Investigation of the Roles of Non-neuronal Acetylcholine in Chronic Myeloid Leukemic Cells and their Erythroid or Megakaryocytic Differentiated Lines.

BACKGROUND: Many studies suggested that Acetylcholine (ACh) might serve as an autocrine/ paracrine growth factor in several types of tumors or tumor cell lines. High levels of Acetylcholinesterase (AChE) activity have been reported in primary brain tumors, ovarian, colon and lung tumors. OBJECTIVES: The role of cholinergic signaling needs to be clarified in in leukemia. METHOD: K562 cells were derived from a chronic myelogenous leukemia patient during blast crisis serving as pluripotent hematopoietic stem cells. K562 cells were incubated with various cholinergic agonists or antagonists to in

Effects of restraint and immobilization on electrosensory behaviors of weakly electric fish.

Weakly electric fishes have been an important model system in behavioral neuroscience for more than 40 years. These fishes use a specialized electric organ to produce an electric field that is typically below 1 volt/cm and serves in many behaviors including social communication and prey detection. Electrical behaviors are easy to study because inexpensive and widely available tools enable continuous monitoring of the electric field of individual or groups of interacting fish. Weakly electric fish have been routinely used in tightly controlled neurophysiological experiments in which the animal

[Pharmaco-physiologic correlations of iron-dextran complexes in laboratory animals].

Investigated were the Bulgarian preparation dextrofer-100 (Pharmachim) and miofer-100 (Farbwerke, Hoechst, West Germany)--iron-dextran preparations with 100 mg Fe3+/cm3 each. Use was made of 62 domestic cats, 8 guinea pigs, and 18 albino rats. tI was found that the venous introduction of iron-dextran complexes into uretane-treated cats at rates of 100 to 500 mg Fe3+/kg body mass lowered the blood pressure by 10 to 65 per cent; the depressive effect was more strongly manifested with the use of miofer-100. At 100 mg Fe3+/kg and higher doses led to alfa-adrenolytic manifestations (an inverse effe

Acetylcholine receptor in planar lipid bilayers. Characterization of the channel properties of the purified nicotinic acetylcholine receptor from Torpedo californica reconstituted in planar lipid bilayers.

The properties of the channel of the purified acetylcholine receptor (AChR) were investigated after reconstitution in planar lipid bilayers. The time course of the agonist-induced conductance exhibits a transient peak that relaxes to a steady state value. The macroscopic steady state membrane conductance increases with agonist concentration, reaching saturation at 10(-5) M for carbamylcholine (CCh). The agonist-induced membrane conductance was inhibited by d-tubocurarine (50% inhibition, IC50, at approximately 10(-6) M) and hexamethonium (IC50 approximately 10(-5) M). The single channel conduc

Neuromuscular activity of Bothrops fonsecai snake venom in vertebrate preparations.

The neuromuscular activity of venom from Bothrops fonsecai, a lancehead endemic to southeastern Brazil, was investigated. Chick biventer cervicis (CBC) and mouse phrenic nerve-diaphragm (PND) preparations were used for myographic recordings and mouse diaphragm muscle was used for membrane resting potential (RP) and miniature end-plate potential (MEPP) recordings. Creatine kinase release and muscle damage were also assessed. In CBC, venom (40, 80 and 160μg/ml) produced concentration- and time-dependent neuromuscular blockade (50% blockade in 85±9 min and 73±8 min with 80 and 160μg/ml, respectiv

Emergence of deletions during treadmill locomotion as a function of supraspinal and sensory inputs.

During locomotion, alternating activity of flexor and extensor muscles is largely regulated by a spinal neuronal network, the central pattern generator, the activity of which is modulated by peripheral and supraspinal inputs. In the absence of these modulatory inputs, for example during fictive locomotion after spinalization and curarization, spontaneous failures of motor activation (deletions) in a muscle can occur without perturbing the rhythmic cycle structure of the antagonists on the same side or the contralateral side. This suggests that the central pattern generator can maintain the loc

Studies on the neuromuscular blocking action of MPTP in the mouse phrenic nerve-diaphragm.

The peripheral neurotoxicity of 1-methyl-4-phenyl-1,2,3,6- tetrahydropyridine (MPTP) in the mouse phrenic nerve-diaphragm preparation was studied. The inhibition of nerve-evoked twitches was dependent on the concentrations of MPTP, ranging from 1 to 200 microM, which only slightly increased the amplitude of the twitch evoked by direct stimulation of the muscle. Pretreatment with pargyline did not prevent this inhibitory action of MPTP. The inhibition of the twitch by MPTP was reversible and could be partially reversed by anticholinesterase agents such as neostigmine and physostigmine, as well

Effects of SITS, an anion transport blocker, on CSF ionic composition in metabolic alkalosis.

Disulfonic stilbenes combine with the carrier protein involved in anion transport and inhibit the exchange of Cl- for HCO3- in a variety of biomembranes. Our aim was to determine whether such a mechanism is operative in the regulation of cerebrospinal fluid (CSF) [HCO3-] in metabolic alkalosis. In anesthetized, curarized, and artificially ventilated dogs either mock CSF (group I, 9 dogs) or mock CSF containing SITS, 4-acetamido-4'-isothiocyanostilbene-2,2'-disulfonic acid (group II, 7 dogs) was periodically injected into both lateral cerebral ventricles. During 6 h of isocapnic metabolic alkal

Effect of tubocurarine and decamethonium on voluntary muscle contractions in man.

Six healthy, young male subjects performed repeated brisk maximal voluntary muscle contractions (MVC) with the knee and hip extensors. Three MVCs were performed every minute. On separate days decamethonium 0.03 mg X kg-1 and tubocurarine 0.01 mg X kg-1 were administered intravenously during repeated MVCs. While ordinary MVCs showed a slow rate of rise of tension over approximately 1 s, brisk MVCs showed a steep rate of rise of tension and a biphasic configuration appeared, as a notch was seen 370-480 ms after the initiation of the contraction curve. An arbitrary straight line was drawn connect

Systematic assessment of column selectivity for the isolation of subcordatine and other flavonoid glycosides from Strychnos subcordata spruce ex Benth using off-line single heart-cutting two-dimensional liquid chromatography.

The genus Strychnos has gained worldwide recognition due to its role in the preparation of curare, a traditional arrow poison used by Indigenous peoples in South America, and its potential as a promising source of bioactive natural products. Although the phytochemical content of several Strychnos species has been extensively investigated, others, such as S. subcordata, remain largely unexplored. In this study, we describe for the first time the systematic screening of the selectivity of selected columns (C18, C8, PFP, phenyl-hexyl, CN and biphenyl) for the isolation of target compounds from th

Contralateral-noise effects on cochlear responses in anesthetized mice are dominated by feedback from an unknown pathway.

Suppression of ipsilateral distortion product otoacoustic emissions (DPOAEs) by contralateral noise is used in humans and animals to assay the strength of sound-evoked negative feedback from the medial olivocochlear (MOC) efferent pathway. However, depending on species and anesthesia, contributions of other feedback systems to the middle or inner ear can cloud the interpretation. Here, contributions of MOC and middle-ear muscle reflexes, as well as autonomic feedback, to contra-noise suppression in anesthetized mice are dissected by selectively eliminating each pathway by surgical transection,

Characteristics of pancreatic exocrine secretion produced by venom from the Brazilian scorpion, Tityus serrulatus.

The influence of venom (TSV) from the Brazilian scorpion, Tityus serrulatus, on exocrine pancreatic secretion was studied in relation to known cholinergic and peptidergic secretagogue activity. Pulse-labeling followed by chase incubation in the presence of secretagogues and various pharmacological agents revealed unique physiological characteristics of TSV in guinea pig pancreatic lobules. Exocytotic discharge of newly synthesized 3H-labeled proteins during a 3-h chase incubation showed a marked increase over basal discharge levels using logarithmic TSV doses of 0.10 to 100 micrograms/ml. Clinical spectrum of accidental hemlock poisoning: neurotoxic manifestations, rhabdomyolysis and acute tubular necrosis.

In the past, hemlock poisoning was only known for its neurotoxic effects; quite recently non-neurological features, consisting of rhabdomyolysis and acute renal failure, have been also described. Here we report our experience with these clinical findings, which we frequently observe in accidental hemlock poisoning. Between 1972 and 1990 we studied 18 patients: 17 of them were poisoned by conline (an alkaloid of Conium maculatim) in Apulia (Italy), and one by cicutoxin (the active principle of water hemlock) in New Mexico (USA). In the non-rapidly-fatal cases we tested myoglobinuria, serum musc

EEG-fMRI in awake rat and whole-brain simulations show decreased brain responsiveness to sensory stimulations during absence seizures.

In patients suffering absence epilepsy, recurring seizures can significantly decrease their quality of life and lead to yet untreatable comorbidities. Absence seizures are characterized by spike-and-wave discharges on the electroencephalogram associated with a transient alteration of consciousness. However, it is still unknown how the brain responds to external stimuli during and outside of seizures. This study aimed to investigate responsiveness to visual and somatosensory stimulation in Genetic Absence Epilepsy Rats from Strasbourg (GAERS), a well-established rat model for absence epilepsy.

Schwann cells sense and control acetylcholine spillover at the neuromuscular junction by α7 nicotinic receptors and butyrylcholinesterase.

Terminal Schwann cells (TSCs) are key components of the mammalian neuromuscular junction (NMJ). How the TSCs sense the synaptic activity in physiological conditions remains unclear. We have taken advantage of the distinct localization of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) at the NMJ to bring out the function of different ACh receptors (AChRs). AChE is clustered by the collagen Q in the synaptic cleft and prevents the repetitive activation of muscle nicotinic AChRs. We found that BChE is anchored at the TSC by a proline-rich membrane anchor, the small transmembrane pro
