To evaluate passenger health risks associated with inhalation exposure to carbonyl compounds mainly emitted from decoration materials of vehicles, we tested the carbonyl concentrations in interior air of 20 family cars, 6 metro lines, and 5 buses in the city of Nanjing. To assess non-carcinogenic health risks, we compared the data to the health guidelines of China, US Environmental Protection Agency (EPA), and Office of Environmental Health Hazard Assessment (OEHHA), respectively. To assess carcinogenic risks, we followed a standard approach proposed by the OEHHA to calculate lifetime cancer r

The pathogenesis of idiopathic cardiomyopathies is likely highly complex and remains elusive. Environmental toxins have been hypothesized to possibly cause a subset of cardiomyopathies. Epidemiological, preclinical, and small clinical studies have investigated the role of numerous elements and compounds in the pathogenesis of these myocardial disorders. In this review, we present the evidence implicating elements and environmental compounds in myocardial toxicity, including antimony, cobalt, mercury, aluminum, copper, and acrolein. We discuss their sources, toxic effects, and epidemiology, as

In this work, we have performed a quantum chemical investigation for the selective oxidation of propane toward acrylic acid on the M1 phase of a mixed metal oxide (MMO) catalyst, consisting of Mo-Te-Nb-O. The M1 phase of the catalyst has a complex surface structure, which involves different arrangements of metal sites with variable oxidation states. This complexity makes it inherently difficult to understand its activity and selectivity in catalytic reactions. We have used a multilayer cluster model of the main catalytically active site of M1 and a hybrid DFT methodology to establish the minim

Aldehyde dehydrogenase 2 (ALDH2) is a mitochondrial enzyme that detoxifies multiple aldehyde species in the body. The ALDH2∗2 allele (E487K) is one of the most common gene polymorphisms in humans, resulting in dysfunction of its enzyme activity. This study investigated in vivo mechanism of acute liver injury caused by exposure to allyl alcohol (AA) using Aldh2∗2 knock-in (KI) mice with the same amino acid replacement as human ALDH2∗2. A rapid burst of plasma acrolein as an active metabolite of AA, as well as plasma endogenous reactive aldehydes malondialdehyde and formaldehyde, was observed at

Besides aliphatic gasoline, ethanol-blended gasoline intended for use in small utility engines was recently introduced on the Swedish market. For small utility engines, little data is available showing the effects of these fuels on exhaust emissions, especially concerning aldehydes and ketones (carbonyls). The objective of the present investigation was to study carbonyl emissions and regulated emissions from a two-stroke chain saw engine using ethanol, gasoline, and ethanol-blended gasoline as fuel (0%, 15%, 50%, 85%, and 100% ethanol). The effects of the ethanol-blending level and mechanical

Despite concerns regarding the potential health risks associated with EC use, their design, puff count, and e-liquid formulations have continued to evolve. Aldehydes are a key class of thermal degradation byproducts generated during the vaporization of EC fluids. This study tested the hypothesis that aldehyde concentrations would be elevated in used ECs with high puff numbers. Devices from multiple brands were analyzed to assess the impact of extended use on aldehyde formation. E-fluids were extracted from field-collected and user-donated devices and analyzed using gas chromatography/mass spec

Reactive aldehydes-formaldehyde (FA), malondialdehyde (MDA), 4-hydroxynonenal (4-HNE), and acrolein-occupy a central role in epigenetic regulation, lipid peroxidation, ferroptosis, and cardiovascular disease, yet their transient nature and low intracellular concentrations have long made them difficult to quantify in living systems. Over the past decade, reaction-based small-molecule fluorescent probes have emerged as the principal tool for addressing this challenge, and this review provides a systematic account of that progress. We compare the six main conjugation chemistries that underlie cur

BACKGROUND: A growing literature has reported associations between traffic-related air pollution and breast cancer, however there are fewer investigations into specific ambient agents and any putative risk of breast cancer development, particularly studies occurring in populations residing in higher pollution areas such as Los Angeles.
OBJECTIVES: To estimate breast cancer risks related to ambient air toxics exposure at residential addresses.
METHODS: We examined the relationships between ambient air toxics and breast cancer risk in the Multiethnic Cohort among 48,665 California female parti

An electronic cigarette (e-cigarette) is a product used to smoke aerosol by heating a solution of "e-liquid" that consists of propylene glycol (PG) and glycerol (GLY) containing nicotine and flavors. In this study, thermal decomposition products generated from three brands of e-cigarettes were determined at various electric power levels. When using neat PG or GLY instead of e-liquid, propylene oxide was detected only in the gas phase from PG and not detected from GLY. In contrast, glycidol was detected only from GLY and not from PG. Almost all of the glyoxal and acrolein was detected from GLY,

Cigarette smoking is associated with chronic obstructive pulmonary disease and chronic bronchitis. Acquired ion transport abnormalities, including cystic fibrosis transmembrane conductance regulator (CFTR) dysfunction, caused by cigarette smoking have been proposed as potential mechanisms for mucus obstruction in chronic bronchitis. Although e-cigarette use is popular and perceived to be safe, whether it harms the airways via mechanisms altering ion transport remains unclear. In the present study, we sought to determine if e-cigarette vapor, like cigarette smoke, has the potential to induce ac

Epidemiologic studies can measure exposure to volatile organic compounds (VOCs) using environmental samples, biomarkers, questionnaires, or observations. These different exposure assessment approaches each have advantages and disadvantages; thus, evaluating relationships is an important consideration. In the National Children's Vanguard Study from 2009 to 2010, participants completed questionnaires and data collectors observed VOC exposure sources and collected urine samples from 488 third trimester pregnant women at in-person study visits. From urine, we simultaneously quantified 28 VOC metab

BACKGROUND: Few studies examine biomarkers of exposure to vaping and tobacco products among youth. We compared biomarkers for toxicants between youth who vape, smoke, "dual-use", or neither.
METHODS: Participants ages 16 to 19 years in Canada, England, and the United States completed surveys and self-collected urine samples between September 2019 and January 2022 (N = 364). Urine was tested for metabolites of tobacco-specific nitrosamine NNK (4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone) and six volatile organic compounds. Regression models examined differences in biomarker concentrations b

BACKGROUND: Chronic respiratory diseases (CRDs) are the third leading cause of death worldwide. Data of the associations between specific volatile organic compounds (VOCs), a major component of air pollution and tobacco smoke, and subsequent CRD mortality in the general population are scarce.
METHODS: In a case-cohort analysis within the population-based Golestan cohort study (n = 50045, aged 40-75 years, 58% women, enrollment: 2004-2008, northeastern Iran), we included all participants who died from CRD during follow-up through 2018 (n = 242) as cases and stratified them into 16 strata defin

Data from National Health and Nutrition Examination Survey for the years 2011-2012 were used to evaluate variability in the observed levels of 20 urinary metabolites of volatile organic compounds (VOCs) by age, gender, and race/ethnicity among children aged 6-11 years. Exposure to environmental tobacco smoke was positively associated with the levels of selected metabolites of acrylonitrile, 1,3-butadiene, cyanide, and propylene oxide in a dose-response manner. Levels of the selected metabolites of acrolein, acrylonitrile, 1,3-butadiene, styrene, toluene, and xylene decreased with increase in a

Epidemiological studies demonstrate an association between arrhythmias and air pollution. Aconitine-induced cardiac arrhythmia is widely used experimentally to examine factors that alter the risk of arrhythmogenesis. In this study, Wistar-Kyoto (WKY) and spontaneously hypertensive (SH) rats acutely exposed to synthetic residual oil fly ash (s-ROFA) particles (450 mug/m(3)) were "challenged" with aconitine to examine whether a single exposure could predispose to arrhythmogenesis. Separately, SH rats were exposed to varied particulate matter (PM) concentrations (0.45, 1.0, or 3.5 mg/m(3) s-ROFA)

Umpolung catalysis is studied by a sequence of model reactions (CPCM in THF, B3LYP/6-31G*) with different aldehydes and catalysts. We involved addition of the catalyst to the aldehyde and 1,2-H-migration to form a carbanionic d1-species, which is the crucial intermediate according to the Lapworth- and Breslow-mechanisms. Cyanide, N-methylthiazol-2-ylidene, and a glycol-based phosphite perform as umpolung catalysts, formaldehyde, acetaldehyde, benzaldehyde, and acrolein are substrates in this study. In these aldehyde substrates, alkyl-substitution disfavors but pi-conjugation favors formation o

Some classic explanations for toxicities caused by small molecule drugs implicate reactive metabolites formed during processing by hepatic enzymes. Such metabolites can be deleterious if they evade detoxication and attack electron-dense amino acids to form adducts within cell proteins. Since such damage accompanies many idiosyncratic adverse drug reactions and other toxic outcomes, reducing tendencies for reactive metabolite formation is a key objective in modern drug discovery. Some 2 decades ago our laboratory identified an alternative route to drug-mediated protein modification that differs

Urinary mercapturic acids (MAs) are often used as biomarkers for monitoring human exposures to occupational and environmental xenobiotics. In this study, we developed an integrated library-guided analysis workflow using ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry. This method includes expanded assignment criteria and a curated library of 220 MAs and addresses the shortcomings of previous untargeted approaches. We employed this workflow to profile MAs in the urine of 70 participants─40 nonsmokers and 30 smokers. We found approximately 500 MA candidates in
