Mycotoxins in food and agriculture: from occurrence to economic consequences.

Filamentous fungus (moulds) create secondary metabolites called mycotoxins, which can be dangerous. These minuscule molecules (typically less than 1 000 Da) are almost inescapably found in nature. Aspergillus, Penicillium, Fusarium, and Alternaria are among the fungal species that produce mycotoxins. If these secondary metabolites make their way into the food chain and production, they are harmful and have a big effect. In African countries like Nigeria that rely significantly on staple foods like grains, cereals, and nuts, mycotoxin contamination is especially problematic. Mycotoxins have att

Aflatoxins as risk factors for hepatocellular carcinoma in humans.

On a global basis, primary liver cancer (PLC) is a very prevalent form of cancer. Wide variation of PLC incidence in different areas of the world suggests the involvement of environmental factors in its etiology. Two major classes of risk factors have been identified. Extensive evidence indicates the importance of infection by the hepatitis B virus as a major risk factor for PLC. Because many organic chemicals induce liver cancer in experimental animals, those to which human exposure is known to occur are also of interest with respect to their possible involvement as risk factors for PLC. A Universal Near-Zero-Background Biosensing Platform: Engineered p-MXene/Quantum Dot@Aptamer Nanocomposites for Ultrasensitive Molecular Detection.

MXene-based fluorescent aptasensors detect targets through MXene-mediated adsorption of labeled aptamer probes and subsequent target-induced release of the probe signals, providing a promising programmable detection approach. However, their sensitivity is limited by the low signal output per aptamer or the incomplete quenching of the ultrabright aptamer probes by MXene. This study proposes an advanced MXene-based detection platform that addresses these challenges through material engineering and innovative sensing strategies. Polydopamine-mediated interface engineering endows monolayer MXene n

Porcine intestinal epithelial barrier disruption by the Fusarium mycotoxins deoxynivalenol and T-2 toxin promotes transepithelial passage of doxycycline and paromomycin.

BACKGROUND: The gastrointestinal tract is the first target for the potentially harmful effects of mycotoxins after intake of mycotoxin contaminated food or feed. With deoxynivalenol (DON), T-2 toxin (T-2), fumonisin B1 (FB1) and zearalenone (ZEA) being important Fusarium toxins in the northern hemisphere, this study aimed to investigate in vitro the toxic effect of these mycotoxins on intestinal porcine epithelial cells derived from the jejunum (IPEC-J2 cells). Viability of IPEC-J2 cells as well as the proportion of apoptotic and necrotic IPEC-J2 cells was determined by flow cytometry after 72

Selective inhibition of carbonic anhydrase IX decreases cell proliferation and induces ceramide-mediated apoptosis in human cancer cells.

Recently, carbonic anhydrase (CA) inhibitors have been proposed as a potential new class of antitumor agents. The aim of this study was to evaluate the antitumor activity of three CA inhibitors, namely acetazolamide (AZ) and two newly synthesized aromatic sulfonamides with high affinity for CA IX, 2-(4-sulfamoylphenyl-amino)-4,6-dichloro-1,3,5-triazine (TR1) and 4-[3-(N,N-dimethylaminopropyl)thioreidophenylsulfonylaminoethyl]benzenesulfonamide (GA15), against human tumor cells. The effects of AZ, TR1, and GA15 on cell proliferation and apoptosis were evaluated in CA IX-positive HeLa and 786-O

Modulating effects of mycotoxin and oxidized oil on intestinal microbiota in broiler chickens.

Climatic change and increased use of alternative sources of feed ingredients could influence poultry production. Mycotoxin and oxidized oil are two contaminations that may occur in chicken feed as a result of climate change and use of alternative feed ingredients, and these factors may have differential and potentially additive effects on birds' intestinal microbiota. The study objective was to determine the main effects of corn, oil quality, and their interaction on ileal content, ileal scrapings, cecal content, and whole cecum (content and tissue) microbiota in broiler chickens. Behavioral toxicity of TDCPP in marine zooplankton: Evidence from feeding and swimming responses, molecular dynamics and metabolomics of rotifers.

As new organic flame retardants, chlorinated organophosphate esters (Cl-OPEs) have high water solubility and structural similarity to organophosphate pesticides, posing risks to aquatic organisms. The potential neurotoxicity of Cl-OPEs has attracted attention, especially in marine invertebrates with a relatively simple nervous system. In this study, a marine rotifer with a cerebral ganglion, Brachionus plicatilis, was exposed to tris (1,3-dichloro-2-propyl) phosphate (TDCPP) (two environmental concentrations and one extreme level), and the changes in feeding and swimming behaviors and internal

The SiaABC threonine phosphorylation pathway controls biofilm formation in response to carbon availability in Pseudomonas aeruginosa.

The critical role of bacterial biofilms in chronic human infections calls for novel anti-biofilm strategies targeting the regulation of biofilm development. However, the regulation of biofilm development is very complex and can include multiple, highly interconnected signal transduction/response pathways, which are incompletely understood. We demonstrated previously that in the opportunistic, human pathogen P. aeruginosa, the PP2C-like protein phosphatase SiaA and the di-guanylate cyclase SiaD control the formation of macroscopic cellular aggregates, a type of suspended biofilms, in response t

Indigenous aseel chicken-derived probiotics as biofactories of antifungal metabolites to control mycotoxin contamination in poultry feed.

BACKGROUND: Fungal and mycotoxin contamination threaten poultry health, feed safety, and global food security. Mycotoxins result in reduced poultry growth, nutrient absorption, and a compromised immune system, causing severe financial losses. The rise of antibiotic resistance due to excessive antibiotic use in poultry also necessitates alternative strategies. METHODS AND RESULTS: Total 28 strains were isolated from the gut of indigenous Aseel and broiler chickens. Strains SB1, SB2, and SD2 completely inhibited Aspergillus flavus, Aspergillus ochraceus, and Fusarium proliferatum and were asses

The Pathology and Blood Biochemistry of Juvenile Lates calcarifer on Diets Contaminated With Mycotoxins, Histamines and Rancid Fats-A Case Study.

Mycotoxins, originating from contaminated raw materials or suboptimal feed storage, are a growing concern in tropical aquaculture. Common fungi such as Aspergillus spp. and Fusarium spp. produce mycotoxins including aflatoxin, fumonisin, deoxynivalenol and zearalenone. High doses or prolonged exposure (weeks) to low doses of these mycotoxins (< 20 μg/kg) can depress growth, immunity, and cause mortality. This study investigated poor growth and low survival in juvenile Asian seabass, Lates calcarifer fed two different diets (FM40 and ABS3) for 5 weeks. Analysis of these diets revealed high pero

Mycotoxin exposure and infant and young child growth in Africa: what do we know?

INTRODUCTION: Infant and young child (IYC) growth impairment remains a public health problem in Africa partly because infants are exposed to staple foods (contaminated with mycotoxins) at an early age. Understanding the role of mycotoxins in IYC growth is vital, and this paper systematically reviews the available knowledge. METHODS: Studies were searched and included if they provided information on African IYC mycotoxin exposure rates and/or growth. Studies were excluded if subjects were older than 15 years, if they were animal studies or focusing on other mycotoxins. Relevant search words we

Computer aided boar semen motility analysis for cereulide detection in different food matrices.

Computer Aided Semen Analysis (CASA) study of the boar semen motility has been demonstrated to be an appropriate assay for detection of cereulide (Bacillus cereus emetic toxin). Application of the boar semen bio-assay to detect cereulide directly in foods requires investigation of potential interference of food components, preservatives and other microbial and chemical food contaminants with the bio-assay. Current study provides evidence that none of included Staphylococcus aureus enterotoxins A, B, C and D nor B. cereus Hemolysin BL (HBL) and non-hemolytic enterotoxin (NHE) and three mycotoxi

Cationic amphiphilic drugs inhibit the internalization of cholera toxin to the Golgi apparatus and the subsequent elevation of cyclic AMP.

Cholera toxin (CT) consists of a pentameric B subunit which binds with high affinity to ganglioside GM1, and an A subunit which stimulates adenylate cyclase, resulting in the elevation of cAMP. We now examine the effect of cationic amphiphilic drugs (CADs) on the internalization of rhodamine (Rh)-CT in cultured hippocampal neurons. CADs have recently been shown to inhibit receptor recycling by disrupting the assembly-disassembly of clathrin at the plasma membrane and on endosomes (Wang, L.-H., Rothberg, K. G., and Anderson, R. G. W. (1993) J. Cell Biol. 123, 1107-1117). Rh-CT was internalized

MdUGT88F1 enhances plant resistance to Fusarium proliferatum f.sp. malus domestica MR5 via root exudate-mediated assembly of disease-suppressive rhizosphere microbiota.

BACKGROUND: Apple replant disease (ARD) is a major threat to the sustainable development of China's apple industry. It is primarily caused by the accumulation of phloridzin and the pathogen Fusarium proliferatum f.sp. malus domestica MR5 (Fpmd MR5). MdUGT88F1-mediated phloridzin biosynthesis is known to enhance disease resistance, but its role in shaping the rhizosphere microbiome and conferring resistance against Fpmd MR5 remains unclear. In this study, we used wild-type (WT) and MdUGT88F1 transgenic apple lines to systematically investigate the mechanism by which MdUGT88F1 regulates the rhiz

Efficacy of a mycotoxin-deactivating product to reduce the impact of Fusarium mycotoxin-contaminated rations in dairy cows during early lactation.

Livestock feeds can be contaminated by several fungi. Among these, Fusarium spp. are widespread and produce mycotoxins, such as deoxynivalenol (DON), zearalenone (ZEN), and fumonisins B1 and B2 (FB1 and FB2). We investigated whether feeding a TMR contaminated with moderate to high concentrations of ZEN, DON, FB1, and FB2 to dairy cows in early lactation alters (1) their feeding behavior and rumination time; (2) milk yield, composition, and quality; (3) plasma metabolic profile and biochemical traits; and (4) postpartum uterine involution and resumption of cyclicity. We tested whether adding a

The genome, transcriptome and metabolome of Aspergillus welwitschiae Ocstreb1 from wild halophytic rice, Oryza coarctata, indicates its distinctiveness.

Aspergillus welwitschiae is a widespread fungus with diverse roles as a plant mutualist, opportunistic human pathogen and industrial enzyme producer. The endophytic strain AwOcstreb1, isolated from halophytic rice (Oryza coarctata), promotes growth in commercial rice under normal and saline conditions. Despite its significance, genomic and metabolic resources for A. welwitschiae remain limited, with no complete genome information available for endophytic strains within the species. Moreover, the close relationship of this species to Aspergillus niger complicates its taxonomic resolution. We pe
