Early development of the thymus in Xenopus laevis.

BACKGROUND: Although Xenopus laevis has been a model of choice for comparative and developmental studies of the immune system, little is known about organogenesis of the thymus, a primary lymphoid organ in vertebrates. Here we examined the expression of three transcription factors that have been functionally associated with pharyngeal gland development, gcm2, hoxa3, and foxn1, and evaluated the neural crest contribution to thymus development. RESULTS: In most species Hoxa3 is expressed in the third pharyngeal pouch endoderm where it directs thymus formation. In Xenopus, the thymus primordium

Development of neurogenic placodes in Xenopus laevis.

The development of neurogenic placodes in Xenopus laevis from the time of neural fold closure to larval stages is described. Placodes were reconstructed from camera lucida drawings of serial sections, and the spatiotemporal pattern of placodal neurogenesis was analyzed using in situ hybridization for the genes X-NGNR-1, XNeuroD, X-MyT1, and X-Delta-1, all of which have been implicated in the regulation of neurogenesis. Olfactory, profundal, and trigeminal placodes, a series of dorsolateral placodes (otic placode and five lateral line placodes), a series of epibranchial placodes, and two hypobr

Expression of parasite transporters in Xenopus oocytes.

The Xenopus laevis oocyte heterologous expression system is particularly useful for the study of transporter proteins. We demonstrated the utility of this expression system for studies on Plasmodium falciparum transporters by inducing increased uptake of metabolites or their analogues (nucleosides, nucleobases, lactate and glucose) into oocytes after microinjection of mRNA obtained from asexual stages of P. falciparum. We identified a hexose transporter of P. falciparum (PfHT1) and studied its function. Higher levels of functional activity are obtained when 5' and 3' untranslated Xenopus globi

Development of the interphotoreceptor matrix in Xenopus laevis.

Xenopus laevis interphotoreceptor matrix (IPM) contains a relatively aqueous insoluble wheat germ agglutinin (WGA)-binding component containing unidentified sialoglycoconjugates (Wood et al [1984] J. Comp. Neurol. 228:299-307). The appearance of WGA-binding macromolecules in the IPM was assessed during late embryonic stages (32-45) and in retinal rudiment cultures, using lectin cytochemistry and Western blotting techniques. Metabolic labeling of the neural retina versus retinal pigment epithelium (RPE)-choroid of juvenile Xenopus with 35S-MET was also evaluated in vivo and in vitro. Lectin cyt

Neurogenesis in the vocalization pathway of Xenopus laevis.

We examined possible contributions of neurogenesis to sex differences in the vocalization pathway of the South African clawed frog, Xenopus laevis. Birthdates of neurons were obtained from autoradiograms of animals receiving tritiated thymidine from gastrulation through 1 month after metamorphosis. Thymidine availability studies showed that 80% of the [3H]-thymidine injected into embryos and tadpoles was incorporated into the DNA of dividing cells within 3 hours. We observed 3 patterns of neurogenesis: late-short, a short burst of proliferation occurred late in development in the anterior preo

A natural heterohaemagglutinin in Xenopus laevis serum.

In most adult Xenopus laevis the serum contains a 'natural' factor capable of lysing the erythrocytes from a wide variety of amniote species. The factor has no effect on the erythrocytes of another amphibian, Ambystoma mexicanum, nor will serum from one animal lyse red cells from another Xenopus individual. No lysing factor was present in the serum of larval (tadpole) Xenopus. Heating of Xenopus serum to 56 degrees for 30 min, absorption of the serum with zymosan or inulin, or removal of calcium and magnesium ions results in loss of lytic activity, although haemagglutinating activity remains,

Magnesium deficiency in embryos of Xenopus laevis.

Embryos of Xenopus laevis reared in media with various low (less than or equal to 10(-5) M) magnesium ion concentrations will exhibit differing degrees of a potentially lethal magnesium starvation syndrome depending on the ion concentration and rearing temperature. The higher the rearing temperature or the lower the magnesium ion content of the medium the more severely the syndrome will be expressed. (Normal development can be expected at temperatures 13-30 degrees C and magnesium ion concentrations greater than 10(-5)M to 10(-2) M.). The magnesium deficiency syndrome in Xenopus embryos is des

Effects of Copper on the Neuromasts of Xenopus Laevis.

Fish and aquatic amphibians possess neuromasts on the surface of their body that constitute the lateral line, a sensory system used to detect water displacement. Copper is known to inactivate the neuromast organs of this system. Copper-induced neuromast loss in African clawed frogs, Xenopus laevis, was examined by exposing Nieuwkoop-Faber stage 54-55 larvae to copper concentrations of 0, 100, 200, 300, and 400 µg/L for 96 h, followed by an examination of neuromast counts, staining intensity, and behavioral responses. Neuromasts were counted using a novel imaging method across four different bo

Studies of Limb Regeneration in Larval Xenopus.

A basic protocol is given for animal maintenance and surgery in studies of hindlimb regeneration in larval Xenopus laevis Unlike urodele limbs, those of larval frogs typically show much more variation in the extent of regeneration after amputation. Such variation can be reduced by optimizing the conditions of larval maintenance to regulate the rates of growth and development, by selecting only larvae with normal rates of growth and morphological development for experimental use, and by attention to precision and consistency in the proximo-distal level of surgical amputation.

The structure and development of Xenopus laevis cornea.

The African clawed frog, Xenopus laevis, is a widely used model organism for tissue development. We have followed the process of corneal development closely in Xenopus and examined the corneal ultrastructure at each stage during its formation. Xenopus cornea development starts at stage 25 from a simple embryonic epidermis overlying the developing optic vesicle. After detachment of the lens placode which takes place around stage 30, cranial neural crest cells start to invade the space between the lens and the embryonic epidermis to construct the corneal endothelium. At stage 41, a second wave o

Vertebral Abnormalities Following Heat Shock in Xenopus Embryos: (Xenopus embryos/vertebral abnormality/heat shock/somite disturbance/bone staining.).

Xenopus embryos were heat-shocked at several stages of development and vertebral abnormalities were examined by means of in situ staining. A high incidence of vertebral abnormalities was evident in larvae treated at the neurula stages (stages 15 and 20) and at the tailbud stages (stages 32 and 35). Heat shock at the neurula stages led to malformed vertebrae and their fusion following an altered arrangement of the arcualia. Heat shock at the tailbud stages induced an asymmetric arrangement of the sacrum and a change in the number of vertebrae, but the arrangement of the arcualia was not affecte

Xmab21l3 mediates dorsoventral patterning in Xenopus laevis.

Specification of the dorsoventral (DV) axis is critical for the subsequent differentiation of regional fate in the primary germ layers of the vertebrate embryo. We have identified a novel factor that is essential for dorsal development in embryos of the frog Xenopus laevis. Misexpression of Xenopus mab21-like 3 (Xmab21l3) dorsalizes gastrula-stage mesoderm and neurula-stage ectoderm, while morpholino-mediated knockdown of Xmab21l3 inhibits dorsal differentiation of these embryonic germ layers. Xmab21l3 is a member of a chordate-specific subclass of a recently characterized gene family, all mem

Calcium oscillations in Xenopus egg cycling extracts.

Cell cycle in various types of cells and in early embryos is often accompanied by transient changes in the concentration of free cytosolic calcium. In the present study, using fluorescent indicator fura-2, we demonstrate that Ca(2+) oscillates cyclically with an amplitude of about 100 nM and a period of mitotic cycle in cell-free Xenopus egg cycling extracts. It peaks in early metaphase just preceding mitotic reactivation of Cdc2 kinase and MAPK and reaches a minimum in interphase. The source of Ca(2+) in the extracts is a particulate fraction containing egg intracellular Ca(2+) stores, since

Retinoid receptors promote primary neurogenesis in Xenopus.

Retinoid receptors, which are members of the nuclear hormone receptor superfamily, act as ligand-dependent transcription factors. They mediate the effects of retinoic acid primarily as heterodimers of retinoic acid receptors (RARs) and retinoid X receptors (RXRs). To analyse their function, xRXR beta synthetic mRNA was injected into Xenopus embryos in combination with normal and mutated xRAR alpha transcripts. Two informative phenotypes are reported here. Firstly, over-expression of xRXR beta with xRAR alpha results in the formation of ectopic primary neurons. Secondly, blocking retinoid signa

Trochlear nerve regeneration in Xenopus laevis larvae.

The trochlear nerve of Xenopus laevis larvae was sectioned in the orbit, and the nerve distal to the section was removed, so that it could not serve to guide the return of the regenerating nerve to the superior oblique muscle (SOM). In a second series, in addition to removing the distal nerve segment, the SOM was removed or damaged. The regeneration pattern of the trochlear nerve fibers was recorded in situ at 1 to 5-day intervals with the aid of methylene blue staining. The early growth or regenerating fibers was multidirectional; only some fibers encountered the SOM by day 6 or 7; others rea

Unilateral Gonadectomy in Adult Male Xenopus laevis.

In vitro fertilization is a reliable laboratory technique for generating large numbers of synchronously developing embryos. This synchronicity enhances Xenopus laevis as a powerful model organism for studying vertebrate embryonic development, molecular, cellular, and disease mechanisms. In conventional in vitro fertilization protocols for X. laevis, male frogs are euthanized, and both testes are harvested. In order to reduce the number of required males, a procedure was developed for unilateral gonadectomy in male X. laevis, which allows sequential use of male frogs' gonads for in vitro fertil

Modelling human genetic disorders in Xenopus tropicalis.

Recent progress in human disease genetics is leading to rapid advances in understanding pathobiological mechanisms. However, the sheer number of risk-conveying genetic variants being identified demands in vivo model systems that are amenable to functional analyses at scale. Here we provide a practical guide for using the diploid frog species Xenopus tropicalis to study many genes and variants to uncover conserved mechanisms of pathobiology relevant to human disease. We discuss key considerations in modelling human genetic disorders: genetic architecture, conservation, phenotyping strategy and

Epichordal vertebral column formation in Xenopus laevis.

Although Xenopus Laevis is the most widely used model amphibian, skeletal development of its vertebral column has not been well illustrated so far. The mode of vertebral column development in anurans has been classified into two modes: perichordal and epichordal. Xenopus vertebral column formation is believed to follow the epichordal mode, but this aspect has been underemphasized, and illustrative examples are currently unavailable to the scientific community. This study documents the entire process of vertebral column formation in X. laevis, from the initial neural arch formation to the compl
