Current status of genetic engineering for sophorolipids production.

Sophorolipids are a family of microbial biosurfactants that contain a hydrophilic sophorose head and a hydrophobic fatty acid tail. Sophorolipids have gained prominence within both academia and the commercial biochemical industry due to their potential as a more sustainable and functional substitute for petroleum-based surfactants in many applications, both traditional and novel. However, the sophorolipids market competitiveness and economic viability face significant challenges from the current level of production performance, production costs, as well as selectivity of sophorolipids forms an

Current scenario of emerging pollutants in farmlands and water reservoirs: Prospects and challenges.

Globally, roughly more than 400 million metric tons of plastics are produced annually. Similarly, the pharmaceuticals business is rising exponentially yearly, 5.8 %. It is expected to increase from USD 714 billion to USD 1454 billion by 2029. Beyond their intended uses, these substances are released into the environment as contaminants due to improper usage and management practices. Therefore, pharmaceuticals and microplastics (MPs) are classified as emerging pollutants (EPs), and their existence in agricultural ecosystems adversely affects soil and environmental health, ultimately impacting b

Targeted-Produced Dirhamnolipids from Pseudomonas aeruginosa Induce Antinociception in Mice.

Rhamnolipids are highly effective surface-active glycolipid biosurfactants with enormous market potential. Composed of β-d-(β-d-hydroxyalkanoyloxy)-alkanoic acids attached to mono- or dirhamnose moieties, these green amphiphilic molecules exhibit remarkable biomedical prospects, with the double rhamnose congeners being the most effective ones. Although reports on dirhamnolipid antimicrobial activities and wound healing properties are quite abundant, the antinociceptive effect on inflammatory pain has never been tested before. Here we report a targeted-producing dirhamnolipid process, which rea

Psychrophilic insights into petroleum degradation: Gene abundance dynamics.

Petroleum degradation by psychrophiles can be enhanced on the basis of omics analyses, which offer better sensitivity than traditional biochemical methods do. A metagenomic analysis focusing on gene abundance comparisons may provide new guidance to optimize soil decontamination under cold environmental conditions. The soil used in this study was sampled from Dalian, from which an indigenous consortium was isolated. The degradative soil systems, initially categorized into control (DLC) and experimental (DLD) groups, were kept at room temperature (20 ± 5 °C) for six weeks. The DLD group was subs

Microbial Diversity and Hydrocarbon-Oxidizing Bacteria in Coastal Waters and Sands Contaminated by the Fuel Oil Spill in the Black Sea.

In 2024, an accident involving two tankers in the Kerch Strait resulted in the release of approximately 2400 tons of fuel oil into the Black Sea, causing significant contamination of seawater and the coastal zone. This study presents the first microbiological and molecular-ecological assessment of prokaryotic community composition and hydrocarbon-oxidizing bacteria (HOB) in coastal seawater and sand near Anapa (Russian Federation) following the spill. The taxonomic composition of nine samples was analyzed using high-throughput sequencing of 16S rRNA genes (V3-V4 regions), identifying Bacteria

New Imidazolium Alkaloids with Broad Spectrum of Action from the Marine Bacterium Shewanella aquimarina.

The continuous outbreak of drug-resistant bacterial and viral infections imposes the need to search for new drug candidates. Natural products from marine bacteria still inspire the design of pharmaceuticals. Indeed, marine bacteria have unique metabolic flexibility to inhabit each ecological niche, thus expanding their biosynthetic ability to assemble unprecedented molecules. The One-Strain-Many-Compounds approach and tandem mass spectrometry allowed the discovery of a Shewanella aquimarina strain as a source of novel imidazolium alkaloids via molecular networking. The alkaloid mixture was sho

Biobased surfactant-like molecules from organic wastes: the effect of waste composition and composting process on surfactant properties and on the ability to solubilize Tetrachloroethene (PCE).

In this work, four surfactant-like humic acids (HAs) obtained from garden lignocellulose wastes and kitchen food wastes mixed with garden-lignocellulose wastes, both before and after composting, were tested for surfactant properties and the ability to solubilize tetrachloroethene (PCE). The waste-derived HAs showed good surfactant properties, lowering the water surface tension from 74 mN m(-1) to 45.4 +/- 4.4 mN m(-1), with a critical micelle concentration (CMC) of 1.54 +/- 1.68 g L(-1), which is lower than many synthetic ionic surfactants. CMC was affected by both waste origin and composting

Gastrointestinal Bile Salt Concentrations in Healthy Adults Under Fasted and Fed Conditions: A Systematic Review and Meta-Analysis for Mechanistic Physiologically-Based Pharmacokinetic (PBPK) Modelling.

Bile salts are biosurfactants released into the intestinal lumen which play an important role in the solubilisation of fats and certain drugs. Their concentrations vary along the gastrointestinal tract (GIT). This is significant for implementation in physiologically based pharmacokinetic (PBPK) modelling to mechanistically capture drug absorption. The aims of this meta-analysis were to collate all appropriate data on intestinal bile salt concentrations in healthy adults across all GIT segments in fasted and fed states for the purpose of PBPK modelling. Terms relating to bile composition were s

Screening a 681-membered yeast collection for the secretion of proteins with antifungal activity.

Fungal pathogens pose a threat to human health and food security. Few antifungals are available and resistance to all has been reported. Novel strategies to control plant and human pathogens as well as food spoilers are urgently required. Environmental yeasts provide a functionally diverse, yet underexploited potential for fungal control based on their natural competition via the secretion of proteins and other small molecules such as iron chelators, volatile organic compounds or biosurfactants. However, there is a lack of standardized workflows to systematically access application-relevant ye

Targeting Bacterial Biofilms on Medical Implants: Current and Emerging Approaches.

Biofilms are structured communities of microorganisms encased in a self-produced extracellular matrix, and they represent one of the most widespread forms of microbial life on Earth. Their presence poses serious challenges in both environmental and clinical settings. In natural and industrial systems, biofilms contribute to water contamination, pipeline corrosion, and biofouling. Clinically, biofilm-associated infections are responsible for approximately 80% of all microbial infections, including endocarditis, osteomyelitis, cystic fibrosis, and chronic sinusitis. A particularly critical conce

Bioadhesive Polymeric Films Containing Rhamnolipids, An Innovative Antimicrobial Topical Formulation.

Acne affects most of the world's population, causing an impact on the self-esteem of adolescents and young adults. One of the causes is the presence of the bacteria Cutibacterium acnes which are part of the natural microbiota of the skin. Topical treatments consist of anti-inflammatory and antibiotics, which could select resistant strains. Alternatives to the antibiotic are biocomposites that have antimicrobial activity like biosurfactants which are produced by bacteria. An innovative way of applying these compounds is bioadhesive polymeric films that adhere to the skin and release the active

Thioredoxin reductase from the malaria mosquito Anopheles gambiae.

Human pathogenic diseases received much attention recently due to their uncontrolled spread of antimicrobial resistance (AMR) which causes several threads every year. Effective alternate antimicrobials are urgently required to combat those disease causing infectious microbes. Halophilic actinobacteria revealed huge potentials and unexplored cultivable/non-cultivable actinobacterial species producing enormous antimicrobials have been proved in several genomics approaches. Potential gene clusters, PKS and NRPKS from Nocardia, Salinospora, Rhodococcus, and Streptomyces have wide range coding gene

Quorum quenching pathoblockers from bacteria: an alternative approach for bacterial infection management.

Antibiotic resistance in bacterial pathogens has reached a crisis stage and has led to fears of a “post-antibiotic era”. To combat this, current approach focuses on indirect attack on bacteria by disrupting their communication system, quorum sensing (QS). This system comprises small diffusible signalling molecules activating expression of myriad genes that control an array of functions, including biofilm formation, production of virulence factors and development of antibiotic resistance. Gram-positive bacteria utilise peptide derivatives of these signalling molecules, whereas Gram-negative bac

Updated component analysis method for naturally occurring sophorolipids from Starmerella bombicola.

Sophorolipids (SLs) are promising glycolipid biosurfactants as they are easily produced and functional. SLs from microorganisms are comprised of mixtures of multiple derivatives that have different structures and properties, including well-known acidic and lactonic SL (ASLs and LSLs, respectively). In this study, we established a method for analyzing all SL derivatives in the products of Starmerella bombicola, a typical SL-producing yeast. Detailed component analyses of S. bombicola products were carried out using reversed-phase high-performance liquid chromatography and mass spectrometry. Met

Bioremediation by Brevibacterium sediminis: a prospective pyrene degrading agent to eliminate environmental polycyclic aromatic hydrocarbons.

Environmental abuses and subsequent array of health hazards by petroleum products have emerged as a global concern that warrants proper remediation. Pyrene (PYR), a polycyclic aromatic hydrocarbon, is a xenobiotic by-product during crude petroleum processing. Biodegradation potential of two bacterial isolates (MK4 and MK9) of Brevibacterium sediminis from oil contaminated sites was explored. MK4 and MK9 could degrade PYR up to 23 and 59% (1000 mg.L- 1), respectively. A first-order formalism with the rate constant for MK4 and MK9 were found to be 0.022 ± 0.001 and 0.081 ± 0.005 day- 1, respecti
