[Establishment and evaluation of identification method for Yersinia pestis and Yersinia pseudotuberculosis].

OBJECTIVE: To establish a gene identification method of Yersinia pestis and Yersinia pseudotuberculosis for plague surveillance. METHODS: According to the specific genomic sequences of Y. pestis and Y. pseudotuberculosis, i.e. "pestis Island (PeI)" and "pseudotuberculosis Island (PsI)" and the published genomic sequences of 12 strains of Y. pestis and 4 strains of Y. pseudotuberculosis, the specific identification primers of these sequences were designed. RESULTS: A total of 52 strains of Y. pestis and 57 strains of Y. pseudotuberculosis and other intestinal bacteria strains were tested with

Pyogenic vertebral osteomyelitis caused by Yersinia pseudotuberculosis.

Septic vertebral osteomyelitis caused by Yersinia pseudotuberculosis is extremely rare. We report the first case of pyogenic vertebral osteomyelitis due to Y. pseudotuberculosis. A 65-year-old man presented with low back pain and fever. He had a history of psoriasis vulgaris treated with adalimumab. The patient reported drinking adequate amounts of well water daily. Clinical examination revealed no neurological deficit of the lower limbs. Laboratory findings included increased inflammatory reactions. Magnetic resonance imaging showed diffuse changes in intensity at the T10 and L3 vertebrae, an

[IMMUNOCHEMICAL ACTIVITY OF YERSINIA PSEUDOTUBERCULOSIS B-ANTIGEN].

A hybridoma panel producing monoclonal antibodies to immunochemically non-identical antigenic epitopes of the protein nature located in outer membrane of Yersinia pseudotuberculosis was obtained. It was revealed that the previously identified B-antigen protecting laboratory animals from experimental plague was detected using both monoclonal antibodies against mentioned protein determinants and the determinants of lipopolysaccharide O-side chains. The B-antigen is a component of the Y. pseudotuberculosis outer membrane, egested in the form of the vesicles (OMVs) by bacteria cultivated in fluid

Oral vaccination against plague using Yersinia pseudotuberculosis.

Yersinia pestis, the agent of plague, is among the deadliest bacterial pathogens affecting humans, and is a potential biological weapon. Because antibiotic resistant strains of Yersinia pestis have been observed or could be engineered for evil use, vaccination against plague might become the only means to reduce mortality. Although plague is re-emerging in many countries, a vaccine with worldwide license is currently lacking. The vaccine strategy described here is based on an oral vaccination with an attenuated strain of Yersinia pseudotuberculosis. Indeed, this species is genetically almost i

Caprine enteritis associated with Yersinia pseudotuberculosis infection.

Yersiniosis was prevalent among a caprine herd during the late autumn of 2003 in Iwate Prefecture, Japan. The disease affected 29 of about 100 lactating goats, but not dried or nonparous goats, mature male goats or kids. Four animals died within an epidemic period of 20 days. Affected animals developed decreased milk production with subsequent watery diarrhea, neutrophilia with increased band forms and multiple microabscesses characteristic of yersiniosis in the intestinal mucosa from the jejunum to caecum as well as in the mesenteric lymph nodes. Y. pseudotuberculosis serotype III was isolate

Characterization of chromosomal regions conserved in Yersinia pseudotuberculosis and lost by Yersinia pestis.

The transformation of the enteropathogenic bacterium Yersinia pseudotuberculosis into the plague bacillus, Yersinia pestis, has been accompanied by extensive genetic loss. This study focused on chromosomal regions conserved in Y. pseudotuberculosis and lost during its transformation into Y. pestis. An extensive PCR screening of 78 strains of the two species identified five regions (R1 to R5) and four open reading frames (ORFs; orf1 to orf4) that were conserved in Y. pseudotuberculosis and absent from Y. pestis. Their conservation in Y. pseudotuberculosis suggests a positive selective pressure

Prevalence of pathogenic Yersinia enterocolitica and Yersinia pseudotuberculosis in wild boars in Switzerland.

Between October 2007 and March 2008, 153 wild boars shot in the Canton of Geneva in Switzerland were sampled. Fifty-one percent of the animals were males and 49% were females. The age of most (81%) animals varied between 6months and 2years. Prevalence of enteropathogenic Yersinia in tonsils and faeces was studied using culture and PCR methods and in tissue fluid of tonsils using an ELISA system. Prevalence of anti-Yersinia antibodies in tissue fluid was 65%. Detection rate of enteropathogenic Yersinia in tonsils of 153 wild boars by real-time PCR was 44%. The ability to replicate in macrophages is conserved between Yersinia pestis and Yersinia pseudotuberculosis.

Yersinia pestis, the agent of plague, has arisen from a less virulent pathogen, Yersinia pseudotuberculosis, by a rapid evolutionary process. Although Y. pestis displays a large number of virulence phenotypes, it is not yet clear which of these phenotypes descended from Y. pseudotuberculosis and which were acquired independently. Y. pestis is known to replicate in macrophages, but there is no consensus in the literature on whether Y. pseudotuberculosis shares this property. We investigated whether the ability to replicate in macrophages is common to Y. pestis and Y. pseudotuberculosis or is a

Yersinia pestis, the cause of plague, is a recently emerged clone of Yersinia pseudotuberculosis.

Plague, one of the most devastating diseases of human history, is caused by Yersinia pestis. In this study, we analyzed the population genetic structure of Y. pestis and the two other pathogenic Yersinia species, Y. pseudotuberculosis and Y. enterocolitica. Fragments of five housekeeping genes and a gene involved in the synthesis of lipopolysaccharide were sequenced from 36 strains representing the global diversity of Y. pestis and from 12-13 strains from each of the other species. No sequence diversity was found in any Y. pestis gene, and these alleles were identical or nearly identical to al

Yersinia pseudotuberculosis infection in a group of Macaca fascicularis.

Diarrheal disease associated with abortions and stillbirths occurred in a group of 42 Macaca fascicularis. The group was composed of wild-caught adults and their colony-born offspring housed in a half-acre cage. Thirteen adult females and 1 infant male were affected. All 14 monkeys had diarrhea and all 9 pregnant females either aborted or had a stillbirth. Yersinia pseudotuberculosis was isolated from 7 animals. In addition, Y enterocolitica was isolated from 1 monkey and Shigella flexneri type IV was isolated from 5 monkeys. One adult female had a mixed infection of both Y pseudotuberculosis

An outbreak of infection by Yersinia pseudotuberculosis in nonhuman primates.

Between October 1970 and June 1971, at the National Center for Primate Biology, Yersinia pseudotuberculosis of serotypes I-B and III was isolated from 9 monkeys (one during life and 8 at necropsy) of the following species: Macaca cynomolgus, Macaca nemestrina, Macaca radiata and Cercocebus fulliginosus. All these animals had characteristic gastrointestinal lesions consisting of superficial erosions or ulcerations with masses of gram-negative coccobacilli and an acute inflammatory exudate. Involvement of mesenteric nodes, livers and spleens by similar lesions was common. A more granulomatous re

Yersinia pseudotuberculosis colitis presented with severe gastrointestinal bleeding.

We report an adult case of Yersinia pseudotuberculosis colitis who presented with severe gastrointestinal bleeding. A 25-year-old male had admitted with fever, vomiting, body aches, and massive lower gastrointestinal bleeding. Since diagnostic tests were unremarkable and the patient's hemodynamic condition was unstable, emergency explorative laparotomy was performed. During the operation, localized wall thickening and ulcers were seen in the cecum. Right hemicolectomy was performed. Histological examination showed non-caseation granulomas in mesenteric lymph nodes and transmural inflammation i

Enteritis in cattle due to Yersinia pseudotuberculosis infection.

A selective medium was used to isolate Yersinia sp from the intestinal tract of 222 scouring cattle in Gippsland during 1985 and 1986. Intestinal infection with Y. pseudotuberculosis, particularly of serotype III, was found to be especially prevalent in weaned calves, yearlings and young adult cattle. Clinically affected cattle had a profuse liquid diarrhoea and many were systemically ill. Haematological changes suggestive of infection were present in 38 of 49 of these cattle. At least 35 cattle died and characteristic microabscesses were demonstrated in the intestinal mucosa of 20 of 26 exami

Yersinia pseudotuberculosis infection in four free-ranging odontocetes.

Yersinia pseudotuberculosis is a zoonotic pathogen that has been recovered from a variety of companion, production, laboratory, wildlife and avian species. The prevalence of this bacterium varies geographically but is generally considered widespread within the terrestrial environment with human exposure principally by ingestion of contaminated food or water. Pseudotuberculosis, due to Y. pseudotuberculosis, is a common cause of mortality in captive exotic birds and mammals and, according to the OIE WAHIS-Wild Interface, is the 8th most frequently reported disease/infection for all wildlife wor
