Comparative Antimicrobial Activity of Hp404 Peptide and Its Analogs against Acinetobacter baumannii.

An amphipathic α-helical peptide, Hp1404, was isolated from the venomous gland of the scorpion Heterometrus petersii. Hp1404 exhibits antimicrobial activity against methicillin-resistant Staphylococcus aureus but is cytotoxic. In this study, we designed antimicrobial peptides by substituting amino acids at the 14 C-terminal residues of Hp1404 to reduce toxicity and improve antibacterial activity. The analog peptides, which had an amphipathic α-helical structure, were active against gram-positive and gram-negative bacteria, particularly multidrug-resistant Acinetobacter baumannii, and showed lo

Co-Existence of Carbapenemase-Encoding Genes in Acinetobacter baumannii from Cancer Patients.

INTRODUCTION: Acinetobacter baumannii is an opportunistic pathogen, which can acquire new resistance genes. Infections by carbapenem-resistant A. baumannii (CRAB) in cancer patients cause high mortality. METHODS: CRAB isolates from cancer patients were screened for carbapenemase-encoding genes that belong to Ambler classes (A), (B), and (D), followed by genotypic characterization by enterobacterial-repetitive-Intergenic-consensus-polymerase chain reaction (ERIC-PCR) and multilocus-sequence-typing (MLST). RESULTS: A total of 94.1% of CRAB isolates co-harbored more than one carbapenemase-encod

Repurposing Screen Identifies Unconventional Drugs With Activity Against Multidrug Resistant Acinetobacter baumannii.

Antibiotic-resistant nosocomial infections are an emerging public health issue; carbapenem-resistant gram-negative bacteria such as Acinetobacter baumannii are among the pathogens against which new therapeutic agents are desperately needed. Drug repurposing has recently emerged as an alternative approach to rapidly identifying effective drugs and drug combinations to combat drug resistant bacteria. We performed a drug repurposing screen against a highly virulent, multidrug resistant, Acinetobacter baumannii strain AB5075. This strain, isolated from a patient, is resistant to 25 first-line anti

Acinetobacter baumannii isolated from hospital-acquired infection: biofilm production and drug susceptibility.

Acinetobacter baumannii cause opportunistic nosocomial infections and is often multidrug resistant. It has ability to form biofilm. The possession of drug resistance mechanism and ability of biofilm formation seems to be the different way to enhancement of viability in stressful environment. In this study, we evaluate relation between these two factors. The biofilm formation was investigated in M63 medium with casein in microtiter plates, and the drug susceptibility was performed by disk diffusion methods. We found that 80-98% strains formed a biofilm. Strains showing sensitivity to amikacin a

Molecular detection of aminoglycoside-modifying enzyme genes in Acinetobacter baumannii clinical isolates.

Acinetobacter baumannii is a major opportunistic pathogen in healthcare settings worldwide. In Iran, there are only few reports on the prevalence of aminoglycoside resistance genes among A. baumannii isolates. The aim of this study was to investigate the existence of aminoglycoside-modifying enzyme (AME) genes from A. baumannii strains collected at a university teaching hospital in Iran. One hundred A. baumannii strains were collected between 2014 and 2015 from hospitalized patients at Loghman Hakim Hospital, Tehran, Iran. Antimicrobial susceptibility was determined by disk diffusion method ac

In Vivo Selection of Pan-Drug Resistant Acinetobacter baumannii during Antibiotic Treatment.

PURPOSE: Colistin resistance in Acinetobacter baumannii (A. baumannii) is mediated by a complete loss of lipopolysaccharide production via mutations in lpxA, lpxC, and lpxD gene or lipid A modifications via mutations in the pmrA and pmrB genes. However, the exact mechanism of therapy-induced colistin resistance in A. baumannii is not well understood. MATERIALS AND METHODS: We investigated the genotypic and phenotypic changes that underlie pan-drug resistance mechanisms by determining differences between the alterations in extensively drug-resistant (XDR) A. baumannii (AB001 and AB002) isolate

Carbapenem resistance in Acinetobacter baumannii: laboratory challenges, mechanistic insights and therapeutic strategies.

Unprecedented levels of antimicrobial resistance in bacterial isolates have prompted great concerns globally. In 2012 the WHO released a publication outlining the evolving threat of antimicrobial resistance in order to raise awareness and to stimulate coordinated international efforts. The carbapenem class of antibiotics is largely considered as an antibiotic of last-resort when treating infections. Now carbapenem resistance further limits treatment options. In this article the authors discuss carbapenem resistance in Acinetobacter baumannii, a bacterial isolate often implicated in nosocomial

The mechanism of lentinan in preventing and treating acinetobacter baumannii pneumonia via immunomodulation.

BackgroundAcinetobacter baumannii pneumonia (ABP) presents a clinical challenge due to drug resistance and immune evasion. The specific mechanism by which the natural immunomodulator lentinan (LNT) exerts anti-infective effects remains unclear. This study aimed to investigate whether LNT functions through the PINK1/Parkin-mediated mitophagy pathway.MethodsAn immunosuppressed mouse model of ABP was established, and the mice were treated with LNT alone or in combination with the mitophagy inhibitor mdivi-1. Therapeutic efficacy was evaluated through survival analysis. The bacterial load in lung

Single-cell morphological responses as predictors of ciprofloxacin resistance in Acinetobacter baumannii.

UNLABELLED: The rise of antimicrobial resistance (AMR) coincides with a declining pipeline of new antibiotics, underscoring the urgent need for rapid and accurate antibiotic susceptibility testing (AST). Single-cell AST approaches offer the potential to shorten diagnostic turnaround times; however, reliably distinguishing antibiotic-sensitive from antibiotic-resistant strains at the single-cell level remains challenging. In this study, we employed bacterial cytological profiling (BCP), a single-cell imaging technique previously used for mechanism of action identification, to quantitatively ana

The intestinal microbiota impacts nutritional immunity and resistance to Acinetobacter baumannii pneumonia.

Broad-spectrum antibiotics are frequently administered to intensive care unit patients as part of empiric care. This treatment has been associated with subsequent infections by the emerging nosocomial pathogen Acinetobacter baumannii; however, the mechanisms underlying this linkage remain unclear. Here, we observe an association between antibiotic treatment and microbiota disruption that precedes A. baumannii infection in a hospitalized patient cohort and demonstrate in a murine model that broad-spectrum antibiotic administration drives susceptibility to intranasal infection with this pathogen

Biotransformation-guided production of quinalizarin as an antibacterial agent against Acinetobacter baumannii.

The search for antibacterial agents remains a global priority, particularly against multidrug-resistant pathogens. In this study, microbial transformation of 1,8-dihydroxyanthraquinone (chrysazin) was performed to obtain derivatives with enhanced antibacterial activity. Biotransformation using Absidia corymbifera AUMC 7104 and Beauveria bassiana AUMC 5133 yielded two metabolites: 1,2,5,8-tetrahydroxyanthraquinone (1, quinalizarin) and 1,8-dihydroxy-3-methyl-anthraquinone (2, chrysophanol), respectively. Their structures were elucidated by HRESIMS, 1D/2D NMR analyses. To the best of our knowled

DcaP-Family Porins are Required for Carboxylic Acid Catabolism in Acinetobacter baumannii.

Acinetobacter baumannii is a pathogen of concern and a leading cause of multidrug-resistant healthcare-associated infections. The A. baumannii outer membrane is a barrier to antimicrobials and host defenses but must also allow essential nutrients to permeate. Here, we investigate the functional importance of the putative DcaP-family outer membrane porins, which include a proposed vaccine target in A. baumannii. All A. baumannii genomes surveyed encode multiple DcaP family porins, which we classify in four classes based on protein sequence phylogeny (DcaP1-4). DcaP proteins encoded by species i

Antibiotic-resistant Acinetobacter baumannii can be killed by a combination of bacteriophages and complement.

Infections caused by multidrug-resistant Acinetobacter baumannii are an emerging global health threat. Although phages have shown promising results in treating bacterial infections, the mechanisms of the combined effect of phages and innate immunity on clearing A. baumannii remain unclear. Here, we report a synergistic effect of the complement system and phages on clearing multidrug-resistant A. baumannii. We show that A. baumannii rapidly adapts and becomes resistant to phage or serum complement by modifying the expression of capsule and lipooligosaccharides, which can be regulated through re

The evolution of the multidrug-resistant and globally distributed ST2 Acinetobacter baumannii in China.

BACKGROUND: Emergence and global expansion of the multidrug-resistant (MDR) clones of Acinetobacter baumannii which often cause difficult-to-treat infections, have been increasingly reported in nosocomial and community-acquired infections. However, as the most widely prevalent sequence type of MDR A. baumannii, the distribution and evolution of ST2 A. baumannii remain obscure. In this study, a multicenter epidemiological survey was conducted using A. baumannii isolates obtained from 27 provinces and municipalities across China between 1999 and 2022. RESULTS: It was observed that ST2 was the mo

Acinetobacter baumannii does not affect Candida albicans Susceptibility to Fluconazole in vitro.

Candida albicans is the most common human fungal pathogen and is difficult to treat due to the scarcity of and increasing resistance to fungicidal drugs. C. albicans is commonly isolated with bacterial pathogens such as Pseudomonas aeruginosa and Acinetobacter baumannii that also cause multidrug-resistant hospital-acquired infections. We have recently described how the presence of P. aeruginosa makes the antifungal drug fluconazole (FLC) more effective and switches it from fungistatic to fungicidal against C. albicans , a process sensitive to iron chelation. Although different P. aeruginosa is
