Transcriptome Analysis Reveals an Inhibitory Effect of Dihydrotestosterone-Treated 2D- and 3D-Cultured Dermal Papilla Cells on Hair Follicle Growth.

Dermal papillae are a target of androgen action in patients with androgenic alopecia, where androgen acts on the epidermis of hair follicles in a paracrine manner. To mimic the complexity of the dermal papilla microenvironment, a better culture model of human dermal papilla cells (DPCs) is needed. Therefore, we evaluated the inhibitory effect of dihydrotestosterone (DHT)-treated two-dimensional (2D)- and 3D-cultured DPCs on hair follicle growth. 2D- and 3D-cultured DPC proliferation was inhibited after co-culturing with outer root sheath (ORS) cells under DHT treatment. Moreover, gene expressi

Optimisation of A Sample Preparation Method for the Determination of Multi-Elemental Compositions in Human Hair By Triple Quadrupole ICP-MS Analysis.

Monitoring toxic elements has a long tradition in Slovenia due to historical mining. More recently, attention has shifted to essential elements, since both deficiencies and excesses can harm health. Regular monitoring of (non-)essential elements supports risk assessment and policymaking. While urine and blood are common biomonitoring matrices, hair offers a non-invasive alternative that reflects exposure over several months, though standardised methodologies for hair analysis remain limited. This study aimed to develop and validate a sensitive and robust analytical method for the determination

Compatibility of preparatory procedures for the analysis of cortisol concentrations and stable isotope (δ13C, δ15N) ratios: a test on brown bear hair.

The measurement of naturally occurring glucocorticoids and stable isotopes of several elements has gained importance in wildlife studies in recent decades and opened a myriad of ecological applications. Cortisol and stable isotopes equilibrate in animal tissues over periods of integration related to the growth rate of the tissue, providing information reflecting systemic cortisol secretion and dietary intake. Sample preparation shares the common step of first cleaning the sample of external contamination. However, it is not well understood how different solvents used in sample preparation affe

Non-invasive biomonitoring for PFRs and PBDEs: new insights in analysis of human hair externally exposed to selected flame retardants.

In this study, we investigated the hypothesis whether externally adsorbed and internally deposited flame retardants (FRs) in hair could be distinguished. To this extent, hair samples collected from one volunteer were exposed under controlled conditions to phosphate FR (PFR) and polybrominated diphenyl ether (PBDE) standards to mimic external contamination. Afterwards, suitable washing procedures to selectively remove contaminants from the hair surface were investigated. The samples were measured by GC-(ECNI)-MS for PBDEs and LC-(ESI+)-MS/MS for PFRs. All investigated compounds were transferred

Analysis of drugs of abuse in hair: evaluation of the immunochemical method VMA-T vs. LC-MS/MS or GC-MS.

Hair analysis is an elaborate and time-consuming multi-step process. The immunometric test VMA-T from Comedical has been evaluated as screening assay for hair analysis. From routine work, authentic samples were selected that were positive for opiates, cocaine, MDMA-type drugs, amphetamines, methadone or THC. These hair samples were investigated by LC-MS or GC-MS and the VMA-T procedure, respectively. Using the cut-off values recommended by the Society of Hair Testing, the VMA-T method discriminates with good sensitivity between negative and positive hair samples and is an expedient screening m

Correlation between drugs of abuse and alcohol by hair analysis: parents at risk for having children with fetal alcohol spectrum disorder.

The fatty acid ethyl esters (FAEEs) hair test, a biomarker of excessive alcohol exposure, has demonstrated its potential for use in fetal alcohol spectrum disorder (FASD) diagnosis. FASD may be compounded by polydrug exposure. Our objective was to determine the likelihood of positive FAEE test among parents testing positive for other drugs of abuse. Samples submitted for FAEE hair analysis by Children's Aid Societies between October 2005 and May 2007, also concurrently tested for cocaine, cannabinoids, opiates, methamphetamine, amphetamine, benzodiazepines, methadone, and/or oxycodone, were in

Evidence of gestational heroin exposure by comparative analysis of fetal and maternal body fluids, tissues, and hair in a heroin-related death.

In order to determine the fetal-maternal distribution of heroin and its main metabolites (6-monoacetylmorphine and morphine) the drug concentrations were measured in autopsy material. The heroin-related death of a pregnant drug abuser (approximately the 32nd week of gestation) provided the fetal and maternal material. Fetal and maternal hair was analyzed in order to obtain long-term information on the transplacental opiate transfer. Morphine and 6-monoacetylmorphine were detected in toxic concentrations in maternal as well as in fetal tissues and body fluids. The drug concentrations in the fet

Single-Cell Analysis of Late-Stage Wound-Induced Hair Follicle Neogenesis Reveals Keratinocyte Heterogeneity and Enrichment of Infundibular Basal Keratinocytes.

Scarring alopecia remains a major clinical challenge, characterized by irreversible hair follicle (HF) loss and replacement by scar tissue. Wound-induced hair follicle neogenesis (WIHN) serves as a model in which HFs can regenerate following skin wounding. In this study, we characterized the cellular landscape associated with differences in regenerative capacity between small wound (SW) and large wound (LW) tissues at a late regenerative stage (post-wounding day 22, PWD22), a time point at which newly formed hair follicles are already present. Using single-cell RNA sequencing (scRNA-seq), we p

Predictors of Skin, Hair, and Nail Supplement Use: Analysis of National Health and Nutrition Examination Survey (NHANES) Cross-Sectional Data.

OBJECTIVES: Approximately half of US adults use dietary supplements, often for dermatologic purposes. When survey respondents in a nationally representative population were asked the reasons for their use of a specific supplement, 87% chose "for healthy skin, hair, and nails." Despite widespread use, supplements have potential risks and lack regulatory safeguards. Because most supplement users choose to use them without the advice of a healthcare professional, it is important to identify factors predicting use. This study examined demographic predictors of skin, hair, and nail supplement use a

Hair quantitative PCR outperforms blood analysis for detecting Leishmania DNA in dogs: a non-invasive tool for clinical states of infection.

BACKGROUND: Canine leishmaniosis, caused by Leishmania infantum, is a significant zoonotic disease threatening both canine and human health worldwide. Early and accurate diagnosis is crucial for effective treatment and control. The use of hair samples for molecular diagnosis of L. infantum infection in dogs is a relatively novel approach and has not yet been extensively studied. Despite recent advances in blood-based polymerase chain reaction (PCR), noninvasive alternatives like hair quantitative PCR (qPCR) remain underexplored for different clinical states of infection. This study evaluates t
