[Change of isoforms' spectrum of alpha-L-fucoside secreted by affected cells in some hereditary lysosomal diseases].

In vitro it was studied the isoform spectra of the intracellular and secreted alpha-L-fucosidase from skin fibroblasts of patients with Fabry disease (glycolipidosis), Hurler and Sanfilippo D diseases (mucopolysaccharodosis, types I and III) and in the normal state was studied. It was shown that the multiple form profile of secreted alpha-L-fucosidase in patients fibroblasts was changed as compared to that in control: the pathological cells were characterized by expression of more basic isoforms of alpha-L-fucosidase. The changes were similar to those in sucrose-loaded normal cells, modelling

α-L-fucosidase from Paenibacillus thiaminolyticus: its hydrolytic and transglycosylation abilities.

In this work, focused on possible application of α-L-fucosidases from bacterial sources in the synthesis of α-L-fucosylated glycoconjugates, several nonpathogenic aerobic bacterial strains were screened for α-L-fucosidase activity. Among them Paenibacillus thiaminolyticus was confirmed as a potent producer of enzyme with the ability to cleave the chromogenic substrate p-nitrophenyl α-L-fucopyranoside. The gene encoding α-L-fucosidase was found using the genomic library of P. thiaminolyticus constructed in the cells of Escherichia coli DH5α and sequenced (EMBL database: FN869117, carbohydrate-a

Deciphering α-L-Fucosidase Activity Contribution in Human and Mouse: Tissue α-L-Fucosidase FUCA1 Meets Plasma α-L-Fucosidase FUCA2.

Fucose-containing glycoproteins and glycolipids broadly occur in humans as well as in many other species and are essential for a wide range of physiological processes, such as cell adhesion, fertilization, and tumor development. In humans, the cellular degradation of various fucosylated glycoconjugates depends on the FUCA1-encoded lysosomal tissue α-L-fucosidase (FUCA1). The crucial role of FUCA1 is reflected by the severe lysosomal storage disease fucosidosis, which causes a massive accumulation of fucosylated glycans, glycolipids, and α(1,6)-fucosylated glycoasparagines. Therefore, it is rea

Increased expression of beta-hexosaminidase alpha chain in cultured skin fibroblasts from patients with carbohydrate-deficient glycoprotein syndrome type I.

Carbohydrate-deficient glycoprotein (CDG) syndrome type I is an autosomal recessive multisystem disorder characterized by multiple serum glycoproteins with deficient oligosaccharide chains. This characteristic under-glycosylation is found in several serum glycoproteins. We studied secreted forms of lysosomal enzymes, beta-hexosaminidase and alpha-fucosidase, in serum from the patients and media of cultured fibroblasts. Both beta-hexosaminidase and alpha-fucosidase activities were increased in sera from three CDG patients. The enzyme activity staining using the fluorogenic substrate-4-methylumb

Canine alpha-L-fucosidosis: a storage disease of Springer Spaniels.

Progressive ataxia, proprioceptive deficits, dysphagia and wasting occurred in a female and a male from the same litter of Springer Spaniels after the age of 12 and 19 months. At autopsy both showed marked enlargement of cranial parts of vagus and cervical nerves and dorsal root ganglia, and there was widespread vacuolation of cells of central nervous system (CNS), some peripheral nerves and of epithelial and mesenchymal cells of many organs; these vacuoles were largely empty in histological material, and were assumed to be of lysosomal origin after electron-microscopic study. Fucosidosis: A Review of a Rare Disease.

Fucosidosis is a rare lysosomal storage disease caused by α-L-fucosidase deficiency following a mutation in the FUCA1 gene. This enzyme is responsible for breaking down fucose-containing glycoproteins, glycolipids, and oligosaccharides within the lysosome. Mutations in FUCA1 result in either reduced enzyme activity or complete loss of function, leading to the accumulation of fucose-rich substrates in lysosomes. Lysosomes become engorged with undigested substrates, which leads to secondary storage defects affecting other metabolic pathways. The central nervous system is particularly vulnerable,

A novel endo-α-(1,3)-L-fucanase of the GH107 family from Winogradskyella eximia reveals source-dependent anticoagulant fucoidan oligosaccharides.

Fucoidans are sulfated polysaccharides from brown algae whose diverse biological activities depend critically on their structural features. In particular, 2,4-di-sulfation is considered one of the key determinants of anticoagulant effects. Endo-fucanases of glycoside hydrolase family 107 (GH107) are valuable tools for the structural elucidation and controlled depolymerization of fucoidans into bioactive oligosaccharides. However, the specificity of endo-α-(1,3)-L-fucanases toward defined sulfation patterns remains insufficiently characterized. In the present work, three novel GH107 endo-fucana

Severe hypomyelination as the leading neuroradiological sign in a patient with fucosidosis.

Fucosidosis is a rare autosomal recessive lysosomal storage disease, resulting from a deficiency of alpha- L-fucosidase. We report on the clinical and MRI findings of a girl with this disorder. Developmental delay became obvious at an age between 6 and 12 months. Cranial MRI at 16 months revealed severe global hypomyelination of both supra- and infratentorial white matter but no involvement of basal ganglia or thalamus. No clinical signs typical for fucosidosis were present at this time, and psychomotor development still progressed slowly. Since the age of 2 years, progressive neurological det

[Enzymatic activity of glycoprotein catabolism in the organs of rats with hyper- and hypovitaminosis A].

Activities of glycosidases neuromanidase, alpha-L-fucosidase, beta-galactosidase and beta-N-acetylglucosaminidase were studied in rat small intestine and liver tissue under conditions of hyper- and hypovitaminosis A. Both excessive and insufficient administrations of vitamin A were accompanied by distinct alterations in activity of the enzymes studied in small intestine and (although less distinct) in liver tissue. The most significant dependence on the presence of vitamin A exhibited beta-galactosidase and especially alpha-L-fucosidase, activity of which was decreased in hypervitaminosis and

Pre-fertilization approach using α-l-fucosidase modulates zona pellucida hardening during bovine in vitro embryo production.

The polyspermy occurrence is considerably lower under in vivo compared to in vitro embryo culture conditions, suggesting that the presence of some factors in the maternal environment is responsible for this. The α-L-fucosidase (FUCA) is a natural glycosidase present in the oviductal fluid, therefore, this study aimed at investigating the effect of adding FUCA to the hardening of the zona pellucida (ZP), polyspermy control, and embryonic yield and quality of bovine blastocysts produced in vitro. In the first experiment, the effect of FUCA (0.125 U/mL) was evaluated during the entire in vitro fe

An unusual presentation of fucosidosis in a Chinese boy: a case report and literature review (childhood fucosidosis).

BACKGROUND: Fucosidosis is one of the rare autosomal recessive lysosomal storage diseases (LSDs) attributed to FUCA1 variants causing the deficiency of α-L-fucosidase in vivo. Α-L-fucosidase deficiency will cause excessive accumulation of fucosylated glycoproteins and glycolipids, which eventually leads to dysfunction in all tissue systems and presents with multiple symptoms. Fucosidosis is a rare disease which is approximately 120 cases have been reported worldwide (Wang, L. et al., J Int Med Res 48, 1-6, 2020). The number of reported cases in China is no more than 10 (Zhang, X. et al., J Int

Epididymal α-l-fucosidase and its possible role in remodelling the surface of bull spermatozoa.

The mammalian epididymis provides an appropriate environment for sperm maturation. During the epididymal transit, spermatozoa undergo biochemical and morphological changes that lead to the acquisition of the fertilizing capacity. In this study we analysed the fucosylation status of membrane glycoproteins in the spermatozoa obtained from different regions of the bull epididymis. High amounts of fucose were detected on caput spermatozoa (R.F.I. = 1010 ± 20.35), mostly located in the post-acrosome zone. A significant decrease in the fucose levels was detected toward the cauda (R.F.I. = 540.5 ± 49

Distribution, crypticity, stability, and localization of α-L-fucosidase of mouse cauda epididymal sperm.

Sperm-associated and semen-specific isoforms of α-L-fucosidase are thought to function in fertilization in numerous organisms. Here, we report the localization, distribution, crypticity, and stability of this enzyme in mouse cauda epididymal sperm and cauda fluid. Western analysis revealed that the sperm-associated α-L-fucosidase is present as two isoforms (Mr ∼49 and 56 kDa), whereas the cauda fluid α-L-fucosidase shows a single band at 50 kDa. α-L-Fucosidase activity was detected using the fluorogenic substrate 4-MU-FUC. Of the total α-L-fucosidase activity recovered in the cauda epididymal

In vitro effect of hyaluronate on lysosomal enzyme activities in the bovine ciliary body and iris.

The effect of hyaluronate on lysosomal enzyme activities in bovine ciliary body and iris was studied in vitro. Hyaluronates from both the human umbilical cord and bovine vitreous inhibited the activities of cathepsin B and acid phosphatase. Cathepsin D, beta-N-acetyl-glucosaminidase, beta-D-glucuronidase and alpha-L-fucosidase activities were not affected by the hyaluronate.

Comparison of postnatal development of several acid glycosidases in the rat forebrain and cerebellum.

Changes of activity of several glycosidases (beta-galactosidase, beta-glucuronidase, N-acetyl-beta-D-glucosaminidase, alpha-D-mannosidase and alpha-L-fucosidase) were compared in the forebrain and cerebellum during postnatal development of the rat. Detailed analysis of the data showed similarities, but also substantial differences in their development in both organs. This is interpreted as an indication of the presence of common regulatory mechanisms, as well as of other factors which differently influence development of the glycosidases studied in both CNS parts.

Bioinformatics-aided function exploration of GH29 fucosidases from human gut Parabacteroides.

Gut microbes produce α-l-fucosidases critical for utilizing human milk oligosaccharides, mucosal and dietary glycans. Although gut Parabacteroides have garnered attention for their impact on host health and disease, their CAZymes remain poorly studied. CAZome analysis of eleven gut Parabacteroides type strains revealed their capacity to degrade mucin O-glycans. Their abundance of GH29 fucosidases caught our attention, and we predicted the functional profiles of 46 GH29 fucosidases using in silico approaches. Our findings showed diverse linkages specificities and species-specific distributions,
