Circadian clock and cancer.

Circadian rhythms play a critical role in regulating physiological and behavioral processes, such as the sleep-wake cycle, hormone secretion, and metabolism. These rhythms are controlled by endogenous biological clocks, which are synchronized with environmental cues, primarily the light-dark cycle. Notably, disruption or reprogramming of circadian rhythms is closely associated with cancer initiation, progression, and treatment response. At the molecular level, the dysregulation of core clock genes can disturb normal cellular rhythms, leading to aberrant cell proliferation, impaired DNA repair,

Prenatal lead exposure, telomere length in cord blood, and DNA methylation age in the PROGRESS prenatal cohort.

BACKGROUND: Lead is a ubiquitous pollutant with deleterious effects on human health and remains a major current public health concern in developing countries. This heavy metal may interfere with nucleic acids via oxidative stress or epigenetic changes that affect biological markers of aging, e.g., telomere length and DNA methylation (DNAm). Telomere shortening associates with biological age in newborns, and DNA methylation at specific CpG sites can be used to calculate "epigenetic clocks". OBJECTIVE: The aim of this study was to examine the associations of prenatal lead exposures with telomer

Distinct epigenetic ageing patterns are associated with heterogeneity in kidney function decline in type 2 diabetes.

Biological ageing is a heterogeneous process that shapes susceptibility to chronic disease. However, whether ageing patterns diverge within clinically defined type 2 diabetes (T2D) subgroups remains unclear. We investigated whether epigenetic age acceleration (EAA) differs across T2D phenotypes and whether these patterns relate to subsequent renal vulnerability. We included 607 multi-ethnic Asians with recent-onset T2D previously classified into three clinically distinct subgroups: mild obesity-related diabetes (MOD), mild age-related diabetes with insulin insufficiency (MARD-II), and severe i

Epigenetic Aging Clocks and Incident Cardiovascular Outcomes: Results From the MESA.

BACKGROUND: DNA methylation-based aging clocks capture biological aging processes and may improve cardiovascular risk prognostication. However, evidence about epigenetic aging clocks, incident outcomes, and interactions with clinical biomarkers such as coronary artery calcium (CAC) in diverse cohorts are limited. METHODS: In this retrospective cohort study, we assessed 1264 MESA (Multi-Ethnic Study of Atherosclerosis) participants (mean age, 69 years; 49% men) who provided DNA at examination 5 (2010-2012). Epigenetic clock measures included GrimAge and age acceleration (the residual of GrimAg

Lifetime trauma exposure and accelerated epigenetic aging among midlife women.

OBJECTIVE: Trauma exposure may be linked to accelerated biological aging. However, studies have largely considered childhood abuse, with limited consideration of lifetime trauma exposure, particularly for women. Furthermore, few studies have considered newer epigenetic clocks, which have enhanced links with health outcomes. Among midlife women, we investigated whether lifetime trauma exposure is associated with older epigenetic age with several generations of clocks. We explored associations between childhood maltreatment and epigenetic age and racial differences in associations between trauma

Sleep, circadian rhythms, chronobiology, and neural synchrony: Integrative mechanisms in brain function, health, and disease.

Sleep and circadian rhythms are crucial for regulating metabolism, cognitive function, and nightly brain repair. Chronobiology studies these biological timing systems by focusing on mechanisms such as neural synchrony, which underlies coordinated brain wave patterns that support transitions between sleep stages, memory encoding, and cognitive performance. Recent work in genetics, brain imaging, and computational modeling has uncovered specific pathways connecting circadian clocks to sleep regulation and neural network synchronization. For example, clock genes drive rhythmic cellular activity,

Physical Frameworks and Theories on How Autonomous Biological Oscillators Synchronize.

Rhythms and oscillations are widely observed in biological systems. However, it is unclear how such noisy systems can produce robust and consistent timing, or how individual molecules or time-keeping systems couple together to produce macroscopically observable rhythms. Here, we focus on the synchronization of oscillatory processes at different scales: from the sub-cellular to the cellular and intercellular levels. Using examples from the circadian rhythms in cyanobacteria, cell cycle in eukaryotes, the firing of neurons, and from other emerging findings on the frontier of autonomous clocks, w

Accelerated epigenetic aging and shorter DNA methylation-based telomere length in sarcopenic obesity: an exploratory pilot study.

BACKGROUND: Sarcopenic obesity (SO), defined as the coexistence of excess fat mass and low muscle mass/function, has been linked to adverse outcomes. Epigenetic alterations are central hallmarks of aging. Evaluating how obesity, sarcopenia, and SO are related to epigenetic aging biomarkers may provide insights into cellular aging and disease risk. METHODS: In this cross-sectional study, 30 older women were classified into the control, obesity, sarcopenia, and SO groups and underwent anthropometry measurements, body composition analysis, and handgrip strength. Blood DNA methylation (DNAm) biom

Semaglutide slows epigenetic aging in a randomized trial of HIV-associated lipohypertrophy.

Glucagon-like peptide-1 (GLP-1) receptor agonists have attracted interest as gerotherapeutics, yet clinical-trial evidence for their effects on biological aging is lacking. We report a post hoc exploratory epigenetic age analysis of a 32-week, randomized, double-blind, placebo-controlled phase 2b trial (NCT04019197) of semaglutide in adults with human immunodeficiency virus (HIV)-associated lipohypertrophy (semaglutide n = 45; placebo n = 39). The parent trial's primary endpoint was change in visceral adipose tissue, with secondary cardiometabolic and body-composition endpoints; epigenetic agi

Rhythms of mood: Bidirectional regulation of molecular clocks and neurotransmission in affective disorders.

Mood disorders, primarily major depressive and bipolar disorder, are characterized by significant neurochemical dysregulation and disturbances in biological rhythms. Recent evidence suggests for a complex, bidirectional interplay between the molecular circadian clock and major neurotransmitter systems, including monoamines (dopamine, serotonin, and norepinephrine), glutamate and GABA. While the suprachiasmatic nucleus acts as the master pacemaker, local clocks in limbic regions-such as the prefrontal cortex, hippocampus, ventral tegmental area and nucleus accumbens-maintain semi-autonomous osc

Associations of four biological age markers with child development: A multi-omic analysis in the European HELIX cohort.

BACKGROUND: While biological age in adults is often understood as representing general health and resilience, the conceptual interpretation of accelerated biological age in children and its relationship to development remains unclear. We aimed to clarify the relationship of accelerated biological age, assessed through two established biological age indicators, telomere length and DNA methylation age, and two novel candidate biological age indicators, to child developmental outcomes, including growth and adiposity, cognition, behavior, lung function and the onset of puberty, among European scho

Circadian disruption in colorectal cancer: Promise of chronotherapy as precision medicine.

Circadian disruption promotes colorectal cancer (CRC) at multiple levels that could address CRC-treatment challenges like late detection, tumour heterogeneity and therapeutic resistance. Therefore, chronodisruption-CRC axis gains tremendous research attention. This review illustrates well-rounded mechanistic details and therapeutic perspective of the domain. Deregulated expressions of core clock genes (BMAL1, CLOCK, CRY, PER) disrupt crucial cellular processes like cell cycle control, DNA-repair and apoptotic regulation. They are associated with key CRC signaling pathways Wingless-related inte

Effects of nocturnal light on (clock) gene expression in peripheral organs: a role for the autonomic innervation of the liver.

BACKGROUND: The biological clock, located in the hypothalamic suprachiasmatic nucleus (SCN), controls the daily rhythms in physiology and behavior. Early studies demonstrated that light exposure not only affects the phase of the SCN but also the functional activity of peripheral organs. More recently it was shown that the same light stimulus induces immediate changes in clock gene expression in the pineal and adrenal, suggesting a role of peripheral clocks in the organ-specific output. In the present study, we further investigated the immediate effect of nocturnal light exposure on clock genes

Evaluation of the Circadian Rhythm Component Cipc (Clock-Interacting Pacemaker) in Leukemogenesis: A Literature Review and Bioinformatics Approach.

Circadian rhythms (CRs) are a key biological system regulating physiological processes such as metabolism, cell growth, DNA repair, and immunity, adapting to environmental changes like the light/dark cycle. Governed by internal clocks, it modulates gene expression through feedback loops involving Clock Genes (CGs), with the cycle initiated by CLOCK-BMAL1 and NPAS2-BMAL1 heterodimers. Disruptions in circadian rhythms have been linked to diseases including metabolic disorders, neurodegeneration, and cancer. CIPC (CLOCK-interacting pacemaker) has been studied as a negative regulator of the CLOCK-

Physical activity stimulates clock neurons of the day-active rodent Arvicanthis ansorgei.

Our biological clock, located in the suprachiasmatic nucleus (SCN), controls behavioral activity rhythms by producing circadian rhythms in SCN electrical activity. Behavioral studies in humans suggest that the clock is sensitive not only to light but also to physical activity. Here, we examined the effect of physical activity on the brain's clock in the diurnal rodent, Arvicanthis ansorgei. We found that the electrical activity of SCN neurons in vitro is high during the day and low during the night. Recordings via stationary microelectrodes in freely moving Arvicanthis revealed that the SCN ba

Dysfunction of the circadian clock in the kidney tubule leads to enhanced kidney gluconeogenesis and exacerbated hyperglycemia in diabetes.

The circadian clock is a ubiquitous molecular time-keeping mechanism which synchronizes cellular, tissue, and systemic biological functions with 24-hour environmental cycles. Local circadian clocks drive cell type- and tissue-specific rhythms and their dysregulation has been implicated in pathogenesis and/or progression of a broad spectrum of diseases. However, the pathophysiological role of intrinsic circadian clocks in the kidney of diabetics remains unknown. To address this question, we induced type I diabetes with streptozotocin in mice devoid of the circadian transcriptional regulator BMA

Keeping time with nutrients: integrating circadian rhythms and mineral nutrition in plants.

Plants grow in environments structured by predictable daily cycles of light, temperature and nutrient supply. To anticipate these fluctuations, they rely on the circadian clock, an endogenous timekeeping system that coordinates metabolic and physiological processes in time. This review synthesizes current knowledge on the role of the circadian system in mineral nutrition in plants. Core clock components rhythmically regulate the expression of nutrient transporters and homeostatic pathways. In turn, nutrient status feeds back on circadian timing, revealing tight bidirectional coupling between t

Chronic mild stress-induced alterations of clock gene expression in rat prefrontal cortex: modulatory effects of prolonged lurasidone treatment.

Disruptions of biological rhythms are known to be associated with depressive disorders, suggesting that abnormalities in the molecular clock may contribute to the development of these disorders. These mechanisms have been extensively characterized in the suprachiasmatic nucleus, but little is know about the role exerted by individual clock genes in brain structures that are important for depressive disorders. Using the chronic mild stress model we found a significant reduction of BMAL1 and CLOCK protein levels in the nuclear compartment of the prefrontal cortex of CMS rats, which was parallele

Estimation of biological aging based on T-cell differentiation trajectories: emerging and future avenues.

The relationship between disease onset and chronological age varies between individuals, driving the need for a more accurate, universal biomarker of biological aging. Among the emerging alternatives, the immune system represents a universally shared, complex system that consistently shows aging-related decline across diverse individuals. Specifically, T-cell dynamics, capturing both thymic involution and lifelong antigenic exposure, provide insights into immune system aging. Although existing aging clocks, such as those based on DNA methylation (i.e., Horvath's and GrimAge), offer valuable pr

Fixing the broken clock in adrenal disorders: focus on glucocorticoids and chronotherapy.

The circadian rhythm derives from the integration of many signals that shape the expression of clock-related genes in a 24-h cycle. Biological tasks, including cell proliferation, differentiation, energy storage, and immune regulation, are preferentially confined to specific periods. A gating system, supervised by the central and peripheral clocks, coordinates the endogenous and exogenous signals and prepares for transition to activities confined to periods of light or darkness. The fluctuations of cortisol and its receptor are crucial in modulating these signals. Glucocorticoids and the auton
