Contractile to extensile transitions and mechanical adaptability enabled by activity in cytoskeletal structures.

UNLABELLED: Cytoskeletal network architecture is crucial in determining emergent morphology and dynamics, particularly in processes like stress fiber nucleation, where cells respond to mechanical cues. However, a clear understanding of the connection between architecture, dynamics, and mechanical response remains lacking. In this study, we investigate how self-assembled cytoskeletal structures respond to external mechanical perturbations, focusing on filament and crosslinker mixtures in two dimensions. Using agent-based models complemented by coarse-grained thermodynamic analysis, we reveal ho

Imaging Single-Cell Ca2+ Dynamics of Brainstem Neurons and Glia in Freely Behaving Mice.

In vivo brain imaging, using a combination of genetically encoded Ca2+ indicators and gradient refractive index (GRIN) lens, is a transformative technology that has become an increasingly potent research tool over the last decade. It allows direct visualisation of the dynamic cellular activity of deep brain neurons and glia in conscious animals and avoids the effect of anaesthesia on the network. This technique provides a step change in brain imaging where fibre photometry combines the whole ensemble of cellular activity, and multiphoton microscopy is limited to imaging superficial brain struc

ActA and human zyxin harbour Arp2/3-independent actin-polymerization activity.

The actin cytoskeleton is a dynamic network that is composed of a variety of F-actin structures. To understand how these structures are produced, we tested the capacity of proteins to direct actin polymerization in a bead assay in vitro and in a mitochondrial-targeting assay in cells. We found that human zyxin and the related protein ActA of Listeria monocytogenes can generate new actin structures in a vasodilator-stimulated phosphoprotein-dependent (VASP) manner, but independently of the Arp2/3 complex. These results are consistent with the concept that there are multiple actin-polymerization

Nuclear ataxias.

Historically basic neuroscience research has made several important contributions to the cell biology of the nucleus, in particular the elucidation of nuclear structures and compartments. As research progressed towards elucidating the mechanism of neurological disease at the cellular and molecular levels, it is now providing insight into the importance and basis of coordination of nuclear pathways within the nucleus and with other cellular compartments. Ataxias, lethal neurodegenerative diseases that are distinguished by a progressive loss of motor coordination, stem from disruption of nuclear

Tailored surface topographical scaffolds: a breakthrough in osteoarthritic cartilage and subchondral bone defect repair.

Osteoarthritis, a common degenerative joint disorder, has consistently increased in incidence in recent years. Bioactive scaffolds with specific surface topographies have demonstrated significant therapeutic potential for addressing the complex structures of articular cartilage and subchondral bone. Key signaling pathways, notably Wnt/β-catenin and NF-κB, are critical mediators in tissue repair processes. Developing osteochondral tissue engineering requires a thorough evaluation of parameters such as biocompatibility, biodegradability, and mechanical properties. Advanced manufacturing technolo

Appearance of membranocystic lesion (Nasu)-like changes in Behçet's syndrome. An electron microscopic study of erythema nodosum-like lesions.

Membranocystic lesion (Nasu)-like changes were seen in the subcutaneous fat tissues of erythema nodosum-like lesions in three of 18 patients with Behçet's syndrome. The membranocystic structures, which were derived from degenerative fat cells with a loss of the cell membrane, were observed in the edematous connective tissues accompanied by fibrin deposition and diapedesis of erythrocytes. Electron microscopic evaluation revealed two different patterns of peculiar membranous structures in the cytoplasm of the degenerative fat cells. One was proliferating membrane structures such as well develop

Thiol Specific and Mitochondria Selective Fluorogenic Benzofurazan Sulfide for Live Cell Nonprotein Thiol Imaging and Quantification in Mitochondria.

Cellular thiols are divided into two major categories: nonprotein thiols (NPSH) and protein thiols (PSH). Thiols are unevenly distributed inside the cell and compartmentalized in subcellular structures. Most of our knowledge on functions/dysfunctions of cellular/subcellular thiols is based on the quantification of cellular/subcellular thiols through homogenization of cellular/subcellular structures followed by a thiol quantification method. We would like to report a thiol-specific mitochondria-selective fluorogenic benzofurazan sulfide {7,7'-thiobis( N-rhodamine-benzo[c][1,2,5]oxadiazole-4-sul

Cellular immune response to Ia induction by intraocular gamma-interferon.

BALB/c mice and Lewis rats were injected intraocularly with recombinant gamma-interferon (IFN-gamma). The distribution of Ia antigens and cellular infiltrates was studied over a 21-day time course. In mice, Ia antigens were induced on cells in the corneal stroma, corneal endothelium, iris, ciliary body, retinal pigment epithelium and cells in the retina. No detectable cellular infiltrate accompanied class II major histocompatibility (MHC) antigen induction in mice. In rats, cells in all of the intraocular tissues mentioned above were induced to express Ia antigens. However, in distinct contras

Scalable quantification of dynamic subcellular spatial organization in single cells across tissues.

The polarized organization of cellular constituents is vital for cell migration, fate decisions, and tissue organization and often altered in disease. However, its quantification remains challenging because cells vary in size, shape, marker expression, and change over time. As existing approaches lack throughput, discard spatial information, and cannot measure polarity dynamics, we introduce CellPolariS, a novel image analysis framework to quantify polarity in diverse cell types from different tissues and in living cells. Unlike other methods, CellPolariS uses spatial subcellular organization

Blood microcirculation in the lymph node during the primary immune response.

Microangiography performed after total blood replacement with contrast material provided complete visualization of the vascular structures of the lymph node. Starting of the 2nd day, there is capillary redistribution throughout the cortex of the lymph node. The previously rather avascular nodules dissolve, and the cortical lymphoid tissue becomes uniformly vascular. Beginning on the 2nd day and reaching its peak on the 5th day, there is a significant increase in diameter and density of the subcapsular and medullary cord capillaries. 15 days after the antigenic stimulus, the appearance of the m

Calmodulin-related CML24 interacts with ATG4b and affects autophagy progression in Arabidopsis.

Plants encounter environmental stress challenges that are distinct from those of other eukaryotes because of their relative immobility. Therefore, plants may have evolved distinct regulatory mechanisms for conserved cellular functions. Plants, like other eukaryotes, share aspects of both calcium- and calmodulin-based cellular signaling and the autophagic process of cellular renewal. Here, we report a novel function for an Arabidopsis calmodulin-related protein, CML24, and insight into ATG4-regulated autophagy. CML24 interacts with ATG4b in yeast two-hybrid, in vitro pull-down and transient tob

Polymorphic assembly of virus-capsid proteins around DNA and the cellular uptake of the resulting particles.

Virus-like particles (VLPs), i.e. molecular assemblies that resemble the geometry and organization of viruses, are promising platforms for therapeutics and imaging. Understanding the assembly and cellular uptake pathways of VLPs can contribute to the development of new antiviral drugs and new virus-based materials for the delivery of drugs or nucleic acid-based therapies. Here we report the assembly of capsid proteins of the cowpea chlorotic mottle virus (CCMV) around DNA into defined structures at neutral pH. Depending on the type of DNA used, we are able to create spherical structures of var

Ecological drivers of Promethearchaeati's specialization for deep subsurface environments.

Promethearchaeati, a recently discovered kingdom of archaea, shares striking similarities with eukaryotes, including genes for cellular complexity previously thought unique to eukaryotes. Despite these features, Promethearchaeati thrive almost exclusively in stable subsurface environments, unlike their eukaryotic relatives that thrive on Earth's surface. Here we explore why these microbes remain confined to the subsurface, examining their slow growth, fragile cellular structures, and dependence on microbial partners. We discuss how their eukaryotic-like traits might function in subsurface ecos

Heterocyclic periphery in the design of carbonic anhydrase inhibitors: 1,2,4-Oxadiazol-5-yl benzenesulfonamides as potent and selective inhibitors of cytosolic hCA II and membrane-bound hCA IX isoforms.

A series of novel aromatic primary sulfonamides decorated with diversely substituted 1,2,4-oxadiazole periphery groups has been prepared using a parallel chemistry approach. The compounds displayed a potent inhibition of cytosolic hCA II and membrane-bound hCA IX isoforms. Due to a different cellular localization of the two target enzymes, the compounds can be viewed as selective inhibition tools for either isoform, depending on the cellular permeability profile. The SAR findings revealed in this study has been well rationalized by docking simulation of the key compounds against the crystal st

Channel-linked multi-emitter fitting enables fast and accurate multi-channel single-molecule localization microscopy.

Single-molecule localization microscopy (SMLM) provides nanometer-scale spatial resolution for visualizing cellular structures. Multi-channel SMLM further extends its capabilities by incorporating single-molecule information from multiple channels, enabling more information-rich imaging modalities. However, both SMLM and its multi-channel variants rely on sparse activation, which limits imaging speed and labeling density. Here, we introduce climeLoc, a channel-linked multi-emitter fitter designed to correlate and localize emitters at high density across multiple channels. By integrating pre-ca

RNA Cis-Elements Involved in Animal Virus Stop Codon Readthrough: Stop Codon Context and Downstream RNA Structures.

Stop codon readthrough is a noncanonical translation strategy employed by certain RNA viruses, in which a viral termination codon is either decoded by host near-cognate tRNAs or canonically recognized by the class I release factor (RF, eRF1 in eukaryotes). Ribosomal A-site competition between near-cognate tRNAs and eRF can shift decoding toward near-cognate tRNAs, thereby promoting non-canonical decoding events by transiently pausing termination and favoring readthrough. This review focuses on two viral cis-elements that modulate readthrough across four viral genera in which this decoding even
