The role of clinical, genetic and segregation evaluation in sudden infant death.

Sudden infant death syndrome (SIDS) is the leading cause of death in the first year of life. Several arrhythmogenic genes have been associated with cardiac pathologies leading to infant sudden cardiac death (SCD). Our aim was to take advantage of next generation sequencing (NGS) technology to perform a thorough genetic analysis of a SIDS case. A SIDS case was referred to our institution after negative autopsy. We performed a genetic analysis of 104 SCD-related genes using a custom panel. Confirmed variants in index case were also analyzed in relatives. Clinical evaluation of first-degree famil

The Rise of Fentanyl: Molecular Aspects and Forensic Investigations.

Fentanyl is a synthetic opioid widely used for its potent analgesic effects in chronic pain management and intraoperative anesthesia. However, its high potency, low cost, and accessibility have also made it a significant drug of abuse, contributing to the global opioid epidemic. This review aims to provide an in-depth analysis of fentanyl's medical applications, pharmacokinetics, metabolism, and pharmacogenetics while examining its adverse effects and forensic implications. Special attention is given to its misuse, polydrug interactions, and the challenges in determining the cause of death in

Comprehensive body fluid identification and contributor assignment by combining targeted sequencing of mRNA and coding region SNPs.

Forensic genetic analyses aim to retrieve as much information as possible from biological trace material recovered from crime scenes. While standard short tandem repeat (STR) profiling is essential to individualize biological traces, its significance is diminished in crime scenarios where the presence of a suspect's DNA is acknowledged by all parties. In such cases, forensic (m)RNA analysis can provide crucial contextualizing information on the source level about a trace's composition, i.e., body fluids/tissues, and has therefore emerged as a powerful tool for modern forensic investigations. Use of Panoramic Radiography in the Identification of Human Remains - Significance in Forensic Medical Practice.

Genetic testing, although regarded as the gold standard in the identification of human remains of unknown identity, is subject to significant limitations resulting from the degradation of biological material and the absence of comparative reference samples. This paper presents the case of an unidentified male (NN) whose body exhibited extensive mechanical trauma and advanced thermal alterations. The severity of soft‑tissue destruction precluded visual identification, while the lack of reference material rendered DNA analysis impossible. The key diagnostic tool in this case was a comparative as

[Molecular autopsy in sudden cardiac death].

Currently, there are a significant percentage of autopsies left without a conclusive diagnosis of death, especially when this lethal event occurs suddenly. Genetic analysis has been recently incorporated into the field of forensic medicine, especially in patients with sudden death and where no conclusive cause of death is identified after a complete medical-legal autopsy. Inherited arrhythmogenic diseases are the main cause of death in these cases. To date, more than 40 genes have been associated with arrhythmogenic disease, and causing sudden cardiac death has been described. The main arrhyth

A simple Duplex-PCR to evaluate the DNA quality of anthropological and forensic samples prior short tandem repeat typing.

Typing of DNA from ancient or otherwise highly degraded material, e.g. formalin fixed tissues, can be difficult, time consuming and costly. Very often, genetic typing is not possible at all. We present an inexpensive and easy to use Duplex-PCR that amplifies a 164 bp fragment specific for nuclear DNA together with a 260 bp mitochondrial DNA fragment and that can be employed as a pretest prior to short tandem repeat (STR) typing. All together, we analyzed DNA from 20 ancient bones, 20 formalin fixed tissues and 20 other forensic samples in different concentrations. Each sample that failed in th

Cytochrome-b discriminates civet species: A case study of suspected civets in Tamil Nadu, India.

In India, eight civet species have been documented. Among these, the Asian palm civet and the Small Indian civet are most frequently reported in poaching incidents for bushmeat and other purposes. In this study, we examined four processed leg samples (n = 4) seized by the Tamil Nadu Forest Department and suspected to be of civet origin. DNA was extracted and four partial mitochondrial genes (cytochrome b (cyt-b), 12 S ribosomal RNA (12 S rRNA), 16 S ribosomal RNA (16 S rRNA), and cytochrome c oxidase subunit 1 (COXI)) were amplified for species identification. Minimally invasive skeletal sampling for STR-based human identification in degraded marine remains.

The identification of unknown skeletal remains represents a fundamental humanitarian and ethical obligation, particularly in mass fatality events and migration-related disasters, including routine forensic investigations and missing persons cases. DNA analysis is essential in these contexts, yet current sampling methods often rely on invasive procedures that compromise skeletal integrity and conflict with cultural and ethical norms. Despite major advances in forensic DNA analysis, validated strategies for minimally invasive genomic sampling in humanitarian contexts remain limited. In this stud

Autopsies in pandemics - a perspective on barriers and benefits. Is it time for a revival?

Influenza virus and coronavirus pandemics regularly sweep the globe, at great cost of health and economy. Our aim was to conduct a PubMed search for autopsy studies on influenza and coronavirus to investigate the contribution of autopsies during pandemics, focussing on autopsy methods and procedures and the role of autopsy findings in pandemics. The retrieved autopsy studies generally relied on microscopy, polymerase chain reaction (PCR), immunostaining and electron microscopy. Most were small and reported on lung effects, including diffuse alveolar damage (DAD), pneumonia and tracheobronchiti

Genetic identification and subsequent LC-QTOF MS analysis of plant remains (Oenanthe spp.) could prove the cause of an undetermined sudden death.

In recent years, recorded cases related to forensic botany and, in particular, of plant poisoning have become rare. We report on the medicolegal characteristics of an undetermined sudden death (USD) of a woman in which scene there were remnants of a vegetal peeling. After the autopsy, macroscopic findings reported multiorgan failure and requested the investigation of the cause of death. Postmortem blood was firstly investigated on cyanide toxicity presumptively coming from a yucca-like root; however, found cyanide levels were under normality. Because of the lack of morphological features of th

Anthropological analyses of 30 insertion/deletion autosomal markers in five major ethnic groups of Pakistan.

We investigated the forensic efficacy of the 30 insertion/deletion (Indel) markers included in the Qiagen Investigator® DIPplex kit in 529 Pakistani individuals from five major subpopulations in Pakistan (Punjabi, Pashtun, Sindhi, Saraiki, and Baloch). In the Sindhi population, the distribution of HLD81 and HLD97 alleles deviated from Hardy-Weinberg equilibrium after Bonferroni correction. The combined match probability ranged from 2.0E-12 (Pashtun and Baloch) to 1.0E-12 (Sindhi), and the mean paternity exclusion power varied from 0.995 (Punjabi, Sindhi, and Saraiki) to 0.996 (Pashtun and Balo

A preliminary study on identification of the blood donor in a body fluid mixture using a novel compound genetic marker blood-specific methylation-microhaplotype.

Blood-containing mixtures are frequently encountered at crime scenes involving violence and murder. However, the presence of blood, and the association of blood with a specific donor within these mixtures present significant challenges in forensic analysis. In light of these challenges, this study sought to address these issues by leveraging blood-specific methylation sites and closely linked microhaplotype sites, proposing a novel composite genetic marker known as "blood-specific methylation-microhaplotype". This marker was designed to the detection of blood and the determination of blood don

Some mathematical problems in the DNA identification of victims in the 2004 tsunami and similar mass fatalities.

DNA is a major and essential identification tool for mass fatality incidents including the hundreds of thousands of victims of the 2004 Indian Ocean tsunami. Mathematical complications characteristic of this sort of mass fatality include prevalence of related victims, the many races represented among the victims, and various identification modalities in tandem with DNA. Four mathematical problems of interest are discussed in this paper. (1) Other quantifiable factors (i.e. geography) can be formally accounted for by including a likelihood ratio that can be thought of as reducing the "effective

Inter-population variation in third molar agenesis and its effect on dental age estimation: Evidence from black and white South Africans.

Third molar development is widely used for dental age estimation (DAE) in late adolescence and young adulthood; however, agenesis of one or more third molars can compromise its reliability in forensic practice. The prevalence of third molar agenesis is known to vary across population groups, yet its practical impact on forensic age assessment remains insufficiently explored. This study aimed to assess the prevalence and distribution of third molar agenesis in Black South Africans (BSA) and White South Africans (WSA) and to assess its impact on DAE according to sex, tooth position, and birth co

Applicability of imprinted DIP-SNP markers in DNA mixture analysis: a pilot study.

In recent years, a new compound genetic marker, deletion/insertion polymorphism-single nucleotide polymorphism (DIP-SNP), has been applied in forensic DNA mixture analysis. DIP-SNP genotyping based on capillary electrophoresis (CE) typically requires two separate amplification reactions and is difficult to interpret in complex DNA mixtures. In this study, four candidate imprinted DIP-SNP markers were selected to establish a multiplex polymerase chain reaction (PCR) set based on the amplification refractory mutation system (ARMS) principle for a one-tube reaction. Furthermore, parentally imprin

Invisible impact: 3D scanning sprays do not inhibit forensic STR analysis.

The 3D reconstruction of crime scenes or evidentiary objects is becoming a widely adopted forensic technique. Scanning methods struggle with transparent or reflective surfaces. A solution to this problem is the application of 3D scanning sprays, which create a matte layer on the surface of the examined object, which later sublimes. Our study aimed to examine the effects of different 3D modeling sprays on DNA recovery and forensic autosomal STR analysis. We simulated crime scene samples by depositing blood droplets or by touching separate microscope slides. Following the spray treatment, DNA wa

[Alleles dropout patterns of Y-short tandem repeats in infertile males with Y chromosome microdeletions].

Y chromosomal short tandem repeat (Y-STR) typing is the most commonly used genetic technique in forensic studies. However, there may be a limit to the application of Y-STR in forensic science as Y-STR loci are subject to loss or variation caused by the higher chromosomal structures' spontaneous mutation rate. Located in the long arm of the Y chromosome, azoospermia factor (AZF) have been shown to participate in spermatogenesis and its deletion could cause infertility. However, little is known about the Y-STR dropout pattern in individuals with Y chromosome microdeletions. In this study, 85 inf

Sudden Unexpected Death in Fetal Life Through Early Childhood.

In March 2015, the Eunice Kennedy Shriver National Institute of Child Health and Human Development held a workshop entitled "Sudden Unexpected Death in Fetal Life Through Early Childhood: New Opportunities." Its objective was to advance efforts to understand and ultimately prevent sudden deaths in early life, by considering their pathogenesis as a potential continuum with some commonalities in biological origins or pathways. A second objective of this meeting was to highlight current issues surrounding the classification of sudden infant death syndrome (SIDS), and the implications of variation

HIV forensics: pitfalls and acceptable standards in the use of phylogenetic analysis as evidence in criminal investigations of HIV transmission.

BACKGROUND: Phylogenetic analysis - the study of the genetic relatedness between HIV strains - has recently been used in criminal prosecutions as evidence of responsibility for HIV transmission. In these trials, the expert opinion of virologists has been of critical importance. PITFALLS: Phylogenetic analysis of HIV gene sequences is complex and its findings do not achieve the levels of certainty obtained with the forensic analysis of human DNA. Although two individuals may carry HIV strains that are closely related, these will not necessarily be unique to the two parties and could extend to
