Yersinia pestis Lipopolysaccharide Remodeling Confers Resistance to a Xenopsylla cheopis Cecropin.

Fleas are major vectors of Yersinia pestis, the causative agent of plague. It has been proposed that Y. pestis has developed the ability to overcome the innate immune responses of fleas. Despite the fact that they transmit a number of bacterial infections, very little is known about the immune responses in fleas. In this study, we describe the antimicrobial activities of a cecropin from Xenopsylla cheopis (cheopin), an efficient vector for Y. pestis in the wild. This is the first cecropin-class antimicrobial peptide described from Siphonaptera insects. Cheopin showed potent activity against Gr

Genetic structure and gene flow of the flea Xenopsylla cheopis in Madagascar and Mayotte.

BACKGROUND: The flea Xenopsylla cheopis (Siphonaptera: Pulicidae) is a vector of plague. Despite this insect's medical importance, especially in Madagascar where plague is endemic, little is known about the organization of its natural populations. We undertook population genetic analyses (i) to determine the spatial genetic structure of X. cheopis in Madagascar and (ii) to determine the potential risk of plague introduction in the neighboring island of Mayotte. RESULTS: We genotyped 205 fleas from 12 sites using nine microsatellite markers. Madagascan populations of X. cheopis differed, with

Investigation of Bartonella acquisition and transmission in Xenopsylla ramesis fleas (Siphonaptera: Pulicidae).

Bartonella are emerging and re-emerging pathogens affecting humans and a wide variety of animals including rodents. Horizontal transmission of Bartonella species by different hematophagous vectors is well acknowledged but vertical transmission (from mother to offspring) is questionable and was never explored in fleas. The aim of this study was to investigate whether the rodent flea, Xenopsylla ramesis, can acquire native Bartonella from wild rodents and transmit it transovarially. For this aim, Bartonella-free laboratory-reared X. ramesis fleas were placed on six naturally Bartonella-infected

[Differences in the blocking of the proventriculus in male and female Xenopsylla cheopis (Siphonaptera: Pulicidae)].

Frequency of block formation was studied in Xenopsylla cheopis infected with the triptophan-dependent and the typical strains of Yersinia pestis from the Mountain-Altai natural plague focus. It was shown that the rate of blocked males in comparison with that of females was statistically higher. In experiments with the triptophan-dependent strain the frequency of blocking in males was 40.9% and exceeded that in females at four times. The highest rate of blocked males was marked at 7-10th and that of females at 21st days. The tryptophan-dependent strain did not concede to the typical strain in t

Rickettsia typhi and R. felis in rat fleas (Xenopsylla cheopis), Oahu, Hawaii.

Rickettsia typhi (prevalence 1.9%) and R. felis (prevalence 24.8%) DNA were detected in rat fleas (Xenopsylla cheopis) collected from mice on Oahu Island, Hawaii. The low prevalence of R. typhi on Oahu suggests that R. felis may be a more common cause of rickettsiosis than R. typhi in Hawaii.

Effects of temperature on early-phase transmission of Yersina pestis by the flea, Xenopsylla cheopis.

Sharp declines in human and animal cases of plague, caused by the bacterium Yersinia pestis (Yersin), have been observed when outbreaks coincide with hot weather. Failure of biofilm production, or blockage, to occur in the flea, as temperatures reach 30 degrees C has been suggested as an explanation for these declines. Recent work demonstrating efficient flea transmission during the first few days after fleas have taken an infectious blood meal, in the absence of blockage (e.g., early-phase transmission), however, has called this hypothesis into question. To explore the potential effects of te

Distribution and insecticide resistance of the plague flea Xenopsylla cheopis in Maharashtra State, India.

Distributions are reported for commensal rat fleas, predominantly Xenopsylla cheopis (Rothschild), in the State of Maharashtra, India, including the city of Bombay, during 1965-80. The X.cheopis flea index was high in most parts of Maharashtra, but low in Bombay. Rattus rattus Linnaeus is the principal host of X.cheopis, but the host range includes Bandicota bengalensis Gray, Golunda ellioti Gray, Mus musculus Linnaeus, Rattus blandfordi Thomas, R. norvegicus Berkenhout, Suncus caerulaeus Lerr, S. murinus Linnaeus and Tatera indica Hardwicke. X.cheopis was found to have high degrees of resista

[Effect of the digestive process on the survival of the plague agent in Xenopsylla gerbilli Minax fleas].

Hystological investigations of experimentally infected X. gerbilli minax have shown that at the early stages of the digestion, while in the intestine compact clots of non-digested blood, are preserved, proceeds the reproduction of the plague agent. With the decay of the alimentary clot it changes into the dying off of the microbe population and after the ceasing of digestion the accumulation of bacteria renews again. Simultaneously with the changes in the number of microbes their morphology varies. At the initial stage of blood digestion bacilli typical for the agent are dominant. With the dec

Effects of host diet and thermal state on feeding performance of the flea Xenopsylla ramesis.

We examined feeding performance of the flea Xenopsylla ramesis on three different hosts: its natural, granivorous, rodent host, Sundevall's jird (Meriones crassus); the frugivorous Egyptian fruit bat (Rousettus aegyptiacus); and an insectivorous bat, Kuhl's pipistrelle (Pipistrellus kuhlii). Because these fleas are not known to occur on bats, we hypothesized that the fleas' feeding performance (i.e. feeding and digestion rates) would be higher when feeding on their natural host than on either of the bats that they do not naturally parasitize. We found that mass-specific blood-meal size of both

Genetic differentiation of the oriental rat flea, Xenopsylla cheopis, from two sympatric host species.

BACKGROUND: The oriental rat flea (Xenopsylla cheopis), which infests several mammals, primarily rats (Rattus spp.), is the most notorious vector of human plague. In this study, we measured the genetic differentiation among populations of fleas from the Asian house rat (Rattus tanezumi) and the brown rat (R. norvegicus) using microsatellite markers in order to investigate the extent of host-switching in this parasite. RESULTS: We developed 11 polymorphic microsatellite loci for our study, nine of which showed high potential for inbreeding. AMOVA showed that the majority (84.07%, P < 0.001) of

First detection of the kdr mutation (L1014F) in the plague vector Xenopsylla cheopis (Siphonaptera: Pulicidae).

BACKGROUND: The oriental rat flea, Xenopsylla cheopis, is the most efficient vector of the plague. Pyrethroid insecticides such as cypermethrin, cyhalothrin and deltamethrin have been often used to limit plague transmission via controlling the vector during outbreaks. However, this strategy is threatened by the development of insecticide resistance. Understanding the mechanisms underlying pyrethroid resistance is the prerequisite for successful flea control. METHODS: Partial DNA sequences of X. cheopis voltage gated sodium channel (VGSC) gene were amplified from a total of 111 individuals, co

Effectiveness of Fipronil as a Systemic Control Agent Against Xenopsylla cheopis (Siphonaptera: Pulicidae) in Madagascar.

Fipronil was evaluated as a systemic control agent for the rat flea Xenopsylla cheopis (Rothschild), the main vector of Yersinia pestis (Yersin), the causative agent of plague, in Madagascar. The effectiveness of fipronil as a systemic control agent against X. cheopis was assessed by determining the toxicity values of the "Lethal Dose 50" (LD50). Two techniques were used to evaluate the systemic action of the insecticide on the vector: 1) an artificial feeding device filled with blood-fipronil mixture from which X. cheopis was fed and 2) rodent hosts, Rattus norvegicus (Berkenhout) and Rattus

[Characteristics of the multiplication of a virulent strain of the plague microbe in Xenopsylla cheopis fleas infected parenterally].

Xenopsylla cheopis fleas infected parenterally with the virulent strain of plague microbe of gerbil variant preserved the agent to the end of their lives. In the body cavity the microbes retained their ability for reproduction which was, however, limited. During the first seven days after the infection the number of microbes slightly increased and later became stabilized. Its mean indices (mean g) varied within the limits of 500 to 2000 microbe cells per 1 individual, maximum index rarely exceeded 30 000 microbe cells. Parenteral infection with plague agent did not affect essentially the longe

The host/parasite relationships of Xenopsylla astia and X. cheopis on Bandicota bengalensis in Rangoon, Burma.

In Rangoon, Burma, over half the commensal B. bengalensis taken in live traps were found to be infested with Xenopsylla species fleas. X. astia infestations were twice as common as those of the classic plague vector X. cheopis. In terms of absolute numbers X. astia outnumbered X. cheopis by 10 to 1. X. astia was more commonly found as a monospecific infestation, and X. cheopis was more commonly found mixed with X. astia. The flea indicies were high, with a specific index of 2.4 for X. cheopis and 10.2 for X. astia. The flea indicies and flea sex ratios remained stable across the host populatio

Experimental evidence of early-phase transmission of Yersinia pestis by Synopsyllus fonquerniei and Xenopsylla brasiliensis.

BACKGROUND: Flea-borne transmission of the plague bacillus, Yersinia pestis, occurs through two regurgitation-based mechanisms. One, called early-phase transmission (EPT), takes place within 24-96 hours post-infection through a poorly understood mechanism. The second requires an intrinsic incubation period and relies on the formation of a biofilm obstructing the flea's proventriculus. Although these mechanisms have been described in different flea species, they have never been characterized in Xenopsylla brasiliensis and Synopsyllus fonquerniei, which are two flea vectors suspected and confirm

Protocol for monitoring Yersinia pestis colonization of the proventriculus in the flea Xenopsylla cheopis using microscopy.

Colonization of the flea proventriculus is essential for increasing the likelihood of Yersinia pestis transmission. Here, we present a protocol for visualizing Y. pestis colonization dynamics within the proventriculus of the flea Xenopsylla cheopis. We describe steps for quantifying the number of infected proventriculi, the percentage of the surface colonized by bacteria, and an estimation of the bacterial load. These metrics provide a tool for assessing the role of bacterial genes in establishing a transmissible infection. For complete details on the use and execution of this protocol, please

Bioecology of fleas with a focus on the plague vector Xenopsylla Brasiliensis in Mandritsara district, Northern Madagascar.

The rat flea Xenopsylla brasiliensis, a known plague vector present in East and South Africa, has recently been identified in Mandritsara, Madagascar, a plague-endemic area. However, its national distribution remains poorly documented, and studies on other flea vectors and their small mammal hosts in this district are limited. We surveyed flea species, their mammalian hosts, and flea abundance, with a particular focus on X. brasiliensis, in four sites of Mandritsara District between 2021 and 2022. Small mammals were trapped for three consecutive nights per site, inside and outside houses, off-

Plague risk in vulnerable community: assessment of Xenopsylla cheopis susceptibility to insecticides in Malagasy prisons.

BACKGROUND: Prisons in Madagascar are at high risk of plague outbreak. Occurrence of plague epidemic in prisons can cause significant episode of urban plague through the movement of potentially infected humans, rodents and fleas. Rodent and flea controls are essential in plague prevention, by reducing human contact with plague reservoirs and vectors. Insecticide treatment is the key step available for the control of rat fleas which transmit the disease from infected rodents to human. The implementation of an adapted flea control strategy should rely on the insecticide susceptibility status of
