A mini review of xylanolytic enzymes with regards to their synergistic interactions during hetero-xylan degradation.

This review examines the recent models describing the mode of action of various xylanolytic enzymes and how these enzymes can be applied (sequentially or simultaneously) with their distinctive roles in mind to achieve efficient xylan degradation. With respect to homeosynergy, synergism appears to be as a result of β-xylanase and/or oligosaccharide reducing-end β-xylanase liberating xylo-oligomers (XOS) that are preferred substrates of the processive β-xylosidase. With regards to hetero-synergism, two cross relationships appear to exist and seem to be the reason for synergism between the enzyme

Multiplexed Engineering of Disease Resistance and Lodging Tolerance in Wheat Via a Single Xylan Modifying Glycosyltransferase.

Breeding crops with robust disease resistance often compromises yield, a persistent challenge in agricultural improvement. We report that modifying cell wall architecture through a xylan-specific glycosyltransferase successfully breaks this trade-off in wheat. TaXAX1, a pathogen-induced member of the glycosyltransferase (GT)61 family, catalyses arabinose modification of xylan and is predominantly localized to the Golgi apparatus. CRISPR/Cas9 knockout mutants of Taxax1 exhibited enhanced susceptibility to three major fungal diseases: Fusarium head blight (FHB), Fusarium crown rot (FCR) and stri

Chain-length dependent and synergistic prebiotic effects of xylooligosaccharides and xylan on the fecal microbiota of mice in vitro.

Oligomeric and polymeric prebiotics differ in their structural complexity, which influences microbial accessibility and fermentation kinetics. This study investigated the microbial responses to xylooligosaccharides (XOS), xylan (XY), and their combinations in comparison with inulin (INU) using an in vitro model inoculated with BALB/c mice fecal microbiota. Temporal analyses over 48 h included substrate consumption, acid production, and changes in microbial diversity. XOS was rapidly fermented, yielding high acetate and lactate levels, whereas XY was utilized more slowly due to its polymeric st

Furfural production from xylan using a Pueraria Residues carbon-based solid-acid catalyst.

BACKGROUND: The conversion of biomass into high value-added platform compounds is an important method of biomass utilization. The conversion of hemicellulose represented by xylan into furfural can not only reduce the consumption of fossil fuels, but also promotes the development and utilization of non-edible biomass resources. In this study, a bifunctional solid-acid catalyst prepared from agricultural and forestry waste Pueraria (P. eduli) Residues was used to convert xylan into furfural in a biphasic system. RESULTS: In this study, P. eduli Residues was used as raw material to prepare a P.

Enhanced Activity and Stability of an Acetyl Xylan Esterase in Hydrophilic Alcohols through Site-Directed Mutagenesis.

Current demands for the development of suitable biocatalysts showing high process performance is stimulated by the need to replace current chemical synthesis with cleaner alternatives. A drawback to the use of biocatalysts for unique applications is their low performance in industrial conditions. Hence, enzymes with improved performance are needed to achieve innovative and sustainable biocatalysis. In this study, we report the improved performance of an engineered acetyl xylan esterase (BaAXE) in a hydrophilic organic solvent. The structure of BaAXE was partitioned into a substrate-binding reg

Xylan alleviates dietary fiber deprivation-induced dysbiosis by selectively promoting Bifidobacterium pseudocatenulatum in pigs.

BACKGROUND: Low dietary fiber intake has been shown to disturb the gut microbiome community, damage the mucus barrier, and promote pathogen susceptibility. However, little is known about the temporal response of the gut microbiome to dietary fiber deprivation and the recovery induced by dietary fiber inclusion in pigs. OBJECTIVE: In the present study, temporal responses of ileal and fecal microbiota to dietary fiber deprivation were profiled using an ileum cannulated growing pig model. In addition, the potential of dietary-resistant starch, β-glucan, and xylan to alleviate gut dysbiosis throu

In vitro versus in situ evaluation of xylan hydrolysis into xylo-oligosaccharides in broiler chicken gastrointestinal tract.

Xylan hydrolysis into xylo-oligosaccharides (XOS) was evaluated both in the gizzard and ileum of broiler chickens, and by a 2-step in vitro digestion assay that simulated the pH, temperature and time period of the gastric and small intestine (SI) phases. Twelve dietary treatments with varying soluble and insoluble xylan levels, either with or without supplemental xylanase, were fed to broiler chickens (n = 576) for the in situ analysis, and were exposed to the in vitro assay. Relatedness of the two methods was strong for determination of XOS production in all dietary treatments for X5, X4, X3,

Redox responsive xylan-SS-curcumin prodrug nanoparticles for dual drug delivery in cancer therapy.

Chemotherapeutic agents with different anticancer mechanisms could enhance therapeutic effect in cancer therapy by their combined application. In this study, redox-sensitive prodrug nanoparticles based on Xyl-SS-Cur conjugate were developed for co-delivery of curcumin and 5-FU in cancer therapy. The Xyl-SS-Cur conjugate was synthesized via covalent conjugation of curcumin to xylan through a disulphide (-S-S-) linkage. The Xyl-SS-Cur conjugate could self-assemble in aqueous medium into nanoparticles and the lipophilic 5-fluorouracil-stearic acid (5-FUSA) prodrug was encapsulated into the hydrop

Direct transformation of xylan-type hemicelluloses to furfural via SnCl₄ catalysts in aqueous and biphasic systems.

Direct catalytic transformation of xylan-type hemicelluloses to furfural in the aqueous system and the biphasic system were comparatively investigated under mild conditions. Screening of several promising chlorides for conversion of beech xylan in the aqueous system revealed the Lewis acid SnCl4 was the most effective catalyst. Comparing to the single aqueous system, the bio-based 2-methyltetrahydrofuran (2-MTHF)/H2O biphasic system was more conducive to the synthesis of furfural, in which the highest furfural yield of 78.1% was achieved by using SnCl4 as catalysts under the optimized reaction

Adaptive laboratory evolution and rational engineering enabled xylose utilisation and xylan conversion in natural isolates of Saccharomyces cerevisiae.

Second-generation biofuels produced from renewable lignocellulosic biomass (LCB) are attractive alternatives to environmentally damaging, non-renewable fossil fuels. A key challenge in converting LCB to bioethanol is the incomplete utilisation of all available sugars. To address this, the hemicellulose fraction, consisting mainly of xylan, should be converted to the desired product alongside cellulose. This study aimed to develop natural isolate strains of Saccharomyces cerevisiae capable of xylose utilisation and xylan degradation. Strains YI13, YI59 and FIN1 were selected for potential indus

Reactive xylan derivatives for azid-/alkyne-click-chemistry approaches - From modular synthesis to gel-formation.

A modular synthesis was developed to obtain reactive xylan derivatives that are accessible for further functionalization and chemical crosslinking by click-chemistry approaches. Xylan phenylcarbonates (XPCs) with degrees of substitution (DS) from 0.62 to 1.94 were converted with propargyl amine (PA) or 6-azidohexan-1-amine (AA) to introduce either alkynyl- or azido moieties into the polymer backbone. Quantitative conversion of the XPC derivatives into functional xylan carbamates (XCs) with well-defined DS values was achieved. The molecular structure of the compounds was confirmed by FTIR- and

Production of xylooligosaccharides by xylanase from Pichia stipitis based on xylan preparation from triploid Populas tomentosa.

Xylooligosaccharides (XOS) with DP 2-4 are important synbiotics used as food ingredients based on its prebiotic characteristics. In this work, the production of XOS from lignocellulosic material was performed by combined chemical-enzymatic methods. Xylan was prepared from triploid Populas tomentosa, and bioconverted into XOS by crude xylanase solution obtained from Pichia stipitis. The effects of reaction time, temperature, enzyme dosage, and pH value on the production of XOS were fully evaluated. Under the optimal condition (25U g(-1) substrate, pH 5.4 and 50°C), 36.8% of the xylan preparatio

A formaldehyde-reduced phenolic adhesive featuring multi-scale wood bonding: Utilizing dialdehyde xylan for enhanced performance and sustainability.

As a major carbohydrate derived from agricultural/forestry biomass and paper-making processes, xylan is a promising and environmentally benign feedstock for high-value applications. However, its use in adhesives remains limited, primarily due to its inherently low molecular weight, which results in short molecular chains offering insufficient cross-linking sites, as well as high molecular rigidity, which restricts chain flexibility and effective entanglement, thereby impeding the formation of a stable crosslinked network. In this study, xylan was selectively oxidized with sodium periodate to p
