Developmental expression and function analysis of protein tyrosine phosphatase receptor type D in oligodendrocyte myelination.

Receptor protein tyrosine phosphatases (RPTPs) are extensively expressed in the central nervous system (CNS), and have distinct spatial and temporal patterns in different cell types during development. Previous studies have demonstrated possible roles for RPTPs in axon outgrowth, guidance, and synaptogenesis. In the present study, our results revealed that protein tyrosine phosphatase, receptor type D (PTPRD) was initially expressed in mature neurons in embryonic CNS, and later in oligodendroglial cells at postnatal stages when oligodendrocytes undergo active axonal myelination process. In PTP

GPR37 modulates remyelination following demyelinating injury.

GPR37 has been recognized as a negative regulator of oligodendrocyte myelination. However, its role in demyelination recovery remains unclear. To examine the function of GPR37 in remyelination, we compared wild-type and GPR37-deficient mice of either sex after focal (lysolecithin) or global (cuprizone) toxin-induced demyelination, as well as immune-mediated demyelination (experimental autoimmune encephalomyelitis, EAE). We found that the absence of GPR37 resulted in enhanced recovery in all three models. In focal demyelination, we observed that, while the initial lesioned area was similar in s

A Trem2R47H mouse model without cryptic splicing drives age- and disease-dependent tissue damage and synaptic loss in response to plaques.

BACKGROUND: The TREM2 R47H variant is one of the strongest genetic risk factors for late-onset Alzheimer's Disease (AD). Unfortunately, many current Trem2 R47H mouse models are associated with cryptic mRNA splicing of the mutant allele that produces a confounding reduction in protein product. To overcome this issue, we developed the Trem2R47H NSS (Normal Splice Site) mouse model in which the Trem2 allele is expressed at a similar level to the wild-type Trem2 allele without evidence of cryptic splicing products. METHODS: Trem2R47H NSS mice were treated with the demyelinating agent cuprizone, o

Systemic TLR2 tolerance enhances central nervous system remyelination.

BACKGROUND: Multiple sclerosis (MS) is a central nervous system (CNS) autoimmune disease characterized by both inflammatory demyelination and impaired remyelination. Studies indicate that Toll-like receptor 2 (TLR2) signaling contributes to both the inflammatory component and the defective remyelination in MS. While most MS therapeutics target adaptive immunity, we recently reported that reducing TLR2 signaling in innate immune cells by inducing TLR2 tolerance attenuates adoptively transferred experimental autoimmune encephalomyelitis. Given that previous reports suggest TLR2 signaling also in

Role of copper ions and cytochrome P450 in the vasodilator actions of the nitroxyl anion generator, Angeli's salt, on rat aorta.

Since copper ions catalyse the oxidation of nitroxyl anion to nitric oxide, we investigated whether this might explain the vasodilator actions of the nitroxyl generator, Angeli's salt, in rat aorta. Parallel studies were conducted with S-nitroso-N-acetyl-D,L-penicillamine (SNAP), since Cu ions catalyse the liberation of nitric oxide from this compound. Copper sulphate enhanced relaxation to Angeli's salt and SNAP but this resulted from reduced destruction of nitric oxide by superoxide rather than from enhanced generation of nitric oxide, since it was mimicked by superoxide dismutase and by the

Interferon regulatory factor 8/interferon consensus sequence binding protein is a critical transcription factor for the physiological phenotype of microglia.

BACKGROUND: Recent fate-mapping studies establish that microglia, the resident mononuclear phagocytes of the CNS, are distinct in origin from the bone marrow-derived myeloid lineage. Interferon regulatory factor 8 (IRF8, also known as interferon consensus sequence binding protein) plays essential roles in development and function of the bone marrow-derived myeloid lineage. However, little is known about its roles in microglia. METHODS: The CNS tissues of IRF8-deficient mice were immunohistochemically analyzed. Pure microglia isolated from wild-type and IRF8-deficient mice were studied in vitr

Identifying Lanthionine Ketimine Derivatives for Maturation and Proliferative Effects in Oligodendrocyte Progenitor Cells.

Previous studies have shown that lanthionine ketimine ethyl ester (LKE) reduces clinical scores in the experimental autoimmune encephalomyelitis (EAE) mouse model of Multiple Sclerosis, induces differentiation of oligodendrocyte progenitor cells (OPCs) in vitro, and accelerates remyelination following cuprizone induced demyelination. In a search for derivatives with greater efficacy to induce OPC maturation or proliferation, we screened a panel of 2-alkyl and 3-phosphonate substituted LK derivatives. Incubation of Oli-neu oligodendrocyte cells with 2-n-butyl- or 2-n-hexyl-LKE-phosphonate reduc

Fractalkine enhances oligodendrocyte regeneration and remyelination in a demyelination mouse model.

Demyelinating disorders of the central nervous system (CNS) occur when myelin and oligodendrocytes are damaged or lost. Remyelination and regeneration of oligodendrocytes can be achieved from endogenous oligodendrocyte precursor cells (OPCs) that reside in the adult CNS tissue. Using a cuprizone mouse model of demyelination, we show that infusion of fractalkine (CX3CL1) into the demyelinated murine brain increases de novo oligodendrocyte formation and enhances remyelination in the corpus callosum and cortical gray matter. This is achieved by increased OPC proliferation in the cortical gray mat

Aurka-Bhlhe41 axis prevents premature aging-like microglial dysfunction and promotes remyelination.

Aging accelerates central nervous system remyelination failure and neurodegeneration. Microglia promote remyelination by phagocytosing myelin debris, but this function is impaired by aging-related CD22 upregulation. However, the molecular mechanisms counteracting premature aging-related microglial dysfunction and remyelination impairment remain unclear. Here, we report that Aurka-Bhlhe41 axis prevents premature aging-like microglial dysfunction and promotes remyelination by restraining progressive CD22 upregulation. We identified that microglia-enriched Bhlhe41 was negatively autoregulated and

A comparative transcriptomic analysis of mouse demyelination models and multiple sclerosis lesions.

Demyelinating diseases, including multiple sclerosis (MS), are characterized by loss of myelin and progressive neurodegeneration. It remains unclear if demyelination mouse models, such as cuprizone (CPZ) and lysophosphatidylcholine (LPC) elicit distinct responses or are comparable to human disease. Here, we integrate new and published single-cell transcriptomic datasets from CPZ- and LPC-induced demyelination and compare them with human MS data. We find that CPZ induces a distinct, stressed oligodendrocyte (OL) state, marked by Cdkn1a and Nupr1, that resembles phenotypes in MS lesions. The mod

Plumbagin ameliorates multiple sclerosis by inducing DDX3X-mediated stress granule assembly in mice.

Multiple sclerosis (MS) is a chronic inflammatory demyelinating disease of the central nervous system (CNS) with limited therapeutic options. Stress granules (SGs), membraneless organelles formed via liquid-liquid phase separation (LLPS) in response to cellular stress, function as a cytoprotective mechanism against stress induced apoptosis and pyroptosis. SGs can counteract NLRP3 inflammasome by sequestering dead-box helicase 3 X-linked (DDX3X), thereby promoting the survival of pyroptotic cells. Here, we identified that plumbagin (PL), a natural naphthoquinone with antioxidant and anti-inflam

Slc20a2 Deficiency Increases Susceptibility to CNS Demyelination Possibly Through Th17 Cells.

Primary familial brain calcification (PFBC) is a rare inherited neurodegenerative disorder characterized by abnormal brain calcium-phosphate (Ca-Pi) deposits along microvessels or inside neuronal cells. Eight genes have been linked to PFBC, with the SLC20A2 being the earliest identified. SLC20A2 encodes PiT-2, which is crucial for Pi homeostasis in the cerebrospinal fluid (CSF). In Slc20a2 homozygous knockout (HO) mice, the neurotoxic effects resulting from pathological CSF-Pi accumulation and the unique brain transcriptome suggested that the absence of PiT-2 might lead to myelin abnormalities

Multiple sclerosis risk gene Mertk is required for microglial activation and subsequent remyelination.

In multiple sclerosis (MS) and other neurological diseases, the failure to repair demyelinated lesions contributes to axonal damage and clinical disability. Here, we provide evidence that Mertk, a gene highly expressed by microglia that alters MS risk, is required for efficient remyelination. Compared to wild-type (WT) mice, Mertk-knockout (KO) mice show impaired clearance of myelin debris and remyelination following demyelination. Using single-cell RNA sequencing, we characterize Mertk-influenced responses to cuprizone-mediated demyelination and remyelination across different cell types. Mert

Simple Ultraviolet-Visible Spectroscopy-Based Assay for Fast Evaluation of Magnetic Nanoparticle Selectivity Changes After Doping.

Simple ultraviolet-visible spectroscopy-based methodology was proposed and utilized for the initial characterization of potential changes in selectivity of doped magnetic nanoparticles. Doped and undoped iron(II,III) (Fe3O4) magnetic nanoparticles were synthesized by the coprecipitation method. The doping processes of nanoparticles were confirmed using optical emission spectrometry, while the sizes of undoped and Cu-doped nanoparticles were investigated using a high-resolution field emission scanning electron microscope. The average diameters of nanoparticles were 8.34±1.78 nm and 9.12±1.93 nm

Leonurine suppresses neuroinflammation through promoting oligodendrocyte maturation.

Focal inflammation and remyelination failure are major hallmarks of multiple sclerosis and its animal model, experimental autoimmune encephalomyelitis (EAE). In this study, we found that leonurine, a bioactive alkaloid, alleviated EAE disease severity along with reduced central nervous system inflammation and myelin damage. During the pathogenesis of EAE, leonurine dramatically suppressed the recruitment of encephalitogenic T cells into the central nervous system, whereas did not impair periphery immune responses and microglia activation. Mechanistically, leonurine protected mice against demye

Mitochondrial microprotein MOCCI controls neuroinflammation by altering glial activation states.

Metabolic regulation and its underlying mechanisms play a critical role in controlling and resolving inflammation in the brain, directly shaping glial cell activation and the central nervous system's response to injury and disease. In our screen for microproteins that modify inflammatory outcomes, we discovered MOCCI (protein product of C15orf48/AA467197) as a significant regulator of gut and lung inflammation. However, its involvement in neuroinflammation is unknown. Here, we show that MOCCI is upregulated in microglia and astrocytes in both the mouse and human brain upon inflammation, and is

TREM2 activation on microglia promotes myelin debris clearance and remyelination in a model of multiple sclerosis.

Multiple sclerosis (MS) is an inflammatory, demyelinating, and neurodegenerative disease of the central nervous system (CNS) triggered by autoimmune mechanisms. Microglia are critical for the clearance of myelin debris in areas of demyelination, a key step to allow remyelination. TREM2 is expressed by microglia and promotes microglial survival, proliferation, and phagocytic activity. Herein we demonstrate that TREM2 was highly expressed on myelin-laden phagocytes in active demyelinating lesions in the CNS of subjects with MS. In gene expression studies, macrophages from subjects with TREM2 gen

Terrestrial jumping assay for adult zebrafish as a potential tool to assess motor, affective and drug-evoked phenotypes.

Terrestrial jumping is a typical evolutionarily conserved 'survival' fish behavior. Zebrafish (Danio rerio) are widely used as a model organism in CNS research and drug discovery. Here, we explore the potential utility of the terrestrial jumping assay to characterize behavioral effects of a wide range of pharmacological and other experimental manipulations in adult zebrafish. For this, zebrafish were individually placed in a large glass tank and their jumping behavioral profile was assessed for 60 s by video-recording and subsequent off-line analyses of escape-like jumps and immobility episode

A novel small molecule remyelination therapy for multiple sclerosis.

We conducted a phenotypic screen of 20,000 central nervous system (CNS)-biased small molecules for their ability to stimulate differentiation of mouse oligodendrocyte progenitor cells (OPCs) into oligodendrocytes. We identified a lead hit (CN045) with an EC50 of 40 nM in the OPC differentiation assay and a chemical scaffold conducive to modifications. In OPC differentiation assays, CN045 demonstrated higher potency than a known OPC differentiation compound Triiodothyronine (T3). CN045 promoted myelin-like ensheathment of engineered nanofibers by mouse and human OPCs and significantly increased
