The Influence of Copper Homeostasis Genes copA and copB on Xylella fastidiosa Virulence Is Affected by Sap Copper Concentration.

Xylella fastidiosa is a xylem-limited plant pathogenic bacterium that causes diseases worldwide in crops such as grape, citrus, and olive. Although copper (Cu)-containing compounds are not used for management of X. fastidiosa-caused diseases, they are widely used in X. fastidiosa hosts in vineyards and orchards. The accumulation of Cu in soils and, therefore, plant saps, could be a challenge for X. fastidiosa survival. Here, the molecular basis of Cu homeostasis was studied in relation to virulence. Although homologous Cu-related genes copA (X. fastidiosa loci PD0100) and copB (PD0101) have be

An Evaluation of the Genetic Diversity of Xylella fastidiosa Isolated from Diseased Citrus and Coffee in São Paulo, Brazil.

ABSTRACT Strains of Xylella fastidiosa, isolated from sweet orange trees (Citrus sinensis) and coffee trees (Coffea arabica) with symptoms of citrus variegated chlorosis and Requeima do Café, respectively, were indistinguishable based on repetitive extragenic palindromic polymerase chain reaction (PCR) and enterobacterial repetitive intergenic consensus PCR assays. These strains were also indistinguishable with a previously described PCR assay that distinguished the citrus strains from all other strains of Xylella fastidiosa. Because we were not able to document any genomic diversity in our co

A paratransgenic strategy to block transmission of Xylella fastidiosa from the glassy-winged sharpshooter Homalodisca vitripennis.

BACKGROUND: Arthropod-borne diseases remain a leading cause of human morbidity and mortality and exact an enormous toll on global agriculture. The practice of insecticide-based control is fraught with issues of excessive cost, human and environmental toxicity, unwanted impact on beneficial insects and selection of resistant insects. Efforts to modulate insects to eliminate pathogen transmission have gained some traction and remain future options for disease control. RESULTS: Here, we report a paratransgenic strategy that targets transmission of Xylella fastidiosa, a leading bacterial pathogen

Overexpression of Citrus reticulata SAMT in Nicotiana tabacum increases MeSA volatilization and decreases Xylella fastidiosa symptoms.

Nicotiana tabacum overexpressing CrSAMT from Citrus reticulata increased production of MeSA, which works as an airborne signal in neighboring wild-type plants, inducing PR1 and increasing resistance to the pathogen Xylella fastidiosa. Xylella fastidiosa is one of the major threats to plant health worldwide, affecting yield in many crops. Despite many efforts, the development of highly productive resistant varieties has been challenging. In studying host plant resistance, the S-adenosyl-L-methionine: salicylic acid carboxyl methyltransferase gene (SAMT) from Citrus reticulata, a X. fastidiosa r

Retention sites for Xylella fastidiosa in four sharpshooter vectors (Hemiptera: Cicadellidae) analyzed by scanning electron microscopy.

Xylella fastidiosa is a xylem-limited bacterium that causes citrus variegated chlorosis (CVC), Pierce's disease of grapevine, and leaf scald of coffee and plum and many other plant species. This pathogen is vectored by sharpshooter leafhoppers (Hemiptera: Cicadellidae: Cicadellinae) and resides in the insect foregut. Scanning electron microscopy was used to determine the retention sites of X. fastidiosa for the most common vector species in Brazilian citrus groves, Acrogonia citrina, Bucephalogonia xanthophis, Dilobopterus costalimai, and Oncometopia facialis. After a 48-h acquisition access p

1H NMR Approach for Evaluating the Effects of a Natural Detergent on Olive Trees Infected by Xylella fastidiosa subsp. pauca.

X. fastidiosa subsp. pauca (Xfp) is the etiological agent of "Olive Quick Decline Syndrome" (OQDS). Cellina di Nardò (Olea europaea L., Oleaceae), one of the major Salento cultivars, is highly susceptible to Xfp, usually showing acute symptoms after infection. NuovOlivo® a plant-derived formulation made with vegetal oils and water infusion from multi botanical species has been reported as effective against OQDS in plants affected by Xfp. A non-targeted 1H NMR (Nuclear Magnetic Resonance) fingerprinting approach, with unsupervised and supervised analysis, was applied to observe the possible cha

Towards sustainable management of Xylella fastidiosa vectors: An annotated image dataset for automated in-field detection of Aphrophoridae foam.

Insects feeding on xylem sap, such as adult Aphrophoridae spittlebugs, are vectors of the plant pathogenic xylem-limited bacterium Xylella fastidiosa (Xf), a causal agent of a number of severe diseases, including the Olive Quick Decline Syndrome (OQDS), which has decimated olive trees in the Mediterranean region. The Aphrophoridae life cycle and behaviour feature a weak stage, known as the juvenile stage, in which the insects live solitary on stems covered in a self-produced foamy fluid (froth) that protects them from dehydration and temperature stress. Juvenile vectors are ideal targets for a

Nationwide Survey Using Real-Time PCR in 2024 and 2025 Supports the Absence of Xylella fastidiosa in Korea.

Xylella fastidiosa is a plant-pathogenic bacterium that causes severe diseases in economically important crops, such as citrus and grapevine, thereby posing a significant threat to global agriculture. Although X. fastidiosa has not yet been reported in Korea, the increase in international trade and its presence in neighboring countries highlight the necessity of continued surveillance. The objective of this study was to verify the absence of X. fastidiosa in Korea and to establish a reliable diagnostic framework through a nationwide survey conducted in 2024 and 2025. The sampling design was ge

A metabolic model based on a pangenome core reveals putative conserved biochemical features of the phytopathogen Xylella fastidiosa.

Xylella fastidiosa is a xylem-limited phytopathogenic bacterium responsible for severe diseases in many economically important crops. Despite its impact, its metabolism remains poorly characterized due to fastidious growth and the limited availability of defined culture media. Here, we reconstruct the first pangenome-based genome-scale metabolic model for X. fastidiosa, integrating conserved metabolic functions from 18 strains across five subspecies. The resulting consensus model, iXfcore, is manually curated and used to explore the species' metabolic capabilities. Model simulations predict mi

Identification of an Almond Cultivar Resistant to Xylella fastidiosa subsp. fastidiosa and Insights into Host-Specific Bacterial Behavior.

Xylella fastidiosa (Xf) is a xylem-limited bacterial pathogen causing disease in a variety of important agricultural crops, including almonds. Almond leaf scorch (ALS) disease was first identified in Israel in 2017 and is associated with the subspecies X. fastidiosa subsp. fastidiosa (Xff) sequence type 1. Currently, there is no effective means to cure infected almond trees, and ALS management relies primarily on uprooting infected trees and reducing vector populations, yet these methods are only moderately effective. Thus, identifying resistant almond genotypes is a critical goal for developi

Octahedral ruthenium and magnesium naringenin 5-alkoxide complexes: NMR analysis of diastereoisomers and in-vivo antibacterial activity against Xylella fastidiosa.

Although many copper-based antimicrobial compounds have been developed to control pathogenic bacteria and fungi in plants and applied for crop protection, there is evidence that several plant pathogens have developed resistance to copper-based antimicrobial compounds, including some Xanthomonas species. Xylella is a bacterial genus belonging to the Xanthomonas family; and X. fastidiosa, which is responsible for citrus variegated chlorosis (CVC) in sweet orange, may develop resistance to one or more copper-based antimicrobials. Because of the time required for the development and approval of ne

Identification and characterization of Phi1 and Phi3 bacteriophages targeting Xylella fastidiosa subsp. pauca, the causal agent of olive quick decline in Italy.

The xylem-limited bacterium Xylella fastidiosa (Xf) is increasingly threatening global agriculture due to the absence of effective control strategies. Phage biocontrol represents a promising alternative; however, only a limited number of bacteriophages have been identified against Xf. 1H-NMR Metabolite Fingerprinting Analysis Reveals a Disease Biomarker and a Field Treatment Response in Xylella fastidiosa subsp. pauca-Infected Olive Trees.

Xylella fastidiosa subsp. pauca is a xylem-limited bacterial phytopathogen currently found associated on many hectares with the "olive quick decline syndrome" in the Apulia region (Southern Italy), and the cultivars Ogliarola salentina and Cellina di Nardò result in being particularly sensitive to the disease. In order to find compounds showing the capability of reducing the population cell density of the pathogen within the leaves, we tested, in some olive orchards naturally-infected by the bacterium, a zinc-copper-citric acid biocomplex, namely Dentamet®, by spraying it to the crown, once pe

Tacking the vector of Xylella fastidiosa: geo-statistical analysis of long-term field observations on host plants influencing the distribution of Phylaenus spumarius nymphs.

The meadow froghopper, Philaenus spumarius L., is endemic in Italy and was not considered a harmful species until 2014, when the olive quick decline syndrome (OQDS) showed up in Apulia (southern Italy). It was immediately suspected and then verified as the main vector of Xylella fastidiosa, the bacterium responsible for the disease. Currently, EU Directives consider the fight against P. spumarius compulsory in member states and recommend Integrated Pest Management (IPM), both in uncultivated and cultivated infested areas, to minimise the environmental impact of chemical pesticides. This should

Use of Commercial Fertilizers in an IPDM Protocol to Mitigate Olive Quick Decline Syndrome Caused by Xylella fastidiosa subsp. pauca in Southern Italy.

Management of Xylella fastidiosa subsp. pauca (Xfp) presents significant challenges because of the lack of genetic resistance in major crops, pathogen variability and resistance development against the main antimicrobial compounds, and the lack of effective chemical compounds for control. Traditional bactericides in the EU are restricted, and copper-based products are increasingly limited because of environmental impacts. In this study, the antibacterial side effects of some commercial fertilizers against Xfp were evaluated. These products, containing metal ions (mainly copper and/or zinc) com

Further In Vitro Assessment and Mid-Term Evaluation of Control Strategy of Xylella fastidiosa subsp. pauca in Olive Groves of Salento (Apulia, Italy).

During recent years; Xylella fastidiosa subsp. pauca (Xfp) has spread in Salento causing relevant damage to the olive groves. Measures to contain the spreading of the pathogen include the monitoring of the areas bordering the so-called "infected" zone and the tree eradication in case of positive detection. In order to provide a control strategy aimed to maintain the tree productivity in the infected areas, we further evaluated the in vitro and in planta mid-term effectiveness of a zinc-copper-citric acid biocomplex. The compound showed an in vitro bactericidal activity and inhibited the biofil

Mass Spectrometry-Based Targeted Lipidomics and Supervised Machine Learning Algorithms in Detecting Disease, Cultivar, and Treatment Biomarkers in Xylella fastidiosa subsp. pauca-Infected Olive Trees.

In 2013, Xylella fastidiosa (Xf) was detected for the first time in Apulia and, subsequently, recognized as the causal agent of the olive quick decline syndrome (OQDS). To contain the disease, the olive germplasm was evaluated for resistance to Xf, identifying cultivars with different susceptibility to the pathogen. Regarding this, the resistant cultivar Leccino has generally a lower bacterial titer compared with the susceptible cultivar Ogliarola salentina. Among biomolecules, lipids could have a pivotal role in the interaction of Xf with its host. In the grapevine Pierce's disease, fatty aci

Xylella fastidiosa, a fastidious bacterium causing disease in over 100 plant species, is classified as a single species, although genetic studies support multiple taxons. To determine the taxonomic relatedness among strains of X. fastidiosa, we conducted DNA-DNA relatedness assays and sequenced the 16S-23S intergenic spacer (ITS) region using 26 strains from 10 hosts. Under stringent conditions (Tm -15 degrees C), the DNA relatedness for most X. fastidiosa strains was *70%. However, at high stringency (Tm -8 degrees C), three distinct genotypes (A, B, and C) were revealed. Taxon A included str

A Simple Defined Medium for the Production of True Diketopiperazines in Xylella fastidiosa and Their Identification by Ultra-Fast Liquid Chromatography-Electrospray Ionization Ion Trap Mass Spectrometry.

Diketopiperazines can be generated by non-enzymatic cyclization of linear dipeptides at extreme temperature or pH, and the complex medium used to culture bacteria and fungi including phytone peptone and trypticase peptone, can also produce cyclic peptides by heat sterilization. As a result, it is not always clear if many diketopiperazines reported in the literature are artifacts formed by the different complex media used in microorganism growth. An ideal method for analysis of these compounds should identify whether they are either synthesized de novo from the products of primary metabolism an

However, current understanding of host responses upon Xf-infection is limited to allow early disease prediction, diagnosis, and sustainable strategies for breeding on plant tolerance. Recently, we identified a major complex trait for early de novo programming, named CoV-MAC-TED, by comparing early transcriptome data during plant cell survival with SARS-CoV-2-infected human cells.
