Synthesis mechanism of antifungal active protein in the fermentation of agar waste liquid by Streptomyces rubiginosoheloolus.

Phytopathogenic fungi represent a serious threat to global crop production, food security, and public health. Agar waste liquid as a biomass-rich byproduct, shows great potential for bioconversion into value-added antifungal compounds through microbial fermentation processes. This study demonstrated that fermentation of agar waste liquid by Streptomyces rubiginosoheloolus significantly enhanced antifungal activity, with the 50 % waste liquid group exhibited inhibition rates of 59.7 % and 57.1 % against Alternaria alternata and Alternaria spp. (MICs of 0.98 and 3.90 μg/mL), respectively. Metabo

Sensitive and selective alternariol analysis by a newly developed monoclonal antibody-based immunoassay.

Alternariol is a toxic secondary metabolite produced by Alternaria fungi. Nowadays, this mycotoxin can be found in many products of plant origin at concerning concentrations. The aim of the present study was to develop a highly sensitive and selective immunochemical method for the analysis of alternariol. To this end, hapten synthesis was carried out from the scratch and specific high-affinity monoclonal antibodies to alternariol were generated by using, for the first time, alternariol bioconjugates with unambiguous linker tethering sites. A novel indirect enzyme-linked immunosorbent assay tha

A cysteine-rich antimicrobial peptide from Pinus monticola (PmAMP1) confers resistance to multiple fungal pathogens in canola (Brassica napus).

Canola (Brassica napus), an agriculturally important oilseed crop, can be significantly affected by diseases such as sclerotinia stem rot, blackleg, and alternaria black spot resulting in significant loss of crop productivity and quality. Cysteine-rich antimicrobial peptides isolated from plants have emerged as a potential resource for protection of plants against phytopathogens. Here we report the significance of an antimicrobial peptide, PmAMP1, isolated from western white pine (Pinus monticola), in providing canola with resistance against multiple phytopathogenic fungi. The cDNA encoding Pm

Nutritional Provision of Iron Complexes by the Major Allergen Alt a 1 to Human Immune Cells Decreases Its Presentation.

Alternaria alternata is a common fungus strongly related with severe allergic asthma, with 80% of affected individuals being sensitized solely to its major allergen Alt a 1. Here, we assessed the function of Alt a 1 as an innate defense protein binding to micronutrients, such as iron-quercetin complexes (FeQ2), and its impact on antigen presentation in vitro. Binding of Alt a 1 to FeQ2 was determined in docking calculations. Recombinant Alt a 1 was generated, and binding ability, as well as secondary and quaternary structure, assessed by UV-VIS, CD, and DLS spectroscopy. Proteolytic functions

Analysis of new respiratory allergies in patients monosensitized to airborne allergens in the area north of Milan.

BACKGROUND: A recent study carried out in North of Milan, an area that was recently colonized both by birch and ragweed pollen, showed that subjects developing allergies to these "new" allergens were much older than those becoming allergic to "traditional" airborne allergens, which suggests that predisposition to develop respiratory allergies is probably allergen-specific, persists throughout life, and does not occur until the patient encounters the appropriate allergen for a sufficiently long period. OBJECTIVE: The present study aimed to test this hypothesis by following-up a large cohort of

Biological control of a novel strain Bacillus velezensis BFWR11 against tomato early blight.

As a globally important vegetable crop, tomato suffers significant yield losses due to early blight, a fungal disease caused by Alternaria solani. This study investigated the antifungal activity and plant growth-promoting effects of a novel strain, Bacillus velezensis BFWR11. Genomic analysis identified gene clusters responsible for synthesizing metabolites, including lipopeptides and cell wall-degrading enzymes. Among these were clusters for ribosomally synthesized and post-translationally modified peptides (RiPPs), a class known for their potent antibacterial properties. Additionally, some k

Antibacterial and antioxidant activities of endophytic fungi and nettle (Urtica dioica L.) leaves as their host.

Nettle (Urtica dioica L), as a plant rich in biologically active compounds, is one of the most important plants used in herbal medicine. Studies have shown that this plant has antioxidant, antiplatelet, hypoglycemic and hypocholesterolemia effects. In this study, we characterized three Alternaria endophytic fungi isolated from their host U. dioica. We hypothesized that these endophytic fungi can produce new bioactive metabolites, which may possess the bioactive property with potential application in the medical and pharmaceutical industries. The antibacterial activity was evaluated against ref

Cyclic-di-GMP Induces STING-Dependent ILC2 to ILC1 Shift During Innate Type 2 Lung Inflammation.

Type 2 inflammation is found in most forms of asthma, which may co-exist with recurrent viral infections, bacterial colonization, and host cell death. These processes drive the accumulation of intracellular cyclic-di-nucleotides such as cyclic-di-GMP (CDG). Group 2 innate lymphoid cells (ILC2s) are critical drivers of type 2 lung inflammation during fungal allergen exposure in mice; however, it is unclear how CDG regulates lung ILC responses during lung inflammation. Here, we show that intranasal CDG induced early airway type 1 interferon (IFN) production and dramatically suppressed CD127+ST2+

Biocontrol of Fungal Pathogens of Winter Oilseed Rape (Brassica napus L.) Using Plant Extracts (Allium sativum, Helianthus tuberosus) and Bacterial Fermentation Supernatants (Paenibacillus, Enterobacter).

Winter oilseed rape (Brassica napus L.) production is constrained by a complex of fungal pathogens, including Alternaria alternata, A. brassicicola, Botrytis cinerea, Phoma lingam and Sclerotinia sclerotiorum, which are conventionally managed with synthetic fungicides such as prothioconazole. This study evaluated the biocontrol and biostimulant potential of two plant extracts (garlic, Allium sativum; Jerusalem artichoke, Helianthus tuberosus) and two bacterial fermentation supernatants (Enterobacter sp. and Paenibacillus sp.) against these pathogens under in vitro, greenhouse and field conditi

A closer look at the mycobiome in Alzheimer's disease: Fungal species, pathogenesis and transmission.

Candida spp., Malassezia spp., Cladosporium spp. and Alternaria spp. are among the most common fungi detected in the brain of patients with Alzheimer's disease (AD). These fungi are opportunistic organisms, where they often cause infection among immunocompromised patients. Coincidentally, these fungi can reach the brain and cause fungal meningitis. In general, they enter the brain via systemic infection due to disrupted epithelial barrier from skin and gut colonization. Once it reaches the brain, Candida species has been postulated to induce fungal glial granulomas with amyloid precursor prote

Total Synthesis of Verlamelin A and (5R)-Verlamelin A with Antifungal Activity.

Verlamelin A is a naturally occurring macrocyclic depsipeptide exhibiting high and broad-spectrum antifungal activity. The first unified total synthesis of verlamelin A and its (5R)-verlamelin A (C5-epimer) is reported. This strategy involved homogeneous solution-phase synthesis of Fmoc-l-Val-O-(5S/5R)-tetradecanoic acid, solid-phase peptide synthesis of pentapeptide NH2-d-allo-Thr-d-Ala-l-Pro-l-Gln-d-Tyr-OH, on-resin coupling of these two fragments to form a linear precursor 17, and off-resin cyclization followed by global deprotection to generate the final verlamelin A and (5R)-verlamelin A
