The St George's Classification Algorithm of Primary Lymphatic Anomalies.

Clinicians and scientists at St George's University Hospital have collaborated to develop a classification algorithm for primary lymphatic anomalies. Instruction is offered on how to apply the algorithm in clinical practice to refine the diagnosis of primary lymphedema and guide on genetic testing and management. It can also be used to interpret mutation testing results of uncertain significance. The algorithm has evolved as more genes have been discovered, and it remains a "work in progress" as further discoveries are made. This transformational approach has revolutionized the understanding a

The Value of Histological Algorithms to Predict the Malignancy Potential of Pheochromocytomas and Abdominal Paragangliomas-A Meta-Analysis and Systematic Review of the Literature.

Pheochromocytomas (PCCs) and abdominal paragangliomas (PGLs), collectively abbreviated PPGLs, are neuroendocrine tumors of the adrenal medulla and paraganglia, respectively. These tumors exhibit malignant potential but seldom display evidence of metastatic spread, the latter being the only widely accepted evidence of malignancy. To counter this, pre-defined histological algorithms have been suggested to stratify the risk of malignancy: Pheochromocytoma of the Adrenal Gland Scaled Score (PASS) and the Grading system for Adrenal Pheochromocytoma and Paraganglioma (GAPP). The PASS algorithm was o

Diagnostic laboratory testing for Charcot Marie Tooth disease (CMT): the spectrum of gene defects in Norwegian patients with CMT and its implications for future genetic test strategies.

BACKGROUND: Current genetic test algorithms for Charcot Marie Tooth (CMT) disease are based on family details and comprehensive clinical and neurophysiological data gathered under ideal conditions for clinical assessment. However, in a diagnostic laboratory setting relying on external test requisitions and patient samples, such conditions are not always met. Our objective was therefore to perform a retrospective evaluation of the data given in laboratory request forms and to assess their quality and applicability with regard to the recommended algorithms for CMT diagnostics. As we are the main

PGpath: recommending population-adapted linear reference genomes from pangenome subpaths.

Large-scale population sequencing requires reference genomes that accurately represent the genetic background of target populations. Although high-quality linear references such as T2T-CHM13 have improved the completeness of human genome representation, a single reference genome remains insufficient to capture genetic diversity across populations. Pangenome graphs provide a richer representation of multi-source genomic variation, but translating pangenome diversity into practical reference genomes remains challenging. First, the direct application of pangenome graphs in routine large-scale seq

A fast method for extracting essential and synthetic lethality genes in GEM models.

SUMMARY: Exploring and categorizing essential and synthetic lethality genes is crucial in developing effective and targeted therapies for various diseases. This endeavor hinges upon genetic minimal cut sets, which also find utility in metabolic engineering. Different methods have been suggested for calculating genetic minimal cut sets. Still, with the emergence of numerous new models and their increasing complexity, it has become essential to introduce new algorithms in this field. This paper presents a new algorithmic approach for computing genetic minimal cut sets, which utilizes linear prog

Cherry picking: a characterization of the temporal hybridization number for a set of phylogenies.

Recently, we have shown that calculating the minimum-temporal-hybridization number for a set [Formula: see text] of rooted binary phylogenetic trees is NP-hard and have characterized this minimum number when [Formula: see text] consists of exactly two trees. In this paper, we give the first characterization of the problem for [Formula: see text] being arbitrarily large. The characterization is in terms of cherries and the existence of a particular type of sequence. Furthermore, in an online appendix to the paper, we show that this new characterization can be used to show that computing the min

Clinical Approach to Genetic Cerebral Arteriopathy in the Adult Patient With Ischemic Stroke.

Genetic arteriopathies leading to stroke in adults constitute a diverse group of cerebrovascular disorders with distinct etiologies, pathophysiologic mechanisms, and clinical presentations. As imaging modalities better delineate subtle changes in cerebral vasculature and access to genetic testing increases, the detection rate for these conditions is expected to rise, particularly among young adults with idiopathic cerebral arteriopathy and stroke. Adults with stroke in the setting of a genetic cerebral arteriopathy often present with few traditional stroke risk factors and, in certain cases, h

Crohn's Disease: Genetics Update.

Since the discovery of the first Crohn's disease (CD) gene NOD2 in 2001, 140 genetic loci have been found in whites using high-throughput genome-wide association studies. Several genes influence the CD subphenotypes and treatment response. With the observations of increasing prevalence in Asia and developing countries and the incomplete explanation of CD variance, other underexplored areas need to be integrated through novel methodologies. Algorithms that incorporate specific genetic risk alleles with other biomarkers will be developed and used to predict CD disease course, complications, and

Plan of Action for Inherited Cardiovascular Diseases: Synthesis of Recommendations and Action Algorithms.

The term inherited cardiovascular disease encompasses a group of cardiovascular diseases (cardiomyopathies, channelopathies, certain aortic diseases, and other syndromes) with a number of common characteristics: they have a genetic basis, a familial presentation, a heterogeneous clinical course, and, finally, can all be associated with sudden cardiac death. The present document summarizes some important concepts related to recent advances in sequencing techniques and understanding of the genetic bases of these diseases. We propose diagnostic algorithms and clinical practice recommendations and

Linear quadratic regulator design via metaheuristics applied to the damping of low-frequency oscillations in power systems.

This paper proposes the application of control by state feedback using the linear quadratic regulator (LQR) optimized by metaheuristics to damp low-frequency electromechanical oscillations in electrical power systems. The current sensitivity model was used to represent the single machine infinite bus (SMIB) system in the time domain. The weighting matrices of the LQR were adjusted using four different algorithms: (i) the genetic algorithm, (ii) the differential evolution algorithm, (iii) the particle swarm optimization algorithm, and (iv) the gray wolf optimization (GWO) algorithm. In the case

Artificial intelligence optimized image segmentation techniques for renal cyst detection.

The vast number of image modalities available nowadays has given rise and access to a number of medical images. These images perhaps suffer issues such as low contrast, noise, ill-defined boundaries and poor visualisation. Therefore, a need for effective segmentation arises. Medical image segmentation plays a significant role in identifying a disorder, treatment planning, routine follow ups and computer-guided surgery respectively. The paper presents automatic medical image segmentation to overcome the imaging concerns and demarcate each notch & boundary in an image. The proposed algorithm ide

Clinical Model for Predicting Warfarin Sensitivity.

Warfarin is a widely used anticoagulant with a narrow therapeutic index and large interpatient variability in the therapeutic dose. Complications from inappropriate warfarin dosing are one of the most common reasons for emergency room visits. Approximately one third of warfarin dose variability results from common genetic variants. Therefore, it is very necessary to recognize warfarin sensitivity in individuals caused by genetic variants. Based on combined polymorphisms in CYP2C9 and VKORC1, we established a clinical classification for warfarin sensitivity. In the International Warfarin Pharma

Clinical, immunophenotypic, and genetic analysis of adult lymphomas with morphologic features of Burkitt lymphoma.

A prompt distinction of Burkitt lymphoma (BL) versus diffuse large B cell lymphomas (DLBCL) has important clinical implications; however, this distinction can be difficult. We analyzed 74 adult gray zone and 10 reference pediatric BL using immunohistochemistry (Ki-67, CD10, bcl2, bcl6) and fluorescence in situ hybridization (FISH) for MYC, BCL2, and BCL6 breakpoints. Two algorithms for classification were followed: algorithm A used a two-step review by four hemato-pathologists and algorithm B a set of only biologic markers (Ki-67 > or = 90%, CD10+, bcl6+, bcl2-, MYC breakpoint+, BCL2 and BCL6

A genetic features and gene interaction study for identifying the genes that cause hereditary spherocytosis.

OBJECTIVE: Hereditary spherocytosis (HS) is a hemolytic disorder characterized by the presence of spherical-shaped red blood cells on the peripheral blood smear. Non-dominant HS cases are due to de novo mutations of the type associated with dominant inheritance or recessive genes. This study is aimed to identify HS-related biological mechanisms and predicting HS candidate genes. METHODS: We searched the known HS-related genes from the public databases. By analyzing the gene ontology (GO) and biological pathway of these genes, we extracted the optimal features to encode HS genes. Based on them

A greedy stacking algorithm for model ensembling and domain weighting.

OBJECTIVE: Because it is impossible to know which statistical learning algorithm performs best on a prediction task, it is common to use stacking methods to ensemble individual learners into a more powerful single learner. Stacking algorithms are usually based on linear models, which may run into problems, especially when predictions are highly correlated. In this study, we develop a greedy algorithm for model stacking that overcomes this issue while still being very fast and easy to interpret. We evaluate our greedy algorithm on 7 different data sets from various biomedical disciplines and co
