Beyond monoculture limits: a critical review of yeast co-cultures for xylitol production from lignocellulosic biomass : Yeast co-cultures for xylitol production.

The biotechnological production of xylitol from lignocellulosic biomass residues offers a sustainable alternative to traditional chemical synthesis; however, its industrial implementation is limited by the metabolic constraints of conventional microbial monocultures. This review examines the limitations of monocultures, highlighting how redox cofactor imbalances, glucose-induced catabolic repression, and the toxicity of furanic inhibitors and organic acids in crude hydrolysates create energetic and metabolic challenges that reduce yields and volumetric productivity. To overcome these biologica

Sweeteners: erythritol, xylitol and cardiovascular risk-friend or foe?

Hyperglycaemia harms vascular health and promotes platelet aggregation. Reducing glucose concentration is crucial, and sugar alcohols may aid this effort. Used for over 50 years in food, cosmetic, and pharmaceutical industries, erythritol and xylitol minimally affect plasma glucose and insulin levels while promoting the release of beneficial gastrointestinal hormones such as e.g. glucagon-like peptide-1. These properties make them particularly appealing for individuals with diabetes, obesity, and metabolic syndrome. Recent pilot trials suggest that xylitol and erythritol might temporarily alte

Improved bone biomechanical properties in xylitol-fed aged rats.

Our previous studies have shown that dietary xylitol protects against weakening of bone biomechanical properties in experimental postmenopausal osteoporosis. To study whether xylitol preserves bone biomechanics also during aging, a long-term experimental study was performed with rats. Twenty-four male Sprague-Dawley rats were divided into 2 groups. The rats in the control group (NON-XYL group) were fed a basal rat and mouse no. 1 maintenance (RM1) diet, while the rats in the experimental group (XYL group) were continuously fed the same diet supplemented with 10% xylitol (wt/wt). The rats were

Xylitol production by recombinant Corynebacterium glutamicum under oxygen deprivation.

Wild-type Corynebacterium glutamicum produced 0.6 g l(-1) xylitol from xylose at a productivity of 0.01 g l(-1) h(-1) under oxygen deprivation. To increase this productivity, the pentose transporter gene (araE) from C. glutamicum ATCC31831 was integrated into the C. glutamicum R chromosome. Consequent disruption of its lactate dehydrogenase gene (ldhA), and expression of single-site mutant xylose reductase from Candida tenuis (CtXR (K274R)) resulted in recombinant C. glutamicum strain CtXR4 that produced 26.5 g l(-1) xylitol at 3.1 g l(-1) h(-1). Lipase-catalyzed synthesis of xylitol monoesters: solvent engineering approach.

A solvent engineering strategy was applied to the lipase-catalyzed synthesis of xylitol-oleic acid monoesters. The different esterification degrees for this polyhydroxylated molecule were examined in different organic solvent mixtures. In this context, conditions for high selectivity towards monooleoyl xylitol synthesis were enhanced from 6 mol% in pure n-hexane to 73 mol% in 2-methyl-2-propanol/dimethylsulfoxide (DMSO) 80:20 (v/v). On the contrary, the highest production of di- and trioleoyl xylitol, corresponding to 94 mol%, was achieved in n-hexane. Changes in polarity of the reaction mediu

A mechanism for the thiamin-sparing action of dietary xylitol in the rat.

The changes induced by dietary xylitol in the gastrointestinal tract of the rat were investigated in relation to the phenomenon of vitamin-sparing. Within 18 days of consuming a synthetic diet, deficient in thiamin, riboflavin and pyridoxine, rats ceased to grow and began to lose weight rapidly. If xylitol was then included in the diet (10% w/w), the effect of the vitamin-deficient diet on growth was reversed. Moreover, within 3 days of the rats ingesting xylitol, the metabolism of this sugar polyol by the caecal microflora was increased 17-fold and the caecal concentrations of thiamin and thi

Biological production and downstream separation of xylitol using Candida tropicalis.

Xylitol is one of the platform chemicals identified by the U.S. Department of Energy as a promising candidate for production from biomass. It is an important chemical building block with applications in both the food and pharmaceutical industries. Xylitol is a pentose sugar alcohol with low caloric value, anti-cariogenic properties, and some ability to inhibit microbial growth. The objective of this work is to identify new routes for production of xylitol from lignocellulosic biomass with a focus on post-fermentation downstream processing and separation. Fermentation (via C. tropicalis) condit

Dietary xylitol supplementation prevents osteoporotic changes in streptozotocin-diabetic rats.

The effects of 10% and 20% dietary xylitol supplementation on the biomechanical properties, trabeculation, and mineral content of long bones were studied in streptozotocin-diabetic rats. Forty 3-month-old male Wistar rats were divided randomly into four groups of 10. Rats in three groups were administered a single injection of streptozotocin (50 mg/kg body weight) to induce type I diabetes, while animals in the fourth group were given a sham injection of physiological saline. The sham-injected group and one of the streptozotocin-diabetic groups were fed the basal diet, while the two diabetic g

Retention of calcium from various xylitol-calcium combinations in rats.

Previous studies have shown that xylitol, the five-carbon sugar alcohol, increases the intestinal absorption of Ca in the rat. In this study an optimum xylitol:Ca molar ratio for Ca absorption and retention was determined in 10-week-old Long-Evans male rats. Xylitol was intubated with 20 mg of 45Ca (as 45CaCl2 or 45CaCO3) into the stomach of fasted rats in 1 ml water using xylitol:Ca molar ratios of 0:1, 0.5:1, 1:1, 1.5:1 or 2:1. Radioactivity of urine, feces and humerus was determined from 48-hr samples. The highest retention rate of 45Ca in the whole body or in the humerus was found when 114

Binding affinity of anti-xylitol antibodies to canine hepatic vessels.

Xylitol is used as a sugar substitute in food products. Dogs have been reported to experience lethal liver injury after accidental ingestion of xylitol. Because liver injury may be a serious consequence of canine immune-mediated reactions, antibodies produced against xylitol may attack the liver. Therefore, in the present study, we evaluated whether binding sites for xylitol antibodies are located at the liver or not. Anti-xylitol antibodies were generated by immunization of rabbits with a xylose-bovine serum albumin conjugate. Immunohistological examination showed that binding sites for the a

Improved bone biomechanical properties in rats after oral xylitol administration.

The effects of 5, 10, and 20% dietary xylitol supplementations on the biomechanical properties, histological architecture, and the contents of collagen, pyridinoline, and deoxypyridinoline in long bones of rats were studied. Tibiae were used for the three-point bending test, and femurs were used for the torsion and loading test of the femoral neck. The 10 and 20% oral xylitol administrations caused a significant increase of tibial stress, femoral shear stress, and stress of the femoral neck as compared with the controls. Long-term effect of xylitol chewing gum on dental caries.

About 85% (n = 269) of the subjects who participated in the Ylivieska follow-up studies on the effect of xylitol chewing gum on dental caries during 1982-84 or 1982-85 were re-examined in 1987 for the analysis of possible long-term preventive effects. Further caries reduction was found 2 or 3 yr after the discontinuation of the use of xylitol. The effect was especially marked in girls; the reduction in caries increment in the post-use years was 60% for the 2-yr users, suggesting that more pronounced caries reduction was associated with the most regular use of xylitol. In teeth erupting during

Effect of erythritol and xylitol on Streptococcus pyogenes causing peritonsillar abscesses.

Polyols are effective against caries-causing streptococci but the effect on oropharynx-derived pyogenic streptococci is not well characterised. We aimed to study the effect of erythritol (ERY) and xylitol (XYL) against Streptococcus pyogenes isolated from peritonsillar abscesses (PTA). We used 31 clinical isolates and 5 throat culture collection strains. Inhibition of bacterial growth by polyols at 2.5%, 5% and 10% concentrations was studied and the results were scored. Amylase levels in PTA pus were compared to polyol effectivity scores (PES). Growth curves of four S. pyogenes isolates were a
