KRT14 activated by transcription factor E2F1 promotes bladder cancer progression.

Bladder cancer is one of the most common cancers in the world, and its high malignancy threatens human health. Therefore, it is vital to explore new therapeutic targets for the early diagnosis and prognosis evaluation of bladder cancer. Keratin 14 (KRT14) is the oncogene of numerous cancers, but its role and mechanism in bladder cancer are not completely clear. The present study aimed to assess the molecular mechanism of KRT14 in bladder cancer. Bioinformatics analyses were performed to screen KRT14 as an oncogene in bladder cancer. Moreover, KRT14 small interfering RNA and KRT14 overexpressio

Tetracyclines increase the survival of NSCLC patients treated with EGFR TKIs: a retrospective nationwide registry study.

BACKGROUND: With the first and second-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs), clinical benefit and rash correlate together. EGFR TKI-induced rash can be alleviated with tetracyclines, but it is unknown whether the use of tetracyclines can increase the survival of non-small-cell lung cancer (NSCLC) patients treated with EGFR TKIs. METHODS: We collected all the patients (n=1271) who had reimbursement for EGFR TKIs (gefitinib, erlotinib and afatinib) in Finland 2011-2016, had purchased TKIs, and had data available at nationwide cancer registry. The s

Exploration Novel Therapeutic Targets for Periodontitis via Stress Granules Biomarkers.

BACKGROUND: Periodontitis (PD) is associated with stress granules (SGs), which are involved in cellular stress responses. Identifying biomarkers related to SGs in PD is key to grasping its pathogenesis and devising novel therapeutic approaches. METHODS: Microarray datasets GSE10334 and GSE106090 were downloaded from public databases, and experimental verification was conducted. Differentially expressed genes (DEGs) were determined via differential expression analysis and intersected with SGs-related genes (SGs-RGs). Machine learning methods and gene expression validation were used to further

Fatal Disease Progression Driven by Acquired MET Amplification After EGFR-TKI Therapy in EGFR- and RBM10-Mutant Lung Adenocarcinoma.

Although mesenchymal-epithelial transition proto-oncogene (MET) amplification is a common resistance mechanism in targeted therapy for lung cancer, rapid disease progression associated with this resistance mechanism in patients with epidermal growth factor receptor (EGFR) mutation has scarcely been reported. Herein, we report a fatal case of lung adenocarcinoma that rapidly progressed after failure of EGFR-tyrosine kinase inhibitor treatment following the emergence of MET amplification. A 62-year-old man was diagnosed with metastatic lung adenocarcinoma containing mutations in EGFR exon L858R

Case Report: Neoadjuvant chemoimmunotherapy achieving pathological complete response in two cases of stage III EGFR-mutant NSCLC with high PD-L1 expression.

Resectable locally advanced EGFR-mutant non-small cell lung cancer (NSCLC) represents a distinct therapeutic challenge. Owing to an inherently immunosuppressive tumor microenvironment-frequently characterized by absence of or low PD-L1 expression and sparse CD8+ tumor-infiltrating lymphocytes-patients with EGFR alterations have been routinely excluded from landmark perioperative immunotherapy trials or have demonstrated negligible pathological responses. Consequently, the optimal neoadjuvant strategy for this population remains undefined, and the potential efficacy of immune checkpoint inhibit

Post-marketing safety surveillance of EGFR-targeted agents and disseminated intravascular coagulation risk: a disproportionality analysis based on the FDA Adverse Event Reporting System.

INTRODUCTION: The epidermal growth factor receptor (EGFR) serves as a principal therapeutic target in oncology, with EGFR inhibitors representing essential agents in cancer treatment. Although thrombotic complications related to EGFR inhibitors have been reported in the literature, the evidence remains insufficient. Notably, EGFR inhibitor-associated disseminated intravascular coagulation (DIC) constitutes a potentially fatal condition that currently lacks comprehensive systematic investigation. AIM: To systematically assess the signal and clinical characteristics of DIC potentially associate

[Mining of ancestral lipases and enzymatic synthesis of amides].

The amide bond plays a crucial role in the synthesis of proteins, pharmaceuticals, and agrochemicals. It is particularly significant in the pharmaceutical field as a key structural unit in many drugs, such as afatinib (an anticancer drug), teriflunomide (used for multiple sclerosis treatment), acebutolol (used for hypertension and arrhythmia treatment), and paracetamol (an antipyretic and analgesic agent). However, chemical synthesis methods usually require harsh reaction conditions (e.g., high temperature, high pressure, or strongly acidic or alkaline environments) and are often accompanied b

Real-world osimertinib pretreatment experience in patients with epidermal growth factor receptor T790M mutation-positive locally advanced or metastatic non-small cell lung cancer.

Osimertinib has demonstrated efficacy in patients with epidermal growth factor receptor (EGFR) T790M-positive non-small cell lung cancer (NSCLC) in clinical trials. However, real-world data on its effectiveness remain scarce. Taiwanese patients with T790M-positive locally advanced or metastatic NSCLC and progressive disease following treatment with at least one EGFR tyrosine kinase inhibitor (TKI) were enrolled from the osimertinib early access program. Of the 419 patients (mean age, 63 years; female, 67%), 53% were heavily pretreated (≥ third-line [3L]), making osimertinib a fourth-line (4L)

Mutational Characteristics and Clinical Outcomes for Lung Adenocarcinoma With EGFR Germline Mutations.

INTRODUCTION: Germline mutations driving lung cancer have been infrequently reported in the literature, with EGFR T790M being a known germline mutation identified in 1% of NSCLCs. Typically, a somatic EGFR mutation is acquired to develop lung adenocarcinoma. Osimertinib has become a standard-of-care treatment for EGFR T790M-positive lung cancer. METHODS: We perform a retrospective analysis through the Lung Cancer Moon Shot GEMINI database at the University of Texas MD Anderson Cancer Center. Of the patients that underwent cell-free DNA analysis, germline mutations were identified by those wit

Clinical efficacy of osimertinib in EGFR-mutant non-small cell lung cancer with distant metastasis.

BACKGROUND: Osimertinib-the third-generation epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitor (TKI)-has been widely used as a first-line treatment for patients with metastatic EGFR-mutant non-small cell lung cancer (NSCLC). Osimertinib demonstrated central nervous system activity in patients with brain metastasis; however, its efficacy against other distant metastatic organs, including bone and liver, remains unclear. Therefore, we retrospectively analyzed the clinical efficacy of osimertinib in these patients in comparison to other EGFR-TKIs. METHODS: Clinical data of patien

Clinicopathologic Features and Response to Therapy of NRG1 Fusion-Driven Lung Cancers: The eNRGy1 Global Multicenter Registry.

PURPOSE: Although NRG1 fusions are oncogenic drivers across multiple tumor types including lung cancers, these are difficult to study because of their rarity. The global eNRGy1 registry was thus established to characterize NRG1 fusion-positive lung cancers in the largest and most diverse series to date. METHODS: From June 2018 to February 2020, a consortium of 22 centers from nine countries in Europe, Asia, and the United States contributed data from patients with pathologically confirmed NRG1 fusion-positive lung cancers. Profiling included DNA-based and/or RNA-based next-generation sequenci

Clinical evaluation of dacomitinib for the treatment of metastatic non-small cell lung cancer (NSCLC): current perspectives.

Systemic treatment of advanced non-small cell lung cancer (NSCLC) has undergone remarkable changes in the last decade, with the introduction of targeted therapies and immunotherapy. The identification of activating mutations in the epidermal growth factor receptor (EGFR) gene (deletions in exon 19 [Del19] and point mutation L858R in exon 21) has been the first important step toward molecularly guided precision therapy in lung cancer. Several randomized trials comparing EGFR tyrosine kinase inhibitors (TKIs) (gefitinib, erlotinib, and afatinib) to standard chemotherapy in first-line treatment o

Construction of a survival prognosis model for epithelial-mesenchymal transition-related genes in gastric cancer.

BACKGROUND: Gastric cancer (GC) is a prevalent malignancy with high mortality rate. The process of Epithelial-mesenchymal transition (EMT) significantly contributes to its metastasis and resistance to therapy. This research is designed to develop a survival prediction model for EMT-related genes in GC. METHODS: This study used GC data from public databases and screened core module genes via weighted gene co-expression network analysis (WGCNA). Subsequently, we combined univariate Cox regression and least absolute shrinkage and selection operator (LASSO) regression to identify key feature gene
