Modulating ferroptosis and mycobactericidal activity in lung epithelial cells via YY1/iNOS pathway.

BACKGROUND: Mycobacterium tuberculosis infection triggers various forms of host cell death, including ferroptosis in lung epithelial cells; YY1, a critical transcription factor, plays a pivotal role in regulating ferroptosis, however, the underlying mechanisms are not fully understood. METHODS: To investigate Mycobacterium marinum (M.marinum) infection in lung epithelial cells A549 and H1299, we utilized flow cytometry to evaluate cell death and measure reactive oxygen species (ROS). Colony-forming unit (CFU) assays determined the intracellular bacterial load. Ferroptosis was analyzed using a

Epithelial inactivation of Yy1 abrogates lung branching morphogenesis.

Yin Yang 1 (YY1) is a multifunctional zinc-finger-containing transcription factor that plays crucial roles in numerous biological processes by selectively activating or repressing transcription, depending upon promoter contextual differences and specific protein interactions. In mice, Yy1 null mutants die early in gestation whereas Yy1 hypomorphs die at birth from lung defects. We studied how the epithelial-specific inactivation of Yy1 impacts on lung development. The Yy1 mutation in lung epithelium resulted in neonatal death due to respiratory failure. It impaired tracheal cartilage formation

RNA sequencing identifies fusion of the EWSR1 and YY1 genes in mesothelioma with t(14;22)(q32;q12).

Mesothelioma is a rare but very aggressive tumor derived from mesothelial cells. A number of often complex but nonrandom cytogenetic abnormalities have been found in these tumors, resulting in loss of chromosome bands 14q32 and 22q12 in more than 35% of the cases. In this study, we used RNA sequencing to search for fusion transcripts in a mesothelioma carrying a t(14;22)(q32;q12) as the sole chromosomal aberration and found an EWSR1-YY1 and its reciprocal YY1-EWSR1 fusion transcript. Screening 15 additional cases of mesothelioma from which we had RNA but no cytogenetic information, we identifi

YY1-PVT1 affects trophoblast invasion and adhesion by regulating mTOR pathway-mediated autophagy.

Insufficient trophoblast invasion is the key factor for the occurrence of recurrent spontaneous abortions (RSA). Our previous studies identified Yin Yang 1 (YY1) as a transcription factor involved in the regulation of trophoblast invasiveness at the maternal-fetal interface. Long noncoding RNAs (lncRNAs) can regulate gene expression and autophagy in many ways. The purpose of this study was to explore the relationship between YY1 and lncRNAs and the mechanism by which lncRNAs affect the biological behavior of trophoblasts. Bioinformatic analysis predicted that YY1 had three binding sites in the

CD4+ Tregs Drive Post-Ischemic Sprouting Angiogenesis via Endothelial YY1/MAML1 Reactivation.

Microvascular complications of diabetes are chronic diseases of small vessels. We previously found that CD4+ regulatory T-cells (Tregs) are markedly reduced in type 2 diabetes (T2D) after ischemic injury in both mice and humans, and that Treg deficiency in immunodeficient mice impairs vascular regeneration. However, the mechanisms by which Tregs protect against diabetic vascular disease remain unclear. Here, we demonstrate that endothelial cells (ECs) upregulate the transcription factor YY1 during post-ischemic vascular regeneration, but this response is blunted in diabetic ECs. Endothelial-sp

miRNA-mediated regulation of lineage plasticity in YY1 knockout pro-B cells.

Yin Yang 1 (YY1) is a multifunctional transcription factor involved in chromatin organization and gene regulation. While the mechanisms by which YY1 controls B cell lineage development have been explored at the transcriptional level, little is known at the post-transcriptional level. To address this, we performed integrated analyses combining bulk transcriptomics, miRNA profiling, and single-cell transcriptomics to identify YY1-dependent regulatory networks at the pro-B cell stage. YY1 knockout (KO) in pro-B cells leads to reduced B-lineage commitment and activation of genes associated with al

Single-cell transcriptomic profiling identifies YY1 as a key regulator of fibroblast plasticity in hypertrophic scar formation.

BACKGROUND: Hypertrophic scars (HTSs) are characterized by excessive extracellular matrix deposition and impaired scar remodelling. Fibroblasts are central to HTS pathogenesis, yet clinical strategies remain limited by an incomplete understanding of fibroblast heterogeneity and transcriptional regulation. This study aimed to identify a key fibroblast subpopulation and its regulatory transcription factors to address this translational gap. METHODS: Single-cell RNA sequencing was performed on dermal cells from freshly excised human HTS and normal skin (NS) tissues. Fibroblast subsets and transc

A mutual regulatory loop between transcription factor Yin Yang 1 and hepatitis B virus replication influences chronic hepatitis B.

Hepatitis B virus (HBV) infections pose a major threat to human health. HBV can upregulate the expression of the transcription factor Yin Yang 1 (YY1) in in vitro cytological experiments, suggesting an association between YY1 and HBV infection. However, data on YY1 expression in chronic hepatitis B (CHB) patients are lacking. In this study, we aimed to assess the correlation between YY1 expression and HBV infection. We detected serum YY1 levels in 420 patients with chronic HBV infection, 30 patients with chronic hepatitis C virus infection, and 32 healthy controls using an enzyme-linked immuno

YY1 regulates vascular resistance and blood pressure dynamics through epigenetic control of m6A RNA modifications in vascular smooth muscle cells.

AIMS: Recent genome-wide association study analysis has identified YY1 as a novel locus associated with blood pressure traits; however, whether YY1 directly controls vasoreactivity remains unknown. The principal function of vascular smooth muscle cells (VSMCs) is to contract, which is essential for regulating vascular tone, blood flow, and blood pressure. We hypothesized that YY1, a transcription factor, facilitates vascular function by epigenetically regulating gene expression in VSMCs. METHODS AND RESULTS: The effects of VSMC-specific YY1 loss were studied in mice. Lineage tracing, calcium

YY1-VDR axis-mediated autophagy slows the progression of intervertebral disc degeneration.

BACKGROUND: Aging is accompanied by degeneration of the intervertebral discs (IVDD), which leads to a poor quality of life and a significant socioeconomic burden. The present study aimed to explore the effect of transcription factor Yin and Yang 1 (YY1) on IVDD and to elucidate the potential underlying mechanism. METHODS: Differentially expressed genes in nucleus pulposus (NP) tissues from patients with early or advanced IVDD were screened out using transcriptome sequencing. Mice were subjected to IVDD modeling, and NP cells were treated with H2O2 and lentivirus. The oxidative stress and auto

Yin Yang 1 controls cerebellar astrocyte maturation.

Diverse subpopulations of astrocytes tile different brain regions to accommodate local requirements of neurons and associated neuronal circuits. Nevertheless, molecular mechanisms governing astrocyte diversity remain mostly unknown. We explored the role of a zinc finger transcription factor Yin Yang 1 (YY1) that is expressed in astrocytes. We found that specific deletion of YY1 from astrocytes causes severe motor deficits in mice, induces Bergmann gliosis, and results in simultaneous loss of GFAP expression in velate and fibrous cerebellar astrocytes. Single cell RNA-seq analysis showed that Y

Transgenic over-expression of YY1 induces pathologic cardiac hypertrophy in a sex-specific manner.

YY1 can activate or repress transcription of various genes. In cardiac myocytes in culture YY1 has been shown to regulate expression of several genes involved in myocyte pathology. YY1 can also acutely protect the heart against detrimental changes in gene expression. In this study we show that cardiac over-expression of YY1 induces pathologic cardiac hypertrophy in male mice, measured by changes in gene expression and lower ejection fraction/fractional shortening. In contrast, female animals are protected against pathologic gene expression changes and cardiac dysfunction. Furthermore, we show

Defective mitochondrial morphology and bioenergetic function in mice lacking the transcription factor Yin Yang 1 in skeletal muscle.

The formation, distribution, and maintenance of functional mitochondria are achieved through dynamic processes that depend strictly on the transcription of nuclear genes encoding mitochondrial proteins. A large number of these mitochondrial genes contain binding sites for the transcription factor Yin Yang 1 (YY1) in their proximal promoters, but the physiological relevance is unknown. We report here that skeletal-muscle-specific YY1 knockout (YY1mKO) mice have severely defective mitochondrial morphology and oxidative function associated with exercise intolerance, signs of mitochondrial myopath

Skeletal muscle stem cells modulate niche function in Duchenne muscular dystrophy mouse through YY1-CCL5 axis.

Adult skeletal muscle stem cells (MuSCs) are indispensable for muscle regeneration and tightly regulated by macrophages (MPs) and fibro-adipogenic progenitors (FAPs) in their niche. Deregulated MuSC/MP/FAP interactions and the ensuing inflammation and fibrosis are hallmarks of dystrophic muscle. Here we demonstrate intrinsic deletion of transcription factor Yin Yang 1 (YY1) in MuSCs exacerbates dystrophic pathologies by altering composition and heterogeneity of MPs and FAPs. Further analysis reveals YY1 loss induces expression of immune genes in MuSCs, including C-C motif chemokine ligand 5 (C

YY1 promotes diabetic vascular calcification via m6A-Binding protein HNRNPC.

BACKGROUND: Diabetes is one of the most common and fastest-growing diseases worldwide, and diabetic atherosclerotic calcification is a frequent and fatal complication, the underlying mechanisms of which remain unclear. In this study, we investigated the mechanism by which HNRNPC regulates diabetic vascular calcification. METHODS: We retrieved datasets GSE211722, GSE84012, and GSE74755 from the GEO database and performed probe-to-gene name conversion. Based on expression profile data and aortic transcriptomic data from diabetic and non-diabetic mice, RNA m6A methylation-related regulatory gene

Novel small-molecule positive allosteric modulator 1 with blood-brain barrier penetration activity exerts anti-cellular senescence effects via the PAC1-R/YY1/SIRT6 pathway.

<p indent="0mm">Aging is the primary risk factor for neurodegeneration. SIRT6, a key longevity regulator, is downregulated in aged brain tissue and linked to neurodegeneration. Thus, activating SIRT6 may delay brain aging. Based on this, the present study reveals the molecular mechanism by which the novel small-molecule positive allosteric modulator (SPAM1) antagonizes neuronal senescence through the PAC1-R/YY1/SIRT6 signaling axis, along with its highly efficient brain-targeting properties. Transcriptomic profiling of insulinomas reveals a new "low-endocrine" subtype with low YY1 expressions.

BACKGROUND: Insulinoma is the most common functional pancreatic neuroendocrine tumors, typically originating from pancreatic β cells. However, the transcriptional heterogeneity and the regulatory role of transcription factor YY1 remain incompletely understood. METHODS: High-throughput bulk-RNA sequencing was conducted on insulinoma tumor samples. Unsupervised clustering algorithms were employed to identify molecular subtypes of insulinomas. YY1 mutation status was analyzed by whole-exome sequencing. Functional roles of YY1 were analyzed by Yy1-overexpression and knockdown in INS-1 cell line.
