m6A is a pervasive post-transcriptional RNA modification that regulates RNA splicing, stability, localization, and translation in the brain. In this review, we outline the core m6A regulatory machinery and summarize its spatial organization across neurons and glial cells, highlighting established roles in brain development, synapse formation, and axon growth. We then focus on experience-dependent plasticity, synthesizing evidence that neuronal activity and environmental inputs dynamically reshape m6A to regulate immediate-early transcription and local translation at synapses across sensory, co

Th2-driven allergic responses are characterized by allergic reactions and immune dysregulation with complex pathogenesis that warrants urgent in-depth study. The predominant posttranscriptional modification observed in eukaryotic messenger RNAs, N6-methyladenosine (m6A), dynamically regulates RNA splicing, which is likely regulated during cell fate determination, cytokine production, and epithelial barrier maintenance in context of Th2-driven allergic responses. This review systematically analyzes the effects of m6A modification on the differentiation and functional regulation of major immune

Massive numbers of modified bases in mRNAs sculpt the epitranscriptome and play vital roles in RNA metabolism. The only known acetylated RNA modification, N-4-acetylcytidine (ac4C), is highly conserved across cell types and among species. Although the GCN5-related acetyltransferase 10 (NAT10) functions as an ac4C writer, the mechanism underlying the acetylation process is largely unknown. In this study, the NAT10/PCBP/TDP43 complex mediated mRNA ac4C formation in mammalian cells is identified. RNA-binding proteins (RBPs) are identified, affiliated with two different families, poly(rC)-binding

Nanopore direct RNA sequencing (DRS) has emerged as a powerful tool for RNA modification identification. However, concurrently detecting multiple types of modifications in a single DRS sample remains a challenge. Here, we develop TandemMod, a transferable deep learning framework capable of detecting multiple types of RNA modifications in single DRS data. To train high-performance TandemMod models, we generate in vitro epitranscriptome datasets from cDNA libraries, containing thousands of transcripts labeled with various types of RNA modifications. We validate the performance of TandemMod on bo

RNA plays essential roles in not only translating nucleic acids into proteins, but also in gene regulation, environmental interactions and many human diseases. Nature uses over 150 chemical modifications to decorate RNA and diversify its functions. With the fast-growing RNA research in the burgeoning field of 'epitranscriptome', a term describes post-transcriptional RNA modifications that can dynamically change the transcriptome, it becomes clear that these modifications participate in modulating gene expression and controlling the cell fate, thereby igniting the new interests in RNA-based dru

FTO (Fat mass and obesity-associated protein) is an RNA demethylase crucial for epitranscriptomic regulation, catalyzing the removal of N6-methyladenosine (m6A) modifications. We designed a series of carbazole derivatives and identified compound K27 as a promising FTO inhibitor, exhibiting an IC50 value of 35.58 ± 5.23 μM. Spectroscopic analyses confirmed the binding of K27 to FTO and revealed ligand-induced changes in the protein microenvironment. Moreover, molecular docking and 300 ns molecular dynamics simulations demonstrated stable binding of K27 within the FTO active site. Collectively,

Fibrosis is an abnormal tissue healing process characterized by the excessive accumulation of ECM components, such as COL I and COL III, in response to tissue injury or chronic inflammation. Recent advances in epitranscriptomics have underscored the importance of m6A modification in fibrosis. m6A, the most prevalent modification in eukaryotic RNA, is catalyzed by methyltransferases (e.g., METTL3), removed by demethylases (e.g., FTO), and recognized by reader proteins (e.g., YTHDF1/2). These modifications are crucial in regulating collagen metabolism and associated diseases. Understanding the r

Perfluorooctanesulfonic acid (PFOS), a persistent per- and polyfluoroalkyl substance, has been associated with male reproductive toxicity, but the underlying molecular mechanisms remain incompletely defined. Here, we show that chronic exposure to environmentally relevant doses of PFOS caused dose-dependent testicular injury and spermatogenic dysfunction in mice, as indicated by reduced testis weight, disrupted seminiferous tubule architecture, and impaired sperm quality. Transcriptomic profiling revealed marked enrichment of pathways related to oxidative stress, mitochondrial dysfunction, and

Traumatic brain injury (TBI) occurs when an external mechanical force damages brain tissue, leading to temporary or lasting disturbances in brain structure and function. The heterogeneous molecular and phenotypic nature of TBI poses a major challenge to translating basic research discoveries into clinically effective interventions. Emerging evidence indicates that epigenetic and epitranscriptomic mechanisms, including histone modifications, DNA methylation, and RNA modifications, play pivotal roles in the molecular response to TBI. In this review, we discuss post-TBI epigenomic alterations wit

N6-methyladenosine (m6A), the most abundant modification in eukaryotic RNAs, plays a critical role in regulating RNA stability, expression, and translation. Key m6A regulators are up- or down-regulated in human gastric cancer tissues, by Helicobacter pylori and transcription factors, and the aberrant expression is associated with poor patient prognosis. Dynamic m6A modification, orchestrated by methyltransferases, demethylases, and m6A-binding proteins, profoundly impacts gastric cancer tumorigenesis, metastasis, cancer cell stemness, immune evasion, and resistance to chemotherapy, radiotherap

Acute kidney injury (AKI) is a clinically critical condition with a high mortality rate. Its complex pathophysiological mechanisms remain incompletely understood, and there is a lack of effective targeted therapeutic strategies. In recent years, the role of epigenetic modifications in the initiation and progression of kidney disease has been increasingly clarified. N6-methyladenosine (m6A) is one of the most important and common post-transcriptional modifications among various RNAs, including eukaryotic mRNAs, lncRNAs, and miRNAs. It is dynamically and reversibly regulated by methyltransferase

Spermatogenic dysfunction refers to a pathological condition characterized by impaired sperm production in the testes, leading to male infertility. This disorder manifests through abnormal semen parameters, testicular lesions, endocrine disturbances, clinical infertility, and genetic defects. Emerging evidence highlights N6-methyladenosine (m6A) RNA methylation as a critical regulator in this process, with its "writer-eraser-reader" machinery playing pivotal roles. This review systematically examines the involvement of m6A regulatory proteins in spermatogenic impairment, providing both theoret

Where a transcript goes among a cell's membraneless compartments-stress granules, P-bodies, paraspeckles, nuclear speckles-is an addressable, rewritable property that this article proposes chemical modifications of RNA help write. The code's output depends on the transcript's role: for client messenger RNAs, an apparent partition coefficient (Kpart), its enrichment in a condensate; for the rare long noncoding RNAs acting as architectural scaffolds, the saturation concentration that decides whether the compartment forms at all. This client/scaffold asymmetry is the framework's central proposal,

Pseudouridine (Ψ) is a prevalent RNA modification whose site-resolved quantification is critical for understanding epitranscriptomic regulation. We present pseU-SCOPE, a ligation-assisted rolling circle amplification (RCA) assay that converts a bisulfite-derived Ψ signature into a padlock-probe ligation gate, enabling single-nucleotide resolution without qPCR. The workflow supports fluorescence and gel-based, naked-eye readouts following RCA and operates with low RNA input. We validate pseU-SCOPE on established Ψ sites across human rRNAs and mitochondrial tRNAs, generate calibration curves for

N6-methyladenosine (m6A) RNA modifications and reovirus infections have recently been implicated in the development of celiac disease. While viral infections are known to alter the RNA modification machinery of the host, their interplay in autoimmune tissue damage remains unexplored. In this study we employed an in vitro model combining a viral mimic with gliadin peptides to simulate celiac disease conditions, alongside the analysis of serum and intestinal biopsies from controls and patients to investigate the link between m6A methylation and viral infections in inducing autoimmune inflammatio
