacteraceae in the abomasum of dairy cattle and sheep in Northern Italy.

Although several Helicobacter species have been associated with gastric disease in humans and animals, their role in domestic ruminants is still unclear. This study aimed to investigate the prevalence of H. pylori, "Candidatus H. bovis" and other Helicobacteraceae in the abomasum of dairy cattle and sheep reared in Northern Italy, to shed light on their role as potential reservoir of H. pylori for humans and to evaluate the involvement of Helicobacter spp. in the gastric pathology of these animal species. Abomasa from 25 dairy cows and 24 sheep were collected at slaughter. Following macroscopi

Interspecies interaction and effect of co-contaminants in an anaerobic dichloromethane-degrading culture.

An anaerobic stable mixed culture dominated by bacteria belonging to the genera Dehalobacterium, Acetobacterium, Desulfovibrio, and Wolinella was used as a model to study the microbial interactions during DCM degradation. Physiological studies indicated that DCM was degraded in this mixed culture at least in a three-step process: i) fermentation of DCM to acetate and formate, ii) formate oxidation to CO2 and H2, and iii) H2/CO2 reductive acetogenesis. The 16S rRNA gene sequencing of cultures enriched with formate or H2 showed that Desulfovibrio was the dominant population followed by Acetobact

Molecular and carbon isotopic characterization of an anaerobic stable enrichment culture containing Dehalobacterium sp. during dichloromethane fermentation.

UNLABELLED: Biodegradation of dichloromethane (DCM) under reducing conditions is of major concern due to its widespread detection in contaminated groundwaters. Here, we report an anaerobic enrichment culture derived from a membrane bioreactor operating in an industrial wastewater treatment plant, capable of fermenting DCM and the brominated analogue dibromomethane (DBM). Comparative analysis of bacterial 16S rDNA-DGGE profiles from fresh liquid medium inoculated with single colonies picked from serial dilution-to-extinction agar vials showed that cultures degrading DCM contained a predominant

Periodontal pathogens in the shallow pockets of immigrants from developing countries.

The aim of this study was to examine the distribution of typical periodontitis-associated microorganisms in refugees arriving from non-industrialized countries, and to relate the presence of these organisms to the periodontal condition of the subjects. Thirty males between 35-44 years of age were surveyed. Dental plaque, calculus, gingivitis, loss of attachment, and probing depths were recorded for all surfaces. A total of 90 microbiological samples were taken with paper points from mesial sites of teeth 16, 36 and 41. Microbiological test sites were grouped by probing depths and loss of attac

Dental bacterial plaques. Nature and role in periodontal disease.

Antony van Leeuwenhoek first described oral bacteria. However, not until almost 200 years later was the famous Koch postulate introduced. Since then, research has extensively been performed regarding the development and microbiology of dental plaques. In spite of the complexity of the developing flora of supragingival plaque, culture studies have shown a remarkably orderly succession of organisms. Lately, the concept of microbial specificity in the etiology of periodontal diseases has been widely suggested, i.e., that different forms of periodontal disease are associated with qualitatively dis

Active DNRA and denitrification in oxic hypereutrophic waters.

Since the start of synthetic fertilizer production more than a hundred years ago, the coastal ocean has been exposed to increasing nutrient loading, which has led to eutrophication and extensive algal blooms. Such hypereutrophic waters might harbor anaerobic nitrogen (N) cycling processes due to low-oxygen microniches associated with abundant organic particles, but studies on nitrate reduction in coastal pelagic environments are scarce. Here, we report on 15N isotope-labeling experiments, metagenome, and RT-qPCR data from a large hypereutrophic lagoon indicating that dissimilatory nitrate redu

Pilot-scale inoculum-free valorization of raw chicken feathers: ammonium recovery, keratinase production and community dynamics.

Chicken feathers constitute a major keratin-rich agro-industrial residue whose sustainable valorization remains challenging due to the recalcitrant structure of keratin and the energy-intensive nature of conventional treatments. In this study, we investigated the feasibility of converting raw, mechanically untreated chicken feathers into soluble nitrogen compounds and extracellular keratinases through an inoculum-free microbial process operated at pilot scale. A 50 L horizontal bioreactor was run using only tap water and the indigenous feather-associated microbiota, without sterilization, nutr
