Sensitivity analysis for the probability of benefit in randomized controlled trials with a binary treatment and a binary outcome.

For a comprehensive understanding of the effect of a given treatment on an outcome of interest, quantification of individual treatment heterogeneity is essential, alongside estimation of the average causal effect. However, even in randomized controlled trials, quantities such as the probability of benefit or the probability of harm are not identifiable, since multiple potential outcomes cannot be observed simultaneously for the same individual. We propose a sensitivity analysis for the probability of benefit in randomized controlled trial settings with a binary treatment and a binary outcome,

A comparison of cluster and factor analytic techniques for identifying symptom-based dimensions of obsessive-compulsive disorder.

A growing body of literature suggests that obsessive-compulsive disorder (OCD) is a heterogeneous condition. The studies investigating symptom dimensions have been limited by numerous methodological differences and sample characteristics. The purpose of this study was to compare the two most commonly applied statistical techniques used in addressing this question in the same large cohort of individuals with OCD. Both cluster analysis and factor analysis were used to examine OCD symptom data as measured by the Yale-Brown Obsessive Compulsive Scale (Y-BOCS) Symptom Checklist for 355 individuals

Sample size calculation for agreement between two raters with binary endpoints using exact tests.

In an agreement test between two raters with binary endpoints, existing methods for sample size calculation are always based on asymptotic approaches that use limiting distributions of a test statistic under null and alternative hypotheses. These calculated sample sizes may be not reliable due to the unsatisfactory type I error control of asymptotic approaches. We propose a new sample size calculation based on exact approaches which control for the type I error rate. The two exact approaches are considered: one approach based on maximization and the other based on estimation and maximization.

Joint synthesis of multiple correlated outcomes in networks of interventions.

Multiple outcomes multivariate meta-analysis (MOMA) is gaining in popularity as a tool for jointly synthesizing evidence coming from studies that report effect estimates for multiple correlated outcomes. Models for MOMA are available for the case of the pairwise meta-analysis of two treatments for multiple outcomes. Network meta-analysis (NMA) can be used for handling studies that compare more than two treatments; however, there is currently little guidance on how to perform an MOMA for the case of a network of interventions with multiple outcomes. The aim of this paper is to address this issu

Level-adjusted funnel plots based on predicted marginal expectations: an application to prophylactic antibiotics in gallstone surgery.

Funnel plots are widely used to visualize grouped data, for example, in institutional comparison. This paper extends the concept to a multi-level setting, displaying one level at a time, adjusted for the other levels, as well as for covariates at all levels. These level-adjusted funnel plots are based on a Markov chain Monte Carlo fit of a random effects model, translating the estimated model parameters to predicted marginal expectations. Working within the estimation framework, we accommodate outlying institutions using heavy-tailed random effects distributions. We also develop computer-effic

Misuse of DeLong test to compare AUCs for nested models.

The area under the receiver operating characteristics curve (AUC of ROC) is a widely used measure of discrimination in risk prediction models. Routinely, the Mann-Whitney statistics is used as an estimator of AUC, while the change in AUC is tested by the DeLong test. However, very often, in settings where the model is developed and tested on the same dataset, the added predictor is statistically significantly associated with the outcome but fails to produce a significant improvement in the AUC. No conclusive resolution exists to explain this finding. In this paper, we will show that the reason

Interpretation of cellular proliferation data: avoid the panglossian.

There are several statistics that may be calculated to characterize a cellular proliferation experiment. By far, the most commonly-reported statistic is the percent of cells in the final culture that have divided; however, this statistic has significant limitations. Other statistics provided by software modeling provide a much richer characterization of the biological response; however, their use also comes with caveats. Here, I discuss the practical application of these statistics, including their limitations and interdependencies, using hypothetical data. The goal of this perspective is to p

A marginalized conditional linear model for longitudinal binary data when informative dropout occurs in continuous time.

Within the pattern-mixture modeling framework for informative dropout, conditional linear models (CLMs) are a useful approach to deal with dropout that can occur at any point in continuous time (not just at observation times). However, in contrast with selection models, inferences about marginal covariate effects in CLMs are not readily available if nonidentity links are used in the mean structures. In this article, we propose a CLM for long series of longitudinal binary data with marginal covariate effects directly specified. The association between the binary responses and the dropout time i

A spatial scan statistic for multiple clusters.

Spatial scan statistics are commonly used for geographical disease surveillance and cluster detection. While there are multiple clusters coexisting in the study area, they become difficult to detect because of clusters' shadowing effect to each other. The recently proposed sequential method showed its better power for detecting the second weaker cluster, but did not improve the ability of detecting the first stronger cluster which is more important than the second one. We propose a new extension of the spatial scan statistic which could be used to detect multiple clusters. Through constructing

A flexible strategy for testing subgroups and overall population.

Subgroup analyses in addition to the total study population analysis are common in clinical trials. However, it is well recognized that findings from subgroup analyses do not provide confirmatory evidence for subgroup treatment effects without placing a priori criteria for ensuring that their findings are scientifically sound. In this paper we address some of the common pitfalls of subgroup analyses. Subgroups analyses inherently have low power for detecting treatment effects. We investigate the power interplay for a subgroup analysis and that for the total study population and list factors th

[Investigation of the impact of pterygium on ocular optic system aberration. Communication 1. Relationship of induced aberrations to the stage of pterygium].

The authors made a study in two areas: they investigated corneal aberrations in patients from different age groups in health; analyzed the corneal aberration pattern at different stages of pterygium in the matched age groups; and comparatively estimated values in pterygium and in health in the similar age groups. High-order aberrations (HOA) of the corneal were studied in 45 patients (84 eyes) aged 20 to 76 years. The corneal aberration pattern was analyzed in 44 patients (55 eyes) aged 25 to 74 years who had pterygium of different stages. The study covered patients with Stages II (24 eyes), I

A generalized concordance correlation coefficient for continuous and categorical data.

This paper discusses a generalized version of the concordance correlation coefficient for agreement data. The concordance correlation coefficient evaluates the accuracy and precision between two measures, and is based on the expected value of the squared function of distance. We have generalized this coefficient by applying alternative functions of distance to produce more robust versions of the concordance correlation coefficient. In this paper we extend the application of this class of estimators to categorical data as well, and demonstrate similarities to the kappa and weighted kappa statis

Nonparametric inference on the E/O ratio in model validation.

In preventing diseases such as cancers and osteoporosis, statistical models are often used to identify subjects with high risks. The ratio of the expected (or predicted) number of cases in the target population and the observed numbers of cases (the E/O ratio) is a useful quantity to evaluate the goodness of fit of the model. The model is usually evaluated on a sample taken from the target population and, in the literature, statistical inferences on the E/O ratio often assume that the expected number is a constant and the observed number follows a Poisson distribution. In this paper, we introd

Confidence Intervals for the MADET Index and Its Corresponding Classification Thresholds Using Generalized Pivotal Quantities.

In medical contexts, especially in the diagnosis process of patients into different ordered stages of a disease (e.g., not diseased, early disease, and advanced disease) based on a continuous biomarker, the concept of the receiver operating characteristic (ROC) curve has been extended to that of the ROC surface and, consequently, the corresponding diagnostic performance measures as well. Specifically, the maximum absolute determinant (MADET) has been recently proposed, extending the classic concept of the Youden index. However, no method has been proposed for constructing confidence intervals

Variations of Power Priors in Time-to-Event Analysis With Piecewise Constant Hazards Through Baseline and Outcome Similarity Driven Bounded Weighting.

Patient recruitment in clinical trials, particularly in rare diseases or pediatric populations, remains a significant challenge. Leveraging external data to supplement trial information offers a promising solution, reducing placebo allocation, and enhancing recruitment feasibility while increasing the number of patients receiving active treatment, thus improving safety evaluations. However, the success of this method depends on the alignment of external data with target data from a population characteristics perspective and scientific translation perspective. Prediction intervals for random-effects meta-analysis based on confidence distributions and Edgington's method.

Statistical inference about the average effect in random-effects meta-analysis has been considered insufficient in the presence of substantial between-study heterogeneity. Predictive distributions are well suited for quantifying heterogeneity since they are interpretable on the effect scale and provide clinically relevant information about future events. We construct predictive distributions accounting for uncertainty through confidence distributions from Edgington's p-value combination method and the generalized heterogeneity statistic. Simulation results suggest that 95% prediction intervals

Density and treatment effect estimation under covariate-adaptive randomization with heavy-tailed outcomes.

Randomized experiments are the gold standard for investigating causal relationships, with comparisons of potential outcomes under different treatment groups used to estimate treatment effects. However, outcomes with heavy-tailed distributions pose significant challenges to traditional causal inference approaches. While recent studies have explored these issues under simple randomization, their application in more complex randomization designs, such as stratified randomization or covariate-adaptive randomization, has not been adequately addressed. To fill the gap, we first investigate the perfo

Integration of aggregate data in causally interpretable meta-analysis by inverse weighting.

Obtaining causally interpretable meta-analysis results is challenging when there are differences in the distribution of effect modifiers between eligible trials. To overcome this, recent work on transportability methods has considered standardizing results of individual studies over the case-mix of a target population, prior to pooling them as in a classical random-effect meta-analysis. One practical challenge, however, is that case-mix standardization often requires individual participant data (IPD) on outcome, treatments and case-mix characteristics to be fully accessible in every eligible s

Marginally Interpretable Spatial Logistic Regression With Bridge Processes.

In including random effects to account for dependent observations, the odds ratio interpretation of logistic regression coefficients is changed from population-averaged to subject-specific. This is unappealing in many applications, motivating a rich literature on methods that maintain the marginal logistic regression structure without random effects, such as generalized estimating equations. However, for spatial data, random effect approaches are appealing in providing a full probabilistic characterization of the data that can be used for prediction. We propose a new class of spatial logistic

The impact of coarsening an exposure on partial identifiability in instrumental variable settings.

In instrumental variable (IV) settings, such as imperfect randomized trials and observational studies with Mendelian randomization, one may encounter a continuous exposure, the causal effect of which is not of true interest. Instead, scientific interest may lie in a coarsened version of this exposure. Although there is a lengthy literature on the impact of coarsening of an exposure with several works focusing specifically on IV settings, all methods proposed in this literature require parametric assumptions. Instead, just as in the standard IV setting, one can consider partial identification v
