Taenia crassiceps cysticercosis: humoral immune response and protection elicited by DNA immunization.

The purpose of this study was to evaluate DNA vaccination in cysticercosis prevention by using a Taenia crassiceps cDNA of a recombinant antigen (KETc7) that has been reported as protective against murine cysticercosis. The KETc7 cDNA was cloned into the pcDNA3 plasmid alone or with the betaglycan signal peptide sequence (pTc-7 and pTc-sp7, respectively). Positive expression of the pTc-sp7 product was confirmed by transfection of C33 cells and immunofluorescence using sera of mice infected with T. crassiceps. Immunization of mice with 3 injections of pTc-sp7 DNA at the higher dose (200 microg)

The complete mitochondrial genome of the bird schistosome Trichobilharzia regenti (Platyhelminthes: Digenea), causative agent of cercarial dermatitis.

The complete mitochondrial (mt) genome of the neuropathogenic bird schistosome Trichobilharzia regenti was fully sequenced in order to develop molecular markers for future diagnostic, molecular ecological, population, and phylogenetic studies. The genome was 14,838 bp in length, with a 68.4% AT bias in protein coding regions. A repeat element (3 x 184 bp) between trnV and trnW distinguished a single short noncoding region. As 9 of 14 genera of schistosomes parasitize birds, future characterization of their mt genomes is desirable for species-specific and strain- or population-specific diagnost

Opisthorchis viverrini: identification of a glycine-tyrosine rich eggshell protein and its potential as a diagnostic tool for human opisthorchiasis.

A cDNA encoding a novel eggshell protein (OvESP) with high-glycine (49.2%) and -tyrosine (27.8%) content was cloned from the human liver fluke Opisthorchis viverrini. In the adult parasite, the RNA products of the OvESP gene are limited to the vitelline follicles. They have a size of 800 nucleotides and are already present in 2-week-old juveniles. Immune sera of hamsters, experimentally infected, and humans, naturally infected with O. viverrini, detect bacterially expressed recombinant OvESP (rOvESP). A rabbit anti-rOvESP antiserum only reacts with the shells of intrauterine eggs in tissue sec

Morphology and molecular phylogeny of two bat trematodes from the taxonomically problematic genus Paralecithodendrium (Lecithodendriidae, Digenea).

Trematodes of the genus Paralecithodendrium parasitize bats worldwide. Among them, the identification of Paralecithodendrium longiforme and Paralecithodendrium cryptolecithum is complicated by their high morphological similarity. We studied Paralecithodendrium trematodes from the small intestine of bats inhabiting the Middle Volga region (European Russia). The aim of our study was to analyze these two Paralecithodendrium species using both morphological and molecular phylogenetic approaches. Here, we present the first complete morphological description and molecular phylogenetic analysis of Pa

Morphological and molecular data for two Plagiorchis species (Trematoda: Plagiorchiidae) from the southern Far East of Russia.

Adult trematodes of two species of the genus Plagiorchis were found in the southern Far East of Russia: one species in the intestine of a naturally infected Japanese large-footed bat (Myotis macrodactylus) and the other in a golden hamster (Mesocricetus auratus) experimentally infected with metacercariae collected from a stonefly. On the basis of morphological and molecular studies, the trematode individuals found in the bat were identified as Plagiorchis koreanus, which confirms that this species is cosmopolitan. The trematodes reared in the hamster were similar in their morphological feature

Detection and molecular characteristics of Rhytidodoides sp. (Digenea: Rhytidodidae) from the gall bladder of green sea turtles (Chelonia mydas) in the Ogasawara Islands, Japan.

Trematodes of the genus Rhytidodoides are parasitic in marine turtles. Of the already known species, Rhytidodoides similis Price, 1939, occurs especially in the gall bladder. In this study, we surveyed 73 green sea turtles (Chelonia mydas) in the Ogasawara Islands, Japan, and detected Rhytidodoides sp. from the gall bladders of 18 turtles. A detailed morphological analysis revealed that the forebody of Rhytidodoides sp. differed slightly in shape from that of R. similis. There has been no information on DNA sequences of the family Rhytidodidae. A molecular phylogeny based on 28S rDNA sequences

Molecular identification of Anisakis spp. from fishes collected in the Tyrrhenian Sea (NW Mediterranean).

The accurate identification of anisakid nematodes at any life cycle stage is important both to deepen the knowledge on their taxonomy, ecology, epidemiology and for diagnosis and control, as larval stages cause a clinical disease in humans known as anisakidosis. With the aim to investigate the presence of anisakid larvae, specimens of horse mackerel, Trachurus trachurus (Linnaeus, 1758), silver scabbardfish, Lepidopus caudatus (Euphrasen, 1788), European anchovy, Engraulis encrasicolus (Linnaeus, 1758) and opah fish, Lampris guttatus (Brunnich, 1788), were collected by trawling at depths rangi

Mitochondrial genomes of parasitic flatworms.

Complete or near-complete mitochondrial genomes are now available for 11 species or strains of parasitic flatworms belonging to the Trematoda and the Cestoda. The organization of these genomes is not strikingly different from those of other eumetazoans, although one gene (atp8) commonly found in other phyla is absent from flatworms. The gene order in most flatworms has similarities to those seen in higher protostomes such as annelids. However, the gene order has been drastically altered in Schistosoma mansoni, which obscures this possible relationship. Among the sequenced taxa, base compositio

Sedimentary ancient DNA as part of a multimethod paleoparasitology approach reveals temporal trends in human parasitic burden in the Roman period.

The detection of parasite infections in past populations has classically relied on microscopic analysis of sediment samples and coprolites. In recent years, additional methods have been integrated into paleoparasitology such as enzyme-linked immunosorbent assay (ELISA) and ancient DNA (aDNA). The aim of this study was to evaluate a multimethod approach for paleoparasitology using microscopy, ELISA, and sedimentary ancient DNA (sedaDNA) with a parasite-specific targeted capture approach and high-throughput sequencing. Using 26 samples dating from c. 6400 BCE to 1500 CE that were previously anal

Cloning and analysis of a cDNA encoding a putative serine protease comprising two trypsin-like domains of Trichinella spiralis.

The cDNA encoding a putative serine protease, TsSerP, was cloned by degenerative polymerase chain reaction and screening of the cDNA library from Trichinella spiralis adult-newborn larvae stage. Sequence analysis revealed the presence of two trypsin-like serine protease domains flanking a hydrophilic domain, with the catalytic triad residue histidine in the alpha domain substituted by an arginine residue. Southern blots indicated that this was a single copy gene in the parasite genome. Northern blots demonstrated a single 2.3-kb transcript during the muscle larvae and adult stages of T. spiral

Schistosomes in South African penguins.

During the years 2009-2012, faeces of African penguins (Spheniscus demersus L.) from South African rehabilitation centres were examined for helminths. In total, 46 out 555 samples (8.29 %), mostly belonging to adult birds, were found to contain oval schistosome eggs with a spine on one pole. Their dimensions were 153.21 ± 9.07 × 87.14 ± 8.67 μm. Selected DNA fragments (18S, 28S and ITS rDNA) were sequenced and compared to other schistosome isolates deposited in GenBank. The shape of the eggs suggests that they belong to the genus Gigantobilharzia; however, due to the insufficient stage of know

Application of a Multiplex Quantitative PCR to Assess Prevalence and Intensity Of Intestinal Parasite Infections in a Controlled Clinical Trial.

BACKGROUND: Accurate quantitative assessment of infection with soil transmitted helminths and protozoa is key to the interpretation of epidemiologic studies of these parasites, as well as for monitoring large scale treatment efficacy and effectiveness studies. As morbidity and transmission of helminth infections are directly related to both the prevalence and intensity of infection, there is particular need for improved techniques for assessment of infection intensity for both purposes. The current study aimed to evaluate two multiplex PCR assays to determine prevalence and intensity of intest

Angiostrongylus vasorum in golden jackals (Canis aureus) and red foxes (Vulpes vulpes) from Northern Bosnia and Herzegovina.

In this study, we conducted post-mortem examinations of golden jackals (Canis aureus) and red foxes (Vulpes vulpes) from the northern region of the Republic of Srpska (Bosnia and Herzegovina) to detect the presence of Angiostrongylus vasorum. An epidemiological survey was conducted in the densest golden jackal population in the country. Over two time intervals, we examined a total of 30 jackals and 16 foxes. The presence of A. vasorum was confirmed in two jackals (6.6%) and one fox (6.25%). These findings strongly suggest that the spread of A. vasorum can be facilitated by the active expansion

Human cystic echinococcosis: first molecular identification of Echinococcus canadensis G7 in Brazil.

Echinococcus granulosus sensu lato (s.l.) is a species complex with the potential to cause cystic echinococcosis (CE). Contact with the feces of domestic dogs (Canis familiaris) fed with raw viscera of intermediate livestock hosts is a risk factor for this infection in the southern region of Brazil. Although the region has been considered endemic to CE for many years, molecular data regarding the species of the complex causing CE in humans are scarce. This study aimed to perform a molecular analysis of the biological fluid from a human liver cyst to investigate the species responsible for CE.

Short report: Identification of Echinococcus species from a yak in the Qinghai-Tibet plateau region of China.

The species identification of an echinococcal lesion in the liver of a yak in the Qinghai-Tibet plateau region of China, where both Echinococcus granulosus and E. multilocularis are present, was difficult to determine because of the atypical appearance of the lesion. Polymerase chain reaction-based mitochondrial genotyping allowed us to discriminate the Echinococcus species. Nucleotide sequences of the cytochrome c oxidase subunit 1 (cox1) and cytochrome b (cytb) genes amplified from the echinococcal lesion demonstrated that the yak was infected with the E. granulosus G1 genotype (sheep strain
