Haloperidol-induced dyskinesia is associated with striatal NO synthase suppression: reversal with olanzapine.

The underlying pathophysiological basis of tardive dyskinesia (TD) remains speculative. Haloperidol (HP) inhibits neuronal nitric oxide (NO) synthase (NOS) activity in vitro, but has not to date been studied in an intact animal model. Recent animal studies have found that extrapyramidal dysfunction evoked by chronic HP is associated with suppression of striatal cyclic guanosine monophosphate (cGMP), as well as plasma nitrogen oxides. Striatal dopamine (DA) is central to motor control, while NO plays an important neuroregulatory role in striatal DA function. Recent case reports suggest that aty

A case of child abuse: haloperidol poisoning of a child caused by his mother.

This presentation focused on a hyperactive child admitted to the emergency department because of ataxia and imbalance. It was later discovered that the child had been abused by his mother. In cases where there is conflict between the clinical findings, the patient and the family's history in a clinical state which cannot be explained satisfactorily by the medical staff, the need for taking the history again from other relatives whilst taking into account the parents' physiological well-being, is time well spent.

Compatibility and stability of ternary admixtures of morphine with haloperidol or midazolam and dexamethasone or methylprednisolone.

The concentration range over which compatible admixtures of morphine hydrochloride with haloperidol lactate (Haldol) or midazolam hydrochloride (Dormicum) and dexamethasone-21-sodium phosphate (Decadron and Decadron Pack) or methylprednisolone-21-sodium succinate (Solu-Medrol) can be prepared was determined by visual evaluation of the solutions at 22 degrees C. The compatibility was evaluated for admixtures prepared in a ratio morphine hydrochloride (D1)/drug 2 (D2)/drug 2 (D3) in a ratio 10/1/1 to 10/1/10 (v/v/v). The solutions of morphine hydrochloride used were 10, 20, 30, 40 and 50 mg/ml p

[The influence of calcium channel blockers on the effects of haloperidol and phenamine in mice and rats].

Effects of verapamil (5 and 25 mg/kg), nifedipine (5 and 10 mg/kg), diltiazem (5, 10 and 20 mg/kg), cinnarizine (25 and 50 mg/kg), and fendilin (20 mg/kg) on the haloperidol (3 mg/kg)-induced catalepsy have been studied in rats. In higher doses these drugs attenuate and in lower doses potentiate the action of haloperidol. The bilateral intrastriatal injection of verapamil (5 mcg), diltiazem (5 mcg), and nimodipine (0.4 mcg) also alleviates the haloperidol catalepsy. Intraperitoneal administration of calcium channel blockers potentiated the amphetamine group toxicity in mice. No mediation of th

Lack of major effects of ethanol, chlorpromazine, hexobarbital and haloperidol on brain gangliosides.

Ethanol, hexobarbital, chlorpromazine and haloperidol had no effect on the composition and total amount of gangliosides in rat and mouse brains. The gangliosides seemed to be resistant to ethanol and the above mentioned drugs. None of them either in single or repeated doses had a major effect on the sialic acid-containing glycosphingolipids. The results of this study are at variance with those reported by Klemm & Foster (Life Sci., 1986, 39, 897-902) who described a sharp decrease of the total level of brain gangliosides after injection of a single dose of ethanol.

[Haloperidol decanoate: results of a 6-month open trial in paranoid schizophrenia].

An open clinical trial with Haloperidol Decanoate was carried out in order to determine its therapeutic efficacy, side effects, and tolerance. The sample population was a 30 male-and-female-psychotic-patient group (15 in-patients and 15 out-patients treated at a private psychiatric institution). A marked stabilizing effect with no relapses was observed. Therefore, it can safely be said that Haloperidol Decanoate helped in controling these patients satisfactorily, and getting a positive social reintegration.

Effect of pindolol administration on serum levels of thioridazine, haloperidol, phenytoin, and phenobarbital.

Twenty-six male patients with intermittent explosive disorder secondary to organic brain disease were studied. Each had been treated with long-term administration of thioridazine, haloperidol, phenytoin, and/or phenobarbital, either singly or in combination. The effect of adding increasing doses of pindolol--a beta-adrenergic blocking agent previously demonstrated to be effective in the treatment of intermittent explosive disorder--on serum levels of each long-term medication was determined. Moderate, dose-related increases in serum levels of thioridazine and two of its metabolites were found

Fluperlapine vs haloperidol: a comparison of their neuroendocrinological profiles and the influence on serum lipids.

Twelve male patients suffering from acute paranoid schizophrenia, randomly receiving fluperlapine - a new dibenzodiazepine very similar to clozapine - or haloperidol were included in a double blind study. Plasma levels of triglyceride, cholesterol and HDL-cholesterol, prolactin, thyroxine, triiodothyronine, ACTH and cortisol were determined. There was no evidence, that fluperlapine has any relevant influence on those hypothalamic-pituitary axis mediated hormones with the possible exception of plasma lipids. These findings, together with the clinical experiences, suggest that fluperlapine might

A comparison of parenteral loxapine and haloperidol in hostile and aggressive acutely schizophrenic patients.

In a parallel groups, double-blind study, 54 acutely psychotic schizophrenics were given loxapine or haloperidol parenterally for 24 to 72 hours, then orally for a total study period of up to 10 days. Dosage ratios of loxapine to haloperidol ranged from a minimum of 2.7:1 to a maximum of 4.4:1. Both groups showed significant and rapid improvement from baseline. Forty-eight percent of the loxapine patients and 33% of the haloperidol patients achieved and maintained a global severity of illness rating of mild or better. By the end of the study, 84% of the loxapine patients and 63% of the haloper

The emetic effect of B-HT 920 and apomorphine in the dog: antagonism by haloperidol.

Recent investigations have suggested that the alpha 2-adrenoreceptor agonist B-HT 920 is also a dopamine (DA) agonist with a selectivity for presynaptic receptors. In the present study, the emetic effect of B-HT 920 was investigated. Intravenous injections of B-HT 920 (0.32-10.0 micrograms/kg) and a DA2-agonist apomorphine (3.2-100.0 micrograms/kg) caused dose-dependent emesis. The ED50 of B-HT 920 and apomorphine were 3.2 and 12.3 micrograms/kg, respectively. When haloperidol (10.0-24.5 micrograms/kg i.v.), a DA2-antagonist, was given 5 minutes before B-HT 920 (10 micrograms/kg) or apomorphin

The effects of neuroleptics (haloperidol and chlorpromazine) on the pharmacokinetics of valproic acid in schizophrenic patients.

The steady state trough concentrations (Cmin) and pharmacokinetics of valproic acid were investigated in schizophrenic patients during the treatment of valproic acid, 200 mg, twice daily and after the one dose of 400 mg, respectively, with and without haloperidol (6 to 10 mg/day, N = 6) or chlorpromazine (100 to 300 mg/day, N = 6). Four to 5 Cmin were monitored just before the morning valproic acid dose for 3 to 4 days preceding the kinetic study. The overall Cmin of valproic acid with chlorpromazine (33.2 +/- 1.7 micrograms/ml, N = 25) was significantly (p less than 0.01) greater than without

The influence of haloperidol and aminooxyacetic acid on etonitazene, alcohol, diazepam and barbital consumption.

Four groups of rats were given free choice between water and solutions of either 3 micrograms/ml etonitazene, 5% ethanol (v/v), 0.1 mg/ml diazepam or 3 mg/ml barbital for 10-14 days. With the exception of barbital, some rats spontaneously preferred the drug solutions to water. This preference was reduced by addition of 7 micrograms/ml haolperidol. In a forced drug fluid consumption procedure, the daily administration of 15 mg/kg i.p. of the gamma-aminobutyric acid (GABA)-transaminase blocker aminooxyacetic acid (AOAA) led to a reduction of ethanol and diazepam intake, but not of etonitazene an

Effects of chlorprothixene, haloperidol, and trifluoperazine on the delayed-matching-to-sample performance of pigeons.

The effects of chlorprothixene (4,6,8, and 10 mg/kg), haloperidol (0.13, 0.25, 0.38, and 0.5 mg/kg), and trifluoperazine (0.5, 1,2, and 3 mg/kg) were examined in pigeons responding under a delayed-matching-to-sample procedure in which delays of 0.5-, 1-, 2-, 4-, and 8-sec duration were arranged during each experimental session. Both chlorprothixene and trifluoperazine typically reduced accuracy (percent correct responses); the magnitude of this effect was generally largest at the longest delay values. Chlorprothixene was associated with an increased rate of responding to the sample stimulus in

School avoidance and social phobia triggered by haloperidol in patients with Tourette's disorder.

Fifteen patients with Tourette's disorder developed school and work avoidance syndromes when treated with low doses (mean 2.5 mg/day) of haloperidol for short periods of time (mean, 8 weeks). The phobic syndromes disappeared completely with discontinuation or reduction of the haloperidol dose. Haloperidol's effects on dopaminergic functioning support a role for catecholamines in the pathogenesis of phobic syndromes. It is not known whether phobias are precipitated by haloperidol only in patients with Tourette's disorder as a consequence of the specific metabolic alterations in this disorder or

Effects of Intravenous Hydroxyzine Versus Haloperidol Monotherapy for Delirium: A Retrospective Study.

Objective: Although antipsychotics are used commonly for delirium, they increase the risk of mortality in elderly patients and those with dementia. As hydroxyzine has sedative and anxiolytic effects, it can be used in the treatment of delirium. We performed a retrospective study to compare the effects of intravenous hydroxyzine and haloperidol monotherapy on delirium. Methods: Patients who admitted to a university hospital from April 1, 2017, to September 30, 2022, and received either hydroxyzine or haloperidol intravenously as monotherapy for the treatment of delirium were included. The time

Quetiapine Versus Haloperidol in the Management of Hyperactive Delirium: Randomized Controlled Trial.

BACKGROUND: In the population of patients in the intensive care unit (ICU), most studies compared the use of atypical antipsychotics, such as quetiapine, with the use of traditional haloperidol in patients with delirium of various forms and etiologies. The role of such agents in patients with hyperactive delirium is not fully understood. This study compares the effectiveness of quetiapine with haloperidol in treating the hyperactive form of delirium in terms of their effects on the Delirium Rating Scale-Revised-98 (DRS-R-98), length of stay in the ICU, and mortality in critically ill patients.

A Case of Exacerbation of Haloperidol-Induced Rhabdomyolysis Following the Onset of COVID-19.

BACKGROUND Rhabdomyolysis is a condition in which intracellular components are released into the blood and urine. Rhabdomyolysis can be caused by drug-related complications and COVID-19; however, the underlying mechanism is not clear. In this study, we report a case of rhabdomyolysis complicated by COVID-19, in which we presumed that the cause of rhabdomyolysis was related to prior administration of haloperidol by assessment of the drug history and progression of myopathy. CASE REPORT A 52-year-old man with schizophrenia experienced worsening insomnia 10 days before admission. Thus, haloperido
