Catechol-O-methyltransferase inhibition with tolcapone reduces the "wearing off" phenomenon and levodopa requirements in fluctuating parkinsonian patients.

BACKGROUND: More than 50% of patients with Parkinson's disease develop motor response fluctuations (the 'wearing off" phenomenon) after more than five years of levodopa therapy. Inhibition of catechol-O-methyltransferase by tolcapone has been shown to increase levodopa bioavailability and plasma elimination half life, thereby prolonging the efficacy of levodopa. OBJECTIVES: The primary objective was to evaluate the efficacy of tolcapone in reducing "wearing off" in levodopa treated, fluctuating parkinsonian patients. Secondary objectives included assessment of reduction in levodopa requiremen

Ascorbic acid inhibits spinal meningeal catechol-o-methyl transferase in vitro, markedly increasing epinephrine bioavailability.

BACKGROUND: The spinal menings have previously been shown to contain catechol-o-methyl transferase (COMT), the enzyme that metabolizes epinephrine to the inactive metabolite metanephrine. The authors of this study aimed to quantitate the metabolism of epinephrine traversing the spinal meninges and to determine if that metabolism could be inhibited. In addition, they tried to determine the meningeal permeability of epinephrine. METHODS: Macca nemestrina spinal meninges were mounted in a diffusion cell and epinephrine was added to the donor reservoir at time 0. Three hundred minutes later, all

Programmable DNA-mediated coordination polymer nanospheres as highly specific catechol oxidase mimics for biosensing and environmental monitoring.

We leverage adenine-rich single-stranded DNA as both a structural director and a coordination scaffold to synthesize Cu-coordination nanospheres. These nanospheres act as highly selective catechol oxidase mimics, exhibiting no peroxidase-like activity toward TMB or ABTS, thus overcoming the catalytic promiscuity inherent to conventional oxidase nanozymes.

Bioinspired Catechol-Nanocellulose/Chitosan Polymeric Hydrogel with Enhanced Wet Adhesion and Antibacterial Activity for Diabetic Wound Healing.

Diabetic skin injury is a serious clinical challenge due to impaired healing and infection. Here, we developed a sandcastle worm-inspired hydrogel featuring a phosphate-catechol-amine synergistic network based on a poly(acrylic acid-co-acrylamide) network, functionalized with dopamine-grafted cellulose nanofibers (DA-PCNF) and quaternized chitosan (QCS), to accelerate diabetic wound healing. The hydrogel exhibits strong tissue adhesion (78.82 ± 2.55 kPa) and broad-spectrum antibacterial activity (99.67% killing of Escherichia coli and 99.55% killing of Staphylococcus aureus in vitro). Sexually Dimorphic Association of Catechol-O-Methyltransferase (COMT) Functional Polymorphism with Dimensional Personality Pathology: A Preliminary Study.

Objective: Genetic and environmental factors influence the expression of personality pathology and subsequent treatment efforts. This study associates genetics with a contemporary nosology of personality pathology represented in the Alternative Model for Personality Disorders (AMPD). We hypothesized traits from Criterion B of the AMPD would differ between genotypes of the catechol-O-methyltransferase (COMT) polymorphism (rs4680/Val158Met variation), given this genetic marker's role in the metabolism of dopamine and norepinephrine, especially in the prefrontal cortex. Methods: The Personality I

Yersinia ruckeri YRB periplasmic binding protein YiuA selectively recognizes a Fe(III)-mono-catecholate siderophore.

The marine pathogen Yersinia ruckeri synthesizes the tri-catecholate siderophore ruckerbactin, Rb, (DHB-LArg-LSer)3, to acquire iron during infection. Its biosynthetic gene cluster encodes a single periplasmic binding protein, RupB, which surprisingly does not bind Fe(III)-Rb nor the Fe(III) complexes of its hydrolysis products, the di- and mono-catecholate siderophores RbDC and RbMC, with biologically relevant affinities. Instead, the periplasmic binding protein YiuA, encoded in a different region of the chromosome, binds the 1 : 2 Fe(III) complex of the mono-catecholate RbMC, Fe(III)-(RbMC)2

Biocatalytic production of a monoamine oxidase B/catechol-O-methyltransferase inhibitor from piperine by engineered P450 BM3.

The single-step biotransformation of the natural compound piperine into a known dual inhibitor of monoamine oxidase B (MAO-B) and catechol-O-methyltransferase (COMT), was achieved by cytochrome P450 BM3 wild-type and the D251G/Q307H double mutant. This compound is used for research in neurodegenerative disorders, such as Parkinson's disease, and its value in the market is ∼14,000 €/g. Currently, it is produced by chemical synthesis requiring incubation of piperine with boron tribromide (BBr3) in dichloromethane with yield of product not exceeding 55 % and using tedious and long procedure for i

A comparative study on surface-engineered nanoceria using a catechol copolymer design: colloidal stability vs. antioxidant activity.

Nanoceria (NC) are widely studied as potent nanozyme antioxidants, featuring unique multifunctional, self-regenerative, and high-throughput enzymatic functions. However, bare NC are reported to show poor colloidal stability in biological media. Despite this, the nexus between colloidal stability and antioxidant activity has rarely been assessed. Here, a library of three copolymeric stabilising agents was synthesised, each consisting of hydrophilic poly(oligo(ethylene glycol) methyl ether methacrylate) brushes (P(OEGMA)) and a novel catechol anchoring block, and used for surface engineering of

Fluorescence detection of valence speciation of Cr(III) based on the catechol oxidase mimic enzyme activity of CuSeNP nanozymes.

Copper selenide nanoparticles (CuSeNP) were synthesized using histidine, ethylenediamine, and sodium selenate as precursors by one-step microwave digestion methods. The as-prepared CuSeNPs exhibit excellent catechol oxidase mimic enzyme and catalase (CAT)-like activities. Dopamine (DA) can be oxidized to aminochrome with H2O2 by CuSeNPs, and the intermediate product aminochrome can further react with α-naphthol to yield a highly fluorescent derivative. It was confirmed that Cr(III) could adsorb on the surface of CuSeNPs and inhibit the production of semiquinone radicals in the reaction system,

Catechol-containing compounds are a broad class of protein aggregation inhibitors: Redox state is a key determinant of the inhibitory activities.

A range of neurodegenerative and related aging diseases, such as Alzheimer's disease and type 2 diabetes, are linked to toxic protein aggregation. Yet the mechanisms of protein aggregation inhibition by small molecule inhibitors remain poorly understood, in part because most protein targets of aggregation assembly are partially unfolded or intrinsically disordered, which hinders detailed structural characterization of protein-inhibitor complexes and structural-based inhibitor design. Herein we employed a parallel small molecule library-screening approach to identify inhibitors against three pr

Picomolar level electrochemical detection of hydroquinone, catechol and resorcinol simultaneously using a MoS2 nano-flower decorated graphene.

Herein, a graphene-nano-molybdenum disulphide (pGr-MoS2), synthesized from pulverized graphite and using precursors of MoS2, was investigated for the electrochemical sensing of dihydroxy benzene isomers (DHBI): hydroquinone (HQ), catechol (CA), and resorcinol (RE). Interestingly, the material could sense the three isomers simultaneously, with well-defined peaks and an adequate potential difference between each peak. The detection limits (3σ method) of HQ, CA, and RE on the glassy carbon electrode (GCE) modified with pGr-MoS2 are 10-13, 10-12, and 10-8 M (i.e., 0.1 pM, 1 pM, and 10 nM), respect

Working memory dysfunction in fibromyalgia is associated with genotypes of the catechol- O-methyltransferase gene: an event-related potential study.

Recent findings have associated different COMT genotypes with working memory capacity in patients with fibromyalgia. Although it is thought that the COMT gene may influence neural correlates (P2 and P3 ERP components) underlying working memory impairment in this chronic-pain syndrome, it has not yet been explored. Therefore, the aim of the present research was to investigate the potential effect of the COMT gene in fibromyalgia patients on ERP working memory indices (P2 and P3 components). For this purpose, 102 participants (51 patients and 51 healthy control participants) took part in the exp

Para-tertiary butyl catechol induces eryptosis in vitro via oxidative stress and hemoglobin leakage in human erythrocytes.

Exposure of human population to industrial chemicals is believed as a significant contributing factor to the outgrowth of occupational diseases especially in developing countries due to improper safety measures and sanitary conditions. Para-tertiary butylcatechol (PTBC) widely employed in petrochemical, thermofax and phototypesetting industries, induces melanocytotoxicity and contact dermatitis leading to occupational leukoderma/vitiligo. Few vitiligo patients were reported for oxidative stress-induced hemolytic anemia and thrombocytopenia, however its impact on blood components is still not c

Protective Role of Maternal P.VAL158MET Catechol-O-Methyltransferase Polymorphism against Early-Onset Preeclampsia and its Complications.

BACKGROUND: Up until now there have been contradictory data about the association between p.Val158Met catechol-O-methyltransferase (COMT) polymorphism and risk of preeclampsia (PE). The goal of this study was to assess the potential correlation between p.Val158Met COMT polymorphism and risk of early-onset PE, risk of a severe form of early-onset PE, as well as risk of small-for-gestational-age (SGA) complicating PE. METHODS: The study included 47 early-onset PE patients and 47 control cases. Forty-seven early-onset PE patients were grouped by disease severity (33 patients with a severe form a
