We examined the effect of taurine depletion on hepatic sulfur-containing amino acid metabolism and carbon tetrachloride-induced acute liver injury. Mice were supplemented with beta-alanine (3%) in drinking water for one week. beta-Alanine intake significantly reduced hepatic taurine levels, but did not influence S-adenosylmethionine, S-adenosylhomocysteine, glutathione levels or methionine adenosyltransferase activity in liver. However, hepatic cysteine levels were significantly elevated by beta-alanine administration. Hepatotoxicity caused by carbon tetrachloride (50 microl/kg, ip) in mice fe

β-Alanine supplementation has been shown to increase muscle carnosine levels and exercise performance. However, its effects on muscle recovery from resistance exercise (RE) remains unknown. The purpose of this study was to investigate the effects of β-alanine supplementation on muscle function during recovery from a single session of high-intensity RE. Twenty-four untrained young adults (22.1 ± 4.6 years old) were assigned to one of two groups (N = 12 per group): a placebo-supplement group (4.8 g/day) or an β-alanine-supplement group (4.8 g/day). The groups completed a single session of high-i

beta-N-methylamino-l-alanine (BMAA) is a non-protein amino acid implicated in the neurodegenerative disease amyotrophic lateral sclerosis/Parkinson-dementia complex (ALS/PDC) on Guam. BMAA has recently been discovered in the brains of Alzheimer's patients in Canada and is produced by various species of cyanobacteria around the world. These findings suggest the possibility that BMAA may be of concern not only for specific groups of Pacific Islanders, but for a much larger population. Previous studies have indicated that BMAA can act as an excitotoxin by acting on the NMDA receptor. We have show

BACKGROUND/OBJECTIVE: Beta-alanine supplementation increases muscle carnosine content and also improves exercise capacity and performance in young adults, with mixed findings emerging from the few studies investigating its effects on older participants. Therefore, this study aimed to systematically review the evidence regarding the effects of beta-alanine on exercise capacity, muscle strength, and functional performance of older adults.
METHODS: This systematic review was conducted following the specific methodological guidelines of the Preferred Report Items for Systematic Reviews and Meta-A

Ethanol in the presence of disulfiram (N,N,N',N'-tetraethylthiuram disulfide, an inhibitor of aldehyde dehydrogenase) inhibited liver beta-alanine-oxoglutarate aminotransferase (beta-AlaAT I) activity yet activated tyrosine aminotransferase (TAT) in weanling rats in vivo. The effect on beta-AlaAT I was followed by the inhibitory expression of beta-AlaAT I mRNA. The beta-AlaAT I activity was reduced with a pseudo-first-order profile with time, and the half-life was calculated to be 12.3 +/- 0.83 h with the rate constant (Kd) of 0.056 +/- 0.004 h-1. The synthesis of beta-AlaAT I in rat liver was

BACKGROUND: Beta-alanine (βA) is a non-essential amino acid purportedly used to enhance aerobic exercise performance. While previous research indicates the benefits of βA on time to exhaustion (TTE) and aerobic capacity (VO2peak) in adults, evidence is lacking in adolescent athletes. Thus, the purpose of this study was to determine the effects of 4 weeks of βA supplementation on aerobic performance in adolescent runners.
METHODS: Twenty-seven middle- and long-distance runners (aged 17.36 ± 2.17 years) were randomly divided into a βA or placebo (PL) group (maltodextrin). Subjects performed max

The naturally occurring, non-essential amino acid beta-N-methylamino-L-alanine (BMAA) has been recently found in high concentrations in brain tissues of patients with tauopathies such as the Amyotrophic Lateral Sclerosis-Parkinsonism-Dementia Complex (ALS/PDC) in the South Pacific island of Guam and in a small number of Caucasian, North American patients with sporadic Alzheimer's disease. BMAA is produced by cyanobacteria that are present in all conceivable aquatic and/or terrestrial ecosystems and may be accumulated in living tissues in free and protein-bound forms through the process of biom

PURPOSE: High-dose 5-fluorouracil (5-FU) containing chemotherapy occasionally causes hyperammonemia and can be lethal. However, the mechanism of 5FU-associated hyperammonemia has not been known. The aim of this study was to reveal the pharmacokinetics of 5-FU-associated hyperammonemia in a recurrent colorectal cancer patient with end-stage renal disease (ESRD).
METHODS: We experienced a case of hyperammonemia during mFOLFOX6 plus bevacizumab therapy for recurrent colorectal cancer. He was a dialyzed patient due to diabetic nephropathy and was registered to prospective blood sampling for pharm

The dynamics of the axoplasmic transport of free beta-alanine and 3-O-methyl-D-glucose were quantitatively characterized in vivo in the pike olfactory nerve following intranasal application of the tritiated substances. Both compounds were found to be taken up by the epithelium but not metabolized to a significant extent. Axoplasmic transport showed the same dynamics as rapid transport of protein and free leucine in this nerve. No synthesis or transport of carnosine was detectable. The results are in agreement with the view that rapid axoplasmic transport is capable of conveying free, low molec

This study aimed to determine the impact of caffeine (200 mg), beta-alanine (3 g), and their combination on intraocular pressure (IOP), ocular perfusion pressure (OPP), and mean arterial pressure (MAP) at rest and after resistance training. Twenty young men (age = 23.4 ± 4.5 years) took part in this placebo-controlled, triple-blind, balanced crossover study. Participants visited the lab on four different days, with the only difference of the supplement used (caffeine, beta-alanine, caffeine + beta-alanine, and placebo). IOP and blood pressure were measured at baseline after 30 min from supplem

The neurotoxin beta-N-oxalylamino-L-alanine (BOAA), found in Lathyrus sativus seeds, is thought to be the causative agent of neurolathyrism. We have investigated the neuroprotective effects of free radical scavengers on BOAA-induced toxicity following focal injection (1 microliter) of BOAA and comparing the pathological outcome with the effects of injections of alpha-amino-3-hydroxy-5-methyl-isoxazole-4-propionate (AMPA), kainate (KA) or N-methyl-D-aspartate (NMDA) into the dorsal hippocampus of male Wistar rats. Cellular damage was assessed histologically. BOAA (50 nmol) induced a highly sele

beta-Alanine-oxoglutarate aminotransferase (beta-Ala-TI) was found to be distributed mainly in liver, brain, kidney, and testis (decreasing order of enzyme activity in the rat). D-3-Aminoisobutyrate aminotransferase (beta-Ala-T II) was distributed in kidney and liver. Both beta-Ala-T I and beta-Ala-T II were localized in the mitochondrial fraction in rat kidney. beta-Ala-T I in the liver of rats fed on pyridoxine-deficient or control diets was induced by injecting with prednisolone, while beta-Ala-T II in the liver of these rats was unaffected by prednisolone injection. The activities of beta-

We present a family of human sst(4)-selective, high-affinity (IC(50) = 2-4 nM) cyclic somatostatin (SRIF) octapeptides. These peptides result from the substitution of dTrp(8) in H-c[Cys(3)-Phe(6)-Phe(7)-DTrp(8)-Lys(9)-Thr(10)-Phe(11)-Cys(14)]-OH (SRIF numbering) (ODT-8) by one of the four conformationally biased stereoisomers of beta-methyl-3-(2-naphthyl)alanine (beta-Me2Nal). Whereas H-c[Cys-Phe-Phe-DNal-Lys-Thr-Phe-Cys]-OH (ODN-8, 2) has high affinity and marginal selectivity for human sst(3) (Reubi et al., Proc. Natl. Acad. Sci. U.S.A. 2000, 97, 13973-13978), H-c[Cys-Phe-Tyr-D-threo-beta-Me

BACKGROUND: This study aimed to investigate the effects of a high-dose beta-alanine (BA) supplementation on physical performance, blood lactate concentration, and ratings of perceived exertion (RPE) in trained futsal players.
METHODS: Sixteen trained futsal players participated in a randomized, parallel, double-blind experiment, completing a two-week supplementation period with either 12 g/day of BA (n = 8; age: 19 ± 2 years; height: 1.78 ± 0.03 m; body mass: 68.5 ± 5.4 kg) or a placebo (PLA) (n = 8; age: 18 ± 1 years; height: 1.74 ± 0.08 m; body mass: 65.6 ± 6.4 kg). The present study was performed to compare renal injury induced by high acute doses of both the nonacetylated salicylate, salsalate (SSA) and the acetylated salicylate, aspirin (ASA). As a marker of renal injury, urinary excretion of N-acetyl-beta-D-glucosaminidase (NAG) and alanine aminopeptidase (AAP) were measured after oral administration of 600 mg/kg of each drug. Other tests to detect renal injury were also performed. Serum salicylic levels after SSA or ASA were determined in a different experiment. Acetaminophen (APAP) was used as a standard of renal toxicity. Both drugs increase NAG an

The purpose was to investigate the effects of a 7-day creatine (Cr) loading protocol at the end of four weeks of β-alanine supplementation (BA) on physical performance, blood lactate, cognitive performance, and resting hormonal concentrations compared to BA alone. Twenty male military personnel (age: 21.5 ± 1.5 yrs; height: 1.78 ± 0.05 m; body mass: 78.5 ± 7.0 kg; BMI: 23.7 ± 1.64 kg/m2) were recruited and randomized into two groups: BA + Cr or BA + placebo (PL). Participants in each group (n = 10 per group) were supplemented with 6.4 g/day of BA for 28 days. After the third week, the BA + Cr

The excitotoxic amino acid domoate causes anterograde amnesia and memory deficits while the excitotoxin L-beta-oxalyl-amino-alanine (L-BOAA) is considered the causative agent of the motoneurone disorder, neurolathyrism. Employing quantitative autoradiography we investigated the potency of domoate and L-BOAA to inhibit [3H]kainate binding in human hippocampus. Domoate inhibited binding of [3H]kainate with inhibition constants between 5.8 +/- 2.8 nM (deep layers of gyrus parahippocampalis) and 200.9 +/- 247.8 nM (CA1 region of hippocampus). It was about a thousandfold more potent than L-BOAA wit

The reaction of D-amino acid oxidase [EC 1.4.3.3] (DAO) from porcine kidney with beta-cyano-D-alanine (D-BCNA) was studied. DAO was found to catalyze elimination of the cyano group as well as oxidation of D-BCNA. During the course of the reaction in the presence of excess oxygen, an intermediate was observed which exhibited a characteristic absorption spectrum with a broad charge transfer band in the longer wavelength region. The CD spectrum of this intermediate resembles that of DAO-anthranilate complex. The rate of oxygen consumption in the aerobic reaction decreased with time, suggesting pr

BACKGROUND: Beta-alanine (BA) supplementation increases muscle carnosine, an abundant endogenous antioxidant and pH buffer in skeletal muscle. Carnosine loading promotes exercise capacity in healthy older adults. As patients with chronic obstructive pulmonary disease (COPD) suffer from elevated exercise-induced muscle oxidative/carbonyl stress and acidosis, and from reduced muscle carnosine stores, it was investigated whether BA supplementation augments muscle carnosine and induces beneficial changes in exercise capacity, quadriceps function, and muscle oxidative/carbonyl stress in patients wi
