Cryptic diversity and recent diversification in the Pyrrhulina australis species complex (Characiformes, Lebiasinidae) revealed by mitochondrial DNA.

Pyrrhulina australis Eigenmann & Kennedy, 1903, is widely distributed across the main hydrographic basins of South America. The taxonomic integrity of this species is nevertheless challenged by the lack of clear diagnostic characters, its marked geographic variation and the morphological overlap with its congeners. In this context, the present study investigated the genetic diversity of the P. australis group (P. australis, Pyrrhulina marilynae and Pyrrhulina vittata), using sequences of the mitochondrial Cytochrome Oxidase I gene to delimit evolutionary lineages and provide insights into biog

Fall-like acclimation alters mitochondrial function and substrate use in freeze-tolerant crickets.

Acclimation to low temperatures in insects involves complex metabolic adjustments to ensure survival. Notably, cryoprotectants including glycerol, trehalose and proline can accumulate in insects to promote freeze tolerance. Interestingly, these metabolites can also be converted to metabolic fuels sustaining mitochondrial ATP production. This study investigates the metabolic shifts in the freeze-tolerant cricket Gryllus veletis during fall-like acclimation, focusing on mitochondrial function and its possible link with cryoprotectant accumulation. We hypothesized that fall-like acclimation promo

Review of the Progamotaenia macropodis (Cestoda: Anoplocephalidae) species complex from Australian macropodid marsupials, with description of P. mollicula sp. nov.

Genetic evidence indicates that Progamotaenia macropodis Beveridge, 1976 (Cestoda: Anoplocephalidae), found in various macropodid host species, is a complex of cryptic species. However, the genetic data are incomplete, and no morphological re-appraisal of the species has been undertaken since its original description. Here, additional mitochondrial cytochrome c oxidase I sequence data have been added from the type host, Macropus giganteus, as well as other host species, and a morphological study of all available material undertaken. A new species, Progamotaenia mollicula sp. nov., is erected f

Reports of Microthoracius mazzai (Phthiraptera: Anoplura) infestations in alpaca (Vicugna pacos) herds from Austria and Germany.

Sucking lice (Phthiraptera: Anoplura) constitute obligate and permanent insect ectoparasites of mammals and exhibit a hematophagous lifestyle. In South American camelids, three species of the genus Microthoracius have been described, however, reports outside South America are scarce. Here we describe infestations with Microthoracius mazzai in three alpaca (Vicugna pacos) herds from Austria (Styria and Salzburg), and Germany (Bavaria). Overall, lice infestations were detected in eight animals. Four of them exhibited a massive generalized infestation. Lice specimens were identified as M. mazzai

PHYLOGENETIC ANALYSIS OF AEDES AEGYPTI IN TAIF AND JEDDAH GOVERNORATES, SAUDI ARABIA.

With their diverse species, mosquitoes are known to transmit the causal agents of diseases such as malaria, dengue, and yellow fever. Their high adaptability, attraction to humans, and variable adult behaviors make them a significant health concern. The focus on Aedes aegypti is significant for reducing vector-human contacts, monitoring insecticide resistance, and developing innovative vector management strategies. Given the scarcity of studies on Ae. aegypti in the western region of Saudi Arabia, this research is a significant step forward. The study aims to analyze the genetic variations and

Cytochrome oxidase I DNA barcodes of crocodilians meat selling in Hong Kong.

The crocodilians include true crocodiles, alligators, caimans, and gharial, and the trade of crocodilian products is regulated in accordance with the Convention of Wild Fauna and Flora (CITES). Hong Kong does not have her own wild crocodilians; thus, all crocodilians meat available is presumably imported with proper license. Here, we obtained a dataset of cytochrome oxidase I (COI) gene markers of 114 crocodilian meat samples (including frozen and dried crocodilian meat products) available in the contemporary market. We have also validated these barcodes in a phylogenetic approach with other d

A Case of Myiasis Caused by Cordylobia anthropophaga (Diptera: Calliphoridae) in an Italian Traveler Returning from Senegal.

PURPOSE: Myiases are infestations of human and animal tissues by fly larvae. These conditions are widespread in tropical countries and travelers in those areas are at risk of becoming infested. Although Cordylobia anthropophaga (Blanchard & Berenger-Feraud, 1872) is one of the most common myiasis-causing species, few high-quality images and molecular sequences are available for this fly. We present a case of C. anthropophaga infestation in an Italian patient returning from Senegal, with the aim of increasing both visual and molecular data for this species. METHODS: After removal, the larva wa

Investigation of Cerebral Mitochondrial Injury in a Porcine Survivor Model of Carbon Monoxide Poisoning.

INTRODUCTION: Carbon monoxide (CO) is a colorless and odorless gas that is a leading cause of environmental poisoning in the USA with substantial mortality and morbidity. The mechanism of CO poisoning is complex and includes hypoxia, inflammation, and leukocyte sequestration in brain microvessel segments leading to increased reactive oxygen species. Another important pathway is the effects of CO on the mitochondria, specifically at cytochrome c oxidase, also known as Complex IV (CIV). One of the glaring gaps is the lack of rigorous experimental models that may recapitulate survivors of acute C

Identification and molecular characterization of digenean trematode parasites of Aylacostoma chloroticum (Gastropoda: Thiaridae) from a Neotropical Basin.

Thiarids are pantropical gastropods distributed from South America to Australia and found in lentic and lotic freshwater environments. These gastropods act as intermediate hosts for several species of digeneans. There are difficulties in the correct identification of these digeneans in certain stages of the life cycle as the larvae have a reduced size and few morphological characteristics for diagnosis. Because of this, techniques based on molecular markers have become an extremely useful tool. Our objective was to molecularly characterize the digeneans found in gastropods from the Ivaí River,

A NEW GENUS OF DIPLOSTOMIDS (DIGENEA: DIPLOSTOMOIDEA) FROM THE NILE CROCODILE IN SOUTH AFRICA WITH A KEY TO DIPLOSTOMID GENERA.

The Diplostomidae Poirier, 1886 is a large family of digeneans within the superfamily Diplostomoidea Poirier, 1886. Members of the family are distributed worldwide and parasitize a diversity of tetrapod definitive hosts. Notably, only 2 mature diplostomids are known from crocodilians and both are suggested to be accidental infections. In this study, we use morphological and molecular data to describe Neofibricola n. gen. from a Nile crocodile Crocodylus niloticus collected in South Africa. We provide a description of adults and metacercariae of the type species, Neofibricola smiti n. sp., and

Mitochondrial Dysfunction in Cyanotic Congenital Heart Disease: A Promising Therapeutic Approach for the Future.

Congenital heart disease (CHD) is one of the most specific and yet challenging fields of heart surgery. Apart from the known clinical approaches, including surgery, a significant scale of regenerative therapeutic options is available, which increase the number of cardiomyocytes and restore cardiac function. Although it has been revealed in recent years that mitochondrial transplantation can be used as a promising treatment option in this disease group, there is no clinical evidence for the significance of mitochondrial function in myocardial tissue of patients with CHD regarding cardiac surger

Morphometry and genetic diversity pattern of Cysticercus tenuicollis, an important food-borne taeniid metacestode in goats in Bangladesh.

Cysticercus tenuicollis is a larval stage of Taenia hydatigena resulting in cysticercosis, and responsible for enormous economic loses, especially in livestock production. Here, we planned to determine the prevalence and explore genetic variation of C. tenuicollis isolated from goats based on small subunit ribosomal RNA (rrnS) and cytochrome oxidase subunit 1 (cox1). To do this, samples were collected from different slaughter houses of municipal areas such as Bramhapalli slaughterhouse, Jubileeghat slaughterhouse and Mesuabazar slaughterhouse at Mymensingh sadar, and tentatively identified by

Detection of the Japanese encephalitis vector mosquito Culex tritaeniorhynchus in Australia using molecular diagnostics and morphology.

BACKGROUND: Culex (Culex) tritaeniorhynchus is an important vector of Japanese encephalitis virus (JEV) affecting feral pigs, native mammals and humans. The mosquito species is widely distributed throughout Southeast Asia, Africa and Europe, and thought to be absent in Australia. METHODS: In February and May, 2020 the Medical Entomology unit of the Northern Territory (NT) Top End Health Service collected Cx. tritaeniorhynchus female specimens (n = 19) from the Darwin and Katherine regions. Specimens were preliminarily identified morphologically as the Vishnui subgroup in subgenus Culex. Molec

Succinate Can Shuttle Reducing Power from the Hypoxic Retina to the O2-Rich Pigment Epithelium.

When O2 is plentiful, the mitochondrial electron transport chain uses it as a terminal electron acceptor. However, the mammalian retina thrives in a hypoxic niche in the eye. We find that mitochondria in retinas adapt to their hypoxic environment by reversing the succinate dehydrogenase reaction to use fumarate to accept electrons instead of O2. Reverse succinate dehydrogenase activity produces succinate and is enhanced by hypoxia-induced downregulation of cytochrome oxidase. Retinas can export the succinate they produce to the neighboring O2-rich retinal pigment epithelium-choroid complex. Th

Geographical Genetic Variation and Sources of Korean Aedes albopictus (Diptera: Culicidae) Populations.

Aedes albopictus (Skuse, 1894) is a mosquito vector raising global health concerns owing to its transmission of dengue, Zika, and chikungunya viruses. This vector accounts for a large proportion of the Korean mosquito community; however, autochthonous clinical cases resulting from this species remain unreported in South Korea. This study aimed to examine the geographical genetic variations and sources of Ae. albopictus populations in South Korea exclusively on the basis of COI gene analysis from 292 samples collected from 37 localities in 2016-2018 and 290 reference sequences from GenBank. Phylogenetic characterization of Setaria equina and its association with other filarids.

Molecular characterization studies on Setaria equina are limited. The present study aimed to characterize S. equina at the cytochrome c oxidase gene and to examine its phylogenetic relationships with other filarid species. Sequence analysis showed 100% nucleotide homology with an S. equina sequence from Italy (AJ544873). However, both sequences exhibited 7 nucleotide substitutions from a S. equina donkey isolate from Egypt (MK541847). Overall, S. equina formed a monophyletic sister group to Setaria tundra. All Setaria spp. examined formed a separate group on the phylogenetic tree that was rela

G6036A substitution in mitochondrial COX I gene compromises cytochrome c oxidase activity in thiamine responsive Leigh syndrome patients.

Cytochrome c oxidase (COX) deficiency is known to be associated with Leigh syndrome (LS), however there are limited studies on genetic screening of mitochondrial (mt) DNA encoding COX genes as well as the functional validation of identified variants. In our previous studies, we cared for total 165 LS patients and analyzed the nucleotide variations across entire mt genome. We observed a high level of genetic heterogeneity in these patients. We identified various reported and novel variation across entire genome including COX genes. In our present study we have further studied and functionally v

DNA barcoding and fauna of phlebotomine sand flies (Diptera: Psychodidae: Phlebotominae) from Los Tuxtlas, Veracruz, Mexico.

Mexico has great diversity of phlebotomine sand flies related to cases of leishmaniasis, yet few studies have dressed the molecular taxonomy of these sand fly species. The use of the cytochrome oxidase subunit 1 (COI) gene, as a DNA Barcode has facilitated the molecular identification of sand flies species worldwide. We use the DNA barcode as a useful tool for the identification of phlebotomine sand flies of the natural reserve Los Tuxtlas from Veracruz, México. A fragment of 536 bp of the COI gene was obtained from 36 individuals belonging to eight species of five genera (Dampfomyia, Lutzomyi
