Fucosylation deficiency enhances imiquimod-induced psoriasis-like skin inflammation by promoting CXCL1 expression.

Psoriasis is a multifaceted chronic inflammatory skin disease; however, its underlying molecular mechanisms remain unclear. In this study, we explored the role of fucosylation in psoriasis using an imiquimod-induced psoriasis-like mouse model. ABH antigen and fucosyltransferase 1 (Fut1) expression was reduced in the granular layer of lesional skin of patients with psoriasis. In particular, the blood group H antigen type 2 (H2 antigen)-a precursor of blood group A and B antigens-and FUT1 were highly expressed throughout the spinous layer in both patients with psoriasis and the skin of imiquimod

Enhanced targeted DNA methylation of the CMV and endogenous promoters with dCas9-DNMT3A3L entails distinct subsequent histone modification changes in CHO cells.

With the emergence of new CRISPR/dCas9 tools that enable site specific modulation of DNA methylation and histone modifications, more detailed investigations of the contribution of epigenetic regulation to the precise phenotype of cells in culture, including recombinant production subclones, is now possible. These also allow a wide range of applications in metabolic engineering once the impact of such epigenetic modifications on the chromatin state is available. In this study, enhanced DNA methylation tools were targeted to a recombinant viral promoter (CMV), an endogenous promoter that is sile

Novel Antibodies Reactive with Sialyl Lewis X in Both Humans and Mice Define Its Critical Role in Leukocyte Trafficking and Contact Hypersensitivity Responses.

Sialyl Lewis X (sLe(x)) antigen functions as a common carbohydrate determinant recognized by all three members of the selectin family. However, its expression and function in mice remain undefined due to the poor reactivity of conventional anti-sLe(x) monoclonal antibodies (mAbs) with mouse tissues. Here, we developed novel anti-sLe(x) mAbs, termed F1 and F2, which react well with both human and mouse sLe(x), by immunizing fucosyltransferase (FucT)-IV and FucT-VII doubly deficient mice with 6-sulfo-sLe(x)-expressing cells transiently transfected with an expression vector encoding CMP-N-acetyln

Prognostic implications of CD15 antigen expression in patients with acute myeloid leukemia.

The prognostic significance of CD15 antigen in acute myeloid leukemia has been evaluated with conflicting results in various studies. However, existing results are limited and further investigation into additional prognostic parameters is necessary. This study aimed to evaluate the impact of CD15 antigen on survival prognosis in patients with de novo AML (dn-AML). We analyzed CD15 antigen expression on diagnostic samples from 676 patients with dn-AML by flow cytometry. Utilizing a 20% positivity threshold, the cohort was stratified into CD15 positivity (56.07%) and CD15 negativity (43.93%) gro

Fertility in classical galactosaemia, a study of N-glycan, hormonal and inflammatory gene interactions.

BACKGROUND: Classical Galactosaemia (CG) (OMIM #230400) is a rare inborn error of galactose metabolism caused by deficiency of the enzyme galactose-1-phosphate uridylyltransferase (GALT). Long-term complications persist in treated patients despite dietary galactose restriction with significant variations in outcomes suggesting epigenetic glycosylation influences. Primary Ovarian Insufficiency (POI) is a very significant complication affecting females with follicular depletion noted in early life. We studied specific glycan synthesis, leptin system and inflammatory gene expression in white bloo

Endothelial POFUT1 controls injury-induced liver fibrosis by repressing fibrinogen synthesis.

BACKGROUND & AIMS: NOTCH signaling in liver sinusoidal endothelial cells (LSECs) regulates liver fibrosis, a pathological feature of chronic liver diseases. POFUT1 is an essential regulator of NOTCH signaling. Here, we investigated the role of LSEC-expressed POFUT1 in liver fibrosis. METHODS: Endothelial-specific Pofut1 knockout mice were generated and experimental liver fibrosis was induced by chronic carbon tetrachloride exposure or common bile duct ligation. Liver samples were assessed by ELISA, histology, electron microscopy, immunostaining and RNA in situ hybridization. LSECs and hepatic

Effect and Mechanism Analysis of Pig FUT8 Gene on Resistance to Escherichia coli F18 Infection.

Post-weaning diarrhea caused by enterotoxigenic Escherichia coli F18 (E. coli F18) causes significant economic losses for pig producers. Fucosyltransferase 8 (FUT8) is a glycosyltransferase that catalyzes core fucosylation; however, its role in mediating the resistance to E. coli F18 infection in pigs remains unknown. In this study, we systematically verified the relationship between FUT8 expression and E. coli resistance. The results showed that FUT8 was expressed in all detected tissues of Meishan piglets and that its expression was significantly increased in the duodenum and jejunum of E. c

Norovirus and histo-blood group antigens.

Norovirus (NoV), a member of the family Caliciviridae, is a major cause of acute water- and food-borne nonbacterial gastroenteritis and forms antigenically diverse groups of viruses. Human NoVs are divided into at least three genogroups, genogroups I (GI), GII, and GIV, which contain at least 15, 18, and 1 genotypes, respectively. Except for a few genotypes, all NoVs bind to histo-blood group antigens (HBGAs), namely ABH antigens and Lewis antigens, in which carbohydrate core structures, (types 1 and 2) constitute antigenically distinct phenotypes. Volunteer challenge studies have indicated th

Genetic and biochemical evidence that gastrulation defects in Pofut2 mutants result from defects in ADAMTS9 secretion.

Protein O-fucosyltransferase 2 (POFUT2) adds O-linked fucose to Thrombospondin Type 1 Repeats (TSR) in 49 potential target proteins. Nearly half the POFUT2 targets belong to the A Disintegrin and Metalloprotease with ThromboSpondin type-1 motifs (ADAMTS) or ADAMTS-like family of proteins. Both the mouse Pofut2 RST434 gene trap allele and the Adamts9 knockout were reported to result in early embryonic lethality, suggesting that defects in Pofut2 mutant embryos could result from loss of O-fucosylation on ADAMTS9. To address this question, we compared the Pofut2 and Adamts9 knockout phenotypes an

Profile of human milk oligosaccharides and maternal FUT2 polymorphism: genotype-phenotype correlation and proposed cut-off for secretor status.

BACKGROUND: Human milk oligosaccharides (HMO) are complex carbohydrates which is undigestible to humans and confer health benefits. The synthesis of HMOs is determined by fucosyltransferase enzymes, which are encoded by FUT2/FUT3 genes. Polymorphisms of the FUT2 result in various secretor status. This study aimed to identify the proportion of secretor and FUT2 genotype-phenotype correlation in Indonesian mothers. METHODS: This was a cross-sectional study, recruiting 120 healthy mothers aged at least 18 years who exclusively breastfed. Breastmilk samples were obtained at infant's age 2-5 weeks

Prognostic Value of Combined Carbohydrate Antigen 19-9 and Duke Pancreatic Monoclonal Antigen Type 2 Assessment in Biliary Tract Cancer.

BACKGROUND: Carbohydrate antigen 19-9 (CA19-9) represents an established prognostic biomarker for biliary tract cancers (BTCs). Clinical utility remains limited among patients with Lewis antigen negativity or incomplete fucosyltransferase 3 (FUT3) gene function, conditions associated with absent or reduced CA19-9 production. Duke pancreatic monoclonal antigen type 2 (DUPAN-2), a biosynthetic precursor of CA19-9, shows compensatory elevation under such conditions. This study evaluated the prognostic significance of the combined preoperative assessment of CA19-9 and DUPAN-2 among patients with r

Core fucosylation regulates the ovarian response via FSH receptor during follicular development.

INTRODUCTION: Ovarian low response to follicle-stimulating hormone (FSH) causes infertility featuring hypergonadotropic hypogonadism, ovarian failure, and/or defective ovarian response. OBJECTIVES: N-glycosylation is essential for FSH receptor (FSHR). Core fucosylation catalyzed by fucosyltransferase 8 (FUT8) is the most common N-glycosylation. Core fucosylation level changes between individuals and plays important roles in multiple physiological and pathological conditions. This study aims to elucidate the significance of FUT8 to modulate FSHR function in female fertility. METHODS: Samples

Vesicular Integral-Membrane Protein 36 Is Involved in the Selective Secretion of Fucosylated Proteins into Bile Duct-like Structures in HepG2 Cells.

Fucosylated proteins are widely used as biomarkers of cancer and inflammation. Fucosylated alpha-fetoprotein (AFP-L3) is a specific biomarker for hepatocellular carcinoma. We previously showed that increases in serum AFP-L3 levels depend on increased expression of fucosylation-regulatory genes and abnormal transport of fucosylated proteins in cancer cells. In normal hepatocytes, fucosylated proteins are selectively secreted in the bile duct but not blood. In cases of cancer cells without cellular polarity, this selective secretion system is destroyed. Here, we aimed to identify cargo proteins

Gnotobiotic transgenic mice reveal that transmission of Helicobacter pylori is facilitated by loss of acid-producing parietal cells in donors and recipients.

Helicobacter pylori is acquired during childhood, but its mode of transmission remains unclear. A genotyped H. pylori isolate (Hp1) that expresses two classes of adhesins was introduced into the stomachs of three types of germ-free FVB/N mice to model factors that may affect spread of H. pylori in humans. Normal mice represented human hosts with normal gastric acid production. Transgenic animals expressing human alpha-1,3/4-fucosyltransferase in their gastric pit cells represented humans with normal acid production and the commonly encountered Lewis(b) histo-blood group receptor for the bacter

Leukocyte rolling velocities and migration are optimized by cooperative L-selectin and intercellular adhesion molecule-1 functions.

Selectin family members largely mediate initial tethering and rolling of leukocytes on vascular endothelium, whereas integrin and Ig family members are essential for leukocyte firm adhesion. To quantify functional synergy between L-selectin and Ig family members during leukocyte rolling, the EA.hy926 human vascular endothelial line was transfected with either fucosyltransferase VII (926-FtVII) cDNA to generate L-selectin ligands alone or together with ICAM-1 cDNA (926-FtVII/ICAM-1). The ability of transfected 926 cells to support human leukocyte interactions was assessed in vitro using paralle

Structures, function, and transformational changes of the sugar chains of glycohormones.

Human chorionic gonadotropin (hCG), human luteinizing hormone, human thyroid-stimulating hormone, and human follicle-stimulating hormone are closely related family of proteins which share a common alpha-subunit. However, their sugar moieties are quite different. hCG contains five acidic asparagine-linked sugar chains. These five sugar chains are derived by sialylation from three neutral oligosaccharides: two biantennary (N-1 and N-2) and one monoantennary (N-3) complex-type oligosaccharides. Although hCG purified from the urine of pregnant women is more enriched in sialylated sugar chains than

Usefulness of CD56, BRAF and CD15 in the differential diagnosis of thyroid lesions with nuclear features of papillary carcinoma.

INTRODUCTION: Differential diagnosis of thyroid lesions with papillary carcinoma-like nuclear features includes low-risk entities, such as follicular thyroid neoplasia with papillary-like nuclear features (NIFTP), and high-risk entities, such as the tall cell subtype of papillary thyroid carcinoma (TCS). Morphological criteria are essential to distinguish them, but having immunohistochemical techniques to discriminate between these entities would be desirable. MATERIALS AND METHODS: 8 TCS, 27 papillary carcinomas of classic subtype (CS) and 20 NIFTP were selected. Immunohistochemical staining

CD15 as a marker of fetoplacental endothelial immaturity in IUGR placentas.

BACKGROUND: Structural or functional defects in the placenta, are the primary cause of growth restriction of the fetus. Morphological examination of such placentas from intrauterine growth restricted (IUGR) fetuses often appears deceptively normal. Evaluation of angiogenesis and fetoplacental vasculature is critical to understand the underlying pathogenesis of fetal growth restriction in both idiopathic as well as cases where it is thought to be secondary to complications like preeclampsia (PE). We analyzed the immaturity of fetoplacental vasculature using CD15, which is a stage specific embry

Lysophosphatidic acid-induced Arf6-driven macropinocytosis of CD147+ extracellular vesicles promotes sorafenib resistance of hepatocellular carcinoma.

Background: Transarterial chemoembolization (TACE) combined with sorafenib is a common therapeutic strategy for hepatocellular carcinoma (HCC). However, sorafenib resistance (SFR) remains a major clinical obstacle. Evidence suggest that TACE reshapes the tumor microenvironment (TME), creating an external high-glucose (HG) and internal low-glucose (LG) niche. In this context, hyperglycemia-driven lysophosphatidic acid (LPA) production accelerates HCC progression. Moreover, intercellular communication via extracellular vesicles (EVs) has been linked to drug resistance. Despite these insights, th
