Thymic epithelial tumors (TETs) sit at a unique intersection of cancer, central tolerance, and autoimmunity. These tumors arise in the thymus, an immune-rich organ responsible for T-cell education, and despite their low tumor mutational burden (TMB) and microsatellite stability, they display paradoxical behavior under immune checkpoint inhibition. This review synthesizes data on thymic biology, paraneoplastic autoimmunity, molecular profiling, and clinical trials of immune checkpoint inhibitors (ICIs) in thymoma and thymic carcinoma. We describe how disruption of autoimmune regulator (AIRE)/Fe

Malignant tumors remain a global public health challenge with high morbidity and mortality. Conventional therapies are limited by severe adverse effects and drug resistance. Chlorogenic acid (CGA), a widely distributed phenolic compound, has emerged as a promising antitumor agent due to its excellent biocompatibility and multi-targeted bioactivities, including antioxidant, anti-inflammatory, immunomodulatory, and pro-apoptotic. This review systematically summarizes the molecular mechanisms underlying the antitumor activity of CGA, focusing on three core aspects: (1) regulation of key signaling

PURPOSE: Cell-cycle-checkpoint kinase 2 (CHEK2) is an important moderate-penetrance breast cancer predisposition gene; however, recurrent CHEK2 mutations found in Caucasian women are very rare in Chinese population. We investigated the mutation spectrum and clinical relevance of CHEK2 germline mutations in Chinese breast cancer patients.
METHODS: The entire coding regions and splicing sites of CHEK2 were screened in 7657 Chinese BRCA1/2-negative breast cancer patients, using 62-gene panel-based sequencing.
RESULTS: Out of 7657 BRCA1/2-negative breast cancer patients, 26 (0.34%) carried CHEK2

The CHK2 gene encodes a protein kinase that is important for the regulation of cell cycle arrest after DNA damage. CHK2 acts downstream of ataxia teleangiecstasia mutated (ATM), modulates the function of p53 and may help mediate cell cycle arrest at G2/M by phosphorylation of Cdc25C. Recently, the human homolog of the checkpoint kinase Cds1 (CHK2) has been suggested to be a tumor suppressor gene. Heterozygous germline mutations have been reported in Li-Fraumeni syndrome (LFS), a highly penetrant familial cancer phenotype, and in sporadic colon cancer. LFS is associated with the development of

Hand and foot skin reaction (HFSR), usually associated with tyrosine kinase inhibitors, is characterized by painful erythema and hyperkeratosis at palmoplantar pressure points. HFSR differs from the more widely recognized chemotherapy-induced hand-foot syndrome (HFS) in terms of both clinical and histopathological features. Here we present the case of a 79-year-old man with advanced lung adenocarcinoma who developed grade 3 HFSR after receiving pembrolizumab, a PD-1 inhibitor. The patient initially presented with erythema, desquamation, and hyperkeratotic plaques on the palms and soles, follow

Epigenetic regulation is essential for development and adaptation across eukaryotes. However, a comprehensive overview of the molecular framework of chromatin-mediated regulation, particularly in non-model organisms, is lacking. Here, we present a systematic proteomic characterization of the chromatin domains in P. falciparum, an ancient human pathogen with a unique genome composition and epigenetic blueprint. We adapted and employed three proximity-labeling approaches to provide a high-confidence and comprehensive proteome of heterochromatin, euchromatin, and (peri)centromeric chromatin compr

Tuberculosis (TB) is a critical global health concern, with about one-quarter of the world's population having an immune response against Mycobacterium tuberculosis (MTB). These individuals are at risk of developing TB disease when their immune function is reduced. Given the increasing use of immunosuppressive agents, including biological and non-biological immunomodulators, the risk of TB infection (TBI) reactivation has significantly increased, highlighting the importance of screening and preventive treatment. The aim of this review is to summarize the current evidence on TB infection (TBI)

HER2-directed therapies have transformed the treatment landscape of breast and gastric cancers, but targeting HER2 alterations in non-small cell lung cancer (NSCLC) has proven more challenging. HER2 mutations, found in 2-4% of NSCLC cases, define a unique molecular subset characterized by poor responses to conventional chemotherapy and the frequent development of brain metastases. Recent advances in novel therapies, particularly antibody-drug conjugates and selective tyrosine kinase inhibitors, have renewed interest in HER2 as a therapeutic target. These therapies have different efficacy and s

A man in his mid-70s presented with haematuria and haematospermia and experienced a multiyear diagnostic delay due to initially benign impressions. A diagnosis of Gleason 8 prostate adenocarcinoma was eventually rendered, despite consistently normal prostate-specific antigen levels. A prostate-specific membrane antigen-positron emission tomography scan, negative for prostatic uptake, unexpectedly revealed a lung lesion, subsequently confirmed as metastatic adenocarcinoma. Expert pathology review at a tertiary cancer centre, using comprehensive immunohistochemistry-including paired box gene 8 (

PURPOSE: Human papillomavirus (HPV)-negative head and neck squamous cell carcinomas (HNSCC) remain challenging due to their reduced radiosensitivity. Targeting the DNA damage response (DDR) with kinase inhibitors may enhance tumor susceptibility to radiotherapy (RT). While RT is known to alter the immune phenotype of HNSCC cells, its impact on innate immune responses, particularly on natural killer (NK) cells, remains unclear. We therefore investigated how RT combined with ATM or ATR inhibition-key DDR kinases-affects NK cell-mediated killing and activation.
METHODS: The tumor cell lines HSC4

BACKGROUND: A truncating variant, 1100delC, in check point-kinase CHEK2, has been identified as a risk factor for familial and sporadic breast cancer. The prevalence in healthy non-breast cancer cases is low and varies between populations.
METHODS: We analyzed the prevalence of CHEK2 1100delC in 763 breast cancer patients with a defined family history and 760 controls from the Stockholm region. The breast cancer patients originated from; a population-based cohort (n = 452) and from a familial cancer clinic (n = 311), the detailed family history was known in both groups.
RESULTS: The variant

Cultured cells from patients inheriting the rare cancer-prone and radiotherapy-sensitive disorder ataxia-telangiectasia (A-T) exhibit anomalies in cell cycle control and protein kinase C (PKC)-mediated upregulation of p53 protein following exposure to ionizing radiation. It remains unclear, however, as to whether this irregularity in a p53-dependent signal transduction pathway controlling the G1/S checkpoint is causally linked to the most consistent molecular hallmark of A-T-namely, marked attenuation in the inhibition of replicative DNA synthesis at early times (< or = 2 h) after irradiation

MicroRNAs (miRNAs) inhibit protein function by silencing the translation of target mRNAs. However, in primary central nervous system lymphoma (PCNSL), the expression and functions of miRNAs are inadequately known. Here, we examined the expression of 847 miRNAs in 40 PCNSL patients with a microarray and investigated for the miRNA predictors associated with cancer immunity-related genes such as T helper cell type 1/2 (Th-1/Th-2) and regulatory T cell (T-reg) status, and stimulatory and inhibitory checkpoint genes, for prognosis prediction in PCNSL. The aim of this study is to find promising prog

The aim of the current study was to investigate the expression and role of microRNA‑486‑5p (miR‑486‑5p) in hypertrophic scar (HS) formation, and to examine the associated mechanisms. First, miR‑486‑5p expression was detected in HS tissues and human hypertrophic scar fibroblasts (hHSFs) by reverse transcription‑quantitative polymerase chain reaction. Target genes of miR‑486‑5p were predicted using TargetScan and verified by dual‑luciferase reporter assays. To investigate the role of miR‑486‑5p in HS formation, miR‑486‑5p was overexpressed in hHSFs through transfection with miR‑486‑5p mimics. MT

Poly(ADP-ribose) polymerases 1 and 2 (PARP1/PARP2), and poly(ADP-ribose) glycohydrolase (PARG), modulate the level of poly(ADP-ribose) (PAR), a post-translational protein modification, in response to DNA damage or replication stress. Here, we find that replication-dependent and PARP1/PARP2-mediated PARylation recruits the base excision repair (BER)/single-strand break repair (SSBR) scaffold protein XRCC1 and the associated factors DNA polymerase β (POLB), aprataxin (APTX), and DNA ligase isoform 3 (LIG3). Further, these BER/SSBR proteins promote resistance to inhibitors of PARP1/PARP2 and PARG

RATIONALE: The therapeutic value of immune checkpoint inhibitors (ICIs) in a variety of tumors has been found and recognized, and although ICIs have improved the prognosis of many patients with advanced tumors, these drugs sometimes cause immune-related adverse events (irAEs).

The 24-h circadian rhythm handles a wide variety of physiological needs. Clock genes, in coordination with other tissue-specific factors regulate various processes and often turns responsible for the pathological conditions when altered. Cancer is one such disease where the clock genes have been shown to contribute at multiple levels modulating key hallmarks of cancer. Most importantly, adding to this complication, noncoding RNAs (ncRNAs) have emerged as one of the major post-transcriptional regulators of gene expression and many recent studies have indicated about involvement of microRNAs or
