Fabrication of starch/zein-based microcapsules for encapsulation and delivery of fucoxanthin.

As a drug carrier, starch-based microparticle (SMP) has attracted widespread attention. However, because SMP is commonly formed in aqueous media, it is facing the challenge of encapsulation of hydrophobic bioactive. Here, we present an effective method for encapsulating fucoxanthin (Fx), a model hydrophobic bioactive, within SMP formed by self-assembly of short-chain glucans (SCG), using zein nanoparticles as intermediate vectors. SMP exhibited higher encapsulation efficiency to chitosan-coated Fx-zein nanoparticles (∼91%) at a given concentration of 50 μg/mL compared to soybean polysaccharide

Zein-based hybrid nanocarriers for non-viral gene delivery: A review.

Gene therapy and RNA-based therapeutics are increasingly limited by the challenge of delivering fragile, anionic nucleic acids to the correct intracellular compartment. Zein, the principal maize prolamin, has attracted interest as a biodegradable, self-assembling carrier that can protect cargo and be combined with polymeric, lipidic, targeted and biomimetic components. This review critically evaluates zein-based systems for plasmid DNA, siRNA, antisense oligonucleotides and related nucleic-acid cargos. Native zein is best regarded as a protective and sustained-release matrix rather than a stan

Adaptive immune cell reactivity to polycaprolactone, alginate, chitosan, and zein nanofibers.

This study explores how nanofiber chemical composition influences human immune cell responses, a key factor in the body's defense against foreign substances. Nanofiber mats composed of polycaprolactone (PCL), alginate (ALG), chitosan (CS), and zein were incubated with both unstimulated and phytohemagglutinin-L-stimulated peripheral blood mononuclear cells (PBMCs), as well as immature and mature monocyte-derived dendritic cells (MoDCs). We evaluated viability, metabolic activity, and cytokine release of PBMCs while in MoDCs, we assessed surface marker expression, phagocytic capacity, and alloge

Analysis of zein by matrix-assisted laser desorption/ionization mass spectrometry.

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI/MS) was used to analyze the protein composition of corn prolamine (zein). Mass spectra were obtained from commercial zein and zein extracted with aqueous 2-propanol and aqueous ethanol from consumer corn meal. For the commercial zein, three major zein fractions with m/z 26.8k, 24.1k, and 23.4k were clearly seen with two minor fractions (m/z 14.5k and 20.4k) also present. As compared with the results from sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), these three fractions were identified as alpha-zeins

Extraction and solubility characteristics of zein proteins from dry-milled corn.

Zein isolation by aqueous ethanol extraction from dry-milled corn produces a mixture of zeins, covalently linked polymers (dimers, tetramers, etc.) and higher-molecular-weight aggregates, some of which were not soluble in aqueous alcohol. The insoluble particles were identified as protein aggregates which form when the extraction solution is heated, particularly under alkaline conditions. The insoluble protein aggregates were not present in zein isolated by the same method from corn gluten meal. Zeins extracted from corn gluten meal and dry-milled corn were fractionated (by differential solubi

Structural transitions and molecular interactions of zein induced by ethanol and extrusion.

Ethanol improves the zein network in gluten-free dough, but the molecular mechanisms underlying this improvement remain incompletely elucidated. This study investigated ethanol plasticization (0-50 %, v/v) on commercial zein (C-zein) and extrusion-modified zein (E-zein, 140 °C). Both self-assembled into spherical microparticles merging into membranes at 50 % ethanol. Ethanol increased the porous structure and reduced the glass transition temperature (T g) of zein, whereas extrusion showed the opposite trend. Analysis showed ethanol bonded to zein primarily through hydrophobic interactions, hyd

Field applications of zein as a precise nanoscale delivery system for methoxyfenozide.

When insecticides are applied in the environment, much of the product does not reach the target pest. Biopolymeric nanoparticles as nanocarriers have the potential to improve insecticide efficacy by improving absorption, coverage, and permeability while protecting the insecticide active ingredient from abiotic conditions and extending efficacy through controlled release. We conducted a series of experiments using a biopolymeric nanoparticle synthesized from zein, a biodegradable maize protein, to compare efficacy of a nanodelivered hydrophobic insect growth regulator (methoxyfenozide) against

Preparation and characterisation of zein/polyphenol nanofibres for nerve tissue regeneration.

Bridging strategies are required to repair peripheral nerve injuries that result in gaps >5-8 mm. Limitations such as donor-site morbidity and size mismatches with receptor sites for autografts, together with immunological problems associated with allografts and xenografts, have created an increased interest in the field of manufactured nerve guide conduits. In this study, zein, a plant protein-based polymer, was electrospun to prepare nanofibrous mats. An important challenge with zein mats is the rapid change from fibre to film under aqueous conditions. Tannic acid (TA), which is a polyphenol

Injectable Zein-Based Photothermal Composite Hydrogel Enhances Osteoporotic Tendon-Bone Healing.

Osteoporotic rotator cuff tears represent a major clinical challenge owing to impaired tendon-bone interface (TBI) healing. To address this issue, we engineered an injectable Zein-based composite hydrogel (ZDC hydrogel) using electron-beam irradiation and, incorporated niacin-loaded mesoporous carbon nanospheres to achieve photothermal regulation and niacin release. The ZDC hydrogel exhibited favorable self-assembly, mechanical properties, photothermal performance, degradability, and biosafety. In vitro, ZDC hydrogel extracts enhanced the osteogenic differentiation of MC3T3-E1 cells and the te

Curcumin-loaded zein and shellac composite nanoparticles for ulcerative colitis treatment.

This study highlights the efficacy of microfluidic technology in creating curcumin (Cur) loaded zein + shellac (Z + S) hybrid nanoparticles (NPs), presenting a promising avenue for enhancing Cur's availability in the food industry, especially in beverages, and positioning it as a potent antioxidant strategy for applications such as the treatment of enteritis. The study revealed that an increase in the proportion of shellac led to a gradual increase in the particle size of Z + S NPs, while the polydispersity index (PDI) initially decreasing and then increasing. When Cur is encapsulated, an incr

Hydrophobic corn zein-modified cellulose nanofibril (CNF) films with antioxidant properties.

Cellulose nanofibrils (CNFs) can form strong biodegradable films; however, due to their hydrophilicity, moisture can degrade their mechanical and barrier properties. Corn zein (CZ) is a hydrophobic protein that when covalently linked with CNF films through peptide bonds, may improve their hydrophobicity. CZ was covalently linked to aminophenylacetic acid and aminobenzoic acid esterified CNF films which were then assessed for evidence of modification, hydrophobicity, mechanical properties, and antioxidant activity. Upon modification, an increase in hydrophobicity and an increase in antioxidant

Protein-lipid interactions in zein films investigated by surface plasmon resonance.

Experiments on the adsorption of alpha-zein (characterized by SDS-PAGE) from aqueous ethanol and 2-propanol solutions onto hydrophilic and hydrophobic surfaces are reported. Zein adsorption onto self-assembled monolayers (SAMs) was detected by surface plasmon resonance (SPR). Gold substrates were prepared by thermal evaporation on glass slides. Gold-coated surfaces were modified by depositing SAMs of either a long-chain carboxylic acid terminated thiol [COOH(CH2)(10)SH] or a methyl-terminated alkanethiol [CH3(CH2)(7)SH]. Experimental measurements indicated that zein interacted with both hydrop

Optimization and evaluation of curcumin-loaded zein microneedles for potential treatment of melanoma.

Treatment of melanoma is challenging due to limited bioavailability of chemotherapeutic drugs, side effects, and poor patient compliance with conventional intravenous administration. We evaluated the potential of synthesizing zein microneedles (ZMNs) using curcumin (Cur) as an anticancer model compound as a non-invasive approach for the local treatment of melanoma. A QbD approach was employed to create ZMNs with optimal mechanical strength and membrane penetrability, using zein as the matrix and PVP as a plasticizer at a concentration of 50% (w/v) and 20% (w/v), respectively. ZMNs loaded with

A novel zein-based dry coating tablet design for zero-order release.

The purpose of this study was to design zero-order release of dry-coated tablets using pure zein powder, zein granule and zein blend containing two common pharmaceutical excipients such as microcrystalline cellulose (MCC) or starch in different proportions as coating material. The 5-fluorouracil (5-FU) was used as a model drug. The physical characterization and drug release behaviors of dry-coated tablets were investigated. The surface structure of the tablets was examined by a scanning electron microscopy. The correlation coefficient (R) was used as indicator of the best fitting of the zero-o

Solvent-free mechanochemical synthesis of superabsorbent polymers derived from paramylon and zein.

Bio-based superabsorbent polymers (SAPs) were successfully prepared through a sustainable solvent-free and catalyst-free mechanochemical esterification of paramylon and zein using succinic anhydride as the esterification reagent. By optimizing the reaction temperature and raw material composition, a paramylon-to-zein weight ratio of 4:6 exhibited a porous morphology and achieved a maximum water absorption capacity of approximately 60 g/g, comparable to that of a commercial bio-based SAP. The optimized SAP also exhibited favorable biodegradability and showed no observable phytotoxicity in seed

Electrospun zein nanofibres incorporating NADES-phytochemical systems as sustainable antimicrobial wound dressings.

Bioactive wound dressings with antibacterial properties are attractive for infection control. Phytochemicals, products from plant secondary metabolism, like gallic acid (GA), ferulic acid (FA), coumaric acid (CouA) and perillyl alcohol (PA) show promising antimicrobial activity, but their poor water solubility and the high concentrations required for efficacy limit their application. To overcome this, Natural Deep Eutectic Solvents (NADES) provide a sustainable and biocompatible alternative to conventional solvents. NADES were used at sub-minimum bactericidal concentrations (MBC) to enhance ph

Bioactive Zein-Nondigestible Oligosaccharide Nanofibers Enable Rapid Tissue Regeneration and Antibacterial Defense.

Chronic wounds pose a significant clinical challenge due to persistent inflammation, microbial infection, and impaired tissue regeneration. Conventional wound dressings often lack the multifunctionality needed for comprehensive wound management. In this study, we engineered a nanofibrous wound patch composed of zein (ZM) integrated with nondigestible oligosaccharides (ZM-NDOs), such as Chito-oligosaccharides (ZMC), guluronic and mannuronic-rich alginate-oligosaccharides (ZMAGM), and guluronic-rich alginate-oligosaccharides (ZMAG), to address these limitations. Fabrication of zein nanofibrous scaffold containing Scrophularia striata extract for biomedical application.

Skin wounds have the potential to rapidly become infected, with bacteria having the ability to quickly penetrate to the skin's deeper layers. Then they enter the lymph nodes and spread throughout the body; therefore, all wounds should be cleaned and have a permanent cover. Modern wound dressings with effective antibacterial and therapeutic properties are required to create a sterile environment for the acceleration of healing. The aim of this work was to prepare zein electrospun nanofibers containing Scrophularia striata extract for wound healing promotion. Electrospun nanofibers made of zein,
