Procedural separation of appetitive and consummatory behaviors in operant ethanol self-administration: A review and open-source analytical framework.

Despite decades of research, no new FDA-approved medications for alcohol use disorder (AUD) have emerged in over 25 years. Enhancing the translational relevance of preclinical models by more precisely capturing the behavioral and neurobiological features of AUD offers a promising path toward identifying novel therapeutic targets. Operant self-administration paradigms are essential for modeling voluntary ethanol intake in rodents, yet traditional approaches often confound appetitive (seeking) and consummatory (intake) behaviors. A biphasic sipper model developed by Hank Samson's laboratory addr

The Heart-Microbiome Axis: Emerging Relevance in Pediatric Cardiology.

The gut microbiome has emerged as an important regulator of cardiovascular physiology through interconnected metabolic, immune, inflammatory, and endocrine pathways. Although the gut-heart axis has been extensively investigated in adult cardiovascular disease, its relevance in paediatric cardiology is only beginning to be recognised. The dynamic development of the gut microbiome during early life, together with the unique physiological characteristics of congenital heart disease, chronic hypoxia, altered intestinal perfusion, repeated antibiotic exposure, and perioperative stress, creates a se

Maternal Stress and Ethnic Disparities in Pre-Eclampsia: The Significance of a Migrant Perspective.

Persisting ethnic disparities in pre-eclampsia (PE), cardiovascular disease (CVD), and maternal mortality call for a paradigm shift in how ethnicity is understood as a risk factor for PE. Starting from a migrant perspective, we argue that the transgenerational experience of maternal stress within shared, yet dynamic ecosocial contexts can be linked to core pathophysiological features of PE. A growing body of evidence suggests how a vicious cycle of chronic maternal stress, cardiovascular dysfunction, placental ER stress, and endothelial dysfunction may serve as a catalyst for the transmission

Biomaterial-mediated blood clot formation: a key to unlocking osteogenic synergy for bone regeneration.

Bone hemorrhage presents serious clinical challenges due to its high morbidity and mortality. Autologous coagulation forms blood clots that both halt bleeding and provide a natural scaffold for bone repair. The dynamic formation and dissolution of blood clots maintain a delicate balance between hemostasis and regeneration. Clot architecture and cytokine composition play pivotal roles in directing bone repair. These insights inspire biomimetic strategies using engineered biomaterials to regulate clot behavior, a central theme in designing bone-replacement systems that integrate hemostasis with

Gout Inflammation Time Programming: Molecular Clock from Crystal Triggering to Tissue Remodeling.

This review introduces and elaborates a novel temporal paradigm, the "Gout Inflammation Time Programming" model, conceptualized through the Gout-STAT™ framework. This model redefines gout inflammation as a dynamic continuum progressing through three precisely timed phases: an acute Perception phase (0-24 h) initiated by monosodium urate (MSU) crystal recognition, triggering the NOD-like receptor thermal protein domain associated protein 3 (NLRP3) inflammasome and neutrophil-driven burst; a critical Adaptation phase (24-72 h) where outcomes are determined by immunometabolic reprogramming of mac

SynchDP: A correlation based sequence alignment algorithm for synchronizing longitudinal clinical data.

MOTIVATION: Analogous to the sequence-to-sequence alignment problem in discrete settings, two time-series can be aligned based on their longitudinal similarity. In disease contexts, aligning time-series that represent patient severity levels allows quantifying the similarity in their progression. However, in real-world clinical settings, patients are hospitalized at varying stages, and measurements are taken at irregular intervals, leading to sequences of different lengths and sampling rates. This complicates direct comparison of gene expression profiles and hinders biomarker discovery. Here,

Unveiling the impact of anthropogenic wastes on greenhouse gas emissions from the enigmatic mangroves of Indian Sundarban.

The greenhouse gas (GHG) emissions from the mangrove ecosystem due to climate change have been an emerging environmental issue in the present scenario. However, the GHGs, emitted through anthropogenic causes in these vulnerable regions are often neglected. The level of soil pollution has increased due to the uncontrolled disposal of wastes from ports, ferry services, plastics, and metals, emitting huge amounts of GHGs. Here, a novel dynamic model on GHG emission was proposed for the simulation of carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O) emissions using R programming languag

Living materials for gas therapy.

The clinical translation of gas therapy, which employs medical gases such as nitric oxide (NO), carbon monoxide (CO), hydrogen sulfide (H2S), hydrogen (H2), and sulfur dioxide (SO2), is mainly limited by the absence of delivery systems that can provide precise spatiotemporal control in complex pathological environments. While conventional nanocarriers have improved in gas delivery, they often suffer from limited biocompatibility, poor targeting, and insufficient responsiveness. Recently, living materials emerged as a promising and innovative paradigm. Engineered from biological entities such a

ReAlign-N: an integrated realignment approach for multiple nucleic acid sequence alignment, combining global and local realignments.

Ensuring accurate multiple sequence alignment (MSA) is essential for comprehensive biological sequence analysis. However, the complexity of evolutionary relationships often results in variations that generic alignment tools may not adequately address. Realignment is crucial to remedy this issue. Currently, there is a lack of realignment methods tailored for nucleic acid sequences, particularly for lengthy sequences. Thus, there's an urgent need for the development of realignment methods better suited to address these challenges. This study presents ReAlign-N, a realignment method explicitly de

Object Motion Detection Enabled by Reconfigurable Neuromorphic Vision Sensor under Ferroelectric Modulation.

Increasing the demand for object motion detection (OMD) requires shifts of reducing redundancy, heightened power efficiency, and precise programming capabilities to ensure consistency and accuracy. Drawing inspiration from object motion-sensitive ganglion cells, we propose an OMD vision sensor with a simple device structure of a WSe2 homojunction modulated by a ferroelectric copolymer. Under optical mode and intermediate ferroelectric modulation, the vision sensor can generate progressive and bidirectional photocurrents with discrete multistates under zero power consumption. This design enable

Infinite hidden Markov models for multiple multivariate time series with missing data.

Exposure to air pollution is associated with increased morbidity and mortality. Recent technological advancements permit the collection of time-resolved personal exposure data. Such data are often incomplete with missing observations and exposures below the limit of detection, which limit their use in health effects studies. In this paper, we develop an infinite hidden Markov model for multiple asynchronous multivariate time series with missing data. Our model is designed to include covariates that can inform transitions among hidden states. We implement beam sampling, a combination of slice s

A lymphocyte-microglia-astrocyte axis in chronic active multiple sclerosis.

Multiple sclerosis (MS) lesions that do not resolve in the months after they form harbour ongoing demyelination and axon degeneration, and are identifiable in vivo by their paramagnetic rims on MRI scans1-3. Here, to define mechanisms underlying this disabling, progressive neurodegenerative state4-6 and foster development of new therapeutic agents, we used MRI-informed single-nucleus RNA sequencing to profile the edge of demyelinated white matter lesions at various stages of inflammation. We uncovered notable glial and immune cell diversity, especially at the chronically inflamed lesion edge.

Target-directed enzyme-free dual-amplification DNA circuit for rapid signal amplification.

Dynamic DNA circuits have shown promising potential for amplified biosensing and bioengineering applications at the molecular level. Here, an enzyme-free, single-step and rapid signal amplification DNA circuit was developed by integrating target-directed entropy-driven catalysis (EDC) and hybridization chain reaction (HCR) for analysis of nucleic acids and small molecules. The target catalyzes the self-assembly of the EDC premade substrate complex and fuel strands to release the hidden amplicon trigger (T), which was encoded with trigger sequences for the downstream HCR circuit. The released T

Atlas-Free Cervical Spinal Cord Segmentation on Midsagittal T2-Weighted Magnetic Resonance Images.

An automatic atlas-free method for segmenting the cervical spinal cord on midsagittal T2-weighted magnetic resonance images (MRI) is presented. Pertinent anatomical knowledge is transformed into constraints employed at different stages of the algorithm. After picking up the midsagittal image, the spinal cord is detected using expectation maximization and dynamic programming (DP). Using DP, the anterior and posterior edges of the spinal canal and the vertebral column are detected. The vertebral bodies and the intervertebral disks are then segmented using region growing. Then, the anterior and p

RhoDyn: A ρ-TD-RASCI Framework to Study Ultrafast Electron Dynamics in Molecules.

This article presents the program module RhoDyn as part of the OpenMOLCAS project intended to study ultrafast electron dynamics within the density-matrix-based time-dependent restricted active space configuration interaction framework (ρ-TD-RASCI). The formalism allows for the treatment of spin-orbit coupling effects, accounts for nuclear vibrations in the form of a vibrational heat bath, and naturally incorporates (auto)ionization effects. Apart from describing the theory behind and the program workflow, the paper also contains examples of its application to the simulations of the linear L2,3

Integrative analysis of mRNA stability regulation uncovers a metastasis-suppressive program in breast cancer.

Heterogeneity in cancer gene expression is typically linked to genetic and epigenetic alterations, yet the extent of contribution from posttranscriptional regulation remains unclear. Here, we systematically measured messenger RNA (mRNA) dynamics across diverse breast cancer models, revealing that mRNA stability substantially shapes gene expression variability. To decipher these dynamics, we developed GreyHound, an interpretable multimodal deep-learning framework integrating RNA sequence features and RNA binding protein (RBP) expression. GreyHound identified an extensive network of RBPs and the

A modular synthetic promoter toolkit for programmable gene control in activated and chronically stimulated T cells.

Synthetic circuits that couple transgene expression to endogenous transcription factor (TF) activity enable cellular response to changing functional states. In T cells, antigen stimulation results in dynamic changes of transcription factor expression programs. Harnessing these endogenous TF dynamics for programmable gene control remains an important challenge in engineered T cells. Here, we develop a library of synthetic promoters (SPs) responsive to seven TFs identified through transcriptomic analysis of T-cell activation programs. These promoters couple endogenous TF activity to transcriptio

Yersinia pestis: examining wildlife plague surveillance in China and the USA.

Plague is a zoonotic disease caused by the bacterium Yersinia pestis Lehmann and Neumann, 1896. Although it is essentially a disease of rodents, plague can also be transmitted to people. Historically, plague has caused massive morbidity and mortality events in human populations, and has recently been classified as a reemerging disease in many parts of the world. This public health threat has led many countries to set up wild and domestic animal surveillance programs in an attempt to monitor plague activity that could potentially spill over into human populations. Both China and the USA have pl
