Hydra and Niccolo Paganini (1782-1840)--two peas in a pod? The molecular basis of extracellular matrix structure in the invertebrate, Hydra.

The body wall of Hydra is organized as an epithelial bilayer with an intervening extracellular matrix (ECM). Molecular and biochemical analyses of Hydra ECM have established that it contains components similar to those seen in more complicated vertebrates such as human. In terms of biophysical parameters, Hydra ECM is highly flexible; a property that facilitates continuous movements along the organism's longitudinal and radial axis. A more rigid ECM, as in vertebrates, would not be compatible with this degree of movement. The flexible nature of Hydra ECM can now be explained in part by the uni

Disentangling micropollutant toxicity from major ion effects in treated water effluents: Advancing invertebrate biotests for enhanced ecotoxicological assessment.

Water treatment systems generate effluents with varying compositions of major geochemical ions and diverse micropollutants. Well-established procedures exist to assess the toxicity of effluents as a complete and unique matrix. Although informative, these approaches may fail to identify the toxicity specifically attributable to micropollutants. This review provides a bibliographic overview of the physicochemical and ecotoxicological characterization of 181 effluents reported across 88 publications. It highlights a general lack of information on effluent ionic composition while assessments are p

The potential virulence of Listeria monocytogenes strains isolated from fresh produce processing facilities as determined by an invertebrate Galleria mellonella model.

Listeria monocytogenes, a bacterium responsible for listeriosis, is an environmental and food-borne pathogen that poses a particular risk to pregnant women and the elderly. While traditionally associated with animal products, ready-to-eat salads are increasingly recognised as a source of Listeria outbreaks. However, little is known about the potential virulence of Listeria isolates from the fresh produce environment. This study assessed the virulence potential of nine L. monocytogenes strains from the fresh produce chain using the Galleria mellonella invertebrate infection model. Larvae were i

The relevance of European Biota Quality Standards on the ecological water quality as determined by the multimetric macro-invertebrate index: A Flemish case study.

European Biota Quality Standards (EQSbiota), for compounds with low water solubility and high biomagnification, were created to sustain water quality and protect top predators and humans from secondary poisoning. In reality, for multiple compounds, an exceedance of these standards is often reported in literature without a decrease in ecological water quality determined by biotic indices. In the present study, threshold concentrations were defined in biota (from 44 sampling locations throughout Flanders (Belgium)), above which a good ecological water quality, assessed by the Multimetric Macroin

Small Molecule Analysis of Extracellular Vesicles Produced by Cryptococcus gattii: Identification of a Tripeptide Controlling Cryptococcal Infection in an Invertebrate Host Model.

The small molecule (molecular mass <900 Daltons) composition of extracellular vesicles (EVs) produced by the pathogenic fungus Cryptococcus gattii is unknown, which limits the understanding of the functions of cryptococcal EVs. In this study, we analyzed the composition of small molecules in samples obtained from solid cultures of C. gattii by a combination of chromatographic and spectrometric approaches, and untargeted metabolomics. This analysis revealed previously unknown components of EVs, including small peptides with known biological functions in other models. The peptides found in C. ga

Engineered expression of the invertebrate-specific scorpion toxin AaHIT reduces adult longevity and female fecundity in the diamondback moth Plutella xylostella.

BACKGROUND: Previous genetic pest management (GPM) systems in diamondback moth (DBM) have relied on expressing lethal proteins ('effectors') that are 'cell-autonomous', that is, they do not leave the cell in which they are expressed. To increase the flexibility of future GPM systems in DBM, we aimed to assess the use of a non-cell-autonomous, invertebrate-specific, neurotoxic effector - the scorpion toxin AaHIT. This AaHIT effector was designed to be secreted by expressing cells, potentially leading to effects on distant cells, specifically neuromuscular junctions. RESULTS: Expression of AaHI

[D-Ala2]deltorphin I binding and pharmacological evidence for a special subtype of delta opioid receptor on human and invertebrate immune cells.

The effects of the opioid neuropeptide [D-Ala2]deltorphin I, isolated from amphibian skin, on immunoregulatory activities were studied in representatives of vertebrates and invertebrates. The high potency of this compound parallels that of [Met]enkephalin, which was previously demonstrated in vertebrate plasma and invertebrate hemolymph. The addition of [D-Ala2]deltorphin I at 10(-11) M to human granulocytes or immunocytes of the mollusc Mytilus edulis resulted in cellular adherence and conformational changes indicative of cellular activation. This value is in line with the concentrations obta

Incorporating Neglected Insect Larvae in Species Inventories: DNA Barcoding as an Effective Tool for All-Stage Invertebrate Identification in Tree Holes.

Invertebrates, especially insects, are an integral part of biodiversity. Many species live in forest ecosystems where they play a key role in decomposing wood and maintaining ecosystem functions. Nevertheless, global changes, like fires, storms, and pest outbreaks, are impacting insect diversity, reinforcing the need for long-term biomonitoring to understand and tackle these issues. Forests are heterogeneous ecosystems with tree-related microhabitats (TReMs) such as tree holes, which are important for ecosystem diversity. Conventional identification approaches for species inventories are frequ

Individual and combined impacts of carbon dioxide enrichment, heatwaves, flow velocity variability, and fine sediment deposition on stream invertebrate communities.

Climate change and land-use change are widely altering freshwater ecosystem functioning and there is an urgent need to understand how these broad stressor categories may interact in future. While much research has focused on mean temperature increases, climate change also involves increasing variability of both water temperature and flow regimes and increasing concentrations of atmospheric CO2, all with potential to alter stream invertebrate communities. Deposited fine sediment is a pervasive land-use stressor with widespread impacts on stream invertebrates. Sedimentation may be managed at the

Fit for purpose? Evaluating benthic invertebrate DNA metabarcoding for ecological status class assessment in streams under the Water Framework Directive.

The ecological state of aquatic ecosystems is systematically monitored using various bioindicators in many countries worldwide. In the European Union, freshwater biomonitoring is the central component of the EU Water Framework Directive (WFD, 2000/60/EC) and currently based on morpho-taxonomic methods. DNA metabarcoding is a novel approach to assess the ecological state fast and efficiently based on organismal DNA signatures and thereby support and upscale biomonitoring. However, compliance of metabarcoding with existing morpho-taxonomic methods must be ensured prior to official implementation

Size-dependent and sex-specific negative effects of micro- and nano-sized polystyrene particles in the terrestrial invertebrate model Drosophila melanogaster.

Microplastic pollution is believed to be one of the most widespread and long-lasting changes on a global scale. Our understanding that microplastics significantly impact terrestrial systems and are a global change stressor continues to grow. In the present study, we investigated the negative effect of long-term (28 days of exposure in food) polystyrene particles of micro (1.0-1.9 µm, 0.4-0.6 µm) and nano (0.04-0.06 µm) scale, in low doses, on the fruit fly - representing a common, globally distributed terrestrial invertebrate, and a model species in many fields. Lactobacillus rhamnosus Lcr35 as an effective treatment for preventing Candida albicans infection in the invertebrate model Caenorhabditis elegans: First mechanistic insights.

The increased recurrence of Candida albicans infections is associated with greater resistance to antifungal drugs. This involves the establishment of alternative therapeutic protocols, such as probiotic microorganisms whose antifungal potential has already been demonstrated using preclinical models (cell cultures, laboratory animals). Understanding the mechanisms of action of probiotic microorganisms has become a strategic need for the development of new therapeutics for humans. In this study, we investigated the prophylactic anti-C. albicans properties of Lactobacillus rhamnosus Lcr35® using

The Rift Valley fever accessory proteins NSm and P78/NSm-GN are distinct determinants of virus propagation in vertebrate and invertebrate hosts.

Rift Valley fever virus (RVFV) is an enzootic virus circulating in Africa that is transmitted to its vertebrate host by a mosquito vector and causes severe clinical manifestations in humans and ruminants. RVFV has a tripartite genome of negative or ambisense polarity. The M segment contains five in-frame AUG codons that are alternatively used for the synthesis of two major structural glycoproteins, GN and GC, and at least two accessory proteins, NSm, a 14-kDa cytosolic protein, and P78/NSm-GN, a 78-kDa glycoprotein. To determine the relative contribution of P78 and NSm to RVFV infectivity, AUG

Virome analysis of Amblyomma americanum, Dermacentor variabilis, and Ixodes scapularis ticks reveals novel highly divergent vertebrate and invertebrate viruses.

UNLABELLED: A wide range of bacterial pathogens have been identified in ticks, yet the diversity of viruses in ticks is largely unexplored. In the United States, Amblyomma americanum, Dermacentor variabilis, and Ixodes scapularis are among the principal tick species associated with pathogen transmission. We used high-throughput sequencing to characterize the viromes of these tick species and identified the presence of Powassan virus and eight novel viruses. These included the most divergent nairovirus described to date, two new clades of tick-borne phleboviruses, a mononegavirus, and viruses w

Antidepressant fluoxetine suppresses neuronal growth from both vertebrate and invertebrate neurons and perturbs synapse formation between Lymnaea neurons.

Current treatment regimes for a variety of mental disorders involve various selective serotonin reuptake inhibitors such as Fluoxetine (Prozac). Although these drugs may 'manage' the patient better, there has not been a significant change in the treatment paradigm over the years and neither have the outcomes improved. There is also considerable debate as to the effectiveness of various selective serotonin reuptake inhibitors and their potential side-effects on neuronal architecture and function. In this study, using mammalian cortical neurons, a dorsal root ganglia cell line (F11 cells) and id

egl-17 encodes an invertebrate fibroblast growth factor family member required specifically for sex myoblast migration in Caenorhabditis elegans.

The proper guidance of the Caenorhabditis elegans hermaphrodite sex myoblasts (SMs) requires the genes egl-15 and egl-17. egl-15 has been shown to encode the C. elegans orthologue of the fibroblast growth factor receptor (FGFR). Here we clone egl-17 and show it to be a member of the fibroblast growth factor (FGF) family, one of the first functional invertebrate FGFs known. egl-17 shares homology with other FGF members, conserving the key residues required to form the distinctive tertiary structure common to FGFs. Genetic and molecular evidence demonstrates that the SM migration defect seen in

Contaminant Concentrations in Sediments, Aquatic Invertebrates, and Fish in Proximity to Rail Tracks Used for Coal Transport in the Pacific Northwest (USA): A Baseline Assessment.

Railway transport of coal poses an environmental risk, because coal dust contains polycyclic aromatic hydrocarbons (PAHs), mercury, and other trace metals. In the Pacific Northwest of the United States, proposed infrastructure projects could result in an increase in coal transport by train through the Columbia River corridor. Baseline information is needed on current distributions, levels, and spatial patterns of coal dust-derived contaminants in habitats and organisms adjacent to existing coal transport lines. To that end, we collected aquatic surface sediments, aquatic insects, and juvenile

Homologous tropomyosins from vertebrate and invertebrate: Recombinant calibrator proteins in functional biological assays for tropomyosin allergenicity assessment of novel animal foods.

BACKGROUND: Novel foods may provide new protein sources for a growing world population but entail risks of unexpected food-allergic reactions. No guidance on allergenicity assessment of novel foods exists, while for genetically modified (GM) crops it includes comparison of sequence identity with known allergens, digestibility tests and IgE serum screening. OBJECTIVE: As a proof of concept, to evaluate non-/allergenic tropomyosins (TMs) regarding their potential as new calibrator proteins in functional biological in vitro assays for the semi-quantitative allergy risk assessment of novel TM-con
