Novel distamycin analogues that block the cell cycle of African trypanosomes with high selectivity and potency.

Polyamides-based compounds related to the Streptomycetal distamycin and netropsin are potent cytostatic molecules that bind to AT-rich regions of the minor groove of the DNA, hence interfering with DNA replication and transcription. Recently, derivatives belonging to this scaffold have been reported to halt the proliferation of deadly African trypanosomes by different and unrelated mechanisms. Here we describe the synthesis and preliminary characterization of the anti-trypanosomal mode of action of new potent and selective distamycin analogues. Two tri-heterocyclic derivatives containing a cen

Experiences with an in-training community service model in the control of zoonotic sleeping sickness in Uganda.

By 2006, the acute and zoonotic Tripanosoma brucei rhodesiense sleeping sickness in Uganda was spreading northward, leading to fear of a merger with the chronic Tripanosoma brucei gambiese type that affects people in the northwest of the country. Eliminating infection in cattle was urgent because they had been confirmed to be spreading the zoonotic type, and eliminating infection would reduce the animal reservoir and subsequently reduce transmission of sleeping sickness. In this article, we describe how the staff and students of the Faculty of Veterinary Medicine, Makerere University, adjusted

Evaluation of efficacy of bruceine A, a natural quassinoid compound extracted from a medicinal plant, Brucea javanica, for canine babesiosis.

Bruceine A, a natural quassinoid compound extracted from the dried fruits of Brucea javanica (L.) Merr., was evaluated for its antibabesial activity in vitro and in vivo. Bruceine A inhibited the in vitro growth of Babesia gibsoni in canine erythrocytes at lower concentration compared with the standard antibabesial drug diminazene aceturate and killed the parasites within 24 hr at a concentration of 25 nM. Oral administration of bruceine A at a dosage of 6.4 mg/kg/day for 5 days resulted in no clinical findings in a dog with normal ranges of hematological and biochemical values in the blood. T

Equine piroplasmosis an update on diagnosis, treatment and prevention.

Two haemoprotozoan parasites, Babesia caballi and Babesia equi, can cause equine piroplasmosis. Due to the presence of potential tick vectors in areas so far unaffected by equine babesias, import and export regulations often require the serum testing of animals for evidence of infection. Although the complement fixation test (CFT) has been recommended for detecting the presence of antibodies to Babesia spp., it has been demonstrated to have several disadvantages, including false-positive results and low sensitivity for detecting latent infections. An enzyme-linked immunosorbent assay (ELISA) m

Simultaneous control of ticks and tsetse flies in Satiri, Burkina Faso, by the use of flumethrin pour on for cattle.

Treatments of 2000 cattle at monthly intervals with flumethrin pour on (1 mg active ingredient/kg b.w.) resulted in a rapid decrease of African Animal Trypanosomosis (AAT). After an initial curative treatment of the sentinel herd with diminazene aceturate (7 mg/kg b.w.) only positive cases were treated. Three applications of flumethrin were sufficient to reduce the prevalence of AAT to below 5%. Apart from a slight increase at the end of the first year the infection rate varied between 0 and 5%. A tsetse population disappeared from a heavily infested habitat after six treatments. The average t

How rational drug use reduces trypanosome infections in cattle in chemo-resistance hot-spot villages of northern Togo.

The study assessed an integrated trypanosomosis control strategy in drug-resistant hotspot villages of northern Togo. This strategy comprised (i) rational trypanocidal drug use in symptomatic cattle, (ii) vectors and ticks control by targeted bi-monthly insecticidal spraying of the lower body parts of cattle and (iii) strategic deworming with Albendazole in the beginning and the end of the rainy season. The program was implemented between June 2014 and October 2015 in four villages in northern Togo, which had been previously identified as drug resistant hotspots for diminazene diaceturate (DA)

Babesia canis and Babesia rossi co-infection in an untraveled Nigerian dog.

A sexually intact 6-month-old female Alsatian dog was presented to the Veterinary Clinic of the National Veterinary Research Institute, Vom, Plateau State, Nigeria, for the following complaints: anorexia, hemoglobinuria, fever, tick infestation and general malaise. Microscopy revealed piroplasms with a wide range of sizes (1-5 μm in length) in red blood cells, raising a suspicion of a co-infection with two or more Babesia species. Specific PCR assays for canine Babesia spp. and DNA sequencing revealed the presence of Babesia canis and Babesia rossi co-infection. This study constitutes the firs

The effect of Trypanosoma brucei brucei infection on rabbit plasma iron and zinc concentrations.

Changes in plasma iron and zinc concentration were studied in rabbits following a needle challenge with Trypanosoma brucei brucei clone ILTat 2.1. The infection resulted in a decrease of the concentration of both trace elements. Plasma iron concentrations decreased gradually and were decreased maximally to 52.3% of pre-infection levels on day 18 post-inoculation. Plasma zinc concentrations, on the other hand, decreased more rapidly and were decreased maximally to 27.4% of pre-infection levels on day 3 post-inoculation. The onset of these decreases coincided with the appearance of parasites in

Trypanosoma brucei, T. congolense and T. vivax infections in horses on a farm in Kenya.

Equines are particularly susceptible to infection with Trypanosoma evansi and T. brucei, but rarely is natural T. congolense and T. vivax infection seen in horses. An outbreak of trypanosomosis occurred in a herd of horses used for patrolling the pineapple fields on the Del Monte Farm, Thika, Kenya initially involving 6 horses. On subsequent screening of the entire group, T. brucei, T. congolense and T. vivax infections were detected in 16 of the 35 horses. The tests used for diagnosis included microscopic examination of stained blood smears, buffy coat technique, mouse inoculation and antigen

Natural and acquired resistance to Trypanosoma vivax in cattle.

Zebu x European (Z x E) crossbred cattle suffered a more severe course of disease than Boran cattle when infected with Trypanosoma vivax (Likoni) by Glossina morsitans. All Z x E animals in this study required Berenil treatment while all Borans self-cured the infection without treatment. The more severe disease in Z x E animals was characterized by longer periods of patent infection and fever, more severe anaemia and greater likelihood of haemorrhage. Cattle previously infected and cured with Berenil showed resistance and self-cured challenge infections. After self-cure cattle remained immune

The complement fixation test and African trypanosomiasis. II. The complement fixation test as an aid for assessing therapy.

Complement-fixing antibody titres decline after successful treatment of trypanosoma infected cattle during a fixed period. For investigating the practicability of the resulting double test procedure in the field, cattle under controlled drug regimes were tested at treatment and again 3 weeks later. Alternatively, the results from groups of treated and untreated animals from the same area were compared. Both serological and parasitological test results indicated an insuffieicnt effectiveness of therapy in the area under investibation.

EVALUATION OF THE INHIBITORY EFFECTS OF SIX NATURAL PRODUCT EXTRACTS AGAINST BABESIA GIBSONI IN VITRO AND IN VIVO.

New antibabesial drugs are required to fight resistant parasites, and plant-derived natural products are a robust source. Six kinds of natural product extracts derived from herbal medicines that are traditionally used for the treatment of malaria were selected to test the antibabesial effect on Babesia gibsoni in vitro and in vivo. Parasitized blood was collected from dogs infected with B. gibsoni to evaluate the inhibitory effect of verbenalin, catechin hydrate, dihydrolycorine, embelin, ursolic acid, agrimol B, and bruceine H in vitro. The expression levels of the 18S rRNA gene in all drug-t

In vitro and in vivo growth inhibitory activities of cryptolepine hydrate against several Babesia species and Theileria equi.

Piroplasmosis treatment has been based on the use of imidocarb dipropionate or diminazene aceturate (DA), however, their toxic effects. Therefore, the discovery of new drug molecules and targets is urgently needed. Cryptolepine (CRY) is a pharmacologically active plant alkaloid; it has significant potential as an antiprotozoal and antibacterial under different in vitro and in vivo conditions. The fluorescence assay was used for evaluating the inhibitory effect of CRY on four Babesia species and Theileria equi in vitro, and on the multiplication of B. microti in mice. The toxicity assay was eva
